6. Mixing the biological enzyme composition, the Chinese medicinal extract chelating solution, nisin, calcium propionate and silver ion water according to mass fraction, adjusting the pH value to 6.5-7.5, stirring and filling to obtain the biological disinfectant containing silver ion water. When the biological disinfectant containing silver ion water is used, the stock solution is diluted by 100 times.
Nisin is a food additive produced by fermentation, is an antimicrobial peptide, has a strong inhibitory effect on spores of bacillus, and is useful for the growth and reproduction of many gram-positive bacteria such as clostridium botulinum, staphylococcus aureus, streptococcus hemolyticus, listeria, and clostridium stearothermophilus. The antibacterial effect of nisin is to cause cell membrane penetration, nutrient loss and membrane potential drop by interfering with the normal function of the cellular v-membrane, leading to the death of pathogenic and putrefying bacterial cells.
The calcium propionate is a food additive, has strong inhibiting effect on mould, gram-negative bacillus or aerobic bacillus, and can also inhibit the production of aflatoxin.
The scheme is that silver ion water, microbial metabolites such as biological enzyme, nisin, Chinese herbal medicine extracts and chemically synthesized antibacterial agent calcium propionate are completely combined through a scientific experimental formula to play a role respectively, and the antibacterial agent is a safe and efficient biological bacteriostatic agent, does not generate irritant gas, is convenient to use, is low in price and is easy to obtain.
Example 1
The preparation method of the biological disinfectant containing silver ion water is characterized in that the formula comprises the following components in percentage by mass: 40 parts of biological enzyme composition, 43 parts of Chinese medicinal extract chelating liquid, 5 parts of nisin, 1 part of calcium propionate and 11 parts of silver ion water.
The preparation method of the biological disinfectant comprises the following steps:
1. preparing a biological enzyme composition: firstly, 5 parts of lysozyme, 1 part of carboxypeptidase, 1 part of collagen hydrolase, 1 part of catalase and 2 parts of glucose oxidase are mixed, silver ion water with the amount of 3 times of the mass of the biological enzyme composition is added, and the filtrate is obtained for later use after filtration;
2. traditional Chinese medicine extraction: mixing 3 parts of folium artemisiae argyi, 4 parts of honeysuckle, 1 part of dandelion, 1 part of fructus forsythiae and 1 part of liquorice, adding 95% ethanol, extracting for 3 times, each time for 1 hour, combining extracting solutions, cooling, filtering, adding activated carbon with 2 thousandth of mass fraction of the extracting solution for decoloring, recovering a solvent, concentrating until the density is 1.3g/ml, and naming the extracting solution as extracting solution I;
3. adding 3 times of water into the dregs of a decoction extracted in the step (2), extracting for 3 times at 90 ℃ for 1 hour each time, combining the extracting solutions, cooling, filtering, adding activated carbon with 5 per mill of the mass fraction of the extracting solution for decoloring, and naming the extracting solution as extracting solution II;
4. mixing the extracting solution I and the extracting solution II according to the mass fraction of 7:3 to obtain a traditional Chinese medicine extract;
5. adding chitosan with deacetylation degree of 90% in an amount of 2% by mass into the Chinese medicinal extract, chelating with the Chinese medicinal extract for 1 hr at 60 deg.C, centrifuging, and collecting supernatant to obtain Chinese medicinal extract chelating solution;
6. mixing and stirring 40 parts of biological enzyme composition, 43 parts of Chinese medicinal extract chelating liquid, 5 parts of nisin, 1 part of calcium propionate and 11 parts of silver ion water according to mass fraction for 30 min. Adjusting the pH value to 6.5, and filling to obtain the biological disinfectant containing silver ion water, wherein the stock solution of the biological disinfectant containing silver ion water is diluted by 100 times for use.
Example 2
A preparation method of a biological disinfectant containing silver ion water comprises the following components in parts by mass: 20 parts of biological enzyme composition, 30 parts of Chinese medicinal extract chelating liquid, 5 parts of nisin, 5 parts of calcium propionate and 40 parts of silver ion water.
The preparation method of the biological disinfectant comprises the following steps:
1. preparing a biological enzyme composition: mixing 3 parts of lysozyme, 3 parts of carboxypeptidase, 1 part of collagen hydrolase, 1 part of catalase and 2 parts of glucose oxidase, adding silver ion water which is 5 times of the amount of the biological enzyme composition, filtering, and taking filtrate for later use;

Adjusting the pH value to 7.0, and filling to obtain the biological disinfectant containing silver ion water, wherein the stock solution of the biological disinfectant containing silver ion water is diluted by 100 times for use.
Example 3
A preparation method of a biological disinfectant containing silver ion water comprises the following components in parts by mass: 20 parts of biological enzyme composition, 50 parts of Chinese medicinal extract chelating liquid, 1 part of nisin, 1 part of calcium propionate and 28 parts of silver ion water.
The preparation method of the biological disinfectant comprises the following steps:
1. preparing a biological enzyme composition: mixing 4 parts of lysozyme, 2 parts of carboxypeptidase, 2 parts of collagen hydrolase, 1 part of catalase and 1 part of glucose oxidase, adding silver ion water in an amount which is 4 times that of a biological enzyme composition, filtering, and taking filtrate for later use;
2. traditional Chinese medicine extraction: mixing 4 parts of folium artemisiae argyi, 2 parts of honeysuckle, 1 part of dandelion, 2 parts of fructus forsythiae and 1 part of liquorice, adding 95% ethanol, extracting for 3 times, each time for 1 hour, combining the extracting solutions, cooling, filtering, adding activated carbon with the mass fraction of 3 per thousand of the filtrate for decoloring, recovering the solvent, concentrating until the density is 1.3g/ml, and naming the extracting solution as extracting solution I;
3. adding 3 times of water into the dregs of a decoction extracted in the step (2), extracting for 3 times at 90 ℃ for 1 hour each time, combining the extracting solutions, cooling, filtering, adding activated carbon with the mass fraction of 2 per mill of the filtrate for decoloring, and naming the mixture as extracting solution II;

A preparation method of a biological disinfectant containing silver ion water comprises the following components in parts by mass: 30 parts of biological enzyme composition, 30 parts of Chinese medicinal extract chelating liquid, 5 parts of nisin, 5 parts of calcium propionate and 30 parts of silver ion water.
The preparation method of the biological disinfectant comprises the following steps:
1. preparing a biological enzyme composition: mixing 3 parts of lysozyme, 3 parts of carboxypeptidase, 1 part of collagen hydrolase, 1 part of catalase and 23 parts of glucose oxidase, adding silver ion water which is 5 times of the amount of the biological enzyme composition, filtering, and taking filtrate for later use;

A preparation method of the biological disinfectant comprises the following steps:
1. extracting the traditional Chinese medicine composition: mixing 3 parts of folium artemisiae argyi, 4 parts of honeysuckle, 1 part of dandelion, 1 part of fructus forsythiae and 1 part of liquorice, adding 95% ethanol, extracting for 3 times, each time for 1 hour, combining extracting solutions, cooling, filtering, adding activated carbon with 2 thousandth of mass fraction of the extracting solution for decoloring, recovering a solvent, concentrating until the density is 1.3g/ml, and naming the extracting solution as extracting solution I;
2. adding 3 times of water into the dregs of a decoction extracted in the step (2), extracting for 3 times at 90 ℃ for 1 hour each time, combining the extracting solutions, cooling, filtering, adding activated carbon with 5 per mill of the mass fraction of the extracting solution for decoloring, recovering the solvent, and concentrating to the density of 1.2g/ml, which is named as extracting solution II;
3. mixing the extracting solution I and the extracting solution II according to the mass fraction of 7:3 to obtain a traditional Chinese medicine composition extract;
4. mixing 43 parts of the traditional Chinese medicine composition extract, 1 part of chitosan, 5 parts of nisin, 1 part of calcium propionate and 50 parts of distilled water according to mass fraction, and stirring for 30 min. Adjusting pH to 6.5, and bottling to obtain biological disinfectant, wherein the disinfectant is used by diluting stock solution by 100 times.
Sterilization test
Inoculating Escherichia coli, Staphylococcus aureus, Candida albicans, Bacillus subtilis, Aspergillus niger, yeast, rhinovirus, and adenovirus in culture medium, and culturing for 20 hr. Washing thallus Porphyrae in the culture medium with sterile physiological saline, adding 0.5mL of experimental bacterial suspension into sterile test tube with 30g/L bovine serum albumin, mixing, keeping temperature to 20 + -1 deg.C in water bath, adding 10mL of disinfectant (0.1mL of the inventive disinfectant is diluted to 10mL with sterile water) into the test tube, mixing, and timing. Sucking 0.5mL of mixed liquid of the bacterial suspension and the disinfectant prepared in the embodiment 1 of the invention, mixing the mixed liquid with 4.5mL of neutralizing agent TSB for 5-10min, sucking 1mL of mixed liquid, measuring the number of viable bacteria according to a viable bacteria culture counting method, inoculating 3 plates in each tube, counting the number of viable bacteria, calculating the killing rate, and unifying the sterilization time to 5 min. The results are shown in Table 1:

As can be seen from Table 1: the sterilization formula has the advantages that the sterilization formula is well optimized, and the water-soluble and fat-soluble components of the biological enzyme and the Chinese herbal medicine extract, the chitosan, the nisin and the calcium propionate are scientifically matched, so that the product has a good effect on various harmful bacteria.
Product stability test
As the product is a biological disinfectant containing silver ion water and the biological enzyme is an active substance, the product and the comparative example are mainly considered to be placed for 60 days at 0-40 ℃, the stability of the product is considered from the aspects of appearance and enzyme activity measurement, and the enzyme activity measurement of the product is determined according to the method of food additive lysozyme of GB1886.257-2016 food safety national standard.
2 parts of the products of the examples 1 to 4 and the products of the comparative examples 1 to 4 are respectively taken and placed at 0 ℃, 20 ℃ and 40 ℃ for 60 days, the appearance change of the products is observed, the products are taken out after being placed for 60 days to determine the activity of lysozyme, and the activity of the lysozyme added in the biological disinfectant containing silver ion water is not lower than 1000 u/ml.

From table 2, it can be seen that: the examples 1-4 are respectively placed at 0-40 ℃ for 60 days, the appearance is not changed, which shows that the stability is good, the comparative examples 1-5 respectively have the phenomenon of layering at 10-40 ℃, which shows that the quality is unstable and the quality guarantee period is short, and the non-layering of the examples is mainly because the chitosan and the fat-soluble traditional Chinese medicine extract are added, the emulsification effect is realized in the formula, and the quality guarantee period is greatly prolonged.

As can be seen from Table 3, the lysozyme activity changes greatly after being placed at 0-40 ℃ for 60 days, the lysozyme activity hardly changes after being placed at 40 ℃ for 60 days in the embodiment, but the lysozyme is almost lost almost in the comparative examples 1-4 after being placed at 40 ℃ for 60 days, which shows that the product has a very good enzyme activity protection effect, and the product has good stability and can be stably stored for 60 days.

The preparation method of the biological disinfectant containing silver ion water is characterized in that the formula comprises the following components in percentage by mass: 10-40 parts of biological enzyme composition, 30-50 parts of traditional Chinese medicine extract chelating liquid, 1-5 parts of nisin, 1-5 parts of calcium propionate and 10-40 parts of silver ion water.

The method for preparing a biological disinfectant containing silver ion water according to claim 1, wherein said biological enzyme composition comprises, in mass fraction: 3-5 parts of lysozyme, 1-3 parts of carboxypeptidase, 1-3 parts of collagen hydrolase, 1-3 parts of catalase and 1-3 parts of glucose oxidase.
3. The method for preparing biological disinfectant containing silver ion water according to claim 1, wherein the traditional Chinese medicine comprises the following components by mass percent: 3-5 parts of folium artemisiae argyi, 1-4 parts of honeysuckle, 1-3 parts of dandelion, 1-3 parts of fructus forsythiae and 1-3 parts of liquorice.
4. The method for preparing a biological disinfectant containing silver ion water according to claim 1, wherein the silver ion water is water prepared by electrolyzing silver ions, has a sterilizing effect, and has a silver ion content of 50-80 ppm.
5. The method of preparing a biological disinfectant from biological silver-ion-containing water according to claim 1, wherein:
(1) preparing a biological enzyme composition: mixing lysozyme, carboxypeptidase, collagen hydrolase, catalase and glucose oxidase according to mass percent, adding silver ion water, filtering, and taking filtrate for later use;
(2) traditional Chinese medicine extraction: mixing folium artemisiae argyi, honeysuckle, dandelion, fructus forsythiae and liquorice according to mass percentage, adding 95% ethanol, extracting for 3 times, extracting for 1 hour each time, combining the extracting solutions, cooling, filtering, adding a decolorizing agent for decolorizing, recovering the solvent, concentrating until the density is 1.3g/ml, and naming the extracting solution as extracting solution I;
(3) adding 3 times of water into the dregs of a decoction extracted in the step (2), extracting for 3 times at 90 ℃ for 1 hour each time, combining the extracting solutions, cooling, filtering, adding a decoloring agent for decoloring, and naming the extracting solution as an extracting solution II;
(4) mixing the extracting solution I and the extracting solution II according to the mass fraction to obtain a traditional Chinese medicine extract;
(5) adding chitosan with deacetylation degree of 90% in an amount of 2% by mass into the Chinese medicinal extract, chelating with the Chinese medicinal extract for 1 hr at 60 deg.C, centrifuging, and collecting supernatant to obtain Chinese medicinal extract chelating solution;
(6) mixing the biological enzyme composition, the Chinese medicinal extract chelating solution, nisin, calcium propionate and silver ion water according to mass fraction, adjusting the pH value to 6.5-7.5, stirring and filling to obtain the biological disinfectant containing silver ion water.
6. The method for preparing a biological disinfectant containing silver ion water according to claim 5, wherein the silver ion water in step (1) is added in an amount of 3-5 times the mass fraction of the biological enzyme composition.
7. The method for preparing biological disinfectant containing silver ion water according to claim 5, wherein the decolorizing agent in steps (2) and (3) is activated carbon, and the adding amount of the decolorizing agent is 2-5 per mill of the mass fraction of the extracting solution.
8. The method for preparing a biological disinfectant containing silver ion water according to claim 5, wherein in the step (4), the extract I and the extract II are mixed according to the mass fraction of 7: 3.
9. The method for preparing a biological disinfectant containing silver ion water according to claim 5, wherein the mixing and stirring time in step (5) is 30 min.
10. The method for preparing a biological disinfectant containing silver ion water according to claim 1, wherein the stock solution is diluted 100 times for use.
