Dicer enzyme belongs to endoribonuclease-III (RNase-III) family members, and was first found in Drosophila. The Dicer family of drosophila includes both Dicer1 and Dicer2 genes, whereas nematodes and humans only function with Dicer 1. Dicer1 enzyme is a key enzyme for synthesis and maturation of miRNAs, and if the function of Dicer1 enzyme is deficient, the function of the enzyme can affect the maturation of miRNAs in cells, thereby affecting the expression of downstream target genes. Thus, Dicer1 is likely to play a very important role in the development of tumors. However, no studies have shown that the expression of Dicer1 is associated with alzheimer's disease.

The materials used in the present invention can be obtained commercially or by conventional methods in the art. For example, wild-type mouse C57BL/6J mouse was purchased from Wenzhou university of medicine Laboratory animal center, APPSwe/PSEN1dE9 (APP/PS 1) mouse was purchased from Jackson Laboratory, RNA isolation kit was purchased from Beijing Tiangen Biotech, Inc., and over-expression Dicer1 vector preparation and virus packaging were accomplished by Cyagen Biosciences Inc. (Guingzhou, China).
Example 1 Dicer1 immunoblotting and quantitative PCR analysis in mouse hippocampal gyrus tissue
WT at 4 or 11 months of age, AD mice hippocampal gyrus tissue homogenate, and immunoblot examination of Dicer1, caspase3, activated caspase3, GAPDH was used as internal reference. Each group consisted of 6 mice, three males, and three females. Mice were fasted overnight, the next day, under deep anesthesia with isoflurane, 4oC phosphate buffer solution cardiac perfusion. Hippocampal gyrus tissues were isolated and frozen in liquid nitrogen. Solid form of A β 42 (GenScript, Nanjing, China) was dissolved in 1,1,1,3,3,3-Hexafluoro-2-propanol (Sigma-Aldrich) solvent, the insolubles were removed, and the dissolved A β 42 was placed at 4 ℃ for 30 minutes, followed by drying at room temperature and then dissolving in DMSO (Sigma-Aldrich) to prepare a 1mM solution. The solubilized polypeptide was diluted to 500 nM in a phenol red-free F12 solution, and after 24 hours at 4 ℃, the solution was centrifuged at 14,000 Xg for 30 minutes at 4 ℃, and the supernatant was collected as an A β 42 oligomer solution at a concentration of 100 nM.
Mouse primary neuron culture
Neonatal mice were used to culture cortical or hippocampal gyrus neurons. Minced mouse cerebral cortex or hippocampal gyrus tissue was isolated blunt, treated with 0.25% trypsin and 0.1% DNaseI, and incubated at 37 ℃ for 15 minutes in a carbon dioxide incubator. DMEM/F12 containing 10% calf serum was added, centrifuged at 120 Xg for 3 minutes by a centrifuge, the supernatant was collected, and the pellet was removed. When the mouse starts to train, the mouse is respectively put into water from four quadrants of the water maze, and the training mouse explores and searches for an underwater platform (the diameter is 1 cm and 10 cm underwater) for 60 seconds at most. Training was performed 4 times a day with 10 minutes intervals between each training and 6 days. The time it took to find the underwater platform was tested on day 7 (60 seconds maximum). Claims (4)

1. The Dicer1 gene or the product thereof is applied to the preparation of medicaments for treating early and middle-stage Alzheimer's disease, and the nucleotide sequence of the Dicer1 gene is shown as SEQ ID No. 1.
2. The use of claim 1, wherein said product is a viral vector comprising the Dicer1 gene.
3. The use of claim 2, wherein the viral vector is an adenoviral vector.
4. The use of claim 3, wherein the vector has the sequence shown in SEQ ID No. 2.
