Abstract
本发明涉及一种用于预测应激后焦虑抑郁等精神心理症状的生物标志物及其检测试剂盒。本发明提供了一组在潜在患者的血液中可预测应激后焦虑抑郁等精神心理症状发生的生物标志物,其选自包括丁酰胆碱酯酶、过氧化氢酶、多巴胺‑β‑羟化酶、***结合蛋白2、神经纤毛素1或者与它们具有80%同源性的氨基酸序列中的一种或几种。本发明也提供了在血液样品中检测这些蛋白的检测试剂盒和检测方法;以及上述抗原和试剂盒的应用;以及提供了这些蛋白的应激后焦虑抑郁高发生风险的参考浓度阈值。由其制成的免疫检测试剂盒可以快速简单准确地检测血液中的生物标志物浓度,适于大规模推广应用。
Description
技术领域
本发明涉及在应激后患有焦虑抑郁等精神心理症状的对象中产生的生物标志物的检测和鉴定,并提供用于预测潜在的患有焦虑抑郁的对象中区分高发生风险的衡量标准。具体是指一组可预测应激后焦虑抑郁等精神心理症状发生的生物标志物及其在血液样品中检测这些标志物的检测试剂盒和检测方法,并提供了这些标志物的高发生风险的参考浓度阈值,使对于辅助应激后焦虑抑郁等精神心理症状的诊断和预防的生物标志物的可靠检测和鉴定成为可能。
背景技术
近年来,突发公共卫生事件、各类重大事故、自然和人为的灾害等应激性事件的发生率逐步升高,在世界范围内超过70%的人在一生中会至少经历一次创伤性事件,而事件经历者和参与救助的医护人员等救助者在应激事件后往往出现焦虑、抑郁等精神心理症状,部分个体甚至发展成严重的精神疾病。其中,创伤后应激障碍(post-traumatic stressdisorder,PTSD)是其中一种较为严重的精神疾病。创伤后应激障碍是指个体经历、目睹或遭遇到一个或多个涉及自身或他人的实际死亡,或受到死亡的威胁,或严重的受伤,或躯体完整性受到威胁后,所导致的个体延迟出现和持续存在的精神障碍。临床上表现为创伤记忆侵入、创伤相关刺激躲避、认知消极改变、唤醒增强和行为过激等。根据不同国家地区的文化背景以及经济发展水平,PTSD的终身患病率大概在1.3%-12.2%,年患病率大概在0.2%-3.8%。而当今社会,包括PTSD在内的应激后精神疾病已成为备受关注的疾病,尤其是在当前COVID-19大流行时期。此类疾病病程较长且容易复发,患者往往会出现自残、***、药物滥用等行为,严重影响患者的生命质量,且对个人、家庭和社会造成严重的心理、生理和经济负担。
应激后精神类疾病中PTSD是较严重且最受关注的,其临床诊断标准目前被广泛接受的是美国精神医学学会的DSM-5和世界卫生组织的ICD-11。在DSM-5中,PTSD的诊断标准共有8大条目,分为4个症状群和一个亚型,其中4大症状群为创伤记忆侵入、躲避创伤相关刺激、认知和情绪发生消极改变、唤醒增强和行为过激。而在ICD-11中,PTSD的诊断被简化为3个症状群,共6类症状,包括不断重新经历创伤事件、创伤刺激回避和高度警惕感。尽管PTSD的诊断标准略有不同,但其治疗方式主要包括心理干预、药物治疗和其他一些创新性疗法。心理干预作为大部分指南推荐的一线治疗方法,主要包括延长暴露疗法、认知加工疗法、认知行为疗法、眼动脱敏和再加工疗法等。而药物治疗一般作为二线疗法,包括选择性5-羟色胺再摄取抑制剂、血清素和去甲肾上腺素再摄取抑制剂、单胺氧化酶、交感神经阻滞剂、抗惊厥类、苯二氮草类等药物。
尽管PTSD在经历一次或者多次严重创伤的人群中发病率较高,但并非所有经历过创伤的个体均成为PTSD患者,实际PTSD终身患病率远低于理论推测值,提示PTSD的发生率具有显著的个体差异。基于不同种族人群的PTSD发病率研究发现白色人种的发病率显著低于其他人种,而基于双胞胎的研究也发现在经历创伤事件后同卵双胞胎的发病率显著高于异卵双胞胎,提示遗传因素显著影响PTSD的发生率。而目前对PTSD的诊断主要是以患者的病史、精神状态判定、症状持续时间、临床精神量表检测和患者的自述为主要依据进行,缺乏对PTSD客观有效的生物标志物的检测。考虑到大多数创伤事件的发生是不能避免的,因而创伤发生后预测PTSD发生的危险因素则变得尤为重要。但PTSD客观、有效的、特异性的生物学标记物的筛选研究较少,目前仍然缺乏作为对PTSD预警诊断和治疗疗效评估的客观指标。血液中生物标志物的检测因其便利、微创等优势,被认为是有助于PTSD诊断的最有潜力、最方便的方式。有研究报道创伤后血液中皮质醇水平的降低可能与PTSD的发生相关,血液中部分神经递质的失衡、促肾上腺皮质激素(ACTH)、糖皮质激素(GC)、肾上腺素以及儿茶酚氨水平的高低等也都被认为是与PTSD相关的标志物。此外,可能与PTSD相关的其他生物标志物包括胆囊收缩素(CCK)、一氧化氮合成酶(NOS)、神经肽Y(NPY)和p11蛋白等。但这些标志物的特异性和敏感性还未被很好的验证。同时PTSD以外的应激后精神疾病的特异性标志物更鲜有报道,因此急需发现和开发利用针对应激后焦虑抑郁等精神心理症状的特异性生物标志物。
因此,为了实现在应激后有效的预测焦虑抑郁等精神心理症状的发生,有必要鉴定全新的生物标志物,并开发能特异性检测这些生物标志物的检测试剂盒。
发明内容
为了解决上述存在的问题,本发明的目的是提供一组可预测应激后焦虑抑郁等精神心理症状发生的生物标志物及其在血液样品中检测这些标志物的检测试剂盒和检测方法,基于该组生物标志物的检测试剂盒可用于在潜在的应激患者血液中高灵敏度、高特异性和高稳定性地检测该组生物标志物的浓度,并根据本发明给出的浓度阈值提示焦虑抑郁等精神心理症状的高发生风险,从而可以为医生准确预测应激病人罹患焦虑、抑郁、PTSD等精神疾病带来依据,进而及时采取治疗,以预防此类疾病的发生,及时减轻病人痛苦和负担,由其制成的免疫检测试剂盒可以快速简单准确地检测血液的生物标志物浓度,适于大规模推广应用。
为了实现上述目的,在本发明的第一方面,提供了一种用于预测应激后焦虑抑郁等精神心理症状发生的免疫检测试剂盒,所述的免疫检测试剂盒包含定量检测生物标志物表达水平的试剂,所述的生物标志物选自:丁酰胆碱酯酶(Butyrylcholinesterase,BCHE)、过氧化氢酶(Catalase,CAT)、多巴胺-β-羟化酶(Dopamine Beta-Hydroxylase,DBH)、***结合蛋白2(Insulin Like Growth Factor Binding Protein 2,IGFBP2)、神经纤毛素1(Neuropilin 1,NRP1)、与BCHE的氨基酸序列具有至少80%同源性的氨基酸序列、与CAT的氨基酸序列具有至少80%同源性的氨基酸序列、与DBH的氨基酸序列具有至少80%同源性的氨基酸序列、与IGFBP2的氨基酸序列具有至少80%同源性的氨基酸序列以及与NRP1的氨基酸序列具有至少80%同源性的氨基酸序列中的一种或几种。
较佳的,所述的应激后焦虑抑郁等精神心理症状包括应激后焦虑、应激后抑郁、创伤后应激障碍、中风后焦虑、中风后抑郁、脑外伤后焦虑、脑外伤后抑郁及其他应激后相关精神疾病。
在本发明的第二方面,提供了一种生物标志物在制备检测预测应激后精神心理症状发生的试剂的应用,所述的试剂为定量检测所述的生物标志物表达水平的试剂,所述的生物标志物选自:丁酰胆碱酯酶、过氧化氢酶、多巴胺-β-羟化酶、***结合蛋白2、神经纤毛素1、与丁酰胆碱酯酶的氨基酸序列具有至少80%同源性的氨基酸序列、与过氧化氢酶的氨基酸序列具有至少80%同源性的氨基酸序列、与多巴胺-β-羟化酶的氨基酸序列具有至少80%同源性的氨基酸序列、与***结合蛋白2的氨基酸序列具有至少80%同源性的氨基酸序列以及与神经纤毛素1的氨基酸序列具有至少80%同源性的氨基酸序列中的一种或几种。
在本发明的第三方面,提供了一种免疫检测试剂盒的检测方法,检测在全血、中枢神经***组织、血清、血浆、脑脊液、唾液、汗液、泪液、尿、口腔样本、或它们的组合物中的上述生物标志物蛋白的水平。较佳地,所述体液是血液。检测上述蛋白的方法可以通过常规的酶联免疫吸附测定(Enzyme-linked immunosorbent assay,ELISA)进行,优选使用夹心ELISA法。
较佳地,所述免疫检测试剂盒还包括载体,用于检测上述生物标志物蛋白的抗原包被在所述载体上。
较佳地,所述免疫检测试剂盒还包括一抗,一抗为用于检测上述生物标志物蛋白的一抗。
较佳地,所述免疫检测试剂盒还包括标记的上述生物标志物蛋白一抗的二抗。所用标记物可以是辣根过氧化物酶,碱性磷酸酶,荧光分子FITC(或其它荧光标记),化学发光检测。检测方法可以是显色法,荧光方法,化学发光,电化学发光法进行定性、定量分析。
更佳地,所述的标记的上述生物标志物蛋白一抗的二抗是辣根过氧化物酶标记的抗上述蛋白一抗的二抗。
在本发明的第四方面,提供了上述生物标志物蛋白的免疫检测试剂盒在检测人血液中上述生物标志物蛋白的应用,通过将受试者(例如,经历过创伤事件的患者)的上述生物标志物蛋白的测量水平与定量质谱技术的上述生物标志物蛋白的测量水平进行对比,确定出上述蛋白的高风险浓度阈值。
本发明的有益效果在于:本发明提供的一组可预测应激后焦虑抑郁等精神心理症状发生的生物标志物是丁酰胆碱酯酶(Butyrylcholinesterase,BCHE)、过氧化氢酶(Catalase,CAT)、多巴胺-β-羟化酶(Dopamine Beta-Hydroxylase,DBH)、***结合蛋白2(Insulin Like Growth Factor Binding Protein 2,IGFBP2)、神经纤毛素1(Neuropilin1,NRP1)、与BCHE的氨基酸序列具有至少80%同源性的氨基酸序列、与CAT的氨基酸序列具有至少80%同源性的氨基酸序列、与DBH的氨基酸序列具有至少80%同源性的氨基酸序列、与IGFBP2的氨基酸序列具有至少80%同源性的氨基酸序列以及与NRP1的氨基酸序列具有至少80%同源性的氨基酸序列中的一种或几种,潜在的患者在经历应激事件后,可在血液中高特异性、高灵敏度、高特异性地检测上述生物标志物蛋白的浓度,从而可以为医生准确预测应激病人罹患焦虑、抑郁、PTSD等精神疾病带来依据,进而及时采取治疗,以预防此类疾病的发生,及时减轻病人痛苦和负担,由其制成的免疫检测试剂盒可以快速简单准确地检测血液中的生物标志物浓度,适于大规模推广应用。

为了能够更清楚地理解本发明的技术内容,特举以下实施例详细说明。
蛋白生物标志物
用于预测应激后焦虑抑郁等精神心理症状发生的生物标志物是丁酰胆碱酯酶(Butyrylcholinesterase,BCHE)、过氧化氢酶(Catalase,CAT)、多巴胺-β-羟化酶(Dopamine Beta-Hydroxylase,DBH)、***结合蛋白2(Insulin Like GrowthFactor Binding Protein 2,IGFBP2)、神经纤毛素1(Neuropilin 1,NRP1)。
BCHE是具有广泛底物特异性的酯酶,其有助于神经递质乙酰胆碱的失活。BCHE能降解神经毒性有机磷酸酯。如图1所示,本发明鉴定与经历创伤后无焦虑抑郁症状的患者的水平相比,BCHE蛋白在经历创伤后有焦虑抑郁症状的患者的全血、血浆、血清中的水平显著降低(p<0.05)。
CAT存在于几乎所有有氧呼吸的生物体中,用于保护细胞免受过氧化氢的毒性作用。CAT能够促进细胞生长,包括T细胞、B细胞、髓系白血病细胞、黑色素瘤细胞、肥大细胞瘤细胞以及正常和转化的成纤维细胞。如图2所示,本发明鉴定与经历创伤后无焦虑抑郁症状的患者的水平相比,CAT蛋白在经历创伤后有焦虑抑郁症状的患者的全血、血浆、血清中的水平显著降低(p<0.05)。
DBH蛋白在肾上腺髓质的神经分泌囊泡和嗜铬颗粒中表达,催化多巴胺转化为去甲肾上腺素,去甲肾上腺素既是一种激素,也是交感神经***的主要神经递质。DBH以可溶和膜结合形式存在,分别取决于信号肽的缺失或存在。该基因突变导致人类患者多巴胺-β-羟酸缺乏,其特征是自主神经和心血管功能缺陷,包括低血压和上睑下垂。本发明鉴定与经历创伤后无焦虑抑郁症状的患者的水平相比,DBH蛋白在经历创伤后有焦虑抑郁症状的患者的全血、血浆、血清中的水平显著降低(p<0.05)。
IGFBP2抑制IGF介导的生长和发育速率。IGFBP2延长IGF的半衰期,并被证明能抑制或刺激IGF对细胞培养的生长促进作用。IGFBP2能改变IGF与其细胞表面受体的相互作用。本发明鉴定与经历创伤后无焦虑抑郁症状的患者的水平相比,IGFBP2蛋白在经历创伤后有焦虑抑郁症状的患者的全血、血浆、血清中的水平显著降低(p<0.05)。
NRP1是一种细胞表面受体,参与心血管***的发育、血管生成、神经回路的形成以及神经***外的器官发生。NRP1介导信号素的化学排斥活性。NRP1按C端规则识别其配体上的基序R/KXXR/K,导致细胞内化和血管渗漏。它结合信号素3A,PGF的PLGF-2亚型,VEGFA和VEGFB的VEGF165亚型。NRP1与KDR共表达导致VEGF165与KDR结合增加以及趋化性增加。NRP1调节VEGF诱导的血管生成。NRP1与VEGFA结合启动运动神经元轴突导向和细胞体迁移所需的信号通路,包括胚胎发育期间面部运动神经元从菱形4向菱形6的尾侧迁移(通过相似性)。NRP1通过与ABCB8/MITOSUR相互作用调节线粒体铁转运。NRP1结合VEGF-165并可能抑制其与细胞的结合。NRP1可通过隔离VEGF-165诱导细胞凋亡。NRP1也可以绑定信号素家族的各个成员。NRP1表达对血管数量和完整性具有负面影响。NRP1作为人类冠状病毒SARS-CoV-2感染的宿主因子。NRP1以C端规则识别并结合SARS-CoV-2棘突蛋白S1上的基序RRAR以增强SARS-CoV-2的感染。本发明鉴定与经历创伤后无焦虑抑郁症状的患者的水平相比,NRP1蛋白在经历创伤后有焦虑抑郁症状的患者的全血、血浆、血清中的水平显著降低(p<0.05)。
试剂盒
在本发明中,通过五种酶联免疫测定试剂盒分别检测5种血清中应激后焦虑抑郁等精神心理症状发生的生物标志物(BCHE、CAT、DBH、IGFBP2、NRP1)。优先夹心ELISA方法进行测定。
1、夹心ELISA方法的建立
1)根据检测样本数量取已包被抗应激后焦虑抑郁等精神心理症状发生的生物标志物一抗的costar板条若干。
2)加入样本稀释液100ul/孔。
3)加入标准样本、空白对照和已稀释的血清样本,50ul/孔,37度2小时,摇床200rpm。
4)PBST洗涤,400ul/孔,5次,拍干。
5)加入另一辣根过氧化物酶标记的相应生物标志物一抗,200ul/孔,37度孵育2小时,摇床200rpm。
6)PBST洗涤,400ul/孔,5次,拍干。
7)TMB显色:TMB A液和B液等体积混合,100ul/孔,37度20min。
8)终止液,50ul/孔终止,用酶标仪450nm读数。
2、试剂盒组成:
本试剂盒由以下几个部分组成:
1).包被用于检测本发明的抗应激后焦虑抑郁等精神心理症状发生的生物标志物一抗的costar酶标板各一块
2).PTSD生物标志物标准样品各一管(1ml)
3).样品稀释液一瓶(100ml)
4).辣根过氧化物酶标记的抗生物标志物抗体各一管(20ml)
5).10*PBST一瓶(100ml)
6).TMB显色液A液一瓶(60ml),B液一瓶(60ml)
7).终止液一瓶(60ml)
3、应激后焦虑抑郁等精神心理症状的生物标志物的鉴定
1)、患者:选取11位经历过脑外伤的门诊患者,这些患者在脑外伤后6个月由2位精神科医生根据DSM-5的诊断标准对其进行是否出现焦虑抑郁症状进行评估,其中8位患者未出现焦虑抑郁症状且未被诊断为PTSD,3位患者出现焦虑抑郁症状且被诊断为PTSD。11位患者的人口学资料如表一:

2)、血液样本的采集:血液样本通过静脉穿刺的方式采集于含肝素抗凝的真空管中,每位患者采集10ml血样。血样经3000rpm离心5分钟后,分离成血清和血浆。所有全血、血清、血浆分装后存储于-80℃冰箱中以用于后续的测定。
3)、在血清样本中鉴定蛋白质含量:血清样本采用4D-Label-free琥珀酰化定量蛋白质组学方法进行蛋白质含量测定。非标(Label-free)蛋白质组定量技术是一种不依赖于同位素标记的新型蛋白质定量技术,该技术通过液质联用对蛋白质酶解肽段进行分析,无需昂贵的稳定同位素标签做内部标签,只需分析大规模鉴定蛋白质时所产生的质谱数据,比较不同样本中相应肽段的信号强度,就能对相应的蛋白质进行相对定量。将血清样本从-80℃取出后,称取适量样本至液氮预冷的研钵中,加液氮充分研磨至粉末。粉末中加入裂解缓冲液后,各样本取等量蛋白进行酶解。酶解得到的肽段用NanoElute超高效液相***进行分离后,被注入Capillary离子源中进行电离然后进timsTOF Pro质谱进行分析。基于质谱检测得到的raw文件采用分析软件进行数据库搜索,基于搜索结果进行肽段和蛋白质水平的质控后,进行蛋白质注释,随后进行蛋白质定量分析。
4)、结果:将经历过脑外伤的有焦虑抑郁症状和无焦虑抑郁症状的患者的蛋白含量进行比较,鉴定到了一组在有焦虑抑郁症状的患者中差异表达的蛋白质,结果如表二:

图1显示相比于无焦虑抑郁症状的患者,有焦虑抑郁症状的患者血清中BCHE的蛋白水平显著降低(p<0.05)。图2显示相比于无焦虑抑郁症状的患者,有焦虑抑郁症状的患者血清中CAT的蛋白水平显著降低(p<0.05)。图3显示相比于无焦虑抑郁症状的患者,有焦虑抑郁症状的患者血清中DBH的蛋白水平显著降低(p<0.05)。图4显示相比于无焦虑抑郁症状的患者,有焦虑抑郁症状的患者血清中IGFBP2的蛋白水平显著降低(p<0.05)。图5显示相比于无焦虑抑郁症状的患者,有焦虑抑郁症状的患者血清中NRP1的蛋白水平显著降低(p<0.05)。
5)、总结:发现了5种显著区别应激后有焦虑抑郁症状的患者和无焦虑抑郁症状的患者的蛋白质。包括(1)丁酰胆碱酯酶BCHE(SwissProt P06276),根据DIA质谱的结果,与无焦虑抑郁症状的患者相比,在有焦虑抑郁症状的患者的血清样本中BCHE减少约44%。(2)过氧化氢酶CAT(SwissProt P04040),根据DIA质谱的结果,与无焦虑抑郁症状的患者相比,在有焦虑抑郁症状的患者的血清样本中CAT减少约47%。(3)多巴胺-β-羟化酶DBH(SwissProtP09172),根据DIA质谱的结果,与无焦虑抑郁症状的患者相比,在有焦虑抑郁症状的患者的血清样本中DBH减少约52%。(4)***结合蛋白2IGFBP2(SwissProt P18065),根据DIA质谱的结果,与无焦虑抑郁症状的患者相比,在有焦虑抑郁症状的患者的血清样本中IGFBP2减少约75%。(5)神经纤毛素1NRP1(SwissProt O14786),根据DIA质谱的结果,与无焦虑抑郁症状的患者相比,在有焦虑抑郁症状的患者的血清样本中NRP1减少约23%。
4、应激后焦虑抑郁等精神心理症状的生物标志物免疫检测试剂盒的应用
1)、受试者:招募16位健康受试者和15位经历过创伤事件的门诊患者进行试剂盒检测的应用,这些受试者的人口学资料如表三:

2)、血液样本的采集:血液样本通过静脉穿刺的方式采集于含肝素抗凝的真空管中,每位患者采集10ml血样。血样经3000rpm离心5分钟后,分离成血清和血浆。所有全血、血清、血浆分装后存储于-80℃冰箱中以用于后续的测定。
3)、在血清样本中检测应激后焦虑抑郁等精神心理症状的生物标志物的蛋白质含量:血清样本采用本发明的试剂盒,用ELISA的方法分别检测丁酰胆碱酯酶、过氧化氢酶、多巴胺-β-羟化酶、***结合蛋白2、神经纤毛素1的蛋白质含量。
4)、结果:31位受试者血清检测的结果如表四:

5)、数据分析:根据表四结果,分别计算出健康对照和经历过创伤事件受试者的应激后焦虑抑郁等精神心理症状的生物标志物蛋白含量平均值、标准方差和95%CI区间,结果如表五:

6)、总结:应激后焦虑抑郁等精神心理症状的标志物在健康对照受试者和经历过创伤事件受试者的血清中的表达量分别是:BCHE:2748±158.4ng/ml(健康对照)和3064±237.8ng/ml(创伤组);CAT:59.69±4.083pg/ml(健康对照)和67.24±3.718pg/ml(创伤组);DBH:24.15±5.577ng/ml(健康对照)和25.29±7.149ng/ml(创伤组);IGFBP2:142.2±19.85ng/ml(健康对照)和276.6±34.95ng/ml(创伤组);NRP1:265.2±11.31ng/ml(健康对照)和239.2±17.66ng/ml(创伤组)。
5、应激后焦虑抑郁等精神心理症状的生物标志物高风险阈值的确定
1)、根据定量蛋白质组学组测定的应激后有焦虑抑郁症状的患者相比无焦虑抑郁症状的患者的BCHE、CAT、DBH、IGFBP2和NRP1的表达量下降率,结合由本试剂盒确定的经历过创伤事件受试者血清中上述蛋白表达量,折算出上述标志物的高风险cutoff阈值,计算公式如下:
Cutoff计算公式为:cutoff=由ELISA确定的创伤组平均表达量×(1-由定量质谱确定的有焦虑抑郁症状的患者表达下降率)
根据上述公式和表四中创伤组的平均表达量计算得出每个应激后焦虑抑郁等精神心理症状的生物标志物高风险阈值分别是小于以下表达量:
BCHE:3064×(1-44%)=1716ng/ml;
CAT:67.24×(1-47%)=35.64pg/ml;
DBH:25.29×(1-52%)=12.14ng/ml;
IGFBP2:276.6×(1-75%)=69.15ng/ml;
NRP1:239.2×(1-23%)=184.2ng/ml。
2)、通过cutoff值,判断检测的应激患者的发病风险,低于这个cutoff值的即认为是高风险,且只要一个蛋白检测为高风险,即认为有焦虑、抑郁、PTSD等精神疾病高发的风险。
本发明通过非标(Label-free)蛋白质组定量技术,对比经历脑外伤后有焦虑抑郁的和无焦虑抑郁的患者的血液样本中蛋白质的含量,筛选得到了一组可有效预测应激后焦虑抑郁等精神心理症状的发生风险的生物标志物:BCHE、CAT、DBH、IGFBP2和NRP1。本发明根据鉴定得到的应激后焦虑抑郁等精神心理症状的生物标志物制备了在血液样本中检测表达水平的免疫试剂盒,并建立了检测方法。本发明的血液夹心酶标免疫分析法(夹心ELISA)以免疫生化原理为基础,提供高灵敏度,高特异性和高稳定性的抗原-抗体检测方法。通过应用健康对照和经历过创伤性事件患者的血清,对制备的试剂盒进行反复筛选、验证,获得了有高灵敏度,高特异性和高稳定性的应激后焦虑抑郁等精神心理症状的生物标志物检测试剂盒。并通过结合分析质谱定量数据和ELISA结果,给出了在目标人群中预测焦虑、抑郁、PTSD等精神疾病高发生风险的生物标志物浓度阈值。本发明的血液ELISA检测试剂盒操作更为快速,简单;大大提高了检测结果的精确度和准确度;费用便宜,容易推广,适合市场。ELISA检测方法经各种参数和实验程上的优化,可作为大规模筛选的工具用于应激后精神疾病的早期筛查。
综上,本发明提供一组了可预测应激后焦虑抑郁等精神心理症状的发生的生物标志物及其在血液样品中检测这些标志物的检测试剂盒和检测方法,潜在的患者在经历应激性事件后,可应用本发明的试剂盒在血液中高特异性、高灵敏度、高特异性地检测上述蛋白的浓度,并依据本发明所给出的高风险阈值范围,从而可以为医生准确预测应激病人罹患焦虑、抑郁、PTSD等精神疾病带来依据,进而及时采取治疗,以预防此类疾病的发生,及时减轻病人痛苦和负担,由其制成的免疫检测试剂盒可以快速简单准确地检测血液中的生物标志物浓度,适于大规模推广应用。
在此说明书中,本发明已参照其特定的实施例作了描述。但是,很显然仍可以作出各种修改和变换而不背离本发明的精神和范围。因此,说明书应被认为是说明性的而非限制性的。
Claims (2)
1.一种检测生物标志物的试剂在诊断创伤事件受试者的应激后焦虑抑郁症状发生的试剂盒的应用,其特征在于,所述的生物标志物包括***结合蛋白2。
2.根据权利要求1所述的应用,其特征在于,所述的生物标志物还包括丁酰胆碱酯酶、过氧化氢酶、多巴胺-β-羟化酶、神经纤毛素1中的一种或几种。
