Abstract

Background
Dandruff is the most common disease of scalp, is called as pityriasis capitis medically, and is clinically represented by excessive fine, grey, dry or slightly greasy pityrospermy-like scraps on the scalp or hair, which can be accompanied by pruritus and has high morbidity of people. the invention discovers that the natamycin has specific inhibition effect on malassezia furfur and human scalp bacteria microorganisms through an in-vitro drug sensitive experiment, thereby providing a new application of the natamycin in preparing a shampoo and hair care product for removing dandruff and relieving itching. The natamycin can effectively inhibit the main fungus malassezia furfur which causes dandruff, has obvious dandruff removing effect, low using dose and is safe and nontoxic. The invention provides a new choice for the development of dandruff removing hair washing and protecting products, can achieve the effects of removing dandruff and relieving itching under the condition of reducing scalp irritation, and has obvious practical value.

The natamycin provided by the invention is used in shampoo products with small dosage, stable effect and safe and nontoxic characteristics, can effectively replace some existing anti-dandruff medicines, and provides a new choice for the development of anti-dandruff products.
The following examples are intended to illustrate specific embodiments of the present invention, but are not intended to limit the scope of the invention to the examples.
Example 1 drug susceptibility test of Natamycin on Malassezia furfur
The specific operation steps are as follows:
1. preparing a culture medium: preparing a liquid culture medium according to the following proportion (1% peptone, 2% -4% glucose, 0.1% yeast powder, 0.4% bovine bile salt, 0.1% glycerol, 0.25% glyceryl monostearate, 0.2% tween 40, 1.5% agar and 2% olive oil), and carrying out autoclaving treatment at 121 ℃ for 15min for later use; a solid medium was prepared by adding 2% agarose to a liquid medium.
2. Activating strains: taking a standard strain (purchased from ATCC, 14521);
the bacterial powder was dissolved in 2mL of the medium, and the resulting solution was added to 50mL of the liquid medium and cultured on a shaker at 220rpm and 28 ℃ for 24 hours.
3. Drug sensitivity test
Taking 0.8mL of malassezia furfur solution in logarithmic phase, uniformly coating the solution in a solid culture medium, adding a sterile Oxford cup, adding 150 mu L of natamycin solution with different concentrations (2, 4, 8 and 16mg/mL) into each hole, placing the mixture in a biochemical incubator at 28 ℃, culturing for 48h, and observing the size of a bacteriostatic colony.
4. Minimum bactericidal concentration detection
A, 0.5mL of logarithmic growth batch malassezia furfur is taken and added into 5mL of fresh liquid culture medium, and the mixture is cultured for 4 hours at 220rpm and 28 ℃.

The experimental results show that: the natamycin has an inhibiting effect on the malassezia furfur, the malassezia furfur has sensitivity to the natamycin, and the natamycin can be used for inhibiting the malassezia furfur and can be applied to preparing a product for removing dandruff and relieving itching.
The natamycin with different concentrations is treated, the inhibition zone of the bran malassezia is increased along with the increase of the drug concentration, the drug concentration dependence is achieved, and the most suitable natamycin sterilization concentration is more than or equal to 16 mg/mL.
The natamycin can effectively inhibit the growth of main fungus bran malassezia furfur for dandruff and has the function of removing dandruff. It was further found that natamycin at low concentration (16mg/mL) was effective in killing Malassezia furfur. The invention provides a powerful basis for preparing the efficient and safe anti-dandruff shampoo, and can effectively solve the trouble of patients with dandruff.
