Abstract
The invention provides a flocculant production agent, which belongs to the technical field of aquaculture and comprises the following components in parts by weight: 70-90 parts of carbon source, 5-15 parts of plant extract, 0.05-0.15 part of microbial agent, 0.03-0.08 part of baking soda, 0.03-0.08 part of calcium superphosphate, 0.05-0.15 part of vitamin C phosphate, 1-3 parts of amino acid fertilizer, 1-3 parts of mannan oligosaccharide and 5-6 parts of montmorillonite. The flocculant provided by the invention can quickly and continuously generate biological floccules to reduce ammonia nitrogen, nitrite nitrogen and sulfide in water; meanwhile, digestion and absorption can be promoted, and the generated biological floccules can be used as baits to reduce the bait coefficient; can inhibit the growth of harmful bacteria, promote the reproduction of beneficial bacteria, enhance the immunity of fish bodies and reduce the occurrence of fish diseases.

Technical Field

Background
With the intensive high-density aquaculture in China, a large amount of waste, residual bait, animal and plant residues and the like are continuously accumulated in aquaculture water bodies and the bottom of a pond, so that the water quality is deteriorated, the structure of an ecological system is damaged, the biodiversity is damaged, and the fish and the shrimp are restricted in ingestion and growth, even are ill, poisoned or dead. The toxicity of the sulfide mainly refers to the toxicity of hydrogen sulfide, and the hydrogen sulfide is a soluble gas with the odor of a smelly egg and is a highly toxic substance harmful to aquatic animals. The sulfide can enter tissues and blood of fishes, shrimps and crabs through permeation and absorption, the structure of heme is destroyed, and the hemoglobin loses the capability of combining oxygen molecules; meanwhile, the fish mask has strong stimulation and corrosion effects on the skin and mucous membrane of fish, so that tissue generates coagulation necrosis to cause dyspnea and even death.

The flocculant provided by the invention comprises 5-15 parts by weight of plant extract, preferably 10 parts by weight. In the present invention, the plant extracts include yucca extract, matrine, stemona extract, ginkgo biloba extract and echinacea extract; the mass ratio of the yucca extract, the matrine, the stemona root extract, the ginkgo leaf extract and the echinacea purpurea extract is preferably (2-3): (0.1-1): (0.5-1).
In the invention, the yucca extract contains saponin, polysaccharide, yucca phenol and resveratrol, has strong adsorption capacity on harmful gases, can adsorb harmful gases generated by baits, putrefying bodies and the like in water, and also has the functions of adjusting intestinal microenvironment, enhancing resistance, promoting ingestion and nutrient absorption, reducing the discharge amount of nitrogen and phosphorus in feces of fish bodies, improving the environment of aquaculture water and the like. The source of the yucca extract is not particularly limited, but in the present invention, the yucca extract is preferably a product of Xian Lutian Biotechnology Co., Ltd, the product is LT02-80, and the yucca saponin content is 80%.
In the invention, the matrine has antibacterial and antiviral effects, can effectively inhibit the propagation and growth of harmful bacteria, can resist pathogenic fungi when being used with radix stemonae, and reduces the occurrence of fish diseases. The source of the matrine is not specially limited, the matrine can be obtained by adopting conventional commercial products, the matrine is preferably a product of Xian Lvtian Biotechnology Limited company in the specific implementation mode of the invention, and the content of the matrine is more than or equal to 98 percent.
In the invention, the stemona extract contains stemonine, has an inhibitory effect on partial pathogenic bacteria and fungi, and has an antiprotozoal antifungal effect when being combined with matrine. The source of the stemona sessilifolia extract is not particularly limited, and the stemona sessilifolia extract can be prepared from conventional commercially available products, and in a specific embodiment of the invention, the stemona sessilifolia extract is preferably a product of Sanyuan Tianyu biological products, Inc., and has the following product specifications: 10:1 (%).
In the invention, the ginkgo leaf extract contains flavonoids, lactones, phenols and the like, has multiple functions of resisting oxidation and inflammation, reducing the disease incidence of animals, improving immunity, enhancing disease resistance and the like, and can absorb and decompose hydrogen sulfide. The source of the ginkgo biloba extract is not specially limited, and the ginkgo biloba extract is preferably a product of the Sierra-Relin Biotechnology Co., Ltd, and the product specification is as follows: the total ginkgo flavone content is more than or equal to 24.0 percent and the ginkgolide content is 6 percent.
In the invention, the echinacea purpurea extract contains echinacea purpurea polyphenol, and the echinacea purpurea extract obviously improves and maintains the autonomous immunity and has the functions of antibiosis and antivirus. The source of the echinacea purpurea extract is not particularly limited in the invention, and the echinacea purpurea extract can be obtained by adopting conventional commercial products, and in the specific embodiment of the invention, the echinacea purpurea extract is preferably purchased from the company of Xian Lythtian Biotechnology limited, and the product specification is as follows: the content of echinacea polyphenol is 4%.
The flocculant comprises 0.05-0.15 part by weight of a microbial preparation, preferably 0.1 part by weight of a microbial preparation, and the microbial preparation preferably comprises bacillus subtilis, bacillus amyloliquefaciens, EM bacteria, bifidobacterium longum, clostridium butyricum and lactobacillus acidophilus. The invention has no special limitation on the strain source of the microorganism, and the conventional strain is adopted. In the invention, the superphosphate supplements P, Ca microelements in the water body, adjusts the N/P ratio in the water body, stabilizes the water quality and cultures excellent algae phases.
The flocculant provided by the invention comprises 0.05-0.15 part by weight of vitamin C phosphate, preferably 0.1 part by weight. In the invention, the vitamin C phosphate is beneficial to the absorption and utilization of biological floccules, and can improve the survival rate and the weight gain rate; reducing stress reaction of fish, etc.
The flocculant provided by the invention comprises 1-3 parts of amino acid fertilizer by weight, and the commercially available fisherman amino acid fertilizer water paste contains 18 free amino acids, active peptides, organic factors, multiple vitamins, mineral substances, monadian growth promoting elements and the like, so that nutrient substances are provided for algae, fishes and the like, and the immunity of the fishes is improved; promoting the quick propagation of beneficial algae phases, zooplankton and the like and maintaining the ecological balance of the water body.
The flocculant comprises 1-3 parts by weight of mannan oligosaccharide, preferably 2 parts by weight of mannan oligosaccharide, and in the flocculant, the mannan oligosaccharide can improve the micro-ecological environment of intestinal tracts of fishes, promote the growth and reproduction of beneficial bacteria in the intestinal tracts, inhibit the adhesion and colonization of harmful bacteria to intestinal walls and enhance the nonspecific immunity of the fishes; improving the activity of endogenous digestive enzyme, promoting the digestion and absorption of fish to nutrient substances, and improving the utilization rate of bait.
The flocculant comprises 5-6 parts by weight of montmorillonite, preferably 5.7 parts by weight of montmorillonite, and the montmorillonite has the adsorption capacity and can promote the formation of biological flocs.
In the present invention, the method for using the flocculant preferably comprises: mixing the carbon source with aerated water, mixing with a microbial preparation, an amino acid fertilizer, vitamin C phosphate and mannan oligosaccharide, and performing closed fermentation for 2-3 d to obtain a fermentation product; mixing baking soda, calcium superphosphate, plant extract and aerated water to obtain mixture; the mixture and the leavening are mixed and then are sprinkled in the water body for raising aquatic animals, and the dissolved oxygen in the water body is ensured to be more than 4 mg/L. In the invention, water is aerated for 2-3 days to obtain aerated water. In the invention, the splashing should be performed on a sunny day. In the invention, the carbon source, the baking soda and other substances are mixed with water, and the required water amount is not particularly limited and can be completely dissolved.
In the invention, the addition amount of the flocculant is preferably 7-15 kg/667m3。

The flocculant-producing component comprises: 35 parts of glucose, 35 parts of red cane sugar, 8 parts of yucca extract, 0.5 part of matrine, 0.5 part of stemona root extract, 3 parts of ginkgo leaf extract, 3 parts of echinacea extract, 0.04 part of bacillus subtilis, 0.04 part of bacillus amyloliquefaciens, 0.02 part of EM bacteria, 0.03 part of bifidobacterium longum, 0.01 part of clostridium butyricum, 0.01 part of lactobacillus acidophilus, 0.05 part of baking soda, 0.05 part of calcium superphosphate, 0.1 part of vitamin C phosphate, 3 parts of amino acid fertilizer, 3 parts of mannooligosaccharides and 6 parts of montmorillonite.
Comparative example 1
The flocculant-producing component comprises: 40 parts of glucose and 40 parts of red cane sugar.
Example 4
The bighead carps used in the experiment are domesticated, good in food intake in an indoor aquarium with sufficient illumination, and strong and orderly-sized fingerlings are selected for the experiment. The feeding experiment period is 8 weeks, the experiment is divided into 5 groups, which are respectively a control group, an experiment 1 group, an experiment 2 group, an experiment 3 group and an experiment4 groups, each group is provided with 3 repetitions, and 150 fish tails are placed. The control group was fed with no other substances than the normal feed. Test 1, 2, 3 and 4 groups were fed with the flocculant prepared in example 1, 2, 3 and 1 in addition to the normal feed, and the amount of flocculant added was 8kg/667m3. Tests 1, 2 and 3, mixing the carbon source with aerated water, mixing with a microbial preparation, an amino acid fertilizer, vitamin C phosphate and mannan oligosaccharide, and performing closed fermentation for 2-3 days to obtain a fermentation product; mixing baking soda, calcium superphosphate, plant extract and aerated water to obtain mixture; after the mixture and the fermentation product are mixed, uniformly splashing is carried out in the morning on a sunny day; test 4 groups selected the flocculant produced in comparative example 1 to be sufficiently dissolved in aerated water and uniformly sprinkled in the morning of a fine day; the oxygen increasing equipment is required to be started simultaneously during each group of tests to ensure that the dissolved oxygen is not less than 4mg/L all day.
During the test period, all groups are fed normally, water is not changed, and the lost water is supplemented by aerated and temperature-regulated tap water at regular intervals. The results are shown in tables 1 and 2.

As can be seen from tables 1 and 2, the ammonia nitrogen, nitrite nitrogen and sulfide in the water body of the test group are greatly reduced compared with the control group through determination during the test period. When the ammonia nitrogen is added for 4 days, the ammonia nitrogen content in the groups 1, 2 and 3 has no significant difference, and the reduction degree of the group 1 is the largest, and the concentration is reduced by 81.9 percent compared with the concentration at the beginning of the test; at 6 days, the nitrite nitrogen in the test 1, 2 and 3 groups is respectively reduced by 68.4%, 47.4% and 45.0%; the sulfide contents of the 1, 2 and 3 groups of experiments have no significant difference, and are respectively reduced by 42.9%, 31.3% and 25.0%; the bait coefficients of the groups 1, 2 and 3 are not obviously different, and the reduction degree of the group 1 is the maximum and is reduced by 0.27; the values of serum lysozyme and superoxide dismutase in the fish serum of the groups 1, 2 and 3 are larger than those of the control group and the group 4, and the groups 1, 2 and 3 have no morbidity and no mortality, so that the invention can effectively reduce the morbidity and enhance the immunity.
Example 5
The flocculant-producing component comprises: 38 parts of glucose, 48 parts of red cane sugar, 4 parts of yucca extract, 0.2 part of matrine, 0.2 part of stemona root extract, 1.25 parts of ginkgo leaf extract, 1.25 parts of echinacea extract, 0.01 part of bacillus subtilis, 0.01 part of bacillus amyloliquefaciens, 0.01 part of EM bacteria, 0.01 part of bifidobacterium longum, 0.005 part of clostridium butyricum, 0.005 part of lactobacillus acidophilus, 0.05 part of baking soda, 0.05 part of calcium superphosphate, 0.05 part of vitamin C phosphate, 1 part of amino acid fertilizer, 1 part of mannooligosaccharides and 5 parts of montmorillonite.
Example 6
The flocculant-producing component comprises: 40 parts of glucose, 50 parts of red cane sugar, 4 parts of yucca extract, 0.1 part of matrine, 0.1 part of stemona root extract, 0.5 part of ginkgo leaf extract, 0.5 part of echinacea extract, 0.01 part of bacillus subtilis, 0.01 part of bacillus amyloliquefaciens, 0.01 part of EM bacteria, 0.01 part of bifidobacterium longum, 0.005 part of clostridium butyricum, 0.005 part of lactobacillus acidophilus, 0.08 part of baking soda, 0.08 part of calcium superphosphate, 0.15 part of vitamin C phosphate, 1 part of amino acid fertilizer, 1 part of mannooligosaccharides and 6 parts of montmorillonite.
Comparative example 2
The flocculant-producing component comprises: 30 parts of glucose, 30 parts of red cane sugar, 4 parts of yucca extract, 2 parts of matrine, 2 parts of stemona root extract, 1 part of ginkgo leaf extract, 1 part of echinacea extract, 0.04 part of bacillus subtilis, 0.02 part of bacillus amyloliquefaciens, 0.01 part of EM bacteria, 0.02 part of bifidobacterium longum, 0.005 part of clostridium butyricum, 0.005 part of lactobacillus acidophilus, 0.05 part of baking soda, 0.05 part of calcium superphosphate, 0.1 part of vitamin C phosphate, 2 parts of amino acid fertilizer, 2 parts of mannooligosaccharides and 5.7 parts of montmorillonite.
Example 8
The yellow river carps used in the experiment are domesticated, good food intake is realized in an indoor aquarium with sufficient illumination, and fingerlings with strong physique and uniform specification are selected for the experiment. The feeding experiment period is 8 weeks, the experiment is divided into a control group, an experiment 5 group, an experiment 6 group and an experiment 7 group, each group is provided with 3 replicates, and 150 fish tails are placed in each group. The control group was fed with no other substances than the normal feed. In addition to normal feed feeding, the flocculant produced in example 5, example 6 and comparative example 2 were added to the test 5, test 6 and test 7, respectively, in an amount of 8kg/667m3. Mixing the carbon source with aerated water, mixing with a microbial preparation, an amino acid fertilizer, vitamin C phosphate and mannan oligosaccharide, and performing closed fermentation for 2-3 d to obtain a fermentation product; mixing baking soda, calcium superphosphate, plant extract and aerated water to obtain mixture; and after the mixture and the fermented product are mixed, uniformly splashing the mixture in the morning on a sunny day. Meanwhile, the oxygen increasing equipment is required to be started to ensure that the dissolved oxygen is not less than 4mg/L all day.
During the test period, the feeding is normally carried out, water is not changed, and the lost water is supplemented by tap water subjected to aeration and temperature regulation at regular intervals. The results are shown in tables 3 and 4.

As can be seen from tables 3 and 4, the ammonia nitrogen, nitrite nitrogen and sulfide in the water body of the test group are greatly reduced compared with the control group through determination during the test period, the beneficial algae species in the water body are rich, and the bait coefficients of the test groups 5 and 6 are not different and are reduced by 0.16; four days after the addition, the ammonia nitrogen ratio in the water body is respectively reduced by 80.4 percent and 73.9 percent compared with the ammonia nitrogen ratio at the beginning of the test, the sulfide is respectively reduced by 33.6 percent and 30.4 percent, and the nitrite nitrogen is respectively reduced by 61.2 percent and 51.0 percent in 6 days; the fish in the 5 and 6 groups have no morbidity and mortality, and the values of the lysozyme and the superoxide dismutase in the fish serum are larger than those in the control group and the 7 groups, so that the invention can effectively reduce the morbidity and enhance the immunity.
The embodiment can show that the flocculant provided by the invention can quickly and continuously generate biological floccules to reduce ammonia nitrogen, nitrite nitrogen, hydrogen sulfide and the like in the water body; meanwhile, digestion and absorption can be promoted, and the generated biological floccules can be used as baits, so that the bait coefficient is reduced; can inhibit growth of putrefying bacteria, promote reproduction of beneficial bacteria, enhance fish immunity, and reduce fish diseases. And no culture sewage is discharged in the culture process, so that water resources are saved, the pollution to the surrounding environment is avoided, and the economic, ecological and social benefits are obvious.
The foregoing is only a preferred embodiment of the present invention, and it should be noted that, for those skilled in the art, various modifications and decorations can be made without departing from the principle of the present invention, and these modifications and decorations should also be regarded as the protection scope of the present invention.
Claims (10)
1. The flocculant is characterized by comprising the following components in parts by weight: 70-90 parts of carbon source, 5-15 parts of plant extract, 0.05-0.15 part of microbial agent, 0.03-0.08 part of baking soda, 0.03-0.08 part of calcium superphosphate, 0.05-0.15 part of vitamin C phosphate, 1-3 parts of amino acid fertilizer, 1-3 parts of mannan oligosaccharide and 5-6 parts of montmorillonite;
the carbon source is glucose and red sucrose;
the plant extracts are yucca extract, matrine, stemona extract, ginkgo biloba extract and echinacea extract;
the yucca extract contains saponin, polysaccharide, yucca phenol and resveratrol;
the stemona root extract contains stemonine;
the folium Ginkgo extract contains flavonoids, lactones and phenols;
the echinacea purpurea extract contains echinacea purpurea polyphenol;
the microbial preparation is bacillus subtilis, bacillus amyloliquefaciens, EM bacteria, bifidobacterium longum, clostridium butyricum and lactobacillus acidophilus.
2. The flocculant according to claim 1, which is composed of the following components in parts by weight: 80 parts of carbon source, 10 parts of plant extract, 0.1 part of microbial agent, 0.05 part of baking soda, 0.05 part of calcium superphosphate, 0.1 part of vitamin C phosphate, 2 parts of amino acid fertilizer, 2 parts of mannan oligosaccharide and 5.7 parts of montmorillonite.
3. The flocculant of claim 1, wherein the mass ratio of glucose to red sucrose is (0.5-1.5): (0.5-1.5).
4. The flocculant of claim 1, wherein the weight ratio of yucca extract, matrine, stemona root extract, ginkgo biloba extract and echinacea extract is (2-3): 0.1-1: (0.5-1): 0.5-1.
5. The flocculant of claim 1, wherein the mass ratio of Bacillus subtilis, Bacillus amyloliquefaciens, EM bacteria, Bifidobacterium longum, Clostridium butyricum and Lactobacillus acidophilus is (0.02-0.04): (0.01-0.02): (0.02-0.03): (0.005-0.01).
6. The flocculant of claim 1, wherein the mass ratio of glucose, sucrose red, yucca extract, matrine, stemona root extract, ginkgo biloba extract, echinacea extract, bacillus subtilis, bacillus amyloliquefaciens, EM bacteria, bifidobacterium longum, clostridium butyricum and lactobacillus acidophilus is 40:40:4:2:2:1:1:0.04:0.02:0.01:0.02:0.005: 0.005.
7. The flocculant for producing flocculant according to claim 1, 5 or 6, wherein the effective viable count of Bacillus subtilis is (80-100) hundred million cfu/g, and the effective viable count of Bacillus amyloliquefaciens is (100-200) hundred million cfu/g.
8. The flocculant according to claim 1, 5 or 6, wherein the effective viable count of the EM is (200-300) hundred million cfu/g, and the effective viable count of the Bifidobacterium longum is (80-100) hundred million cfu/g.
9. The flocculant of claim 1, 5 or 6, wherein the Clostridium butyricum has an effective viable count of (50-100) hundred million cfu/g.
10. The flocculant according to claim 1, 5 or 6, wherein the effective viable count of the lactobacillus acidophilus is (80-100) hundred million cfu/g.
