Description
Technical Field
The application relates to the field of skin care products, in particular to an anti-aging and repairing lithospermum composition and application thereof in skin care products.
Background
The urban life with fast rhythm and high intensity brings intangible pressure and challenges while creating comfort and convenience for people. Factors such as air pollution, working pressure, seasonal changes, etc. challenge the fragile skin of the urban person at all times. Common symptoms are dryness, desquamation, acne, collagen loss and breakage, wrinkles, sagging, sensitive skin, red blood streaks, skin damage after sun repair and medical arts, and the final appearance is fine lines of skin and aging.
In order to repair the cell vitality of the 'problem skin', increase the skin compactness and enable the skin to present the full outline, polypeptide is added into a plurality of skin care products at present. The polypeptide is milder than vitamin A and glass color, has no side effect basically, does not need to build tolerance, and can be used for sensitive muscles. The whole and non-fragment proteins (such as collagen) cannot be absorbed through the epidermis layer of the skin, the essence of the polypeptide is small fragments or basic components of the proteins, and part of the polypeptide can break through the skin barrier and be absorbed by the skin relatively to the whole protein, so that the polypeptide has the effect of relieving skin aging and injury to a certain extent.
The variety of polypeptides is large, and each peptide has its own specific repair mechanism. For example, pentapeptides can stimulate collagen, spandex, and hyaluronic acid proliferation, increase skin thickness, and reduce wrinkles. Hexapeptide is involved in competing SNAP-25 at the site of the fusogenic complex, affecting the formation of the fusogenic complex, and if the fusogenic complex is slightly unstable, vesicles are unable to effectively release neurotransmitters, thereby causing muscle contraction to diminish and preventing the formation of wrinkles. If different polypeptides can be compounded, the skin can be repaired in multiple directions, and the synergistic effect is achieved.
However, in practice, it has been found that the polypeptides cannot be simply mixed and compounded, and the skin repairing effect is reduced after the polypeptide is compounded. Because the variety and the addition amount of the polypeptide and the compounded components in the skin care product relate to thousands of combination experiments, enterprises cannot verify the effect brought by the compound polypeptide through limited times of experiments. Because of this, there are few products of polypeptide complexation on the market today, and further research and improvement are needed.
Disclosure of Invention
In order to provide a synergistic composite polypeptide, the application provides an anti-aging and repairing lithospermum composition and application thereof in skin care products.
In a first aspect, the application provides an anti-aging and repairing lithospermum composition, which is prepared from the following components in parts by mass:
0.02-0.7 part of composite polypeptide;
0.003-5.0 parts of lithospermum cell mixture;
0.002-2.0 parts of black bean extract;
0-2.6 parts of other conditioning agents;
the compound polypeptide is two or more than two of tripeptide-1, hexapeptide-9, palmitoyl tripeptide-1, palmitoyl tripeptide-5, palmitoyl tetrapeptide-7 and palmitoyl pentapeptide-4.
Tripeptide-1 is a carrier peptide with antioxidant effect and integrated detoxification and other biological activities; hexapeptide-9 is a very stable collagen peptide, and has excellent effects on strong repairing, especially on repairing acne marks; palmitoyl tripeptide-5 can increase the moisture content of skin, improve skin color, and enhance luster and transparency; palmitoyl tripeptide-1 stimulates collagen and glycosaminoglycan synthesis, reduces skin wrinkles, and improves skin quality. Palmitoyl pentapeptide-4 can greatly promote the generation of skin collagen, enable facial skin to be full and endow elasticity, improve the water content and moisture retention of the skin, enable the skin to feel soft and smooth, and lighten the complexion from inside to outside; palmitoyl tetrapeptide-7 can stimulate proliferation of spandex and hyaluronic acid, increase moisture content and moisture retention of skin, increase skin thickness and reduce fine wrinkles, can enhance absorption, and can increase hyaluronic acid content in skin by acting as cellular messenger to stimulate regeneration of collagen fibers in dermis, thereby helping to tighten skin by drawing moisture into epidermis.
The polypeptide can act on skin from different aspects so as to achieve the aim of repairing dry skin, acne marks, dark yellow skin, fine lines, wrinkles and other skin problems. If the polypeptides can be sufficiently fused, the application field of the polypeptides is technically changed. Through continuous exploration of the inventor, the lithospermum cell mixture, the black bean extract and the compound polypeptide are mixed according to the proportion, and different kinds of polypeptides can be well fused together under the synergistic addition effect of the lithospermum cell mixture and the black bean extract. In addition, the lithospermum cell mixture and the black bean extract can effectively eliminate and remove free radicals, reduce aging, accelerate blood circulation, dilate capillaries and promote the absorption and conversion of polypeptides. Meanwhile, the collagen can be combined with protein, so that healthy collagen is protected from being damaged, new collagen regeneration is promoted, acne marks and wrinkles are effectively reduced, and the skin repairing and repairing agent has an excellent skin repairing effect.
More preferably, the complex polypeptide consists of tripeptide-1, hexapeptide-9, palmitoyl tripeptide-5, palmitoyl tripeptide-1, palmitoyl pentapeptide-4 and palmitoyl tetrapeptide-7 at 1: (1.3-2): (0.3-1): (0.7-1): (1-1.3): (3-4.3) and the mass ratio is compounded.
The ratio of polypeptides is the subject of a long-term study, and the scheme of compounding more than five polypeptides together is not studied at present. The main reasons are that the selection of the types of the polypeptides is unclear whether the polypeptides can be compounded to achieve a synergistic effect, and the related types of the polypeptides are multiple, and the existing enterprises are difficult to verify by making experiments on each proportion, so that the method is a very realistic problem. The application adopts the six polypeptides and is matched with the mixture ratio, so that the skin repairing effect of the anti-aging lithospermum composition is broken through, and acne marks and wrinkles are further reduced.
Preferably, the rest of the conditioning agent is fibronectin 0.002-0.1 part.
Because fibronectin is macromolecular protein, the skin has poor absorptivity to macromolecular protein, and the fibronectin can only play a small repairing role by directly using the fibronectin. However, in the anti-aging and repairing lithospermum composition system, the compound polypeptide, the lithospermum cell mixture and the black bean extract are matched, so that sodium-potassium ATPase beta subunit on the surface of the skin can be activated, the beta subunit is used for regulating gaps among skin cells in the skin, and the cell gap is slightly enlarged, so that the degree is enough to guide fibrin to migrate to basal layers through skin barriers, promote cell growth and proliferation, and play an important role in vital activities of organisms such as skin repairing and tissue regeneration.
Preferably, the remaining conditioning agents are fibronectin, arginine and allantoin at 1: (2-10): the mass ratio of (1-8) is compounded.
When the complex polypeptide, lithospermum cell mixture and black bean extract are first introduced into the skin for repair, metabolic stratum corneum is produced. In order to further assist the absorption of fibronectin by skin, arginine and allantoin are selected and the ratio of the arginine to the allantoin is limited, so that the preparation has obvious effects in the aspects of maintaining skin moisture, improving skin roughness, improving the moisture content of horny layer, reducing the adhesive force of horny layer cells, accelerating the regeneration of epidermal cells, and timely cleaning generated waste, so that the fibronectin entering at the back can be better absorbed, thereby further helping the skin repair and reducing wrinkles.
In a second aspect, the application provides an application of an anti-aging and repairing lithospermum composition in skin care varieties, wherein the addition amount of the anti-aging and repairing lithospermum composition in skin care products is 0.2-15%; wherein the addition amount of the cream in the skin care lotion is 1-13%, the addition amount of the cream in the essence is 3-15%, the addition amount of the cream in the emulsion is 1-14%, and the addition amount of the cream in the eye cream is 0.2-14%.
In a third aspect, the application provides an anti-aging and repairing lithospermum essence, which is prepared from the following components in percentage by mass:
21-33.5% of humectant;
0.01-0.1% of emulsifying agent;
3-15% of an anti-aging and repairing lithospermum composition;
0-1.7% of other auxiliary agents;
adding water to 100%;
the humectant is any two of glycerol, butanediol, 1, 2-hexanediol, sodium hyaluronate, propylene glycol, urea and sucrose.
The rest auxiliary agents are preservative, chelating agent and antioxidant. The preservative system is built up from methylparaben, octyl hydroxamic acid, bis (hydroxymethyl) imidazolidinyl urea and iodopropynyl butyl carbamate.
Because of the plurality of polypeptides and plant extracts with a plurality of active ingredients in the anti-aging and repairing lithospermum composition, the proportion of the anti-aging and repairing lithospermum composition to be added into the essence formed by compounding the rest ingredients needs to be controlled, because the cost of the ingredients used in the anti-aging and repairing lithospermum composition is high, the problem of cost is considered in industrial production, and the use of a small amount of the anti-aging and repairing lithospermum composition can also play a role in achieving a good repairing effect, which is a place needing to be further improved.
The components in the essence are in the addition proportion, the anti-aging and repairing lithospermum composition can exert better repairing effect under the condition of adding 3-11%, the humectant can assist the anti-aging and repairing lithospermum composition to repair skin, and other auxiliary agents have no negative influence on the activity of the anti-aging and repairing lithospermum composition, so that the prepared anti-aging and repairing lithospermum composition has better repairing effect and lower cost.
More preferably, the humectant is any three of glycerin, butylene glycol, 1, 2-hexanediol, sodium hyaluronate, propylene glycol, urea, and sucrose.
Most preferably, the humectant is compounded by glycerin, butanediol, 1, 2-hexanediol, sodium hyaluronate, propylene glycol, sucrose and urea, and the compounding ratio is 100: (70-72): (6-8): (2-3): (3-5): (2-4): (4-5).
The moisture loss is increased during skin aging, so that cells shrink, age, tissues lack and shrink, and the skin gradually has fine wrinkles due to the change of tissue structure and morphology, and the humectant plays an important role in keeping skin moisture and repairing skin barrier function. The moisturizing agent is added into the anti-aging and repairing lithospermum essence system according to a specific proportion, so that the skin barrier is in a relatively softened state, the skin can keep better capability of cleaning cutin, and metabolites of the anti-aging and repairing lithospermum composition after repairing skin can be better cleaned by the skin, thereby accelerating the repairing speed and reducing wrinkles, acne marks and the like more in the same time.

2. Test instrument: face image analyzer VISIA, canfield, usa.
3. The test principle is as follows: a facial image analyzer VISIA was used to take a picture of the face and the image analysis software analyzed the UV spot area of the skin.
Note that: the acne marks are formed by pigment deposition on the skin, and are obtained by VISIA ultraviolet color spot detection and ultraviolet shooting with the peak value of 365nm, and the potential pigment deposition area under the epidermis is objectively reflected, so that the acne mark area is reflected.
4. Experiment design: 66 volunteers with acne marks were selected and divided into 22 groups of 3 volunteers, and each group of volunteers was applied with the test sample (applied to eyes in example 15) on the face after daily cleansing in the morning and evening, using the essences prepared in examples 1 to 18 and comparative examples 1 to 4, respectively. The face image analyzer VISIA is adopted to shoot the face images of the volunteer before and after 8 weeks of using the test sample, and the area of the facial acne marks is obtained through analysis, so that the area of the acne marks is obviously reduced after the use compared with that before the use, and the test sample has the effects of repairing skin and removing the acne marks.
5. Data analysis:
area of acne marks: taking the average value of the test results of each group of volunteers;
acne mark reduction rate= (S 0 -S 1 )/S O ×100%
S 0 : average of the acne mark area of each group of volunteers before use.
S 1 : average of the acne mark area of each group of volunteers after use.
Test 2
Detecting the effect of repairing wrinkles
1. Test instrument: the rapid three-dimensional imaging system of skin PRIMOS, canfield, usa.
2. The test principle is as follows: and obtaining a digital three-dimensional image of the surface of the tested skin by testing the position change of the stripe light irradiated to the surface of the skin and the gray values of all image points, and analyzing by system software to obtain parameters such as the volume of wrinkles.
3. The testing method comprises the following steps: 54 volunteers with fine lines and wrinkles on the face were selected and divided into 18 groups of 3 volunteers, and the essences prepared in examples 1 to 14 and comparative examples 1 to 4 were applied to the face after cleansing the face every day in the morning and evening. The rapid three-dimensional imaging system PRIMOS of the skin is adopted to shoot the face images of the volunteer before and after 6 weeks of using the test sample, and the wrinkle volume value V is obtained by analysis, the wrinkle volume after using is obviously reduced compared with that before using, which indicates that the test sample has the effects of repairing the skin and reducing the wrinkles.
4. And (3) data processing:
wrinkle volume value V value change rate (%) = (V) 0 -V 1 )/V O ×100%
V O : the V value of the skin before use of the test sample;
V 1 : v value of skin after use of the test sample. The data for runs 1 and 2 are collated in table 3.

According to the comparison of the data of the comparative examples 1 and 2 with the data of the examples 1 to 4 in Table 3, the reduction rate of the acne mark area and the reduction rate of the wrinkle of the comparative examples 1 and 2 are far less than those of the examples 1 to 4, which shows that even though the polypeptides cannot be fully fused, different kinds of polypeptides can be well fused together through the preparation of the anti-aging and repairing lithospermum composition and the black bean extract, free radicals can be effectively eliminated and removed, ageing can be reduced, blood circulation can be accelerated and capillaries can be expanded, absorption and conversion of the polypeptides can be promoted, the polypeptide can be combined with protein, healthy collagen can be protected from damage, new collagen regeneration can be promoted, acne marks and wrinkles can be effectively reduced, and the skin repairing effect is excellent.
As can be seen from the comparison of the data of examples 1-4 and examples 5-6 in Table 3, the reduction rate of the acne mark area and the reduction rate of the wrinkle of examples 5 and 6 are far greater than those of examples 1-4, which shows that more than five compound polypeptides can be fully fused together to act on the skin together under the coordination of the lithospermum cell mixture and the black bean extract, effectively reduce the acne mark and the wrinkle, and have excellent skin repairing effect.
As can be seen from the comparison of the data of examples 7 and 8 with the data of comparative examples 3 and 4 in Table 3, the reduction rate of the acne mark area and the reduction rate of the wrinkle of examples 7 and 8 are far greater than those of comparative examples 3 and 4, which shows that the skin can be well repaired only within the range of the input amount of the composite polypeptide of the present application, and the ratio of the components in the cosmetics is sufficient to affect the compatibility as well, and is not adjusted randomly.
As can be seen from comparison of the data in Table 3 for examples 9, 10 and example 8, the reduction rate of the acne mark area and the reduction rate of the wrinkles in examples 9, 10 were both greater than those in example 8, indicating that the present application employs a composition consisting of tripeptide-1, hexapeptide-9, palmitoyl tripeptide-5, palmitoyl tripeptide-1, palmitoyl pentapeptide-4 and palmitoyl tetrapeptide-7 in a ratio of 1: (1.3-2): (0.3-1): (0.7-1): (1-1.3): the compound polypeptide compounded in the mass ratio of (3-4.3) can break through the skin repairing effect of the anti-aging lithospermum composition, thereby further reducing acne marks and wrinkles.
As can be seen from the comparison of the data of examples 11 and 12 and example 9 in Table 3, the reduction rate of the acne mark area and the reduction rate of the wrinkles of examples 11 and 12 are both larger than those of example 9, which shows that the complex polypeptide, the lithospermum cell mixture and the black bean extract are matched to activate sodium-potassium ATPase beta subunits on the skin surface, wherein the beta subunits are used for regulating gaps among skin cells in the skin, and the cell gap is slightly enlarged enough to guide fibrin to migrate to basal layers through skin barriers, so that the effects of promoting cell growth and proliferation are important in the vital activities of the organism such as skin repair and tissue regeneration and the like are realized.
According to the comparison of the data of examples 13 and 14 and example 12 in Table 3, the reduction rate of the acne mark area and the reduction rate of the wrinkles of examples 13 and 14 are both larger than those of example 12, which shows that the ratio of the amino acid to the allantoin is defined, so that the skin moisture is maintained, the skin roughness is improved, the moisture content of the horny layer is improved, the adhesion of the horny layer cells is reduced, the updating of the epidermal cells is accelerated, and the generated waste is cleaned in time, so that the fibronectin entering from the back can be absorbed better, thereby further helping the skin repair and reducing the wrinkles.
The present embodiment is only for explanation of the present application and is not to be construed as limiting the present application, and modifications to the present embodiment, which may not creatively contribute to the present application as required by those skilled in the art after reading the present specification, are all protected by patent laws within the scope of claims of the present application.
