本发明属于原料制备领域,具体涉及一种酶解骨粉和异麦芽酮糖醇混合颗粒的制备方法。1)计算酶解骨粉溶液所含的干物质量。2)根按比例称取市售异麦芽酮糖醇,添加纯化水,得到异麦芽酮糖醇水溶液;3)将异麦芽酮糖醇水溶液加入待浓缩的酶解骨粉溶液,减压浓缩;4)设定喷雾干燥参数,离心喷雾干燥;5)将干燥室下部收集桶和收集袋中的酶解骨粉和异麦芽酮糖醇混合颗粒收集装袋密封,称重,取样送检。本申请使酶解骨粉和异麦芽酮糖醇混合溶液快速干燥,获得钙含量和粒度均一的酶解骨粉和异麦芽酮糖醇混合颗粒,解决了酶解骨粉的可压性及流动性问题,可直接用于压制含酶解骨粉的压片糖果。

Description

酶解骨粉和异麦芽酮糖醇混合颗粒的制备方法
技术领域
本发明属于原料制备领域,具体涉及一种酶解骨粉和异麦芽酮糖醇混合颗粒的制备方法。
背景技术
酶解骨粉是一种动物源钙原料,通过对牛骨的蒸煮、粉碎、酶解、中和、喷雾干燥,制得颗粒状酶解骨粉,用于营养高钙冲剂等产品。
现有市售原辅料制备含酶解骨粉的压片糖果,为保证补钙效果,配方中添加量达40%以上,因酶解骨粉原料细度达100目以上,导致混合后的压片物料可压性及流动性很差,成品片剂的硬度和片重差异均无法保证。同时,因酶解骨钙粉含钙量≥17%,密度很大,压片过程中,酶解骨钙粉与其他辅料因密度差异大,容易发生物料分层,成品的含量均匀度受到很大影响。基于以上原因,无法采用直压工艺,只能采用湿法制粒,将酶解骨粉与辅料混合后加润湿剂,再通过干燥、过筛工序,制成可用于压片的颗粒,能耗和人工成本造成了生产成本的增加。
发明内容
本发明的目的在于提供一种酶解骨粉和异麦芽酮糖醇混合颗粒的制备方法。
本发明为实现上述目的,采用以下技术方案:
一种酶解骨粉和异麦芽酮糖醇混合颗粒的制备方法,包括下述步骤:
1)取待浓缩的酶解骨粉溶液,测定相对密度,根据储液罐所示的酶解骨粉溶液体积,计算酶解骨粉溶液所含的干物质量。
2)根据计算所得的酶解骨粉干物质量,按比例称取市售异麦芽酮糖醇,添加等量的纯化水,加热至全部溶解得到异麦芽酮糖醇水溶液;
3)将上述异麦芽酮糖醇水溶液加入待浓缩的酶解骨粉溶液,减压浓缩;
4)设定喷雾干燥参数,将酶解骨粉和异麦芽酮糖醇混合浓缩溶液经均质机混合,离心喷雾干燥。
5)将干燥室下部收集桶和收集袋中的酶解骨粉和异麦芽酮糖醇混合颗粒收集装袋密封,称重,取样送检。
具体的,步骤2)中酶解骨粉干物与异麦芽酮糖醇的质量比为45:55。
具体的,步骤3)中浓缩后的浓缩液相对密度为1.05-1.08。
具体的,步骤4)离心喷雾干燥的参数为进风口温度150℃,出风口温度100℃,干燥室温度100℃,预热10分钟。
与现有技术相比,本发明的有益效果是:
本申请在酶解骨粉的喷雾干燥工序中,在酶解骨粉溶液中添加一定量的异麦芽酮糖醇,通过设定适当的喷雾口温度和喷雾流速,使酶解骨粉和异麦芽酮糖醇混合溶液快速干燥,获得钙含量和粒度均一的酶解骨粉和异麦芽酮糖醇混合颗粒,解决了酶解骨粉的可压性及流动性问题,可直接用于压制含酶解骨粉的压片糖果,成品片剂的硬度、片重差异、含量均匀度符合质量标准。同时,因不需采用湿法制粒工艺的干燥、过筛工序,能耗和人工成本减少,大幅降低了生产成本。
具体实施方式
为了使本技术领域的技术人员更好地理解本发明的技术方案,下面结合最佳实施例对本发明作进一步的详细说明。
实施例1:一种酶解骨粉和异麦芽酮糖醇混合颗粒的制备方法,包括下述步骤:1)取待浓缩的酶解骨粉溶液,测定相对密度,根据储液罐所示的酶解骨粉溶液体积,计算酶解骨粉溶液所含的干物质量。2)根据计算所得的酶解骨粉干物质量,按45:55称取市售异麦芽酮糖醇,添加等量的纯化水,80℃至全部溶解。3)将上述异麦芽酮糖醇水溶液加入待浓缩的酶解骨粉溶液,减压浓缩,至浓缩液相对密度为1.05-1.08。4)设定喷雾干燥进风口温度150℃,出风口温度100℃,干燥室温度100℃,预热10分钟。将酶解骨粉和异麦芽酮糖醇混合浓缩溶液经均质机混合,离心喷雾干燥。5)将干燥室下部收集桶和收集袋中的酶解骨粉和异麦芽酮糖醇混合颗粒收集装袋密封,称重,取样送检。
技术效果:
1、降低成本,表1示出采用传统湿法制粒与本申请的实施例方式得到的酶解骨粉和异麦芽酮糖醇混合颗粒的成本对比。

2、改善流动性:用BT-1000型粉体综合特性测定仪测定喷雾干燥法制得的酶解骨粉与异麦芽酮糖醇混合颗粒的流动性见表2。

结论:采用喷雾干燥法制得的酶解骨粉与异麦芽酮糖醇混合颗粒原料,休止角为26°,压缩度22,表明颗粒的可压性及流动性均适合用于颗粒直压工艺。
3、改善片剂硬度:用YD-35智能片剂硬度仪测定采用喷雾干燥法制得的酶解骨粉与异麦芽酮糖醇混合颗粒直接压制的片剂见表3。

结论:采用喷雾干燥法制得的酶解骨粉与异麦芽酮糖醇喷混合颗粒直压制得的片剂,硬度适中,符合片剂要求。
4、改善含量均匀度:采用2015版药典附录0941含量均匀度检查法,检测喷雾干燥法制得的酶解骨粉与异麦芽酮糖醇混合颗粒直接压制的片剂的含量均匀度见表5。

结论:A+2.20S=1.0,根据片剂含量均匀度判断标准,结果≤15,含量均匀度符合规定,说明喷雾干燥法制得的酶解骨粉与异麦芽酮糖醇混合颗粒满足产品含量均匀度要求,适合直接压片。

1.一种酶解骨粉和异麦芽酮糖醇混合颗粒的制备方法,其特征在于,包括下述步骤:
1)取待浓缩的酶解骨粉溶液,测定相对密度,根据储液罐所示的酶解骨粉溶液体积,计算酶解骨粉溶液所含的干物质量;
2)根据计算所得的酶解骨粉干物质量,按比例称取市售异麦芽酮糖醇,添加等量的纯化水,加热至全部溶解得到异麦芽酮糖醇水溶液;酶解骨粉干物与异麦芽酮糖醇的质量比为45:55;
3)将上述异麦芽酮糖醇水溶液加入待浓缩的酶解骨粉溶液,减压浓缩;浓缩后的浓缩液相对密度为1.05-1.08;
4)设定喷雾干燥参数,将酶解骨粉和异麦芽酮糖醇混合浓缩溶液经均质机混合,离心喷雾干燥;离心喷雾干燥的参数为进风口温度150℃,出风口温度100℃,干燥室温度100℃,预热10分钟;
5)将干燥室下部收集桶和收集袋中的酶解骨粉和异麦芽酮糖醇混合颗粒收集装袋密封,称重,取样送检。
