技术领域
本发明属于祛痘产品领域,具体涉及一种含黄酮化合物与有机混合酸的美白修复祛痘水。
背景技术
痤疮属于慢性炎症性皮肤病,其发病机制相对复杂,发病因素较多,因此临床治疗有一定的难度。痤疮的治疗主要是减少皮脂分泌、溶解毛囊角栓、杀灭痤疮丙酸杆菌和减轻炎症。目前临床上一线治疗多采用局部或***应用抗生素和维A酸类制剂,但是痤疮丙酸杆菌对抗生素的耐药性日趋严重,而长期口服或外用维A酸类制剂由于存在对皮肤屏障功能的破坏、肝肾损坏及生育影响等严重不良反应,导致患者依从性较差,因此,如何采用安全、有效的方法治疗痤疮,是亟需解决的一个问题。
本发明利用黄酮化合物(黄芩苷、积雪草苷)的抗敏、抑菌消炎、美白祛斑作用与有机混合酸(水杨酸、甘草次酸、三甲基甘氨酸、左旋乳酸、左旋乳酸钠、杜鹃花酸)的抗菌、美白、修复作用复配,以有效治疗痤疮,并改善刺激性和毒性造成的皮肤过敏症状(红肿、过度脱皮),达到美白、修复的效果。
发明内容
本发明的目的在于提供一种含黄酮化合物与有机混合酸的美白修复祛痘水,其以左旋乳酸和左旋乳酸钠的复配体系作为缓冲剂,使含有水杨酸、杜鹃花酸的祛痘水的pH值可控制在5.5-6.5范围内,避免了现有产品酸性低对皮肤造成的刺激及带来的角质剥脱等损伤,且其制得的祛痘水可有效治疗痤疮,并达到美白、修复肌肤的效果。
可有效治疗痤疮,并改善皮肤过敏症状,达到美白、修复的效果。
为实现上述目的,本发明采用如下技术方案:
一种含黄酮化合物与有机混合酸的美白修复祛痘水,按重量百分数之和为100%计,其各原料成分的重量百分数为:水杨酸0.5-1.5%、甘草次酸0.2-1.0%、三甲基甘氨酸0.5-2.0%、左旋乳酸0.5-3.0%、左旋乳酸钠0.5-3.0%、杜鹃花酸1.0-5.0%、黄芩苷0.2-1.0%、积雪草苷0.2-1.0%、聚乙烯吡咯烷酮0.3-2.0%、75wt%酒精20.0-50.0%、薄荷脑0.2-0.5%、PEG-40氢化蓖麻油2.0-5.0%、余量为水。
优选地,一种含黄酮化合物与有机混合酸的美白修复祛痘水,其各原料成分按重量百分数计为:水杨酸1.0%、甘草次酸0.5%、三甲基甘氨酸1.0%、左旋乳酸1.0%、左旋乳酸钠1.8%、杜鹃花酸3.5%、黄芩苷0.5%、积雪草苷0.5%、聚乙烯吡咯烷酮0.8%、75wt%酒精35.0%、薄荷脑0.35%、PEG-40 氢化蓖麻油4.0%、水50.05%。
所述美白修复祛痘水的制备方法为:
1)将水杨酸、甘草次酸、杜鹃花酸、薄荷脑、PEG-40 氢化蓖麻油依次加入75wt%酒精中溶解,得澄清透明溶液A;
2)将三甲基甘氨酸、左旋乳酸、左旋乳酸钠、黄芩苷、积雪草苷、聚乙烯吡咯烷酮依次加入水中溶解,得澄清透明溶液B;
3)在搅拌条件下将溶液B缓慢加入溶液A中,混合均匀即得。
水杨酸是一种多功能天然活性成分,它存在于许多植物中,如柳树皮,属于非甾体类抗炎物质,具有抗炎、光谱抗菌能力,低浓度下双向调节角质细胞、高浓度具有松懈剥脱角质细胞能力以及美白的作用(抑制酪氨酸酶活性),故能使皮肤变得光滑细腻,并减少细纹,从而全面改善皮肤外观和质地。水杨酸不仅具有促进角质剥脱、溶解粉刺角栓的作用,还具有抗炎作用,对痤疮丙酸杆菌具有靶向作用,对痤疮后色素沉着亦有淡化作用。
杜鹃花酸又名壬二酸,对痤疮丙酸杆菌和表皮葡萄球菌具有抗菌作用,在皮肤表层具有显著的渗透性,渗透效率是二甲基亚砜的数倍,且毒性低,可作为渗透剂与多种活性物共用,有利于促进水杨酸、黄芩苷和积雪草苷等活性物在皮肤的渗透,且其可使角质形成细胞的粘连性减弱,使真皮中的肥大细胞脱颗粒、真皮浅层毛细血管扩张,从而达到保湿、加快皮肤新陈代谢、减少黑色素、减少皱纹等功效,因此,对于痤疮引起的炎症后色素沉着斑亦有改善作用。
左旋乳酸作为天然保湿因子存在于人体肌肤中,其相对分子量较小,可以有效渗入皮肤,软化表皮角质层,使角质层细胞间的黏着力降低,从而剥落老化死细胞,使角质层变薄,同时可促使表皮细胞的生长,有效去除痘印的细纹,润滑皮肤、增加肌肤弹性、改善皮肤。
甘草次酸具有抗炎和抗过敏作用,可用于治疗过敏性或职业性皮炎、皮下肉囊性炎症等。与左旋乳酸根复配具有美白肌肤淡化痘印色泽的作用。
三甲基甘氨酸又名甜菜碱,分子小,易吸收。保湿性约是丙三醇的12倍、山梨醇的3倍,具有很好的稳定性、水溶性和保湿性,可以迅速改善肌肤水分保持力,保持皮肤角质层充足水分,激发细胞活力,使皮肤柔软,具有保持肌肤滋润、光滑、降低刺激、防止皮肤干燥和发暗的效果,同时有利于化妆品中功效成分的吸收。
薄荷脑的添加可以促进药物透过皮肤,并起到调节油脂分泌、抑菌消毒、舒缓松弛神经、使肌肤清爽止痒、减轻浮肿等功效。
黄芩苷是从黄芩的干燥的根中提取并分离出来的一类黄酮类物质,黄芩苷具有甲硝唑式样原料特点,起植物激素作用,具有抗炎、抑菌、抗***等作用。黄芩苷在相同浓度下抑制痤疮丙酸杆菌的作用是甲硝唑的2倍,在抑制痤疮丙酸杆菌引起的皮肤发炎、囊肿、变红等炎症和过敏现象同时还具有显著的美白、祛斑和祛痘的疗肤功效,能有效抑制黑色素的形成、清除氧自由基,是一种很好的功能性美容化妆品原料。

本发明基于痤疮产生机理,采用水杨酸、甘草次酸、三甲基甘氨酸、左旋乳酸、左旋乳酸钠、杜鹃花酸、黄芩苷、积雪草苷、聚乙烯吡咯烷酮、薄荷脑为原料制备美白修复祛痘水的机制为:
1. 水杨酸、杜鹃花酸在pH>3的条件下易析出而影响使用效果,因此,含水杨酸、杜鹃花酸的产品的pH往往低于3,但这容易对皮肤产生刺激性,导致皮肤角质剥脱。本发明针对亚洲国家黄种人适宜表浅及轻度剥脱的特点,首次提出以左旋乳酸和左旋乳酸钠的复配体系作为缓冲剂,使含有水杨酸、杜鹃花酸的产品pH值控制在5.5-6.5之间,这与皮肤pH值相适应,大大降低了对皮肤的刺激性和角质剥脱过度带来的损伤;同时,通过将水杨酸、杜鹃花酸与甘草次酸、三甲基甘氨酸结合使用,以利用其所含基团的缔合作用,形成结构相对稳定的“大分子”有机酸,可进一步抑制水杨酸、杜鹃花酸的析出,使其稳定存在于溶液中。此外,“大分子”有机酸分子量大、结构大,可从根本上降低含有机酸的祛痘水对皮肤产生的红肿,刺痛、过敏等刺激性。
2. 利用黄芩苷具有甲硝唑式样、积雪草苷具有***的特点,与有机混合酸相互相成,软化表皮角质层,使角质层细胞间的黏着力降低,从而剥落老化死细胞,使角质层变薄,同时促使表皮细胞的生长,去除痘印的细纹,润滑皮肤、增加肌肤弹性、改善皮肤双向调节角质细胞、及美白的作用(抑制酪氨酸酶活性),故能使皮肤变得光滑细腻,并减少细纹,从而全面改善皮肤外观和质地。黄芩苷、积雪草苷的“植物激素”作用可大大提高所得祛痘水的祛痘效果,减少过敏、红肿等刺激性症状,增强对色素沉着、暗瘢的修复,达到美白、修复肌肤的效果。

实施例
一种美白修复祛痘水,其各原料成分按重量百分数计为:水杨酸1.0%、甘草次酸0.5%、三甲基甘氨酸1.0%、左旋乳酸1.0%、左旋乳酸钠1.8%、杜鹃花酸3.5%、黄芩苷0.5%、积雪草苷0.5%、聚乙烯吡咯烷酮0.8%、75wt%酒精35.0%、薄荷脑0.35%、PEG-40 氢化蓖麻油4.0%、水50.05%。
所述美白修复祛痘水的制备方法为:
1)将水杨酸、甘草次酸、杜鹃花酸、薄荷脑、PEG-40 氢化蓖麻油依次加入75wt%酒精中溶解,得澄清透明溶液A;
2)将三甲基甘氨酸、左旋乳酸、左旋乳酸钠、黄芩苷、积雪草苷、聚乙烯吡咯烷酮依次加入水中溶解,得澄清透明溶液B;
3)在搅拌条件下将溶液B缓慢加入溶液A中,混合均匀即得。
所得产品pH在5.5-6.5范围内,品质稳定。
抑菌试验
1. P.acnes的鉴定
将痤疮丙酸杆菌在脑心浸液肉汤琼脂培养基中活化48h后,在培养基上划线分离,挑出单个灰白色、圆形、不透明的细小菌落,涂于载玻片上,革兰氏染色镜检,在油镜100×100下见细菌呈紫色,棒状,直或微弯,排列呈X、Y、V及栅状,无荚膜,无鞭毛即为痤疮丙酸杆菌。
2. 菌液制备
厌氧平板传代细菌:将P.acnes菌株接种到厌氧脑心浸液肉汤,并置于厌氧袋中,迅速将厌氧产气袋打开放入厌氧袋,密封,在37℃培养箱中培养48h,振荡后按照麦氏比浊法调整菌液浓度为0.5麦氏比浊度(1×108cf/mL)备用。
3. 药液的制备
将水杨酸和杜鹃花酸分别用DMSO配制成浓度为10%的溶液,将黄芩苷用无菌水配制成浓度为10%的溶液,再将各溶液与实施例制备的祛痘水分别用0.22μm微孔滤膜过滤,避光,20℃保存。
4. 含药纸片的制备
用打孔器将定性滤纸制成直径6mm的圆纸片,放入洁净干燥的100mL锥形瓶中,瓶口用锡箔纸盖紧,再以单层牛皮纸包扎,121℃下高温灭菌30min,并在100℃烘箱中烘至干燥,密闭保存备用。在无菌条件下,取所需纸片至灭菌洁净平板中,每张纸用移液器给予制备好的药液10μL,待药液完全被纸片吸收后,备用。
5. 实验操作
在超净工作台中无菌操作,将灭菌后的脑心浸液肉汤琼脂培养基倒入平皿内,每个培养皿约20mL,待培养基凝固。同时用无菌棉球沾取浓度为0.5麦氏比浊度(1×108cf/mL)的菌液,并挤压去多余的菌液至无液滴流下。在已凝固平板上,从三个方向Z字划线(每次转60°),平均涂抹琼脂表面、圆周两圈,以使菌液均匀分布,制成含菌平板。室温2~5min微干后,用无菌镊子将含药纸片5片置于平板上(其中1片为含去离子水的空白对照)。将平板倒置放入培养箱中37℃培养48h后观察生长情况并测量、记录抑菌圈直径(mm,包含滤纸片直径)。
样品对痤疮丙酸杄菌菌抑制效果记载方法:5级:抑菌圈直径径大于20mm有明显抑菌效果。4级:抑菌圈直径16~20mm,有较强抑菌效果。3级:抑菌圈直径13~16mm,有抑菌效果。2级:抑菌圈直径10~13mm,抑菌效果不明显。1级:抑菌圈直径7~10mm,抑菌效果微弱。0级:小于7mm(滤纸片直径)。测量抑菌圈直径的方法是十字交叉测量2次,取平均值。

临床疗效
痤疮患者每日在彻底清洁面部皮肤后,将本发明所制备的祛痘水局部涂覆于患处,停留15~20min后用清水洗净,连续使用2-3天后,患者面部痤疮、痘印可明显消除,其有效率可达80%以上。
本发明以黄酮化合物搭配有机混合酸可以有效避免因水杨酸具有溶解油脂的能力而导致的皮肤出现不同程度的干燥和瘙痒等症状,且通过有机混合酸的协同作用,可减轻对皮肤的刺激性和水分流失,两者相辅相成,可有效护理肌肤,达到美白、修复的效果。
以上所述仅为本发明的较佳实施例,凡依本发明申请专利范围所做的均等变化与修饰,皆应属本发明的涵盖范围。

Claims (3)

1. 一种含黄酮化合物与有机混合酸的美白修复祛痘水,其特征在于:按重量百分数之和为100%计,各原料成分的重量百分数为:水杨酸0.5-1.5%、甘草次酸0.2-1.0%、三甲基甘氨酸0.5-2.0%、左旋乳酸0.5-3.0%、左旋乳酸钠0.5-3.0%、杜鹃花酸1.0-5.0%、黄芩苷0.2-1.0%、积雪草苷0.2-1.0%、聚乙烯吡咯烷酮0.3-2.0%、75wt%酒精20.0-50.0%、薄荷脑0.2-0.5%、PEG-40 氢化蓖麻油2.0-5.0%、余量为水。
2. 根据权利要求1所述的含黄酮化合物与有机混合酸的美白修复祛痘水,其特征在于:各原料成分按重量百分数计为:水杨酸1.0%、甘草次酸0.5%、三甲基甘氨酸1.0%、左旋乳酸1.0%、左旋乳酸钠1.8%、杜鹃花酸3.5%、黄芩苷0.5%、积雪草苷0.5%、聚乙烯吡咯烷酮0.8%、75wt%酒精35.0%、薄荷脑0.35%、PEG-40 氢化蓖麻油4.0%、水50.05%。
3.根据权利要求1所述的含黄酮化合物与有机混合酸的美白修复祛痘水,其特征在于:其制备方法为:
1)将水杨酸、甘草次酸、杜鹃花酸、薄荷脑、PEG-40 氢化蓖麻油依次加入75wt%酒精中溶解,得澄清透明溶液A;
2)将三甲基甘氨酸、左旋乳酸、左旋乳酸钠、黄芩苷、积雪草苷、聚乙烯吡咯烷酮依次加入水中溶解,得澄清透明溶液B;
3)在搅拌条件下将溶液B缓慢加入溶液A中,混合均匀即得。
