1.2 clinical trials
1.2.1 Effect on Hartley guinea pig histamine phosphate itching response
The experimental method comprises the following steps:
experimental test article: liquid soap for eliminating turbid and relieving itching.
Three-ingredient heat-clearing itching-relieving lotion, produced by Guangxi Baiyunshan Yingkang pharmaceutical industry Co.
Histamine phosphate, manufactured by national pharmaceutical group chemical company, inc.
Experimental animals: hartley guinea pigs weighing 250g-400g.
Method and results
Effect on the itching response of histamine phosphate in Hartley guinea pigs.
80 healthy Hartley guinea pigs with the weight of 250-400 g are selected and randomly divided into 10 groups of 10 male and female parts. Blank control group (distilled water 0.1ml/100 g), positive control test sample three-flavor heat-clearing and itching-relieving lotion group (three-flavor heat-clearing and itching-relieving lotion 0.1ml/100 g), turbid-discharging itching-relieving liquid soap group (0.1 ml/100 g). Shaving the backs of the right back feet of the Hartley guinea pigs of each group 2 days before the experiment, and externally coating corresponding experiment test objects at the shaving positions of the animals of each group for 2 days for 1 time a day; on day 3, on the experimental day, animals of each group scratch the shaved parts of the backs of the right feet with coarse sand paper to damage the epidermis, the skin is slightly permeated with blood to the extent that the area is 1cm 2 After the corresponding experimental test objects are coated on the parts of the animals of each group for 1 time, the experimental test objects are coated for 10 minutes at last time, 0.05 ml/dose of 0.01% histamine phosphate solution is started to be dripped on the wound surface, the concentration is gradually increased by 0.02%, 0.03% and 0.04% … … … … every 3 minutes, each time, the concentration is 0.05 ml/dose until the Hartley guinea pig licks the right hind foot, the total amount (mug) of histamine phosphate which is given when the Hartley guinea pig licks the right hind foot last occurs is taken as an itching threshold, each group of data is recorded, and the data are subjected to statistical t and t' tests, and the results are shown in Table 1.
TABLE 3 influence of 10 groups of turbid discharge antipruritic liquid soap samples on the itching response of Hartley guinea pig histamine phosphaten=10)
Note that: comparison to the blank control group: * P < 0.05, P < 0.01, P < 0.001.
Experimental results show that 0.1ml/100g of the liquid soap group (example 3, example 4 and example 5) for eliminating turbid and relieving itching can obviously improve the itching threshold value of the histamine phosphate on guinea pigs when being externally used, and has obvious effects of eliminating turbid and relieving itching. However, in example 1, the effect is obviously inferior to that of examples 3, 4 and 5 because the adding ratio of the sample is too small, and the itching threshold of the single medicinal materials of the wampee tree extract, the pomelo leaf extract, the lantana camara leaf extract, the loquat leaf extract and the honeysuckle extract on guinea pigs is relatively low. The experimental results show that the mixture of the five traditional Chinese medicine extracts has stronger turbidity-removing and itching-relieving effects than the single traditional Chinese medicine extract. The experimental results show that the mixing ratio of the five traditional Chinese medicines is between 0.5-2:1.5-3:1.5-3:0.5-2:0.5-2, and a good turbidity-removing and itching-relieving effect can be achieved. The antipruritic effect of examples 3 to 5 is far greater than the one-way effect of examples 6 to 10.
1.2.2 antibacterial Property experiments
The test was performed using the QB/T2738-2012 (7.3) method. The action concentration and time are as follows: 10% strength and 20 minutes of action time. The experimental results are as follows:
table 4 results of antibacterial property test
Experiments show that the antibacterial performance of the five traditional Chinese medicine mixed extract solutions of examples 1-5 containing lantana camara, naria leaf, loquat leaf extract, honeysuckle and wampee is obviously higher than that of examples 6-10, and the antibacterial effect is obviously reduced by only using a single traditional Chinese medicine extract solution. When the implementation concentration of the mixed extract of the traditional Chinese medicines containing lantana camara, naringin leaf, loquat leaf extract, honeysuckle and wampee tree is gradually increased from 1% to 10%, the antibacterial rate is also increased from 50% to 90%, and the antibacterial effect is related to the used concentration. In examples 3 to 5, the composition ratio of the five traditional Chinese medicines is within the range (0.5-2:1.5-3:1.5-3:0.5-2:0.5-2), so that the antibacterial performance of the product can be ensured.
Experiment 2 application of turbidity-expelling and itching-relieving traditional Chinese medicine composition in shampoo
Table 5 1 application of extract in shampoo
In examples 11-13, the shampoo was prepared using the conventional shampoo formulation: adding APG into deionized water, further adding citric acid, adding fatty alcohol disodium succinate, CAB, triisostearate PEG-120 methyl glucose, olive oil, jojoba oil ester, camellia seed oil and the traditional Chinese medicine composition for removing turbid and relieving itching, sequentially adding the raw materials, stirring while adding, heating to about 75 ℃, cooling to about 40 ℃ in the next step, and adding preservative and essence.
