发明内容

具体实施方式
诸位发明人现已发现,可生成经修饰的NK细胞,其产生的重组IL-15的量足以提供不依赖于IL-2/IL-15的生长和刺激,同时维持重组CD16和/或CAR的细胞毒性和功能性表达。此外,在至少一些实施例中,考虑到的经修饰的NK细胞将不仅产生足够的细胞内IL-15(且尤其是erLSP-IL-15),以允许在没有细胞因子的情况下生长和扩增,但也允许分泌一定量的IL-15,当肿瘤微环境(TME)中存在此类细胞时,该量的IL-15有助于TME中的免疫刺激(或免疫抑制的逆转)。因此,根据本发明主题的NK细胞将有利地允许简化培养和扩增,同时通过CD16和/或CAR提供重组靶向细胞毒性的途径。
先前制备的表达具有内质保留序列的IL-2的NK-92衍生物被怀疑支持免疫抑制细胞(诸如骨髓源性抑制细胞(MDSC)和T调节细胞(T-regs))的生长和扩增。这些负调节因子可以减轻免疫细胞且尤其是aNK、haNK和t-haNK以及供体NK细胞的任何抗肿瘤作用。另一方面,重组IL-15仅支持免疫活性细胞的功能,而对erIL-2没有负面影响。
然而,应当理解,IL-15及其变体的表达可能对NK细胞且尤其是NK-92细胞的生长和/或功能产生不利影响。也不能预期IL-15的生物活性形式及其变体的表达,尤其是在不对靶向NK细胞(诸如CD16和/或CAR)也需要的重组蛋白产生不利干扰的情况下支持细胞扩增和活性的量。仍进一步地,超过免疫刺激水平的IL-15及其变体的过表达和分泌可能导致此类细胞的受体的***性副作用。从不同的角度来看,优选的经修饰的NK细胞将产生和分泌足够低的IL-15,以避免***性副作用,但维持TME内经修饰细胞和免疫细胞的所需有益效果,生长和刺激特性。值得注意的是,诸位发明人现在发现,可以制备在细胞内形成和保留的以自分泌方式(即,不需要外源性细胞因子)刺激生长和扩增的IL-15与对TME内的其他免疫感受态细胞提供免疫刺激作用的分泌IL-15之间具有理想平衡的NK细胞。
关于合适的NK细胞,通常考虑到NK细胞可以是来自将接受经基因修饰的NK细胞的受试者的自体NK细胞。此类自体NK细胞可从全血中分离,或使用本领域公知的方法由前体细胞或干细胞培养。然而,还应当理解,NK细胞不需要是自体的,而可以是同种异体或异种NK细胞。在本发明主题的特别优选的方面中,基因工程化NK细胞是NK-92细胞或其衍生物。例如,在本发明主题的一个特别优选的方面,基因工程化NK细胞是NK-92衍生物,其经修饰以具有减少或消除的至少一种杀伤细胞免疫球蛋白样受体(KIR)的表达,这将典型地使此类细胞组成性活化(经由缺乏抑制或降低抑制)。
NK-92细胞表现出不寻常的受体表达谱,表达相对大量的活化受体(例如,NKp30、NKp46、2B4、NKGD、E、CD28)。相反,NK-92细胞还表达很少的抑制性受体(例如,NKGA/B、低水平的KIR2DL4、ILT-2),并缺少正常NK细胞上克隆表达的大多数杀伤抑制性受体(KIR)。此外,NK-92表达相对高水平的参与穿孔素-颗粒酶细胞溶解途径的分子以及另外的细胞毒性效应分子(包括肿瘤坏死因子(TNF)-超家族成员FasL、TRAIL、TWEAK、TNF-α),表明经由可替代的机制杀伤的能力。而且,NK-92细胞还表达关于免疫效应细胞调节的其他分子(CD80、CD86、CD40L、TRANCE),这些分子在NK杀伤方面的相关性尚不清楚。然而,值得注意的是,这些特别理想的特征不会受到本文所述的修饰的不利影响。事实上,在至少一些实施例中,如下文更详细指出的,上述活化剂和/或效应蛋白中的一种或多种可响应于IL-15的细胞内表达而过表达。
而且,适合的NK细胞可具有一个或多个被突变的经修饰的KIR,以减少或消除与MHC I类分子的相互作用。当然,应当注意,还可以使一个或多个KIR缺失或抑制其表达(例如,通过miRNA、siRNA等)。最典型地,多于一个KIR将被突变、缺失或沉默,并且尤其考虑到的KIR包括具有两个或三个结构域、具有短或长的胞质尾区的那些。从不同的角度来看,经修饰的、沉默的或缺失的KIR将包括KIR2DL1、KIR2DL2、KIR2DL3、KIR2DL4、KIR2DL5A、KIR2DI5B、KIR2DS1、KIR2DS2、KIR2DS3、KIR2DS4、KIR2DS5、KIR3DI1、KIR3DL2、KIR3DL3、和KIR3DS1。可以使用本领域熟知的方案制备此类经修饰的细胞。可替代地,此类细胞还可以作为aNK细胞(‘活化的自然杀伤细胞)从南特圭斯特公司(NantKwest)(参见URLwww.nantkwest.com)商购获得。然后,如下文进一步讨论的,此类细胞可以另外地经基因修饰以表达IL-15或其变体。
在本发明主题的另一个优选的方面,基因工程化NK细胞也可经修饰以表达高亲和力Fcγ受体(CD16)的NK-92衍生物。Fcγ受体的高亲和力变体的序列是本领域熟知的(参见,例如,Blood[血液]2009113:3716-3725),并且认为所有产生和表达的方式都适用于本文。据信这种受体的表达允许使用对患者的肿瘤细胞(例如,新表位)、具体的肿瘤类型(例如,her2neu、PSA、PSMA等)特异性的抗体,或与癌症相关联的抗体(例如,CEA-CAM)特异性靶向肿瘤细胞。有利地,此类抗体是可商购的并且可以与细胞连同使用(例如,与Fcγ受体结合)。可替代地,此类细胞还可以作为haNK细胞(‘高亲和力自然杀伤细胞)从南圭斯特(NantKwest)公司商购获得。然后,如下文进一步讨论的,此类细胞可以进一步经基因修饰以表达IL-15或其变体。
可替代地或另外地,基因工程化NK细胞还可被基因工程化以表达嵌合抗原受体(CAR)。在尤其优选的方面,CAR将具有scFv部分或其他针对肿瘤相关抗原、肿瘤特异性抗原和癌症新表位具有结合特异性的胞外结构域。因此,正如容易理解的那样,合适的CAR将包括第一代、第二代和第三代CAR(参见例如,Immunotherapy[免疫疗法](2015)7(5):487-497)。如前所述,存在许多基因工程化NK细胞表达这种嵌合T细胞受体的方式,并且认为所有方式均适用于本文。可替代地,此类细胞还可以作为taNK细胞(‘靶标-活化的自然杀伤细胞’)从南圭斯特(NantKwest)公司商购获得。然后,如下文进一步讨论的,此类细胞可以进一步经基因修饰以表达IL-15或其变体。
在将细胞工程化以具有针对癌症相关抗原或针对癌症相关抗原具有特异性的抗体的亲和力的情况下,考虑到所有已知的癌症相关抗原被认为适合使用。例如,癌症相关抗原包括CEA、MUC-1、CYPB1等。同样地,在将细胞工程化以具有对癌症特异性抗原的亲和力或具有对针对癌症特异性抗原具有特异性的抗体的情况下,考虑到所有已知的癌症特异性抗原被认为适合使用。例如,癌症特异性抗原包括PSA、Her-2、PSA、brachyury等。在将细胞工程化以具有对癌症新表位的亲和力或具有针对癌症新表位具有特异性的抗体情况下,考虑到所有已知的鉴定新表位的方式将产生适合的靶标。例如,可以通过肿瘤活组织检查(或淋巴活组织检查或转移部位的活组织检查)和匹配的正常组织(即,来自同一患者的非患病组织)的全基因组分析,经由同步比较如此获得的组学信息,在第一步中从患者肿瘤中鉴定新表位。然后可以进一步过滤所鉴定的新表位用于匹配患者的HLA类型,以增加新表位抗原呈递的可能性。最优选地,这种匹配可以经由电脑模拟完成。
关于用于表达的合适的IL-15序列,通常优选的是,IL-15为哺乳动物IL-15序列,且最优选地是人IL-15序列(参见例如,UniProt标识符P40933)。此外,值得注意的是,合适的IL-15序列包括各种变体,且尤其考虑到的变体包括具有长信号肽(LSP)和短信号肽(SSP)的变体。LSP变体具有典型地为48个氨基酸的信号肽,且转录物典型地包括316个碱基的5′-非翻译区(UTR)、486个碱基的编码序列和长度为约400个碱基的C-末端3′-UTR区。SSP变体具有典型地为21个氨基酸的短信号肽,其基于选择性剪接(由外显子4A和5编码)。值得注意的是,两种亚型都产生相同的成熟蛋白质,但细胞运输是不同的。更具体地,在高尔基体、早期内体和内质网中检测到IL-15 LSP亚型,且可以以分泌和膜结合形式出现。另一方面,未分泌IL-15 SSP亚型,且其似乎仅限于细胞质和细胞核,其中该亚型显现参与细胞周期的调节。如下文所讨论的,仍进一步考虑到的IL-15变体包括超级激动剂变体,且尤其是IL-15 N72D突变体,其可能包括也可能不包括额外的信号肽和/或运输信号。
基于明显的差异信号传导,诸位发明人因此考虑使用对重组IL-15的各种修饰,从而影响细胞内的适当表达水平和分布以及分泌量。为此目的,诸位发明人考虑使用可与重组IL-15融合的各个信号传导部分。例如,在将IL-15或IL-15变体输出至内吞体和溶酶体区室的情况下,可以使用前导肽(诸如CD1b前导肽),以从细胞质中隔离(新生的)蛋白质。另外地或可替代地,可以采用靶向前序列和/或靶向肽。可以将靶向肽的前序列添加至N末端和/或C末端,并且典型地包含在6-136个之间的碱性氨基酸和疏水性氨基酸。在过氧化物酶体靶向的情况下,靶向序列可以在C末端处。可以使用其他信号(例如,信号斑),并且包括在肽序列中分离并在适当的肽折叠后起作用的序列元件。另外,诸如糖基化的蛋白质修饰可以诱导靶向。在其他适合的靶向信号中,诸位发明人考虑过氧化物酶体靶向信号1(PTS1)(C末端三肽),和过氧化物酶体靶向信号2(PTS2)(是位于N末端附近的九肽)。
另外,还可以通过蛋白质的胞质溶胶结构域内的信号来介导蛋白质向内体和溶酶体的分拣,这些信号通常包含短的线性序列。一些信号被称为基于酪氨酸的分拣信号,并且符合NPXY或共有基序。称为基于双亮氨酸的信号的其他信号也适合[DE]XXXL[LI]或DXXLL共有基序。所有这些信号都被与膜的胞质溶胶表面***相关的蛋白质外壳的成分所识别。和[DE]XXXL[LI]信号被衔接蛋白(AP)复合体AP-1、AP-2、AP-3和AP-4识别为具有特征性精细特异性,然而DXXLL信号则被另一个称为GGA的衔接蛋白家族识别。还可以添加FYVE结构域,该结构域与液泡蛋白分选和内吞体功能相关联。在仍进一步的方面,使用人CD1尾序列还可以靶向内吞体区室(参见,例如,Immunology[免疫学],122,522-531)。例如,使用LAMP1-TM(跨膜)序列可以实现溶酶体靶向;而循环内体可以经由CD1a尾靶向序列来靶向;且分选内体可以经由CD1c尾靶向序列来靶向。
运输至或保留在胞质溶胶区室中不一定需要一种或多种特定的序列元件。然而,在至少一些方面,可以添加N或C末端细胞质保留信号,包括膜锚定蛋白或膜锚定蛋白的膜锚定结构域,使得该蛋白质被保留在面向胞质溶胶的细胞中。例如,膜锚定蛋白包括SNAP-25、突触融合蛋白、小突触泡蛋白(synaptoprevin)、突触结合蛋白、囊泡相关膜蛋白(VAMP)、突触囊泡糖蛋白(SV2)、高亲和力胆碱转运蛋白、神经毒素、电压门控钙通道、乙酰胆碱酯酶和NOTCH。
在仍进一步考虑的本发明主题的方面,IL-15或IL-15变体还可包含一个或多个跨膜片段,该跨膜区段将在加工后将新表位引导至细胞膜的外部,以此使其对免疫感受态细胞可见。本领域中已知存在众多的跨膜结构域,且所有这些都被认为适用于本文,包括具有单α螺旋、多α螺旋、α/β桶状结构等的那些。例如,考虑到的跨膜结构域可包含T细胞受体的α、β或ζ链、CD28、CD3ε、CD45、CD4、CD5、CD8(例如,CD8α、CD8β)、CD9、CD16、CD22、CD33、CD37、CD64、CD80、CD86、CD134、CD137、CD154、KIRDS2、OX40、CD2、CD27、LFA-1(CD11a、CD18)、ICOS(CD278)、4-1BB(CD137)、GITR、CD40、BAFFR、HVEM(LIGHTR)、SLAMF7、NKp80(KLRF1)、CD160、CD19、IL2Rβ、IL2Rγ、IL7Rα、ITGA1、VLA1、CD49a、ITGA4、IA4、CD49D、ITGA6、VLA-6、CD49f、ITGAD、CD11d、ITGAE、CD103、ITGAL、CD11a、LFA-1、ITGAM、CD11b、ITGAX、CD11c、ITGB1、CD29、ITGB2、CD18、LFA-1、ITGB7、TNFR2、DNAM1(CD226)、SLAMF4(CD244、2B4)、CD84、CEACAM1、CRTAM、Ly9(CD229)、CD160(BY55)、PSGL1、CD100(SEMA4D)、SLAMF6(NTB-A、Ly108)、SLAM(SLAMF1、CD150、IPO-3)、BLAME(SLAMF8)、SELPLG(CD162)、LTBR或PAG/Cbp的一个或多个跨膜结构域。
此外,还考虑到IL-15或IL-15变体也可与IL-15受体α亚基共表达。此种共表达被认为允许IL-15或IL-15变体与受体α链的结合或其他缔合,以稳定IL-15或IL-15变体和/或有助于IL-15或IL-15变体在经修饰的NK细胞的表面的分泌或运输/呈递。类似地,各种其他蛋白质(除CD16和/或CAR外)都可与IL-15或IL-15变体共表达,且合适的共表达蛋白质包括各种免疫调节化合物,且尤其是干扰检查点抑制的化合物(例如,针对PD1、PD-L1、CTLA4等的scFv)、干扰免疫刺激(例如,IFN-γ、IL-12、ILL-21等)的化合物和/或结合/抑制与免疫抑制有关的细胞因子(例如,TGF-β、IL-8等)的化合物。
如将容易理解的,IL-15或IL-15变体和其他共表达的蛋白质将编码在重组核酸上,该重组核酸可以是一种或多种重组RNA或DNA分子。然而,最典型地,重组核酸将是多顺反子DNA构建体,且优选是质粒。然而,各种其他构建体也被认为适合于在此使用,且包括病毒载体(可作为病毒或通过其他方法转染)、线性化DNA、结合到载体的DNA(例如,用于射弹转染)等。
无论核酸的特定配置和类型如何,通常优选的是,经修饰的NK-92细胞将以(a)使如此转染的细胞不依赖于外源性细胞因子以及(b)允许刺激/活化靠近转染的细胞(典型地在TME内)的其他免疫感受态细胞的量表达IL-15或IL-15变体。例如,考虑到分泌的或另外的细胞外(或细胞外呈递)重组IL-15或ILL-15变体将占细胞中产生的总IL-15或IL-15变体的5%-10%、或10%-20%、或20%-30%、或30%-50%。因此,从不同的角度来看,分泌的或另外的细胞外(或细胞外呈递)重组IL-15或IL-15变体的存在量可在约20-60pg/ml、或约60-80pg/ml、或约80-100pg/ml、或约100-150pg/ml、或约150-200pg/ml之间或甚至更高。另一方面,考虑到细胞内保留的IL-15或IL-15变体的存在量可在约100-150pg/ml、或约150-250pg/ml、或约250-500pg/rrl、或约500-750pg/ml之间或更高。因此,应注意,特别优选的经修饰的NK-92细胞将产生足够量的重组IL-15或IL-15变体,以支持自主生长和刺激TME中的免疫感受态细胞,以及刺激CD8+T细胞记忆的建立和维持,但此类量不足以在接受此类细胞的患者中引发***性不良事件。
因此,从功能角度来看,重组IL-15或IL-15变体将用于刺激或增强其他NK细胞(例如,TME中的自体NK细胞)、各种T细胞等的效应子功能和/或增殖,以及增强或触发TME中的细胞中的Jak/STAT信号传导。此外,由于细胞内保留的IL-15或IL-15变体的部分,如下文更详细描述的,在完全没有外源性IL-2和/或IL-15的情况下,此类细胞也将能够增殖。在又进一步考虑的方面中,与未修饰的NK-92细胞相比,此类经修饰的NK-92细胞也将具有增加的对IL-12信号传导的敏感性,其可降低IL-4介导的IFN-γ抑制,这反过来可降低TME中的Th1T细胞抑制。
考虑到的经修饰的NK细胞优选地用于治疗对NK细胞的施用有反应的疾病,尤其是用于治疗各种癌症。如将容易理解的,经修饰的NK细胞可作为唯一治疗剂或作为更复杂方案中的药剂施用。例如,经修饰的NK细胞可以是免疫疗法策略的一部分,其中肿瘤可以首先使用突破到TME中的药物(例如,阿伐沙林(abraxane))、减少免疫抑制的药物(例如,环磷酰胺)、刺激各种免疫感受态细胞的药物(例如,ALT-803)以及癌症疫苗组合物(例如,编码肿瘤新抗原的重组AdV)、和/或与有助于维持免疫应答和促进记忆细胞发育的药物(例如,肿瘤靶向IL-12)治疗。在通过引用并入本文的WO 2018/005973中讨论了示例性的合适治疗方案。
关于合适的剂量和施用方式,通常优选的是,细胞量和输注时间表通常遵循输注NK细胞的已知既定方案。因此,经修饰的NK-92细胞的典型量将介于5×107个细胞/剂量IV和5×108个细胞/剂量IV之间,或介于5×108个细胞/剂量IV和5×109个细胞/剂量IV之间,或介于5×109个细胞/剂量IV和5×1010个细胞/剂量IV之间,且最典型地介于7×108个细胞/剂量IV和7×109个细胞/剂量IV之间(例如,2×109个细胞/剂量IV)。当然,应该理解,在经修饰的NK细胞表达CD16或CD16的高亲和力变体(例如,158V)的情况下,可将细胞与有利地与经修饰的NK细胞结合的一种或多种抗体(在输液前体外或依次,例如,与输注细胞前施用的抗体一起)共同施用。
实例
以下实例仅供于代表性指导,且不应理解为限制本发明主题。除非另有说明,否则所有重组表达构建体均使用如SEQ ID NO:1所示的IL-15的密码子优化版本,在至少一些实施例中其提供了更高的重组蛋白产量(数据未显示)。
在第一组实验中,使用SEQ ID NO:1创建了三种IL-15变体:与野生型IL-15相对应的长信号肽变体(LSP-IL-15)、具有ER保留信号的长信号肽变体(erLSP-IL15)和作为替代剪接变体的短信号肽变体(SSP-IL-15)。更特别地,在一个实施例中,IL-15 LSP具有如SEQID NO:2所示的核苷酸序列;IL-15 LSP具有SEQ ID NO:3的肽序列;erIL-15 LSP具有SEQID NO:4的核苷酸序列;erIL-15 LSP具有SEQ ID NO:5的肽序列;IL-15 SSP具有SEQ IDNO:6的核苷酸序列;IL-15具有SEQ ID NO:7的肽序列。
质粒设计和转染:
通过集成DNA技术合成了三个位于KpnI/NotI限制酶位点两侧的gBlock,且用于将每个序列亚克隆到类似消化的pNEUkv1-CD16-erIL2质粒骨架中,以创建以下质粒。

所有质粒均通过Sanger测序确认,通过SalI限制酶消化而线性化,且通过柱纯化分离。通过使用MaxCyte GT电穿孔器(程序NK-92-2-OC)电穿孔1μg线性化DNA/106个细胞来创建细胞系。类似地制备pNEUkv1-CD16-IRES-[erIL-2]且作为阳性对照电穿孔。将该实验重复两次以确认结果。通过电穿孔以下线性化质粒aNK(NK-92“野生型”)创建共表达CD16和erIL-15的NK-92细胞系:pNEUkv1-CD16(158V)-IRES-[LSP IL-15];pNEUkv1-CD16(158V)-IRES-[erIL-15];和pNEUkv1-CD16(158V)-IRES-[SSPIL-15]。将阳性对照用pNeukv1-CD16-IRES-[erIL-2]转染。所有样品均在没有任何细胞因子的X-vivo-10/5%HS中孵育2周。
为了确定质粒的成功转染,使电穿孔细胞在没有任何细胞因子的具有5%HS的X-vivo-10中生长2周。值得注意的是,测试的三个IL-15克隆中只有一个成功生长回来:具有ER保留信号的长信号肽(erIL-15)。此外,如更详细地在下文中显示的,表达erIL-15(erLSP-IL15)的NK-92细胞保持所有相关NK表面标记物和细胞毒性。此外,这些细胞还提供了理想量的保留了其免疫刺激生物学功能的细胞外erLSP-IL15,很可能是在从成熟的IL-15中去除er/LSP区段之后。
从图1可以看出,与aNK或haNK(CD16+,erIL-2)细胞相比,在没有外源性细胞因子的情况下,erIL-15的表达不会改变表达erLSP-IL-15的细胞的生长速率。因此,erLSP-IL-15的重组表达有利地提供足够的自分泌信号传导以支持细胞扩增。此种结果是意外的,因为ER保留序列可能在细胞内不会被去除,但仍然保持适当的功能。此外,erLSP-IL-15的重组表达也不会对经修饰的N-92细胞的许多表型标记物产生不利影响。新产生的细胞系的免疫表型的示例性结果示于图2。在此,测试的流式细胞术表面受体谱与haNK细胞相同,haNK细胞是共表达erIL2的CD16表达NK-92细胞系。
在这种背景下,应该理解,新创建的细胞系保持了功能上关键的表面蛋白的完全表达:CD16(Fc受体)和活化受体NKG2D。为了测试表达erIL-15的细胞系的功能,并与表达erIL-2的细胞系(haNK)进行比较,使用不同的靶细胞系进行细胞毒性测定:K562、Raji和HL-60。图3-5中所示结果明确证明,与haNK细胞相比,erIL-15的引入并未改变自发细胞毒性特性。这是一个意外的发现,因为整合新的/不同的基因可能会影响NK-92细胞的细胞毒性功能。值得注意的是,从图3-5中也可以看出,在相同的效应细胞与靶细胞比率下,表达erIL-15的细胞优于表达erIL-2的细胞。
为了进一步测试新的基于erIL-15的质粒是否可以在不影响CAR活性的情况下掺入三顺反子CAR表达质粒中,用编码CD19CAR、CD20CAR或CD33CAR的三顺反子CAR_CD16_erIL-15质粒通过电穿孔产生NK-92细胞系。测试表达erIL-15的细胞系的细胞毒性功能,且与相应的t-haNK细胞(erIL-2)对几种靶细胞系的细胞毒性功能进行比较:K562(CD19neg、CD20neg、CD33+)、SUPB15CD20(CD20+)、THP-1(CD33+)。图6-8说明了示例性结果。在此,上图中转染的(三顺反子)细胞系对K562的杀伤显示出在所有受试效应物:靶标比率下的卓越杀伤作用。这一结果很重要,因为新的构建体不会影响标准K562系的杀伤(顶部的图)。重要的是,新创建的CAR可以杀死表达CD20和CD33的靶细胞系,这些靶细胞系对野生型NK-92的杀伤具有抵抗力。
在又进一步的实验中,将评估erLSP-IL-15细胞的恢复力和功能。例如,将解冻先前冷冻的erIL-2CD19t-haNK和erIL-15CD19t-haNK,且然后在不含细胞因子的X-Vivo105%中生长。在没有外源添加的细胞因子的情况下,两种细胞系的存活率和扩增特性都具有可比性。
在又进一步的实验中,将测量IL-15和IL-2分泌。在此,考虑到IL-15分泌将与IL-2分泌相等或优于IL-2分泌至少5%、或至少10%、或至少20%、或至少40%、甚至更高,特别是在细胞也共表达IL-15受体α链的情况下。定量地,预期经修饰的NK=92细胞将分泌至少100pg/ml、或至少150pg/ml、或至少200pg/ml、或甚至更高的IL-15,且细胞内量(由裂解物确定)将为至少150pg/ml、或至少250pg/ml、或至少500pg/ml、或至少750pg/ml、或甚至更高。
此外,预期如本文所述的经修饰的NK-92细胞在扩增和培养期间不会表现出成团或其他聚集。类似地,与使用标准生长测定时aNK细胞或haNK细胞的倍增时间相比,本文所述细胞(且特别是表达CD16(优选高亲和力变体)和/或CAR的细胞)将具有基本相同的倍增时间(即,偏差不超过15%、或不超过10%、或不超过5%)。同样,与haNK细胞和t-haNK细胞相比,表达erLSP-IL-15的细胞通常以基本相似的量表达功能活性重组CAR和/或CD16(即,偏差不超过15%、或不超过10%、或不超过5%)。
类似地,预期表达erLSP-IL-15和重组CAR和/或CD16的细胞对K562的非特异性细胞毒性将与aNK和haNK细胞基本相同。如上所示,预期表达erLSP-IL-15和重组CAR的NK-92细胞对CAR介导的SUP-B15细胞杀伤与ADCC介导的SUPB15CD20+细胞杀伤一样。
从功能角度来看,本文提出的考虑到的经修饰的NK-92细胞在有或无靶标刺激(或细胞因子小组IFNγ/IL-8/IL-10/趋化因子)的情况下将具有可比的或增强的IFNγ分泌。此外,预期本文提出的考虑到的经修饰的NK-92细胞将具有可比的或增强的流式标记物(NKp30、NKp44、NKp46、DNAM-1、NKG2D、NKG2A、NKG2C、CD94、CD96、TIGIT、PD-1、PD-L1、CTLA-4、TIM-3、LAG-3、FasL、TRAIL、T-bet、Eomes、颗粒酶B、穿孔素)表达。
在仍进一步考虑的方法中,将执行RNA seq分析以鉴定表达模式,其可用于跟踪肿瘤内经修饰的NK-92细胞的活性和/或存在,以及其他免疫感受态细胞的存在和/或活性。RNA seq分析预期提供基本上代表活化NK细胞的RNA表达模式。

Claims (25)
1.一种经基因修饰的NK细胞,该经基因修饰的NK细胞包含含有编码erLSP-IL-15的第一区段的重组核酸。
2.如权利要求1所述的经基因修饰的NK细胞,其中该NK细胞是NK-92细胞。
3.如前述权利要求中任一项所述的经基因修饰的NK细胞,其中该重组核酸是DNA。
4.如前述权利要求中任一项所述的经基因修饰的NK细胞,其中该重组核酸是线性化质粒。
5.如前述权利要求中任一项所述的经基因修饰的NK细胞,其中该重组核酸进一步包含编码CD16或高亲和力CD16的第二区段。
6.如前述权利要求中任一项所述的经基因修饰的NK细胞,其中该重组核酸进一步包含编码嵌合抗原受体的第三区段。
7.如前述权利要求中任一项所述的经基因修饰的NK细胞,其中该重组核酸进一步包含编码干扰检查点抑制的蛋白质、提供免疫刺激的蛋白质、结合/抑制与免疫抑制有关的细胞因子和/或IL-15受体α链的蛋白质的第四区段。
8.如前述权利要求中任一项所述的经基因修饰的NK细胞,其中该重组核酸包含具有足够强度的启动子,以便驱动该erLSP-IL-15以足以(a)使该经修饰的NK细胞不依赖于外源性细胞因子以及(b)允许刺激/活化靠近该经修饰的NK细胞的其他免疫感受态细胞的量表达。
9.如前述权利要求中任一项所述的经基因修饰的NK细胞,其中该erLSP-IL-15中的IL-15部分包含密码子优化的人IL-15序列。
10.一种经基因修饰的NK细胞,其进一步
包含经由CD16偶联到该细胞的抗体。
11.一种修饰NK细胞的方法,该方法包括:
将重组核酸引入该细胞中,其中该重组核酸第一区段编码erLSP-IL-15。
12.如权利要求11所述的方法,其中该NK细胞是NK-92细胞。

15.如权利要求11-14中任一项所述的方法,其中该重组核酸进一步包含编码CD16或高亲和力CD16的第二区段。
16.如权利要求11-15中任一项所述的方法,其中该重组核酸进一步包含编码嵌合抗原受体的第三区段。
17.如权利要求11-16中任一项所述的方法,其中该重组核酸进一步包含编码干扰检查点抑制的蛋白质、提供免疫刺激的蛋白质、结合/抑制与免疫抑制有关的细胞因子和/或IL-15受体α链的蛋白质的第四区段。
18.如权利要求11-17中任一项所述的方法,其中该重组核酸包含具有足够强度的启动子,以便驱动该erLSP-IL-15以足以(a)使该经修饰的NK细胞不依赖于外源性细胞因子以及(b)允许刺激/活化靠近该经修饰的NK细胞的其他免疫感受态细胞的量表达。
19.如权利要求11-18中任一项所述的方法,其中该erLSP-IL-15中的IL-15部分包含密码子优化的人IL-15序列。

23.如权利要求21所述的药物组合物,该药物组合物每剂量单位包含至少1 x 109个细胞。
24.如权利要求1-10中任一项所述的经基因修饰的NK细胞在治疗癌症中的用途。
25.如权利要求24所述的用途,其中该治疗进一步包括施用突破到该TME中的药物、施用减少免疫抑制的药物、施用刺激免疫感受态细胞的药物、施用癌症疫苗组合物、和/或施用有助于维持免疫应答和促进记忆细胞发育的药物。
