Abstract
The invention relates to a chitosan-coated plant growth substance sustained-release microcapsule and a preparation method thereof, belonging to the field of plant growth substances, pesticides and agricultural fertilizers. The chitosan-coated plant growth substance sustained-release microcapsule is obtained by coating plant growth substances with chitosan. Under the condition of the same dosage, the chitosan is adopted to coat the plant growth substance sustained-release microcapsule to keep the pesticide effect for a longer time.

Technical Field
The invention relates to a chitosan-coated plant growth substance sustained-release microcapsule and a preparation method thereof, belonging to the field of plant growth substances, pesticides and agricultural fertilizers.
Background
Plant growth substances are small molecular substances which can significantly influence the growth and development and physiological functions of plants under the concentration of a base. The plant growth promoter, the plant growth retardant and the plant growth inhibitor are classified into three categories according to the growth effect on plants, are widely applied to agricultural, forestry and horticultural crops, and achieve remarkable effect.
The use of plant growth substances, the concentration of which on the plant is one of the key factors affecting its effect. If the concentration is too high, the chemical injury is easily caused. At present, the phytotoxicity treatment of plant growth substances mostly adopts clear water for leaching, and is sprayed with a medicament with a relieving function for remediation, but the problem is not solved from the preparation. The reasonable control of the concentration of the plant growth substances on the plants, particularly the local concentration, is the key to prevent the source of the phytotoxicity problem of the plant growth substances.
Disclosure of Invention
The invention adopts the plant growth substance microcapsule coated by chitosan, and solves the problems of easy phytotoxicity generation, poor stability, short duration and the like of the traditional pesticide preparation.
The invention provides a chitosan-coated plant growth substance slow-release microcapsule, which is obtained by coating a plant growth substance with chitosan.
The plant growth substance is preferably indolebutyric acid, sodium naphthylacetate, dichlorophenoxyacetic acid, triclosan, iodophenoxyacetic acid, gibberellin, zeatin nucleoside, forchlorfenuron, 6-benzyladenine, ethephon, abscisic acid or brassin.
The molecular weight of the chitosan is preferably 100-500kDa, and the deacetylation degree is 60-99 percent.
The invention preferably selects the mass ratio of the plant growth substance to the chitosan as 1: 100-1000.
The invention preferably selects the particle size of the chitosan coated plant growth substance slow-release microcapsule to be 0.5-20 μm.
The invention also aims to provide a preparation method of the chitosan-coated plant growth substance sustained-release microcapsule, which comprises the following steps: adding the plant growth substance solution into an acetic acid aqueous solution of chitosan to obtain plant growth substance emulsion; and (3) carrying out spray drying on the plant growth substance emulsion to obtain the chitosan-coated plant growth substance sustained-release microcapsule.
The concentration of the plant growth substance in the plant growth substance solution is preferably 0.1-50 mg/mL; the concentration of the surfactant in the plant growth substance solution is 0-40 mg/mL; the surfactant is at least one of tween 20, tween 40, tween 60, tween 80, lecithin, polyoxyethylene 400 monolaurate, polyoxyethylene 400 monostearate and polyoxyethylene 400 monooleate; the solvent in the plant growth substance solution is methanol, ethanol or ethyl acetate.
The invention preferably selects the chitosan concentration in the acetic acid water solution of the chitosan to be 0.5-20 mg/mL; the mass percentage concentration of the glacial acetic acid in the acetic acid aqueous solution is 0.2-2%.
The invention preferably selects the volume ratio of the plant growth substance solution to the acetic acid aqueous solution of chitosan to be 1: 1-5.
The invention preferably selects the air inlet temperature of the spray drying to be 180-200 ℃ and the air outlet temperature to be 70-100 ℃.
The invention has the beneficial effects that:
the chitosan-coated plant growth substance sustained-release microcapsule prepared by the invention has better sustained-release performance, and can avoid phytotoxicity caused by overhigh local concentration of plant growth substances;
the chitosan is adopted to coat the plant growth substance sustained-release microcapsule, and the once administration dosage can be higher than that of the uncoated plant growth substance;
under the condition of the same dosage, the chitosan is adopted to coat the plant growth substance sustained-release microcapsule to keep the pesticide effect for a longer time;
besides coating plant growth substances, the chitosan also has the activity of inducing the improvement of the plant immune activity;
the invention uses degradable natural polysaccharide chitosan as coating material, the coating effect is good and the degradation product has no pollution to the environment.

The following non-limiting examples are presented to enable those of ordinary skill in the art to more fully understand the present invention and are not intended to limit the invention in any way.
Example 1
A preparation method of a chitosan-coated indolebutyric acid sustained-release microcapsule comprises the following steps:
dissolving an indolebutyric acid original drug and lecithin in ethanol to obtain an indolebutyric acid solution, wherein the concentrations of the indolebutyric acid original drug and the lecithin in the indolebutyric acid solution are 0.1mg/mL and 10mg/mL respectively;
dissolving chitosan in an acetic acid aqueous solution to obtain an acetic acid aqueous solution of chitosan, wherein the concentration of chitosan in the acetic acid aqueous solution of chitosan is 1mg/mL, the molecular weight of chitosan is 100kDa, the degree of deacetylation is 60%, and the mass percentage concentration of glacial acetic acid in the acetic acid aqueous solution is 2%;
slowly dropwise adding 1 volume of indolebutyric acid solution into 2 volumes of chitosan acetic acid aqueous solution under the condition of stirring, and continuously stirring for 30min to obtain indolebutyric acid emulsion;
and (3) carrying out spray drying on the indolebutyric acid emulsion, wherein the air inlet temperature is 200 ℃, and the air outlet temperature is 70 ℃, so as to obtain the chitosan-coated indolebutyric acid sustained-release microcapsule with the particle size of 1-10 mu m.
Example 2
A preparation method of chitosan coated gibberellin sustained-release microcapsules comprises the following steps:
dissolving a gibberellin original drug and Tween 20 in ethanol to obtain a gibberellin solution, wherein the concentrations of the gibberellin original drug and the Tween 20 in the gibberellin solution are respectively 50mg/mL and 5 mg/mL;
dissolving chitosan in an acetic acid aqueous solution to obtain an acetic acid aqueous solution of chitosan, wherein the concentration of chitosan in the acetic acid aqueous solution of chitosan is 20mg/mL, the molecular weight of chitosan is 300kDa, the degree of deacetylation is 75%, and the mass percentage concentration of glacial acetic acid in the acetic acid aqueous solution is 0.2%;
slowly dropwise adding 1 volume of gibberellin solution into 5 volumes of chitosan acetic acid aqueous solution under the condition of stirring, and continuously stirring for 30min to obtain gibberellin emulsion;
spray drying gibberellin emulsion at air inlet temperature of 190 deg.C and air outlet temperature of 80 deg.C to obtain chitosan coated gibberellin sustained-release microcapsule with particle diameter of 0.5-20 μm.
Example 3
A preparation method of a chitosan-coated 6-benzyladenine sustained-release microcapsule comprises the following steps:
dissolving a 6-benzyladenine raw drug in 0.1mol/L hydrochloric acid methanol solution to obtain 6-benzyladenine solution, wherein the concentration of the 6-benzyladenine raw drug in the 6-benzyladenine solution is 20 mg/mL;
dissolving chitosan in an acetic acid aqueous solution to obtain an acetic acid aqueous solution of chitosan, wherein the concentration of chitosan in the acetic acid aqueous solution of chitosan is 10mg/mL, the molecular weight of chitosan is 500kDa, the degree of deacetylation is 85 percent, and the mass percentage concentration of glacial acetic acid in the acetic acid aqueous solution is 1 percent;
slowly dropwise adding 1 volume of 6-benzyladenine solution into 1 volume of chitosan acetic acid aqueous solution under the condition of stirring, and continuously stirring for 30min to obtain 6-benzyladenine emulsion;
and (3) spray-drying the 6-benzyladenine emulsion to obtain the chitosan-coated 6-benzyladenine sustained-release microcapsule with the particle size of 0.5-15 mu m, wherein the air inlet temperature is 185 ℃ and the air outlet temperature is 80 ℃.
Example 4
A preparation method of chitosan-coated ethephon sustained-release microcapsules comprises the following steps:
dissolving ethephon original drug and tween 80 in methanol to obtain ethephon solution, wherein the concentrations of ethephon original drug and tween 80 in the ethephon solution are 20mg/mL and 40mg/mL respectively;
dissolving chitosan in an acetic acid aqueous solution to obtain an acetic acid aqueous solution of chitosan, wherein the concentration of chitosan in the acetic acid aqueous solution of chitosan is 20mg/mL, the molecular weight of chitosan is 200kDa, the degree of deacetylation is 99%, and the mass percentage concentration of glacial acetic acid in the acetic acid aqueous solution is 0.5%;
slowly dropwise adding 1 volume of ethephon solution into 1 volume of chitosan acetic acid aqueous solution under the condition of stirring, and continuously stirring for 30min to obtain ethephon emulsion;
spray drying the ethephon emulsion at air inlet temperature of 190 deg.C and air outlet temperature of 70 deg.C to obtain chitosan coated ethephon sustained release microcapsule with particle diameter of 0.5-20 μm.
Example 5
The product in example 3 was selected for characterization, and the results show that: the chitosan coated 6-benzyladenine slow-release microcapsule has a particle size of 0.5-15 μm as shown in figure 1; the product is dispersed in 0.1mol/L hydrochloric acid methanol solution according to the concentration of 200mg/L completely dissolved by 6-benzyladenine, and the supernatant is taken at different times to measure the content of 6-benzyladenine so as to characterize the release rate, and the result shows that: the chitosan coated 6-benzyladenine slow-release microcapsule has better slow-release performance, as shown in figure 2.
Example 6
The method comprises the steps of respectively treating cucumbers (the concentration of ethephon is 300mg/L) at the later stage of flowers by using chitosan coated ethephon slow-release microcapsule solution and ethephon, wherein the cucumber coated with the chitosan coated ethephon slow-release microcapsule group is superior to a ethephon bare medicine group in growth, the ethephon bare medicine group shows slight phytotoxicity, and the chitosan coated ethephon slow-release microcapsule group does not cause phytotoxicity.

6. A process for preparing the slow-release microcapsule of chitosan-coated plant growth substance as claimed in claim 1, 2, 3, 4 or 5, characterized in that: the preparation method comprises the following steps:
adding the plant growth substance solution into an acetic acid aqueous solution of chitosan to obtain plant growth substance emulsion;
and (3) carrying out spray drying on the plant growth substance emulsion to obtain the chitosan-coated plant growth substance sustained-release microcapsule.
7. The method of claim 6, wherein: the concentration of the plant growth substance in the plant growth substance solution is 0.1-50 mg/mL;
the concentration of the surfactant in the plant growth substance solution is 0-40 mg/mL;
the surfactant is at least one of tween 20, tween 40, tween 60, tween 80, lecithin, polyoxyethylene 400 monolaurate, polyoxyethylene 400 monostearate and polyoxyethylene 400 monooleate;
the solvent in the plant growth substance solution is methanol, ethanol or ethyl acetate.
8. The method of claim 7, wherein: the concentration of chitosan in the acetic acid aqueous solution of chitosan is 0.5-20 mg/mL;
the mass percentage concentration of the glacial acetic acid in the acetic acid aqueous solution is 0.2-2%.
9. The method of claim 8, wherein: the volume ratio of the plant growth substance solution to the acetic acid aqueous solution of chitosan is 1: 1-5.
10. The method of claim 9, wherein: the air inlet temperature of the spray drying is 180-200 ℃, and the air outlet temperature is 70-100 ℃.
