preparing a nutrient solution: adding 100g of water into a nutrient mixture consisting of 1.7g of potassium nitrate, 1.4g of ammonium nitrate, 0.23g of monopotassium phosphate, 0.45g of magnesium sulfate and 0.55g of calcium chloride, and ultrasonically mixing for 20min at the frequency of 28KHZ to obtain a nutrient solution for later use;
material treatment: peeling 80g of potato, cleaning under ultrasonic, cutting into pieces, steaming at 90 deg.C for 30min, peeling the cooked potato and 80g of banana, pulverizing in a wall breaking machine, and stirring to obtain paste;
mixing raw materials: adding 12g of peptone, 5g of activated carbon, 15g of white sugar and 8g of agar into the nutrient solution, adding mashed potato and banana, heating to 85 ℃, stirring, mixing, and filling into a culture bottle after stirring;
and (3) sterilization treatment: placing the culture bottle into a sterilizing pot, sterilizing at 125 deg.C for 45min, placing into a sterilizing cabinet or sterilizing cooling workshop, cooling, and getting the final product.
In summary, according to the culture medium for orchids and the preparation method thereof provided by the embodiments of the present invention, the potato is rich in nutrients such as carbohydrates, plant proteins, vitamin C, dietary fibers, etc., and the banana is rich in nutrients such as vitamin a, phosphorus, thiamine, riboflavin, potassium, magnesium, etc., which can provide nutrients required by orchids for tissue culture, and are beneficial for growth and propagation of orchids; the peptone is rich in organic nitrogen compounds, vitamins and saccharides, contains rich amino acids, particularly more sulfur-containing amino acids, can provide a nitrogen source for tissue culture of orchidaceae plants, and is beneficial to growth and reproduction of plant tissues; the activated carbon has adsorption effect and can adsorb some harmful impurities, thereby reducing the influence of the harmful impurities on the growth of the tissue of the orchid and being beneficial to the growth and the propagation of the tissue of the orchid; the white sugar is used as a carbon source, can provide energy for the growth of the tissue of the orchidaceae plant, and is beneficial to the growth and the propagation of the orchidaceae plant. The agar can be used as a coagulator, which is beneficial to the use of a culture medium; the nutrient mixture contains various nutrient elements, can provide sufficient nutrition for the growth of the tissue of the orchidaceae plant, and is beneficial to the growth and the propagation of the orchidaceae plant. The orchid culture medium composed of the components is adjusted on the formula of the orchid plant, and other common raw materials are used for providing nutrition for tissues of the orchid plant to enable the orchid plant to grow and reproduce, so that the orchid culture medium is good in effect and convenient to use. The preparation method comprises the steps of firstly preparing the nutrient solution through the nutrient solution preparation step, adding water into the nutrient mixture, and mixing the nutrient mixture and the water through ultrasound, so that the nutrient mixture and the water can be more fully mixed, and the obtained nutrient solution is favorable for improving the effect of nutrients on tissue culture of orchids; then, the potatoes and the bananas are processed through the material processing steps, the potatoes are peeled and cut into blocks for cooking, the cutting into blocks can reduce the time required for cooking the potatoes, and some toxic substances in the potatoes can be removed through cooking, so that the growth and the propagation of orchid tissues are facilitated, meanwhile, the texture of the potatoes can be softened, the potatoes and the peeled bananas are conveniently put into a wall breaking machine together for crushing and stirring into mud in the subsequent steps, and the use is more convenient; through the raw material mixing step, peptone, activated carbon, white sugar and agar are added into the nutrient solution, and mashed potatoes and bananas are added, and the nutrient solution, the peptone, the activated carbon, the white sugar, the agar, the mashed potatoes and the bananas can be mixed more fully through heating and stirring, so that the mutual combination and the synergistic effect of the components are facilitated, the culture effect of the orchid plant tissue is improved, and the orchid plant tissue is more convenient to use; finally, through the sterilization treatment steps, the culture bottle is placed into a sterilization pot for sterilization, and is placed into a disinfection cabinet or a disinfection cooling workshop for cooling after the sterilization is finished, so that harmful strains and impurities in the culture bottle are removed, the toxicity of the harmful strains and the impurities is reduced, the culture effect on the orchid tissue is improved, the growth and the propagation of the orchid tissue are facilitated, and the use is more convenient. The whole preparation method has simple flow, the prepared culture medium has good culture effect, is beneficial to the growth and the propagation of the tissue of the orchids, and is convenient to use.

Claims (10)

1. An orchid culture medium is characterized by comprising the following components: banana, potato, peptone, activated carbon, white sugar, agar and nutrient mixture and water.
2. The orchid culture medium according to claim 1, wherein the orchid culture medium comprises the following components in parts by weight: 2-80 parts of banana, 2-100 parts of potato, 1-20 parts of peptone, 0.5-10 parts of activated carbon, 1-100 parts of white sugar, 1-10 parts of agar, 1.6-6.2 parts of nutrient and 800 parts of water-activated sugar.
3. The orchid culture medium according to any one of claims 1-2, wherein the nutrient mixture comprises the following components: potassium nitrate, ammonium nitrate, potassium dihydrogen phosphate, magnesium sulfate and calcium chloride.
4. The orchid culture medium according to claim 3, wherein the nutrient mixture comprises the following components in parts by weight: 0.5-2.5 parts of potassium nitrate, 0.5-2 parts of ammonium nitrate, 0.1-0.3 part of monopotassium phosphate, 0.2-0.6 part of magnesium sulfate and 0.3-0.8 part of calcium chloride.
5. The orchid culture medium as claimed in claim 1, wherein the water is deionized water.
6. A method for preparing the orchid culture medium according to any one of claims 1 to 5, comprising the following steps:
preparing a nutrient solution: adding water into the nutrient mixture, and then mixing the nutrient mixture by ultrasonic to obtain a nutrient solution for later use;
material treatment: peeling potatoes, cleaning the potatoes under ultrasonic waves, cutting the potatoes into blocks, cooking the blocks, putting the cooked potatoes and peeled bananas into a wall breaking machine, crushing and stirring the potatoes and the peeled bananas into mud for later use;
mixing raw materials: adding peptone, activated carbon, white sugar and agar into the nutrient solution, adding mashed potato and banana, heating while stirring for mixing, and putting into a culture bottle after stirring;
and (3) sterilization treatment: and (4) placing the culture bottle into a sterilization pot for sterilization, placing the culture bottle into a sterilization cabinet or a sterilization cooling workshop for cooling after the sterilization is finished, and obtaining a finished product after the cooling is finished.
7. The method for preparing the orchid culture medium according to claim 6, wherein the nutrient solution is ultrasonically mixed at a frequency of 28KHZ for 15-30min during the preparation process.
8. The method for preparing the orchid culture medium according to claim 6, wherein the potatoes are cut into pieces and cooked at 80-95 ℃ for 15-30min during the material treatment.
9. The method for producing an orchid culture medium according to claim 6, wherein the raw materials are mixed while being heated to 80-95 ℃ under stirring.
10. The method for preparing the culture medium of Orchidaceae plant as claimed in claim 6, wherein the sterilization treatment is carried out by placing the culture flask in a sterilization pot and sterilizing at 120-130 deg.C for 15-50 min.
