Abstract
The invention discloses a method for promoting Epimedium sagittatum seed germination, which comprises the following operation steps: after pollination of epimedium arrow leaf is finished, 1/2 of the whole rachis at the upper end of the rachis is cut off, and the epimedium arrow leaf living plant is given appropriate shading conditions; air-drying in a natural state indoors to remove seed coats; soaking the seeds for 2-3 times, diluting the seeds by 500-600 times by using carbendazim or chlorothalonil stock solution, soaking the seeds, and then washing and airing the seeds; mixing the seeds with perlite, adding clean water into the perlite until the water content reaches 75% -82%, and storing the seeds in dark for 120 days until the seeds begin to appear white. The invention uses a physical treatment method to break the morphological and physiological after-ripening of epimedium sagittifolium, promotes the growth and development of seed embryos, shortens the seed germination time and improves the seed germination rate; the perlite is used as a storage matrix, has stable water holding capacity, good air permeability and low phenomena of seed rot and mildew, is not easy to sink, and can improve the germination rate of seeds by 5-15%.
Description
Technical Field
The invention belongs to the field of herbal medicine planting, and particularly relates to a method for promoting epimedium sagittatum seed germination.
Background
Epimedium sagittatum is a plant of Epimedium genus of berberidaceae family, and is a perennial herb, and the rhizome of Epimedium sagittatum is nodulated. 1-3 leaves are grown from roots, 3 compound leaves are grown from small leaves, the small leaves are oval to oval and are in the shape of needles, the length is 4-9 cm, the width is 2.5-5 cm, the tip is pointed or tapered, fine burrs are arranged at the edge, the base part is heart-shaped, the base parts of lateral small leaves are asymmetric, the outer split shape is inclined and larger, the shape of a triangle is triangular, and the inner split shape is smaller and is close to circular; the cauline leaves usually grow on the top, the shape is similar to that of the root leaves, the base part is in a skewed arrow-shaped heart shape, the outer lobe is extremely large, and the tip is gradually sharp.
Epimedium sagittatum is an important medicinal resource in China, is mainly distributed in the south of Yangtze river, not only can tonify kidney yang, strengthen muscles and bones, dispel wind-damp, but also has the effects of resisting aging, improving immunity, inhibiting tumors and the like, also has good treatment effects on poliomyelitis, neurasthenia and chronic tracheitis, and is one of the traditional Chinese medicines with the most development potential.
The existing epimedium is mainly used for collecting wild plant resources, and the collection and consumption of the epimedium exceed the regeneration capacity of natural resources along with the increase of demand, so that species are endangered.
At present, the method for promoting the shape and physiological after-ripening of the seeds of the Epimedium sagittatum plant mainly comprises the following steps: the constant temperature treatment and the variable temperature treatment are carried out, the substrate is mainly river sand, the river sand has the advantages of good heat preservation effect and has the defects of heavy river sand, high requirement on loaded materials, unsatisfactory water preservation effect, frequent manual turning times and water replenishing and preserving times due to water sinking, poor air permeability of the river sand, and serious seed rot and mildew.
Disclosure of Invention
Aiming at the defects of the prior art, the invention provides a method for promoting epimedium sagittatum seed germination, which comprises the following specific operation steps:
1. after pollination of epimedium sagittifolia is finished, 1/2 of the whole rachis at the upper end of the rachis is cut off to ensure that the nutrition of seeds at the middle and lower parts is sufficient, and 50% of shading conditions are given to the plants of the epimedium sagittifolia living body until the seeds are harvested;
2. after the seeds are collected, the seeds are dried in the air for 50 to 60 days in an indoor natural state, and the seed coats are removed;
3. soaking the seeds in clean water for 10-15 hours, changing water for 2-3 times, diluting the seeds with carbendazim or chlorothalonil stock solution by 500-600 times, soaking the seeds for about 120-150 minutes, then washing the seeds for 2-3 times by using clear water and airing the seeds;
4. fully and uniformly mixing the slightly dried seeds and perlite with the particle size of 8-12mm and multiple pores according to the volume ratio of 1:1, wherein the perlite is dried at the temperature of 105-115 ℃ for 3-4 hours in advance, and is used after being cooled to the normal temperature;
5. adding water into perlite until the water content reaches 75% -82%, storing at 6-10 deg.C in dark for 120 days, keeping the relative humidity of the storage environment at 75% -82%, and turning over every 20-30 days to facilitate the ventilation of seeds, and detecting the water content of perlite, if dry, spraying clear water in time to keep moisture, and keeping the water content at 75% -82%;
6. storing for 90-120 days, turning over the seeds during the period, and sowing the seeds in the field after the seeds begin to appear white.
By adopting the technical scheme, the physical treatment method is used for breaking the morphological and physiological after-ripening of epimedium sagittifolium, promoting the growth and development of seed embryos, shortening the seed germination time and improving the seed germination rate; meanwhile, the perlite is lighter, the requirement on loading materials is lower, the perlite has small particle size, is porous and stable in water holding, water is not easy to sink after water is added, the air permeability is better, manual frequent turning and water replenishing are not needed, the phenomena of seed rot and mildew are lower, and experimental statistics shows that compared with the existing method, the method can improve the seed germination rate by 5% -15%.
Detailed Description
100 mu of seedlings are grown in high-pass villages from rivers to the towns.
1. After pollination of epimedium arrow leaf is finished, 1/2 of the whole rachis at the upper end of the rachis is cut off, the seeds at the middle and lower parts are ensured to be sufficient in nutrition and to be normal and full in development, and meanwhile, 50% of shading conditions are given to the living plant of epimedium arrow leaf until the seeds are harvested;
2. after the seeds are collected, the seeds are dried in the air for 50 days in an indoor natural state, and the seed coats are removed;
3. soaking the seeds in clean water for 12 hours, changing water for 2 times, diluting the seeds with 500 times of bactericide stock solution such as carbendazim or chlorothalonil and soaking the seeds for about 120 minutes, then washing the seeds for 2 times by using clear water and airing the seeds;
4. fully and uniformly mixing the slightly aired seeds and perlite with the grain size of 10mm and multiple pores according to the volume ratio of 1:1, wherein the perlite is dried for 3 hours at 110 ℃ in advance and is used after being cooled to normal temperature;
5. adding water into perlite until the water content reaches 80%, storing at 8 deg.C in dark for 120 days with the relative humidity of the storage environment kept at 80%, and turning over every 25 days to facilitate the ventilation of seeds, and detecting the water content of perlite, if dry, spraying clear water to keep moisture in time, and keeping the water content at 80%;
6. storing for 90-120 days, turning over the seeds during the period, and sowing the seeds in the field after the seeds begin to appear white.
