Abstract

本发明涉及医药领域，提供一种流式细胞仪的淋巴细胞样品的制备方法，其包括:取抗凝血液样品离心，得血细胞沉淀，用PBS溶液稀释，然后加入到含有淋巴细胞分离液的离心管中离心，弃掉血浆，得白膜层细胞；然后加入到离心管中，添加PBS溶液，离心弃上清液，再加入PBS溶液调节细胞浓度，将细胞溶液加入流式管内；然后加入抗体充分混均，避光孵育；离心并弃掉上清液，加入预冷的PBS‑EDTA溶液重悬细胞，即得。本发明方法得到的样品富集纯化了淋巴细胞，充分保存了淋巴细胞的活性，尽可能去除粒细胞、红细胞、血小板等其它血液成分，使需要检测的亚群分群明显纯化，分析检测更加准确，节省了分析抗体和试剂成本。
