The embodiment provides a traditional Chinese medicine composition for treating gout, which comprises the following components in parts by weight: 12 parts of angelica sinensis, 10 parts of ligusticum wallichii, 30 parts of Chinese violet, 30 parts of dandelion, 9 parts of corydalis amabilis, 12 parts of achyranthes bidentata, 15 parts of desmodium, 10 parts of notopterygium root, 10 parts of myrobalan and 3 parts of catechu.
The traditional Chinese medicine composition is prepared by adopting the following method:
(1) mixing and crushing the traditional Chinese medicine components into coarse powder, and putting the coarse powder into an extraction container;
(2) sequentially adding 10, 8 and 6 times of water, heating and refluxing for three times, wherein each time lasts for 1.5 hours, combining extracting solutions, filtering, concentrating under reduced pressure at 60 ℃ to obtain a fluid extract with the relative density of 1.40, drying under reduced pressure at 65 ℃ in vacuum, crushing to obtain extract powder, and adding 15g of starch, 5g of sodium carboxymethyl starch, 1.5g of magnesium stearate and 10g of microcrystalline cellulose. After being mixed evenly, the mixture is filled into capsules to obtain the traditional Chinese medicine composition capsules.
Pharmacodynamic experiment
The analgesic, anti-inflammatory and uric acid-reducing effects of the traditional Chinese medicine composition are examined by adopting potassium oxonate-induced mouse hyperuricemia experiments, xylene-induced mouse ear swelling experiments and acetic acid-induced mouse writhing experiments.
1 method of experiment
1.1 uric acid reduction experiment: 70 mice are randomly divided into three dose groups of a normal group, a model group, a gout relieving group, an allopurinol group and a gout composition, each group comprises 10 mice, the normal group and the model group are filled with normal saline with the same volume as the gastric lavage, the other administration groups are administered with corresponding liquid medicine, 20ml/kg of the liquid medicine is administered once every morning for 5 days continuously, 600mg/kg of oteracil potassium is adopted for intraperitoneal injection molding before the last administration for 1h, 0.5% cmc-Na is injected into the normal group, 0.5ml of orbital vein is collected after the administration for 1h, the room temperature is kept still for 1h, 3500rmp centrifugation is carried out for 10min, supernatant is taken, and the phosphotungstic acid method is adopted to detect the serum uric acid level.
1.2 xylene-induced ear swelling experiments in mice (anti-inflammatory): the 60 mice are randomly divided into three dose groups of a model group, a gout easing group, a compound acetylsalicylic acid group and a gout composition, wherein each group comprises 10 mice, the normal group and the model group are filled with physiological saline with the same volume as the stomach, the other administration groups are administered with corresponding liquid medicine of 20ml/kg, the administration is carried out once in the morning every day for 3 days continuously, and the gout easing group and the compound acetylsalicylic acid are administered once in the last day. 1h after the last administration, 50 mul of xylene is used for smearing the right ear of the mouse, the animal is killed after 1h, the left ear and the right ear are cut off, the ear piece is punched along the pinna of the mouse by a puncher with the aperture of 6mm, the weight is weighed, and the swelling degree of the ear (= the weight of the right ear-the weight of the left ear) is calculated.
1.3 acetic acid induced writhing test (analgesia) in mice: the 60 mice are randomly divided into three dose groups of a model group, a gout easing group, a compound acetylsalicylic acid group and a gout composition, 10 mice are in each group, the normal group and the model group are filled with physiological saline with the same volume as the stomach, the other administration groups are administered with corresponding liquid medicine of 20ml/kg, the administration is carried out once in the morning every day for 3 days continuously, and the compound acetylsalicylic acid is administered once in the last day. Animals are fasted for 12h before the last day of administration, and are injected intraperitoneally with 0.2ml of 0.85% glacial acetic acid solution 1h after the last administration to cause animal writhing, and the writhing number of the animals within 20min after acetic acid is injected intraperitoneally is calculated.
2 results of the experiment
2.1 the effect of the gout composition on reducing uric acid: after modeling, the animal blood uric acid in the model group is obviously increased compared with that in the normal group, which shows that the hyperuricemia model is successfully established, the gout composition with high dose and allopurinol in the administration group can obviously reduce the blood uric acid level of the animal, and the commercially available medicine gout relieving tablet has no obvious influence on the blood uric acid level of the animal (the result is detailed in table 1);
2.2 anti-inflammatory action of gout composition: the gout composition can obviously reduce the swelling degree of the mouse ear caused by dimethylbenzene in high and medium dosage, and the marketed drug gout easing tablet has no obvious influence on the swelling degree of the ear (the result is detailed in table 2);
2.3 analgesic action of gout composition: the gout composition can obviously reduce the writhing number of mice caused by acetic acid at high dose, and also has a reduction trend at medium and low dose, while the writhing number of animals of the commercial medicine gout tablet is equivalent to that of a model (the result is detailed in a table 3).

The invention has the beneficial effects that: the traditional Chinese medicine composition provided by the invention can relieve red swelling and hot pain in the acute attack stage of gout, reduce the blood uric acid level, effectively control the recurrence of gout and reduce the occurrence probability of other complications.

A traditional Chinese medicine composition for treating gout is characterized in that: the traditional Chinese medicine composition comprises the following components in parts by weight: 6-12 parts of angelica sinensis, 3-10 parts of ligusticum wallichii, 15-30 parts of Chinese violet, 15-30 parts of dandelion, 3-9 parts of corydalis amabilis, 5-12 parts of achyranthes bidentata, 10-15 parts of desmodium, 3-10 parts of notopterygium root, 3-10 parts of myrobalan and 1-3 parts of catechu.

The traditional Chinese medicine composition for treating gout according to claim 1, wherein: the traditional Chinese medicine composition comprises the following components in parts by weight: 7-10 parts of angelica sinensis, 4-8 parts of ligusticum wallichii, 17-25 parts of Chinese violet, 17-25 parts of dandelion, 4-7 parts of corydalis amabilis, 7-10 parts of achyranthes bidentata, 11-14 parts of desmodium, 4-8 parts of notopterygium root, 4-8 parts of myrobalan and 1-3 parts of catechu.

The traditional Chinese medicine composition for treating gout according to claim 1, wherein: the traditional Chinese medicine composition comprises the following components in parts by weight: 8 parts of angelica sinensis, 6 parts of ligusticum wallichii, 22 parts of Chinese violet, 22 parts of dandelion, 6 parts of corydalis amabilis, 8 parts of achyranthes bidentata, 12 parts of desmodium, 6 parts of notopterygium root, 6 parts of myrobalan and 2 parts of catechu.

The traditional Chinese medicine composition for treating gout according to claim 1, wherein: the traditional Chinese medicine composition comprises the following components in parts by weight: 12 parts of angelica sinensis, 10 parts of ligusticum wallichii, 30 parts of Chinese violet, 30 parts of dandelion, 9 parts of corydalis amabilis, 12 parts of achyranthes bidentata, 15 parts of desmodium, 10 parts of notopterygium root, 10 parts of myrobalan and 3 parts of catechu.
5. The preparation method of the traditional Chinese medicine composition for treating gout according to any one of claims 1 to 4, characterized by comprising the following steps: the method comprises the following steps:
(1) mixing and crushing the angelica, the ligusticum wallichii, the Chinese violet, the dandelion, the corydalis amabilis, the achyranthes bidentata, the desmodium, the notopterygium root, the myrobalan and the catechu in parts by weight into coarse powder, and putting the coarse powder into extraction equipment;
(2) adding an extraction solvent in an amount which is 4-15 times the weight of the traditional Chinese medicinal materials in the step (1), heating and refluxing for extraction for 1-4 times, wherein each time lasts for 1-6 hours, combining the extracting solutions and filtering;
(3) concentrating the extract under reduced pressure to obtain soft extract, drying, and pulverizing to obtain extract powder.
6. The preparation method of the traditional Chinese medicine composition for treating gout according to claim 5, wherein the preparation method comprises the following steps: the extraction solvent in the step (2) is water or 50-90% ethanol water solution.

The preparation method of the traditional Chinese medicine composition for treating gout according to claim 5, wherein the preparation method comprises the following steps: the reduced pressure concentration temperature in the step (3) is 50-70 ℃, the drying method is vacuum reduced pressure drying or spray drying, and the extracting solution is concentrated into thick paste with the density of 1.25-1.40 at 60 ℃.
8. The use of a Chinese medicinal composition for the treatment of gout according to any one of claims 1 to 4, wherein: adding medicinal auxiliary granules into the traditional Chinese medicine composition to prepare a medicinal preparation for treating acute gout attack.
9. The use of a traditional Chinese medicine composition for treating gout according to claim 8, wherein: the medicinal auxiliary granule comprises a filler, a disintegrating agent, a plasticizer, a flavoring agent, a lubricant, an adhesive and an absorption enhancer.
10. The use of a traditional Chinese medicine composition for treating gout according to claim 8, wherein: the dosage form of the pharmaceutical preparation comprises granules, tablets, capsules, medicinal granules or oral liquid preparations.
