技术领域
本发明涉及医用材料技术领域,具体涉及一种具有再生能力的组织工程骨的制备方法。
背景技术
由于某种因素如创伤、感染、肿瘤切除或先天性疾病等而使骨丧失了一些骨质,形成较大的间隙,称为骨缺损。骨缺损在临床上比较常见,其治疗一直是临床治疗的难点。骨移植为解决上述问题提供了可能,目前自体骨仍是骨移植来源的首选,但是其供给有限,应用时容易出现“拆东墙补西墙”的困境,难以满足临床需求。因此,找寻一种合适的骨移植替代物已成为研究和修复骨缺损的首要问题。
组织工程骨技术是将种子细胞接种于各种支架材料上,细胞外基质的分泌和材料的吸收协调进行,最终形成由细胞和细胞外基质组成的与材料形状相同的新生组织,达到结构与功能的良好修复。随着技术的发展,可吸收和组织兼容性较好的材料也被用于骨缺损的治疗,但目前组织工程材料和组织工程骨难以形成血管,并形成骨生成和吸收的平衡状态,再生组织与正常组织在结构和功能上仍存在很大的区别,在骨缺失治疗中难以完成血管的生成和骨髓的重建,不能完全替代缺失的功能。
发明内容
为了解决上述背景技术中存在的问题,本发明的目的在于提供一种具有再生能力的组织工程骨的制备方法,该方法制备出具有血管形成和骨髓生成能力的再生组织工程骨,在体内某些骨质完全缺失的情况下进行重建,植入体内后在一定条件下生长为功能性的具有骨髓和血管的骨质。
为了实现上述目的,本发明采用以下技术方案:
本发明提供一种具有再生能力的组织工程骨的制备方法,包括以下步骤:
S1、胶原基质的制备:将无菌、具有生物活性且未变性的Ⅰ型胶原、Ⅱ型胶原或复合型胶原加入至无菌管中,向其中加入在4℃下预冷的低糖DMEM培养基,在4℃温度下经充分混合后,静置备用;
S2、间充质干细胞的制备:选取状态良好的人间充质干细胞按1×107个细胞/cm2的浓度悬浮至诱导培养基中得到细胞悬液;
S3、骨髓的制备:将新鲜骨髓用5倍量的生理盐水悬浮,离心弃去上清液,将下层骨髓沉淀用生理盐水充分悬浮,即得骨髓悬液备用;
S4、半成品组织工程骨的培养制备:向步骤S2中得到的细胞悬液中加入步骤S1中得到的胶原基质,充分混合后,将其移入至透析袋中,固定透析袋,用注射针头纵向穿透,在穿孔处***管式半透膜进行组装;组装完毕后将透析袋放入培养器皿中,加入足量的低糖DMEM诱导培养基,经培养得到半成品组织工程骨;
S5、具有再生能力的组织工程骨的培养制备:将步骤S4中的培养器皿中的透析袋去除,将透析袋中的管式半透膜抽出,并加步骤S3中得到的骨髓悬液注入至管式半透膜抽出后留下的孔洞处,然后将透析袋置于培养器皿中,加入含有10%血清替代品的低糖DMEM诱导培养基浸没组织,并于37℃、二氧化碳含量为5%的培养箱中继续培养24小时,随后去除外侧透析袋,即得可移植使用的具有再生能力的组织工程骨。
进一步地,所述步骤S4中,具体培养过程为:组装完毕后将透析袋放入培养器皿中,加入足量的低糖DMEM诱导培养基,浸没所有组织,向培养器皿中注入管式半透膜,并吸取诱导培养基,以驱赶气泡;随后将培养器皿置于37℃、二氧化碳含量为5%的培养箱中培养,48小时后缓慢吸去培养上清液,然后加入新鲜的低糖DMEM诱导培养基继续培养,每48小时更换一次新鲜的诱导培养液,连续培养10天,得到半成品组织工程骨。
进一步地,所述步骤S4中,透析袋的长度与所需组织工程骨的长度相匹配,透析袋直径为8mm,透析袋截流分子量为1000KD,管式半透膜直径为5mm。
进一步地,所述步骤S2中,诱导培养基为含有谷氨酰胺、抗坏血酸、丙酮酸钠、***、β-甘油磷酸钠、血清替代品的低糖DMEM培养基。

本发明使用具有生物活性的胶原作为工程支架材料,该材料经过特殊处理,即可被组织吸收重建,又不会诱发免疫反应;组织工程细胞选择间充质干细胞,该类型细胞来源广泛,且不具有免疫原性,同时又可使用自身细胞,进一步降低了免疫及其他生物风险;使用骨髓直接移植入半成品的组织工程骨中,使得其产品具有成骨细胞、破骨细胞、又能重建骨髓造血能力和使其有血管生成,同时在制备组织工程骨过程中内外使用透性膜***,保障了骨生成过程中的营养供应,避免细胞凋亡和死亡。

具体实施方式
下面结合具体实施方式对本发明进行详细的说明。
本发明的实施例公开一种具有再生能力的组织工程骨的制备方法,包括以下步骤:
S1、胶原基质的制备:将200毫克无菌、具有生物活性且未变性的Ⅰ型胶原、Ⅱ型胶原或复合型胶原加入至5mL无菌管中,向其中加入在4℃下预冷的低糖DMEM培养基1mL,在4℃温度下经充分混合后,静置备用;
S2、间充质干细胞的制备:选取状态良好的取自患者脂肪的人间充质干细胞按1×107个细胞/cm2的浓度悬浮至诱导培养基中得到细胞悬液;
S3、骨髓的制备:采用EDTA抗凝抽取人骨髓即得新鲜骨髓,将新鲜骨髓用5倍量的生理盐水悬浮,300g离心5分钟,弃去上清液,将下层骨髓沉淀用生理盐水充分悬浮,即得骨髓悬液备用;
S4、半成品组织工程骨的培养制备:向步骤S2中得到的细胞悬液中加入步骤S1中得到的胶原基质,充分混合后,将其移入至直径为8mm,截流分子量为1000KD的透析袋中,固定透析袋,用注射针头纵向穿透,在穿孔处***直径为5mm的管式半透膜进行组装;组装完毕后将透析袋放入培养器皿中,加入足量的低糖DMEM诱导培养基,浸没所有组织,向培养器皿中注入管式半透膜,并吸取诱导培养基,以驱赶气泡;随后将培养器皿置于37℃、二氧化碳含量为5%的培养箱中培养,48小时后缓慢吸去培养上清液,然后加入新鲜的低糖DMEM诱导培养基继续培养,每48小时更换一次新鲜的诱导培养液,连续培养10天,得到半成品组织工程骨。
S5、具有再生能力的组织工程骨的培养制备:将步骤S4中的培养器皿中的透析袋去除,将透析袋中的管式半透膜抽出,并加步骤S3中得到的骨髓悬液注入至管式半透膜抽出后留下的孔洞处,注满为止,然后将透析袋置于培养器皿中,加入含有10%血清替代品的低糖DMEM诱导培养基浸没组织,并于37℃、二氧化碳含量为5%的培养箱中继续培养24小时,随后去除外侧透析袋,即得可移植使用的具有再生能力的组织工程骨;
S6、组织工程骨的使用:获得的组织工程骨可根据实际需要修整大小和形状,直接移植到体内骨缺损部位;或配合其他组织工程骨等制作成目标形状后移植。经过临床实验验证,也可将此组织工程骨移植入骨缺损动物模型的缺损骨处,或移植入血供丰富的肌肉、肾脏等部位,移植入体内后30天左右可形成功能性的骨组织。
S7、组织工程骨的存放:该组织工程骨组织制作完成后宜立即使用,不宜存储过长时间,4℃条件下可存放12小时。
其中,诱导培养基为含有谷氨酰胺、抗坏血酸、丙酮酸钠、***、β-甘油磷酸钠、血清替代品的低糖DMEM培养基。

S1、胶原基质的制备:将无菌、具有生物活性且未变性的Ⅰ型胶原、Ⅱ型胶原或复合型胶原加入至无菌管中,向其中加入在4℃下预冷的低糖DMEM培养基,在4℃温度下经充分混合后,静置备用;
S2、间充质干细胞的制备:选取状态良好的人间充质干细胞按1×107个细胞/cm2的浓度悬浮至诱导培养基中得到细胞悬液;
S3、骨髓的制备:将新鲜骨髓用5倍量的生理盐水悬浮,离心弃去上清液,将下层骨髓沉淀用生理盐水充分悬浮,即得骨髓悬液备用;
S4、半成品组织工程骨的培养制备:向步骤S2中得到的细胞悬液中加入步骤S1中得到的胶原基质,充分混合后,将其移入至透析袋中,固定透析袋,用注射针头纵向穿透,在穿孔处***管式半透膜进行组装;组装完毕后将透析袋放入培养器皿中,加入足量的低糖DMEM诱导培养基,浸没所有组织,向培养器皿中注入管式半透膜,并吸取诱导培养基,以驱赶气泡;随后将培养器皿置于37℃、二氧化碳含量为5%的培养箱中培养,48小时后缓慢吸去培养上清液,然后加入新鲜的低糖DMEM诱导培养基继续培养,每48小时更换一次新鲜的诱导培养液,连续培养10天,得到半成品组织工程骨;
S5、具有再生能力的组织工程骨的培养制备:将步骤S4中的培养器皿中的透析袋去除,将透析袋中的管式半透膜抽出,并加步骤S3中得到的骨髓悬液注入至管式半透膜抽出后留下的孔洞处,然后将透析袋置于培养器皿中,加入含有10%血清替代品的低糖DMEM诱导培养基浸没组织,并于37℃、二氧化碳含量为5%的培养箱中继续培养24小时,随后去除外侧透析袋,即得可移植使用的具有再生能力的组织工程骨。
2.根据权利要求1所述的具有再生能力的组织工程骨的制备方法,其特征在于,所述步骤S4中,透析袋的长度与所需组织工程骨的长度相匹配,透析袋直径为8mm,透析袋截流分子量为1000KD,管式半透膜直径为5mm。
3.根据权利要求1所述的具有再生能力的组织工程骨的制备方法,其特征在于,所述步骤S2中,诱导培养基为含有谷氨酰胺、抗坏血酸、丙酮酸钠、***、β-甘油磷酸钠、血清替代品的低糖DMEM培养基。
4.根据权利要求1所述的具有再生能力的组织工程骨的制备方法,其特征在于,所述步骤S2中人间充质干细胞选自于骨髓、脂肪、皮肤、牙髓、脐带或羊膜。
5.根据权利要求1所述的具有再生能力的组织工程骨的制备方法,其特征在于,所述步骤S3中新鲜骨髓为采用EDTA抗凝抽取人、大鼠或小鼠的骨髓。
