Abstract

本发明涉及一种壳聚糖包覆赤霉素缓释微囊及其制备方法,属于植物生长调节剂、农药和农肥领域。本发明所述壳聚糖包覆赤霉素缓释微囊为将赤霉素用壳聚糖包覆后得到。在同等剂量条件下,采用壳聚糖包覆的赤霉素微囊保持药效的时间更长。

Description

一种壳聚糖包覆赤霉素缓释微囊及其制备方法
技术领域
本发明涉及一种壳聚糖包覆赤霉素缓释微囊及其制备方法,属于植物生长调节剂、农药和农肥领域。
背景技术
赤霉素,广泛存在的植物激素。化学结构属于二萜类酸,由四环骨架衍生而得。能促进茎、叶的生长,提早抽苔开花,促进种子、块茎、块根发芽,刺激果实生长,增加结果率或形成无籽果实等。对马铃薯、蕃茄、稻、麦、棉花、大豆、豌豆、烟草、果树等均有增产作用。
赤霉素的一大缺点是稳定性差,在弱碱性(pH>9)或碱性溶液中,仅2小时90%以上的赤霉素即水解失效。施用者多采用过量给药或重复给药的方法克服这一问题。但施用过多的赤霉素,植物会出现失绿、徒长现象,甚至还会出现锈果、果实硬化变形等药害问题。目前多采用清水或碱性水淋洗,喷施有缓解功能的药剂补救,而没有从制剂本身解决上述问题。
发明内容
本发明采用壳聚糖包覆的赤霉素微囊,解决了传统农药制剂存在的易产生药害、稳定性差、持效期短等问题。
本发明提供了一种壳聚糖包覆赤霉素缓释微囊,所述壳聚糖包覆赤霉素缓释微囊为将赤霉素用壳聚糖包覆后得到。
本发明优选为所述壳聚糖的分子量为100-500kDa,脱乙酰度60-99%。
本发明优选为所述赤霉素与壳聚糖的质量比为1:16-100。
本发明优选为所述壳聚糖包覆赤霉素缓释微囊的粒径为0.5-20μm。
本发明另一目的为提供一种上述壳聚糖包覆赤霉素缓释微囊的制备方法,所述制备方法包括如下步骤:将赤霉素溶液加入到壳聚糖的醋酸水溶液中,得到赤霉素乳液;将赤霉素乳液喷雾干燥,得到壳聚糖包覆赤霉素缓释微囊。
本发明优选为所述赤霉素溶液中赤霉素浓度为0.5-50mg/mL;所述赤霉素溶液中表面活性剂浓度为0-40mg/mL;所述表面活性剂为吐温20、吐温40、吐温60、吐温80、卵磷脂、聚氧乙烯400单月桂酸酯、聚氧乙烯400单硬脂酸酯和聚氧乙烯400单油酸酯中的至少一种;所述赤霉素溶液中溶剂为甲醇、乙醇或乙酸乙酯。
本发明优选为所述壳聚糖的醋酸水溶液中壳聚糖浓度为0.5-20mg/mL;所述醋酸水溶液中冰醋酸质量百分浓度为0.2-2%。
本发明优选为所述赤霉素溶液与壳聚糖的醋酸水溶液的体积比为1:1-4。
本发明优选为所述喷雾干燥的进风温度为180-200℃,出风温度为70-100℃。
本发明有益效果为:
本发明使用可降解的天然多糖壳聚糖作为包覆材料,其包覆效果好且降解产物对环境无污染;
采用壳聚糖包覆的赤霉素微囊,一次给药剂量可高于未包覆赤霉素的给药剂量;
在同等剂量条件下,采用壳聚糖包覆的赤霉素微囊保持药效的时间更长;
壳聚糖除了能包覆赤霉素外,其本身也具有诱导植物免疫活性提高的活性。

下述非限制性实施例可以使本领域的普通技术人员更全面地理解本发明,但不以任何方式限制本发明。
实施例1
一种壳聚糖包覆赤霉素缓释微囊的制备方法,所述制备方法包括如下步骤:
将赤霉素原药及吐温20溶于乙酸乙酯中,得到赤霉素溶液,使赤霉素溶液中赤霉素原药及吐温20的浓度分别为20mg/mL、40mg/mL;
将壳聚糖溶于醋酸水溶液中,得到壳聚糖的醋酸水溶液,使壳聚糖的醋酸水溶液中壳聚糖浓度为10mg/mL,壳聚糖的分子量为500kDa,脱乙酰度90%,醋酸水溶液中冰醋酸质量百分浓度为1%;
在搅拌条件下,将1体积赤霉素溶液缓慢逐滴加入到1体积壳聚糖的醋酸水溶液中,继续搅拌30min,得到赤霉素乳液;
将赤霉素乳液喷雾干燥,进风温度为185℃,出风温度为80℃,得到壳聚糖包覆赤霉素缓释微囊,其粒径0.5-15μm。
实施例2
一种壳聚糖包覆赤霉素缓释微囊的制备方法,所述制备方法包括如下步骤:
将赤霉素原药及吐温20溶于乙醇中,得到赤霉素溶液,使赤霉素溶液中赤霉素原药及吐温20的浓度分别为50mg/mL、5mg/mL;
将壳聚糖溶于醋酸水溶液中,得到壳聚糖的醋酸水溶液,使壳聚糖的醋酸水溶液中壳聚糖浓度为20mg/mL,壳聚糖的分子量为300kDa,脱乙酰度80%,醋酸水溶液中冰醋酸质量百分浓度为0.2%;
在搅拌条件下,将1体积赤霉素溶液缓慢逐滴加入到4体积壳聚糖的醋酸水溶液中,继续搅拌30min,得到赤霉素乳液;
将赤霉素乳液喷雾干燥,进风温度为190℃,出风温度为80℃,得到壳聚糖包覆赤霉素缓释微囊,其粒径0.5-20μm。
实施例3
一种壳聚糖包覆赤霉素缓释微囊的制备方法,所述制备方法包括如下步骤:
将赤霉素原药及卵磷脂溶于乙醇中,得到赤霉素溶液,使赤霉素溶液中赤霉素原药及卵磷脂的浓度分别为0.5mg/mL、10mg/mL;
将壳聚糖溶于醋酸水溶液中,得到壳聚糖的醋酸水溶液,使壳聚糖的醋酸水溶液中壳聚糖浓度为1mg/mL,壳聚糖的分子量为100kDa,脱乙酰度60%,醋酸水溶液中冰醋酸质量百分浓度为2%;
在搅拌条件下,将1体积赤霉素溶液缓慢逐滴加入到2体积壳聚糖的醋酸水溶液中,继续搅拌30min,得到赤霉素乳液;
将赤霉素乳液喷雾干燥,进风温度为200℃,出风温度为70℃,得到壳聚糖包覆赤霉素缓释微囊,其粒径1-10μm。
实施例4
一种壳聚糖包覆赤霉素缓释微囊的制备方法,所述制备方法包括如下步骤:
将赤霉素原药及卵磷脂溶于甲醇中,得到赤霉素溶液,使赤霉素溶液中赤霉素原药及卵磷脂的浓度分别为20mg/mL、40mg/mL;
将壳聚糖溶于醋酸水溶液中,得到壳聚糖的醋酸水溶液,使壳聚糖的醋酸水溶液中壳聚糖浓度为20mg/mL,壳聚糖的分子量为200kDa,脱乙酰度99%,醋酸水溶液中冰醋酸质量百分浓度为0.5%;
在搅拌条件下,将1体积赤霉素溶液缓慢逐滴加入到1体积壳聚糖的醋酸水溶液中,继续搅拌30min,得到赤霉素乳液;
将赤霉素乳液喷雾干燥,进风温度为190℃,出风温度为70℃,得到壳聚糖包覆赤霉素缓释微囊,其粒径0.5-20μm。
实施例5
选取实施例1制备的产品进行表征,结果表明:壳聚糖包覆赤霉素缓释微囊显微镜表征表明,其粒径为0.5-15μm,见附图1;将该产品按赤霉素完全溶解出的浓度200mg/L分散于去离子水中,不同时间取上清测其270nm处的吸收以表征其释放速率,结果表明:24h释放小于40%,72h释放小于70%,168h释放为80%,见附图2;将该产品按赤霉素完全溶解出的浓度200mg/L分散于pH6.1磷酸缓冲液中,不同时间取上清测其255nm处的吸收以表征赤霉素稳定性,结果表明:壳聚糖包覆的赤霉素稳定性显著高于赤霉素裸药,见附图3。
实施例6
用壳聚糖包覆赤霉素缓释微囊溶液及赤霉素在花后期分别处理葡萄(按赤霉素浓度为200mg/L),壳聚糖包覆赤霉素缓释微囊组锈果率低于5%,而赤霉素裸药组锈果率大于30%。

1.一种壳聚糖包覆赤霉素缓释微囊,其特征在于:所述壳聚糖包覆赤霉素缓释微囊为将赤霉素用壳聚糖包覆后得到。
2.根据权利要求1所述的壳聚糖包覆赤霉素缓释微囊,其特征在于:所述壳聚糖的分子量为100-500kDa,脱乙酰度60-99%。
3.根据权利要求2所述的壳聚糖包覆赤霉素缓释微囊,其特征在于:所述赤霉素与壳聚糖的质量比为1:16-100。
4.根据权利要求3所述的壳聚糖包覆赤霉素缓释微囊,其特征在于:所述壳聚糖包覆赤霉素缓释微囊的粒径为0.5-20μm。
5.权利要求1、2、3或4所述壳聚糖包覆赤霉素缓释微囊的制备方法,其特征在于:所述制备方法包括如下步骤:
将赤霉素溶液加入到壳聚糖的醋酸水溶液中,得到赤霉素乳液;
将赤霉素乳液喷雾干燥,得到壳聚糖包覆赤霉素缓释微囊。
6.根据权利要求5所述的制备方法,其特征在于:所述赤霉素溶液中赤霉素浓度为0.5-50mg/mL;
所述赤霉素溶液中表面活性剂浓度为0-40mg/mL;
所述表面活性剂为吐温20、吐温40、吐温60、吐温80、卵磷脂、聚氧乙烯400单月桂酸酯、聚氧乙烯400单硬脂酸酯和聚氧乙烯400单油酸酯中的至少一种;
所述赤霉素溶液中溶剂为甲醇、乙醇或乙酸乙酯。
7.根据权利要求6所述的制备方法,其特征在于:所述壳聚糖的醋酸水溶液中壳聚糖浓度为0.5-20mg/mL;
所述醋酸水溶液中冰醋酸质量百分浓度为0.2-2%。
8.根据权利要求7所述的制备方法,其特征在于:所述赤霉素溶液与壳聚糖的醋酸水溶液的体积比为1:1-4。
9.根据权利要求8所述的制备方法,其特征在于:所述喷雾干燥的进风温度为180-200℃,出风温度为70-100℃。
