Description

Donkey-hide gelatin peptide buccal tablet rich in free arginine and preparation method thereof
Technical Field
The invention belongs to the field of polypeptide preparation in biotechnology, and particularly relates to a donkey-hide gelatin peptide buccal tablet rich in free arginine and a preparation method thereof.
Technical Field
Arginine is an important di-basic amino acid, known as an important carrier for transporting and storing amino acids in the body, and is extremely important in intramuscular metabolism. Arginine is less synthesized in the human body and needs to be partially supplemented from food, while it is an essential amino acid for the middle-aged and elderly people.
Nitric oxide NO is widely distributed in various tissues in the living body, particularly in nerve tissues. It was evaluated as a star molecule in 1992 by the American Science journal. NO is an extremely unstable biological free radical, whose production depends on Nitric Oxide Synthase (NOs) and has important biological effects in cardiovascular and cerebrovascular regulation, nervous regulation, immune regulation, and the like. Therefore, the method is generally regarded by people.
Nitric oxide in the human body has two sources: one is non-enzymatic, from the body surface or chemical degradation and conversion of ingested inorganic nitrogen; the first is enzymatic, produced by the enzyme nitric oxide synthase catalyzing the removal of guanidino from L-arginine. The enzymatic nitric oxide is derived from arginine, a precursor of nitric oxide. The arginine-rich food taken into human body can produce nitric oxide in vivo through enzyme and exert physiological functions.
Because arginine is a precursor of nitric oxide, supplementing arginine can obviously improve the content of nitric oxide in vivo, and nitric oxide can contact and relax muscle cells in arteries, so that arterial blood vessels are expanded, blood pressure is reduced, blood flow is improved, and the risk of cardiovascular and cerebrovascular diseases can be effectively reduced. On the contrary, when the arginine content in the body is deficient, the nitric oxide content in the body is reduced, namely, the possibility of sudden cardiovascular and cerebrovascular diseases is greatly increased.
Once nitric oxide is produced in the body, it contacts and relaxes muscle cells in the arteries, dilating the arteries, which reduces blood pressure and improves blood flow. In addition, nitric oxide prevents blood from clotting at dangerous sites. If blood clots in the heart or brain, the patient may suffer a heart attack or stroke. As long as the human body produces a sufficient amount of nitric oxide, the risk of cardiovascular and cerebrovascular diseases is greatly reduced, and therefore, the nitric oxide can effectively reduce blood pressure and prevent stroke and heart disease.
Arginine is a natural product, an amino acid from proteins, and is found in many sources, including chicken, fish, and certain vegetables. However, arginine in ordinary food is often low in content, and the amount of generated nitric oxide is small, so that the food does not have related nutritional value.
Relevant studies at home and abroad prove that the food rich in arginine can reduce the harm of hypertension and cardiovascular and cerebrovascular diseases, and the physiological function of the food is related to that arginine can effectively promote the generation of nitric oxide in vivo. High fat diets can lead to a decrease in endothelial dependent vasodilatory function in the body, resulting in increased blood pressure, decreased blood flow, and increased blood viscosity. Arginine can effectively promote the generation of nitric oxide in vivo and can reduce the degradation of nitric oxide by in vivo oxidized lipid, thereby improving the content of nitric oxide in vivo.
The colla Corii Asini is a block prepared by removing hair from skin of donkey of Equidae, and has effects of nourishing yin, tonifying blood, and preventing miscarriage. It is used to treat cough due to blood deficiency, cough due to asthenia, hematemesis, epistaxis, hematochezia, menoxenia, metrorrhagia, and fetal leakage. Modern medical research shows that the content of arginine in donkey-hide gelatin is very high, and can reach 6% -8%, but the arginine exists in a polypeptide form and cannot be directly absorbed and utilized. The protein in the donkey-hide gelatin is mainly collagen, the digestion and absorption of which need the participation of related protease, and the adaptation population is often suffered from weakness of the spleen and the stomach or gastrointestinal diseases, and taking the donkey-hide gelatin not only causes the burden of the digestive system of a patient, but also can not effectively exert the drug effect, and is easy to cause 'excessive internal heat'.
With the development of modern biological enzymolysis technology, in recent years, there are increasing reports on the research of preparing donkey-hide gelatin into donkey-hide gelatin peptide by using biological enzymolysis technology. CN102021215A reports a method for extracting colla Corii Asini peptide, which utilizes alkaline protease, trypsin and flavourzyme, and focuses on the molecular weight of colla Corii Asini peptide. CN101294186A utilizes pepsin and trypsin to carry out enzymolysis on colla Corii Asini, and focuses on the size and distribution of colla Corii Asini peptide molecular weight.
The existing preparation technology of the donkey-hide gelatin peptide mostly concentrates on the size and the distribution of the molecular weight of the donkey-hide gelatin peptide, arginine in the donkey-hide gelatin peptide cannot be well released, the arginine exists in a polypeptide form, the content of free arginine is lower than 1 percent, and the arginine can be absorbed and utilized after being digested, so that the utilization rate of the arginine is influenced.

Aiming at various problems existing in the utilization process of the existing donkey-hide gelatin peptide, the invention provides a simple and convenient preparation method of a donkey-hide gelatin peptide buccal tablet. The method is simple to operate, no chemical addition is needed in the whole process, the content of free arginine in the donkey-hide gelatin peptide buccal tablet is more than 4%, the donkey-hide gelatin peptide buccal tablet can be quickly absorbed and utilized by a human body in a buccal mode, and the defects of low arginine absorption utilization rate, low speed and the like are overcome on the premise of keeping the active function of polypeptide.
