The hydroxytyrosol (3, 4-dihydroxyphenylethanol) has wide application in food, health care products and cosmetics, and has wide market value. 2) The HpaB gene with the PDC1p promoter homologous arm and the GPM1t terminator homologous arm is connected to a vector PRS403-PDC1p-GPM1t with a PDC1p promoter and a GPM1t terminator by homologous recombination to construct an HpaB expression cassette capable of being expressed in a saccharomyces cerevisiae strain; the HpaC gene with the homologous arm of the TEF1p promoter and the homologous arm of the PGK1t terminator is connected to a vector PRS403-TEF1p-PGK1t with the TEF1p promoter and the PGK1t terminator through homologous recombination to construct an HpaC expression cassette (recombinant expression vector) capable of expressing HpaC in a Saccharomyces cerevisiae strain.
3) Different HpaC expression cassettes (with promoters and terminators) are amplified in vitro by PCR and are connected to the HpaB expression cassette to construct co-expression cassettes of different HpaB/C combinations;
4) and (3) introducing the co-expression cassettes with different HpaB/C combinations into a saccharomyces cerevisiae strain for producing the hydroxytyrosol, and successfully integrating the co-expression cassettes into a saccharomyces cerevisiae genome through sequencing verification to obtain the saccharomyces cerevisiae for producing the hydroxytyrosol at high yield.
Example 3: construction of expression of PaHpaBQ212DMutant saccharomyces cerevisiae
1) The Saccharomyces cerevisiae strain constructed in example 2 is mutated into PaHpaB by using CRISPR-Cas9Q212D. The spacer is designed to be 5'-TCAAGGTTCTGCTCAATTGT-3', and the sequence of the homology arm is as follows:

2) transferring Cas9 plasmid with corresponding spacer and homologous arm into a saccharomyces cerevisiae strain expressing a PaHpaB-HpaC combination, amplifying a PaHpaB sequence by colony PCR and genome PCR, and sequencing to verify that the PaHpaB is successfully mutated into the PaHpaBQ212D
Example 4: construction of Saccharomyces cerevisiae strain with weakly-expressed TRP2 gene
On the basis of the strain constructed in example 2 or the strain constructed in example 3, the following steps were carried out:
1) the CRISPR-Cas9 is used for constructing a weakly expressed TRP2 gene, and the TRP2 original promoter is replaced by a weak promoter, and preferably, the TRP2 original promoter is replaced by YEN1p weak promoter. Design the spacer as 5'-CTATGGTCCGTCGTAGCAGA-3', SEQ ID No.27, which is YEN1p weak promoter sequence with homologous arm, wherein 489bp-916bp is YEN1p weak promoter sequence, the rest is homologous arm sequence, which can be adjusted according to the actual situation;
2) the Cas9 plasmid with the corresponding spacer and the homology arm are transferred into a saccharomyces cerevisiae strain, and after colony PCR and genome PCR amplification, the original TRP2 promoter is correctly replaced through sequencing verification.
Example 5: test for the preparation of hydroxytyrosol
1. Fermentation process
1) Culturing a saccharomyces cerevisiae strain with high yield of hydroxytyrosol on a YPD plate, and activating the saccharomyces cerevisiae strain;
2) selecting single clone in 5ml YPD liquid, and culturing for 36 hours;
3) transferring 5ml YPD culture solution to 50ml YPD according to a ratio of 1:50, and culturing at 30 ℃ and 220rpm for 84 hours;
4) collecting supernatant of the fermentation liquor to obtain fermentation liquor containing hydroxytyrosol; the yield of hydroxytyrosol was checked by HPLC.
2. Test results
(1) The method of example 1 was followed to construct various HpaB/C combinations of Saccharomyces cerevisiae producing hydroxytyrosol, wherein the various groups of Saccharomyces cerevisiae producing tyrosol were the same, and the method of example 1 was followed, while the experimental conditions were kept the same:
overexpressing Saccharomyces cerevisiae ARO2, ARO10, TKL1 and RKI1, overexpressing Saccharomyces cerevisiae ARO4K229LAnd ARO7G141SMutants, and knock-out expression of saccharomyces cerevisiae PHA2 and PDC 1;
then the constructed saccharomyces cerevisiae for producing hydroxytyrosol is subjected to a fermentation experiment, and the result is shown in figure 1;
the results in FIG. 1 show that compared to the EcHpaB + SeHpaC combination and the EcHpaB + EcHpaC combination, there was a significant yield increase for both the PaHpaB + PaHpaC, PaHpaB + EcHpaC, PaHpaB + SeHpaC, and EcHpaB + PaHpaC combinations, with the best effect for PaHpaB + EcHpaC, indicating that the introduction of different HpaB/C combinations directly affects the hydroxytyrosol yield of Saccharomyces cerevisiae, while the low yields could not be predicted in advance.
(2) Construction of PaHpaB following the procedure of example 2Q212DThe combination of the mutant saccharomyces cerevisiae HpaB/C is the best PaHpaB + EcHpaC in the step (1), the saccharomyces cerevisiae producing tyrosol is the same as the chassis strain in the step (1), and the fermentation experiment result is shown in a figure 2; FIG. 2 results show the use of PaHpaBQ212DThe mutant can further improve the hydroxyl of the strainTyrosol yield.
(3) Constructing a saccharomyces cerevisiae strain with weak expression of TRP2 gene according to the method of example 3 on the basis of the strain in (2), wherein the fermentation experiment result is shown in fig. 3; FIG. 3 shows that YEN1p weakly expressing the TRP2 gene can again improve the hydroxytyrosol production of the strain.
(4) With reference to the technical method of example 1, tyrosol-producing s.cerevisiae was constructed as follows:
overexpressing Saccharomyces cerevisiae ARO2, ARO10, TKL1 and RKI1, overexpressing Saccharomyces cerevisiae ARO4K229L、ARO7G141SAnd ARO3D154NMutant, overexpressing TyrA from E.coliM53I V354VDouble mutants, and knock-out expression of saccharomyces cerevisiae PHA2 and PDC 1;
then, according to the results of (1) and (3), the TRP2 gene was expressed using a combination of PaHpaB + EcHpaC and a weak promoter YEN1p, and the obtained strain had a fermented yield of 1120 mg/L.
The foregoing is only for the purpose of understanding the method of the present invention and the core concept thereof, and it will be understood by those skilled in the art that various changes and modifications may be made without departing from the principle of the invention, and the invention also falls within the scope of the appended claims.

Claims (10)

1. The saccharomyces cerevisiae capable of producing hydroxytyrosol at high yield is characterized in that HpaB and HpaC which are combined randomly as follows are further expressed on the basis of the saccharomyces cerevisiae capable of synthesizing the hydroxytyrosol:
(1) HpaB from Pseudomonas aeruginosa, and HpaC from Salmonella enterica, Pseudomonas aeruginosa, or Escherichia coli; or
(2) HpaB derived from Escherichia coli and HpaC derived from Pseudomonas aeruginosa.
2. The hydroxytyrosol producing saccharomyces cerevisiae as claimed in claim 1, wherein the HpaB derived from pseudomonas aeruginosa is wild type or mutant with Q212D mutation.
3. The high-yield Saccharomyces cerevisiae of claim 1, wherein the Saccharomyces cerevisiae capable of synthesizing tyrosol is obtained by modification of any one of the following ways:
(1) overexpressing Saccharomyces cerevisiae ARO2, ARO10, TKL1 and RKI1, overexpressing Saccharomyces cerevisiae ARO4K229LAnd ARO7G141SMutants, and knock-out expression of saccharomyces cerevisiae PHA2 and PDC 1;
(2) overexpressing Saccharomyces cerevisiae ARO2, ARO10, TKL1 and RKI1, overexpressing Saccharomyces cerevisiae ARO4K229L、ARO7G141SAnd ARO3D154NMutants, and knock-out expression of saccharomyces cerevisiae PHA2 and PDC 1;
(3) overexpressing Saccharomyces cerevisiae ARO2, ARO10, TKL1 and RKI1, overexpressing Saccharomyces cerevisiae ARO4K229L、ARO7G141SAnd ARO3D154NMutant, overexpressing TyrA from E.coliM53I V354VDouble mutants, and knock-out expression of saccharomyces cerevisiae PHA2 and PDC 1.

The high-yield hydroxytyrosol saccharomyces cerevisiae according to any one of the claims 1-3, which further comprises: the promoter of the TRP2 gene endogenous to the saccharomyces cerevisiae capable of synthesizing tyrosol is a weak promoter, and the weak promoter is a promoter with the promoter capability weaker than that of the original promoter.
5. The application of HpaB and HpaC or their expression vectors combined as follows in constructing Saccharomyces cerevisiae for high yield of hydroxytyrosol:
(1) HpaB from Pseudomonas aeruginosa, and HpaC from Salmonella enterica, Pseudomonas aeruginosa, or Escherichia coli; or
(2) HpaB derived from Escherichia coli and HpaC derived from Pseudomonas aeruginosa.

The method for constructing saccharomyces cerevisiae for high yield of hydroxytyrosol as claimed in claim 1, wherein the saccharomyces cerevisiae for high yield of hydroxytyrosol is obtained by transferring a recombinant vector expressing any combination of HpaB and HpaC into saccharomyces cerevisiae capable of synthesizing hydroxytyrosol;
or integrating the HpaB and HpaC coding genes combined randomly into a Saccharomyces cerevisiae genome capable of synthesizing tyrosol for expression to obtain the Saccharomyces cerevisiae with high yield of hydroxytyrosol;
(1) HpaB from Pseudomonas aeruginosa, and HpaC from Salmonella enterica, Pseudomonas aeruginosa, or Escherichia coli; or
(2) HpaB derived from Escherichia coli and HpaC derived from Pseudomonas aeruginosa.
7. The construction method according to claim 6, wherein the Saccharomyces cerevisiae capable of synthesizing tyrosol is obtained by modification in any one of the following ways:
(1) overexpressing Saccharomyces cerevisiae ARO2, ARO10, TKL1 and RKI1, overexpressing Saccharomyces cerevisiae ARO4K229LAnd ARO7G141SMutants, and knock-out expression of saccharomyces cerevisiae PHA2 and PDC 1;
(2) overexpressing Saccharomyces cerevisiae ARO2, ARO10, TKL1 and RKI1, overexpressing Saccharomyces cerevisiae ARO4K229L、ARO7G141SAnd ARO3D154NMutants, and knock-out expression of saccharomyces cerevisiae PHA2 and PDC 1;
(3) overexpressing Saccharomyces cerevisiae ARO2, ARO10, TKL1 and RKI1, overexpressing Saccharomyces cerevisiae ARO4K229L、ARO7G141SAnd ARO3D154NMutant, overexpressing TyrA from E.coliM53I V354VDouble mutants, and knock-out expression of saccharomyces cerevisiae PHA2 and PDC 1.
8. The construction method according to claim 6 or 7, further comprising: the promoter of TRP2 gene endogenous to saccharomyces cerevisiae capable of synthesizing tyrosol was replaced with a promoter having a weaker promoter ability than the original promoter.
9. Use of the high hydroxytyrosol yielding Saccharomyces cerevisiae according to any of the claims 1-4 for the preparation of hydroxytyrosol.
10. A method for preparing hydroxytyrosol, which is characterized in that the Saccharomyces cerevisiae for high yield of hydroxytyrosol of any one of claims 1 to 4 is used for fermentation culture, and the supernatant of the fermentation liquid is collected to separate hydroxytyrosol.
