Transaminase S142 (from Vibrio Fluvialis JS17, having the amino acid sequence of SEQ ID NO: 1, available from Han enzymes Biotech, Suzhou) catalyzed synthesis of 2- ((trans) -4-aminocyclohexyl) isopropanol
Ethyl 1, 4-dioxaspiro [4,5] decane-8-carboxylate (4g) was dissolved in methyltetrahydrofuran (40mL) and placed in a nitrogen blanketed three-neck flask with a magnetic stir bar. The reaction temperature was controlled to 0 ℃ with ice water, and 3M methyl magnesium bromide (18mL) was added dropwise. Stirred for 20h at 25 ℃. Controlling the temperature at 0-10 ℃, dropwise adding saturated ammonium chloride aqueous solution, and stirring for 10 min. 20mL of methyltetrahydrofuran was added and the product was extracted and centrifuged. The organic phase was taken out and blown dry with nitrogen to yield 3.10g of 4- (1, 4-dioxaspiro [4,5] decyl-8) isopropanol. The obtained 4- (1, 4-dioxaspiro [4,5] decyl-8) isopropanol (200mg) was dissolved in tetrahydrofuran (2mL), and 2M hydrochloric acid (3.5mL) was added dropwise thereto, followed by stirring at 25 ℃ for 16 hours. The obtained 4- (1, 4-dioxaspiro [4,5] decyl-8) isopropanol (200mg) was dissolved in tetrahydrofuran (2mL), and 1M hydrochloric acid (5mL) was added dropwise thereto, followed by stirring at 25 ℃ for reaction for 22 hours. The reaction mixture was mixed and added dropwise to pH 7-7.5 with a saturated aqueous sodium bicarbonate solution. Then, it was extracted twice with 10mL of methyltetrahydrofuran, and the organic phases were combined and blown dry with nitrogen to give 265mg of 4- (2-hydroxyisopropyl) cyclohexanone.
Potassium dihydrogen phosphate (5.3g), dipotassium hydrogen phosphate (106g), 85mL of isopropylamine, and 35mL of 85% phosphoric acid were dissolved in 900mL of deionized water to prepare a buffer (pH 7.5) containing an amino donor, and the obtained 4- (2-hydroxyisopropyl) cyclohexanone (3mg) was dissolved in dimethyl sulfoxide (6 uL). 500uL of buffer, 1mg of pyridoxal phosphate, 10mg of transaminase (R142) were added in this order. The mixed reaction system is placed in a shaker at 30 ℃ and 250rpm for reaction for 48 h. After the reaction was completed, the pH was adjusted to 8-9 using a saturated aqueous sodium bicarbonate solution. 2mL of methyltetrahydrofuran was added and extracted with shaking. After standing, 100uL of the organic phase was dissolved in 900uL of methanol. After filtration, gas chromatography detection was carried out. The result shows that the conversion rate of the substrate 4- (2-hydroxyisopropyl) cyclohexanone in the enzyme catalysis step is 87.51%, and the chiral purity of the product is 100%.

Claims (8)

1. A process for the preparation of 2- ((trans) -4-aminocyclohexyl) isopropanol (D), characterized in that it comprises the following steps:
catalyzing 4- (2-hydroxyisopropyl) cyclohexanone (C) by transaminase and coenzyme in dipotassium hydrogen phosphate-potassium dihydrogen phosphate buffer solution by taking isopropylamine as an amino donor;

the transaminase is derived from Vibrio Fluvialis JS17, and has an amino acid sequence shown in SEQ ID No. 1; the coenzyme is pyridoxal phosphate; the reaction temperature is 25-30 ℃; the reaction time is 20-60 h; the reaction pH is 6.5-8.0; the dosage of the transaminase is 0.1-3.5 g/g; the dosage of the coenzyme is 0.01-0.4 g/g.
2. The method according to claim 1, wherein the method comprises the steps of:
adding transaminase and coenzyme into dipotassium hydrogen phosphate-potassium dihydrogen phosphate buffer solution containing amino donor, and adjusting pH of the reaction system to 6.5-8.0; 4- (2-hydroxy isopropyl) cyclohexanone (C) is dissolved in dimethyl sulfoxide, and is added into a buffer solution system with transamination and coenzyme to carry out transamination reaction.
3. The method of any one of claims 1-2, further comprising the steps of:
under the action of hydrochloric acid, removing glycol protection from 4- (1, 4-dioxaspiro [4,5] decyl-8) isopropanol (B) to obtain 4- (2-hydroxyisopropyl) cyclohexanone (C);

4. the method according to claim 3,
the concentration of the hydrochloric acid is 1-3M;
and/or the dosage of the hydrochloric acid is 5-10 molar equivalents;
and/or the reaction temperature for removing the ethylene glycol protection is 20-35 ℃.

The preparation method according to claim 3, further comprising the following post-treatment steps: after the reaction for removing the glycol protection is finished, adjusting the pH value of the reaction system to 7-7.5 by using a saturated sodium bicarbonate water solution; then extracting with methyltetrahydrofuran, separating the organic phase and concentrating to dryness to obtain 4- (2-hydroxy isopropyl) cyclohexanone.
6. The method of any one of claims 4-5, further comprising the steps of:
methylation reaction is carried out on 1, 4-dioxaspiro [4,5] decane-8-carboxylic acid ethyl ester (A) in the presence of a Grignard reagent to obtain 4- (1, 4-dioxaspiro [4,5] decyl-8) isopropanol (B);

the solvent for the methylation reaction is selected from tetrahydrofuran and/or methyltetrahydrofuran;
and/or the Grignard reagent is selected from one or more of methyl magnesium chloride, methyl magnesium bromide and methyl magnesium iodide;
and/or the Grignard reagent is 2.0-5.0 molar equivalents or 3.0-4.0 molar equivalents of ethyl 1, 4-dioxaspiro [4,5] decane-8-carboxylate.
8. The method according to claim 6,
when the Grignard reagent is dripped in the methylation reaction, the temperature is 0-5 ℃;
and/or, after the dropwise addition is finished, the reaction temperature is 15-30 ℃;
and/or after the methylation reaction is finished, quenching the metal reagent by using saturated ammonium chloride, extracting the product by using methyl tetrahydrofuran, separating an organic phase, and concentrating to be dry to obtain the 4- (1, 4-dioxaspiro [4,5] decyl-8) isopropanol.
