Abstract
The invention discloses a hair growth promoting active peptide composition, which comprises a keratin gene stimulating peptide and a synergistic short peptide; the synergistic short peptide is one or more of leucine-proline-arginine, proline-leucine-arginine, isoleucine-proline-arginine, lysine-isoleucine-proline, proline-isoleucine-lysine or valine-glutamine-proline. The active peptide composition can actively identify keratinocytes and hair follicle immune cells in the skin, stimulate keratin genes, play a plurality of effects of hair regeneration, anchoring the stability of new hair and the like, and synergistically and remarkably increase the growth and thickening effects of the hair, particularly eyelashes. The invention also discloses a hair growth promoting preparation containing the active peptide composition and application thereof in the field of cosmetics.
Description
Technical Field
The invention relates to a hair growth promoting active peptide composition and application thereof.
Background
The hair is a main accessory of the skin, is one of the characteristics of mammals such as human beings and the like, and also has the functions of mechanical protection, ultraviolet protection, body temperature regulation, touch feeling regulation and the like. In addition, hair, which is a human being, is an important element that presents different appearance characteristics. For example: the thick and long eyelashes can make the eyes look more attractive, and are one of the important facial modification parts. Therefore, special cosmetics that make eyelashes appear to be in a thickened state are hot in the cosmetic field, and product forms include various mascaras, false eyelashes, and eyelash perming agents. However, frequent mascara application, and the operations of grafting eyelashes or curling the eyelashes for a long time and continuously do not change the natural number and length of the eyelashes, but may cause certain damage to the eyelashes, become thin and fragile, and may even damage the eyes. Therefore, cosmetic products for nourishing hair from the root, making hair strong, strong and healthy, especially in specific areas, such as the eyelashes around the eyes, which are sensitive, are increasingly in social demand.
At present, most of the components added in eyelash beauty lotion on the market are used for moisturizing and caring. Most of these ingredients are also used in beauty care products such as lotions, essences and the like, such as hyaluronic acid, polyquaternium (artificial cell membrane) and the like, and less active ingredients are used for the targeted activation of skin and hair follicle cells and the activation of keratin. Typical two active ingredients include minoxidil and bimatoprost, but both are reported to have certain side effects. For example: minoxidil (Mi nox i d i l) causes irritative or contact hypersensitivity reactions, and increased hair at inappropriate sites. The bimatoprost used for a long time can accelerate the deposition of melanin around eyes, cause dark eye circles, stimulate conjunctiva, cause symptoms such as retina swelling and eye inflammation and the like, and seriously possibly cause optic nerve damage, and lead to vision loss and even blindness.
With the development of biotechnology, it is disclosed that a variety of active peptides (CN1622797A, CN102596170A, CN1445236A and 200680039713.2) can activate hair follicles, or activate keratin genes, promote keratin production, and have the effect of promoting hair growth. However, except for the series of patents (USA Pat. No. 5538945,5177061,5120831) of ProCyte company, in which the polypeptide (Cu-GHK) involved in stimulating hair follicles and promoting hair growth is a small molecule, the molecular weight of the polypeptide involved in the other patents is generally large. Due to the physiological structure of the skin, it mainly exerts a barrier and protective action. Therefore, it is difficult to directly permeate skin with water-soluble or fat-soluble active ingredients alone, especially with some macromolecular ingredients (molecular weight greater than 500 Da). This problem is also indicated in patent application CN105106028A ("the polypeptide in the patent publication, which has a large molecular weight and is not favorable for skin absorption, and reduces the usage rate of the polypeptide"). However, the patent publication discloses only an invention for improving the anchoring of newly grown hair and preventing hair loss, and does not actually solve the problem of absorption promotion of active peptides that promote hair growth. If the functional components cannot be absorbed through the skin, the functional components cannot enter hair follicles to achieve the effects of promoting hair growth and anchoring hairs.
Disclosure of Invention
In view of the above-mentioned deficiencies of the prior art, the present invention provides an active peptide composition for promoting hair growth, which can actively identify keratinocytes and hair follicle immune cells in the skin, stimulate keratin genes, exert various effects such as hair regeneration and stability of anchoring new hair, and synergistically and significantly increase the growth and thickening effects of hair, especially eyelashes.
The specific technical scheme is as follows:
one of the objects of the present invention is to disclose a hair growth promoting active peptide composition.
A hair growth promoting active peptide composition comprises a keratin gene stimulating peptide and a short synergistic peptide;
the keratin gene stimulating peptide is one or more of copper peptide (GHK-Cu), myristoyl pentapeptide-17, myristoyl hexapeptide-16 and biotin tripeptide-1 (biotin-GHK);
the synergistic short peptide is one or more of leucine-proline-arginine, proline-leucine-arginine, isoleucine-proline-arginine, lysine-isoleucine-proline, proline-isoleucine-lysine or valine-glutamine-proline.
The invention synergistically and remarkably increases the effect of growing and thickening hair, especially eyelashes, by using the two active peptides in combination. Although the reason for the specific synergy is not clear, it is presumed that the active peptide composition for promoting hair growth, which is an active ingredient in the active peptide composition of the present invention, can actively recognize keratinocytes and hair follicle immune cells in the skin and stimulate keratin genes, thereby exhibiting various effects such as hair regeneration and stability in anchoring newly-grown hair.
Preferably, the mass ratio of the keratin gene stimulating peptide to the synergistic short peptide is (5-20): (0.05-5).
More preferably, the mass ratio of the keratin gene stimulating peptide to the synergistic short peptide is (5-10): (0.1-2). In a preferred embodiment of the present invention, the short peptides exert a strong synergistic effect, although the content of the short peptides is much smaller than that of the keratin gene-stimulating peptides.
Preferably, the peptide composition of the present invention can be synergistically enhanced in its effect by adding a signal peptide.
More preferably, the signal peptide is one or more of myristoyl pentapeptide-11, palmitoyl tripeptide-1, palmitoyl tripeptide-5, palmitoyl pentapeptide-3, palmitoyl hexapeptide-6, palmitoyl hexapeptide-9, myristoyl hexapeptide-4 and myristoyl octapeptide-1.
Preferably, the peptide composition of the present invention can synergistically increase its effect by adding a neurotransmitter inhibitory peptoid.
More preferably, the neurotransmitter-inhibiting peptoid is one or more of dipeptide snake toxin, carnosine, acetyl hexapeptide-3, acetyl hexapeptide-8, acetyl octapeptide-1, pentapeptide-3, tripeptide-1, and/or palmitoyl dipeptide-5 complex salt, palmitoyl tetrapeptide-7, tripeptide-4, hexapeptide-10, and the like.
Although the active peptides of the present invention have a synergistic effect, their absorption rate is low, and therefore, it is necessary to use active peptide compositions at high concentrations. In order to solve the technical problem, the inventors of the present invention further research and find that the absorption rate of the active peptide for promoting hair growth can be effectively improved by using a nano stabilizer in the active peptide composition, thereby increasing the efficacy of the active peptide.
The invention also discloses a hair growth promoting preparation containing the active peptide composition.
A hair growth promoting preparation contains 0.01-20 wt% of the active peptide composition and 1-20 wt% of a nano stabilizer.
Preferably, the nano stabilizer contains 1-20 wt% of emulsifier, 1-20 wt% of liquid grease, 0.1-20 wt% of polyol and the balance of water.
Further preferably, the emulsifier is: PEG-40 hydrogenated castor oil, PEG-30 hydrogenated castor oil, PEG-20 hydrogenated castor oil, PEG-10 hydrogenated castor oil, PEG-5 hydrogenated castor oil, polysorbate 80, polysorbate 60, polysorbate 40, polysorbate 20, PEG-30 dipolyhydroxystearate, PEG-8 caprylic capric glyceride, PEG-6 caprylic/capric glyceride, polyglycerol-10 stearate, laureth-23, polyoxyethylene castor oil-20, polyoxyethylene castor oil-30, polyoxyethylene castor oil-35, polyoxyethylene castor oil-40, PEG-40 stearate, polyglycerol-10 myristate, polyglycerol-4 oleate, coco glucoside, ceteareth-20, ceteareth-25, polyoxyethylene castor oil-20 ether, polyoxyethylene castor oil-30 ether, polyoxyethylene castor oil-35 ether, polyoxyethylene castor oil-40 ether, polyethylene glycol 400 monoglyceride, polyethylene glycol 400 diglyceride, polyethylene glycol 600 monoglyceride, polyethylene glycol 600 diglyceride, coco glucoside, ceteareth-20, ceteareth-25, sodium bis (lauramide glutamine) lysine, squalane, buthaneth-26, egg yolk lecithin, and soybean lecithin.
More preferably, the liquid oil is: one or more of isopropyl myristate, isopropyl palmitate, caprylic capric triglyceride, vitamin E, simethicone, caprylic capric macrogol glyceride, macrogol laurate, macrogol stearate, linolic acid glyceride, propylene glycol monocaprylate, propylene glycol dicaprylate, diethyl sebacate, isononyl isononanoate, glyceryl triacetate, squalene and soybean oil.
More preferably, the polyol is: one or more of polyethylene glycol, propylene glycol, glycerol, 1, 3-butanediol, and 1, 2-pentanediol.
The invention discloses a preparation method of a hair growth promoting preparation in the technical scheme, which comprises the following steps:
(1) under the condition of heating and stirring, uniformly mixing the emulsifier, the liquid oil and fat and the polyol.
(2) And (2) after the mixture obtained in the step (1) is stabilized and returns to the room temperature condition, adding the synergistic short peptide and the balance of water into the mixture to obtain the nano stabilizer mixed with the synergistic short peptide.
(3) And (3) after the pH value of the system obtained in the step (2) is adjusted to be 5.5-7.0, adding other components except the synergistic short peptide in the active peptide composition into the active peptide composition, and mixing to obtain the hair growth promoting preparation.
Preferably, the heating temperature in the step (1) is 40-80 ℃, and the stirring speed is 10-3000 rpm.
Preferably, the room temperature in the step (2) is 10-40 ℃.
Preferably, the pH adjusting agent used in step (3) is triethanolamine, citric acid or acetic acid.
The hair growth promoting preparation obtained according to the technical scheme of the invention has a vesicle, tubular or onion-shaped structure.
The size of the composition obtained according to the technical scheme of the invention is 10-1000nm, preferably 10-500nm, more preferably 10-200nm, and even more preferably 10-100 nm.
The preparation for promoting hair growth can also be added with algae and traditional Chinese medicine extracts.
Preferably, the algae and Chinese medicinal extract is one or more of algae extract, wild radix Puerariae extract, Polygoni Multiflori radix root extract, radix Gentianae extract, fructus Vitics Simplicifoliae extract, Nanzhu bamboo shoot bark extract, and radix Arctii extract.
Fructus Viticis is dry mature fruit of Simpleleaf Vitex rotundifolia or Vitex rotundifolia of Verbenaceae, and is the first choice of herbs for hair care in ancient formula; gentian is a plant of gentian of gentianaceae, is one of the oldest plants on earth, and has various hair-benefiting effects. Polygonum multiflorum is recorded in Kai Bao Ben Cao to blacken beard and hair and delight color, and is called four fairy grasses in China together with ginseng, ganoderma lucidum and Cordyceps sinensis.
The composition of the present invention may further comprise said vitamins and derivatives thereof.
Preferably, the vitamin and its derivatives are one or more of vitamin B3, vitamin B5, vitamin B6, Magnesium Ascorbyl Phosphate (MAP), ascorbyl tetraisopalmitate, ascorbyl ethyl ether, vitamin E linoleate, vitamin E succinic acid monoester, vitamin E acetate, vitamin K and vitamin H.
The third purpose of the invention is to disclose the application of the active peptide composition or the hair growth promoting preparation in the cosmetic field. In particular to the application of the hair promoting effect in cosmetics.
Preferably, the compositions of the invention are suitable for administration by the topical route, they can be used as care and/or make-up products for the parts of the head, eyebrows and eyelashes, etc.
The composition of the present invention can be applied to all cosmetic forms comprising it. Can be in liquid form such as cream, oil-in-water emulsion or water-in-oil or multiple emulsion, suspension, gel, milk, paste, foam, lotion, etc., and can also be in solid form such as lyophilized powder, lyophilized floc, lyophilized film, etc.
Furthermore, the composition according to the invention comprises any additives normally used in the given field of application, as well as adjuvants required for its formulation, such as pH adjusters, thickeners, diluents, antioxidants, colorants, sunscreens, pigments, fillers, preservatives, fragrances, odor absorbers, essential oils and the like.
The invention has the following beneficial effects:
the invention synergistically and remarkably increases the effect of growing and thickening hair, especially eyelashes, by using the two active peptides in combination.
In the invention, the nano stabilizer is a thermodynamically stable nano dispersion system, has uniform size, ensures that the hair growth promoting active peptide composition has quick skin absorption, good permeability and good biocompatibility, plays a role of stabilizing active substances, ensures that the content of the hair growth promoting active peptide composition is high, ensures that the concentration of the active ingredient hair growth promoting active peptide in the preparation can reach 20 percent, and can continuously release the hair growth promoting active peptide as an active ingredient storage, thereby being capable of maintaining the effective concentration for a long time.
The preparation method of the active peptide composition for promoting hair growth, provided by the invention, is simple, the process is easy to control, the active peptide composition is suitable for industrial and large-scale production, and an organic solvent is not used in the whole process, so that the active peptide composition has small irritation to skin and high safety.

The principles and features of this invention are described below in conjunction with examples, which are included to illustrate the invention and not to limit the scope of the invention.
The synergistic short peptides described in the examples were prepared by solid phase/liquid phase chemical synthesis with reference to the mature peptide.
Example 1
A hair growth promoting active peptide composition comprises a keratin gene stimulating peptide and a synergistic short peptide, wherein the mass ratio of the keratin gene stimulating peptide to the synergistic short peptide is 5: 0.5; also included is palmitoyl pentapeptide-3, in a mass ratio to the keratin gene stimulating peptide of 3: 5;
wherein the keratin gene stimulating peptide is myristoyl hexapeptide-16;
the synergistic short peptide is valine-glutamine-proline.
A hair growth promoting preparation comprises the active peptide composition, and the specific components (the total amount is 100 wt%):
the preparation method of the hair growth promoting preparation comprises the following steps:
(1) the compound in phase A was heated to 60 ℃ with stirring at 2000 rpm. Heating and stirring are continued until all components are mixed uniformly.
(2) Cooling the mixture to 30 ℃, adding all the components in the phase B, and stirring uniformly.
(3) The pH of the system was adjusted to 7.0 with triethanolamine and the D and E components were added.
And (4) carrying out particle size and shape characterization on the obtained hair growth promoting preparation. The DLS result shows that the particle size is 350nm, and the scanning electron microscope and the transmission electron microscope detect that the particle size presents a vesicle structure, and the results are shown in figure 1 and figure 2.
Example 2
A hair growth promoting active peptide composition comprises a keratin gene stimulating peptide and a synergistic short peptide, wherein the mass ratio of the keratin gene stimulating peptide to the synergistic short peptide is 5: 1; also included is myristoyl pentapeptide-11, in a mass ratio to the keratin gene stimulating peptide of 1: 5;
wherein the peptide stimulating the keratin gene is a copper peptide;
the synergistic short peptide is leucine-proline-arginine.
A hair growth promoting preparation comprises the active peptide composition, and the specific components (the total amount is 100 wt%):
the preparation method of the hair growth promoting preparation comprises the following steps:
(1) the compound in phase A was heated to 80 ℃ with stirring at 800 rpm. Heating and stirring are continued until all components are mixed uniformly.
(2) Cooling the mixture to 40 ℃, adding all the components in the phase B, and stirring uniformly.
(3) The pH of the system was adjusted to 5.5 with citric acid and the D and E components were added.
The resulting hair growth promoting formulation was characterized. The tube-like structure was observed by atomic force microscope, and the results are shown in fig. 3 and 4.
Example 3
A hair growth promoting active peptide composition comprises a keratin gene stimulating peptide and a synergistic short peptide, wherein the mass ratio of the keratin gene stimulating peptide to the synergistic short peptide is 5: 0.2; also included is acetyl hexapeptide-3, in a mass ratio to the keratin gene stimulating peptide of 2: 5.

wherein the synergistic oligopeptide is proline-leucine-arginine and valine-glutamine-proline, and the mass ratio of the proline to the valine-glutamine-proline is 1: 1.

A hair growth promoting active peptide composition comprises a keratin gene stimulating peptide and a synergistic short peptide, wherein the mass ratio of the keratin gene stimulating peptide to the synergistic short peptide is 5: 0.2; also included is myristoyl hexapeptide-4, in a mass ratio to the keratin gene stimulating peptide of 2: 5;
wherein the peptide stimulating the keratin gene is biotin tripeptide-1;
the synergistic short peptide is proline-leucine-arginine and valine-glutamine-proline, and the mass ratio of the proline to the valine-glutamine-proline is 1: 1.

A hair growth promoting active peptide composition comprises a keratin gene stimulating peptide and a synergistic short peptide, wherein the mass ratio of the keratin gene stimulating peptide to the synergistic short peptide is 7: 1.

A hair growth promoting active peptide composition comprises a keratin gene stimulating peptide and a synergistic short peptide, wherein the mass ratio of the keratin gene stimulating peptide to the synergistic short peptide is 10: 1.5.

Comparative example 1
Preparing the hair growth promoting preparation. As a control for examples 1-6, the formulation was made without the polypeptide of component B (i.e., without the short peptide synergist), and the remaining technical features were the same as in examples 1-6, to obtain control groups 1-6 in sequence.
Test 1 evaluation test of safety
The safety of the hair growth promoting preparations obtained in examples 1 to 6 was evaluated.
The safety of the composition of each embodiment is evaluated by selecting 9-day-old chick embryos according to the chorioallantoic membrane test standard of cosmetic eye irritation/corrosiveness published by the entry-exit inspection and quarantine industry standard.
By using the reaction time method, 0.9 wt% NaCl solution is used as a negative control, 0.3 wt% NaOH solution is used as a positive control, and 1 wt% fatty alcohol ether sodium sulfate salt mixture (ASV) is used as a reference substance control. The results are shown in the following table:
according to the standard IS (301-SecH) 5/300+ (301-SecL) 7/300+ (301-SecC) 9/300;
IS <1 IS non-irritating; 1< IS <5 mild stimulation; 5< IS <9 moderate stimulation; IS >9 severe stimulation.
The results show that the composition of the invention has good safety and is suitable for sensitive parts such as the periphery of eyes.
Test 2 evaluation test of effectiveness
The effectiveness of the hair growth promoting formulations obtained in examples 1 to 6 and comparative examples 1 to 6 was evaluated.
75 volunteers were recruited, recording the number and length of upper/lower eyelashes, and randomly divided into 15 groups: placebo (pure water, negative control), 0.1 wt% bimatoprost (positive control), 2 wt% myristoyl hexapeptide-16 (single peptide group), control 1, example 1, control 2, example 2, control 3, example 3, control 4, example 4, control 5, example 5, control 6, example 6. Applying the composition 1 time every morning and evening, and continuously applying for 4 weeks and 8 weeks, and counting the number and length of eyelashes. The results are set forth in the following table:
the above results indicate that simple myristoyl hexapeptide-16 has some hair growth promoting effect, but the effect is slightly lower than that of bimatoprost. After synergizing with other functional peptides, it showed a beneficial effect comparable to bimatoprost (control 1). The compositions provided by the present invention, however, all showed significant gain effects due to the improved synergy and long-lasting delivery, which were higher than bimatoprost and its control.

Claims (8)
1. A hair growth promoting active peptide composition comprising a keratin gene stimulating peptide and a short synergistic peptide;
the keratin gene stimulating peptide is one or more of copper peptide, myristoyl pentapeptide-17, myristoyl hexapeptide-16 and biotin tripeptide-1;
the synergistic short peptide is one or more of leucine-proline-arginine, proline-leucine-arginine, isoleucine-proline-arginine, lysine-isoleucine-proline, proline-isoleucine-lysine and valine-glutamine-proline.
2. The active peptide composition of claim 1, wherein the mass ratio of the keratin gene-stimulating peptide to the short synergistic peptide is (5-20): (0.05-5).
3. The active peptide composition of claim 2, wherein the mass ratio of the keratin gene-stimulating peptide to the short synergistic peptide is (5-10): (0.1-2).
4. The active peptide composition of any one of claims 1 to 3, further comprising a signal peptide, wherein the signal peptide is one or more selected from the group consisting of myristoyl pentapeptide-11, palmitoyl tripeptide-1, palmitoyl tripeptide-5, palmitoyl pentapeptide-3, palmitoyl hexapeptide-6, palmitoyl hexapeptide-9, myristoyl hexapeptide-4, and myristoyl octapeptide-1.
5. The active peptide composition of any one of claims 1 to 3, further comprising a neurotransmitter-inhibiting peptoid, wherein the neurotransmitter-inhibiting peptoid is one or more of the dipeptides snake toxin, carnosine, acetyl hexapeptide-3, acetyl hexapeptide-8, acetyl octapeptide-1, pentapeptide-3, tripeptide-1, palmitoyl dipeptide-5 complex salt, palmitoyl tetrapeptide-7, tripeptide-4, and hexapeptide-10.
6. A hair growth promoting formulation comprising the active peptide composition of any one of claims 1 to 5, wherein 0.01 to 20 wt% of the active peptide composition and 1 to 20 wt% of the nano-stabilizer are contained;
wherein the nano stabilizer contains 1-20 wt% of emulsifier, 1-20 wt% of liquid grease, 0.1-20 wt% of polyalcohol and the balance of water.
7. The hair growth promoting formulation of claim 6, wherein the emulsifier is: PEG-40 hydrogenated castor oil, PEG-30 hydrogenated castor oil, PEG-20 hydrogenated castor oil, PEG-10 hydrogenated castor oil, PEG-5 hydrogenated castor oil, polysorbate 80, polysorbate 60, polysorbate 40, polysorbate 20, PEG-30 dipolyhydroxystearate, PEG-8 caprylic capric glyceride, PEG-6 caprylic/capric glyceride, polyglycerol-10 stearate, laureth-23, polyoxyethylene castor oil-20, polyoxyethylene castor oil-30, polyoxyethylene castor oil-35, polyoxyethylene castor oil-40, PEG-40 stearate, polyglycerol-10 myristate, polyglycerol-4 oleate, coco glucoside, ceteareth-20, ceteareth-25, one or more of polyoxyethylene castor oil-20 ether, polyoxyethylene castor oil-30 ether, polyoxyethylene castor oil-35 ether, polyoxyethylene castor oil-40 ether, polyethylene glycol 400 monoglyceride, polyethylene glycol 400 diglyceride, polyethylene glycol 600 monoglyceride, polyethylene glycol 600 diglyceride, coco glucoside, cetostearyl alcohol polyether-20, cetostearyl alcohol polyether-25, sodium bis (lauramide glutamine) lysine, squalane, buthanol polyether-26, egg yolk lecithin and soybean lecithin;
the liquid grease is as follows: one or more of isopropyl myristate, isopropyl palmitate, caprylic capric triglyceride, vitamin E, simethicone, caprylic capric macrogol glyceride, macrogol laurate, macrogol stearate, linolic acid glyceride, propylene glycol monocaprylate, propylene glycol dicaprylate, diethyl sebacate, isononyl isononanoate, triacetin, squalene and soybean oil;
the polyol is as follows: one or more of polyethylene glycol, propylene glycol, glycerol, 1, 3-butanediol, and 1, 2-pentanediol.
8. Use of the active peptide composition of any one of claims 1 to 5 for the preparation of a hair-stimulating effect-type cosmetic.
