本发明公开了一种具有解酒保肝中药组合物的制备工艺及保存方法,制备方法包括S1.以葛根中有效成分葛根素含量为指标,进行水提取,加水6‑8倍,浸泡30‑90mi n,煎煮30‑90mi n,煎煮1‑3次,取其滤液,得到水提取溶液;S2.将步骤S1的水提取溶液浓缩到既定体积120‑144mL,加50vo l%‑70vo l%乙醇,于10℃下静置6‑18h,过滤,回收乙醇,定容。保存方法还包括S3.将步骤S2得到的溶液用盐酸和氢氧化钠调节pH至3‑9,分装为20ml/支,进行水煮灭菌保存。本发明用水提取工艺和醇沉积工艺来提取有效成分,并研究出具有较佳提取效果所用的提取工艺参数。

背景技术
酒中的乙醇及其代谢物会损害人的中枢神经***,使神经***从兴奋到高度的抑制,严重地破坏神经***的正常功能。过量的饮酒就是损害肝脏。慢性酒精中毒,则可导致酒精性肝硬化。此外慢性酒精中毒,对身体还有多方面的损害。如可导致多发性神经炎、心肌病变、脑病变、造血功能障碍、胰腺炎、胃炎和溃疡病等,还可使高血压病的发病率升高。还有人注意到,长期大量饮酒,能危害生殖细胞,导致后代的智力低下。经常大量饮酒的人,其喉癌及消化道癌发病率明显增加。因此,研制具有解酒醒酒作用的中药组合物具有重大意义。

有鉴于此,本发明的目的在于提出一种具有解酒保肝中药组合物的制备工艺及保存方法,该制备工艺较为简单,但具有较佳的提取效果。
所采用的技术方案为:
一种具有解酒保肝中药组合物的制备工艺,所述具有解酒保肝中药组合物含有枳椇子和葛根,该制备工艺包括如下步骤:
S1.以葛根中有效成分葛根素含量为指标,进行水提取,加水6-8倍,浸泡30-90min,煎煮30-90min,煎煮1-3次,取其滤液,得到水提取溶液;
S2.将步骤S1的水提取溶液浓缩到既定体积120-144mL,加50vol%-70vol%乙醇,于10℃下静置6-18h,过滤,回收乙醇,定容。
优选地,S1中,以葛根中有效成分葛根素含量为指标,进行水提取,加水8倍,浸泡90min,煎煮60min,煎煮3次,取其滤液,得到水提取物;
优选地,S2中,将步骤S1的水提取溶液浓缩到既定体积120-144mL,加60vol%乙醇,于10℃下静置12h,过滤,回收乙醇,定容至60ml。
进一步地,所述具有解酒保肝中药组合物还含有青果和桑葚。

S2.将步骤S1的水提取溶液浓缩到既定体积120-144mL,加50vol%-70vol%乙醇,于10℃下静置6-18h,过滤,回收乙醇,定容。

S2醇沉积的最佳工艺为:将水提取溶液浓缩到既定体积120-144mL,加60vol%乙醇,于10℃下静置12h,过滤,回收乙醇,定容至60ml。
下面通过实验进一步证实:
以加水量、浸泡时间、煎煮时间、煎煮次数为因素,选用L9(34)因素水平进行正交实验。称取样品,按照表中的9种工艺进行水提取工艺,提取液浓缩为60ml。

取中药在不同影响因素下进行正交试验,以葛根素(浓度为0.104272mg/ml)为对照,结果显示在不同影响因素条件下,A3B3C2D3对葛根素含量有显著影响,结果如表2所示,故确定最佳水提取工艺为加水8倍,浸泡90min,煎煮60min,煎煮3次,并以葛根素(浓度为0.05214mg/ml)为对照品进行水提取工艺复核,显示葛根素含量稳定,结果如表3所示。

对水提取溶液浓缩到既定体积,进行醇沉积工艺研究,考察乙醇浓度,静置时间对葛根素含量的影响,结果显示加醇至60%,于10℃下静置12小时,所得葛根素含量最大,结果如表5所示。

本发明的一种具有解酒保肝中药组合物的保存方法,其包括上述步骤S1和S2,还包括如下步骤:
S3.将步骤S2得到的溶液用盐酸和氢氧化钠调节pH至3-9,分装为20ml/支,进行水煮灭菌保存。
其中保存的最佳工艺为:调节pH值为4.5-6进行水煮灭菌保存。
下面通过实验进一步证实:
S3.将步骤S2得到的溶液用盐酸和氢氧化钠调节pH至3-9,分装为20ml/支,然后分别用水煮灭菌和高压蒸汽灭菌。高压蒸汽灭菌由于温度高,导致产品口感出现糊味,故以水煮灭菌作为灭菌方式。
而从色谱分析看出,pH值<4.5或大于6时,灭菌对溶液影响大,从而选择调节pH值为4.5-6进行水煮灭菌保存。

Claims (6)
1.一种具有解酒保肝中药组合物的制备工艺,其特征在于,所述具有解酒保肝中药组合物含有枳椇子和葛根,该制备工艺包括如下步骤:
S1.以葛根中有效成分葛根素含量为指标,进行水提取,加水6-8倍,浸泡30-90min,煎煮30-90min,煎煮1-3次,取其滤液,得到水提取溶液;
S2.将步骤S1的水提取溶液浓缩到既定体积120-144mL,加50vol%-70vol%乙醇,于10℃下静置6-18h,过滤,回收乙醇,定容。
2.根据权利要求1所述的具有解酒保肝中药组合物的制备工艺,其特征在于,S1中,以葛根中有效成分葛根素含量为指标,进行水提取,加水8倍,浸泡90min,煎煮60min,煎煮3次,取其滤液,得到水提取物。
3.根据权利要求1所述的具有解酒保肝中药组合物的制备工艺,其特征在于,S2中,将步骤S1的水提取溶液浓缩到既定体积120-144mL,加60vol%乙醇,于10℃下静置12h,过滤,回收乙醇,定容至60ml。
4.根据权利要求1所述的具有解酒保肝中药组合物的制备工艺,其特征在于,所述具有解酒保肝中药组合物还含有青果和桑葚。
5.一种具有解酒保肝中药组合物的保存方法,其特征在于,其包括权利要求1-4任一所述的具有解酒保肝中药组合物的制备工艺,还包括如下步骤:
S3.将步骤S2得到的溶液用盐酸和氢氧化钠调节pH至3-9,分装为20ml/支,进行水煮灭菌保存。
6.根据权利要求5所述的具有解酒保肝中药组合物的保存方法,其特征在于,调节pH值为4.5-6进行水煮灭菌保存。
