本发明涉及医药,特别是一种治疗黑眼圈的归参花精油。
背景技术
黑眼圈是由于熬夜,情绪波动大,眼疲劳、衰老导致眼部皮肤血管血流速度过于缓慢形成滞流;组织供氧不足,血管中代谢废物积累过多,造成眼部色素沉着。年纪愈大的人,眼睛周围的皮下脂肪变得愈薄,所以黑眼圈就更明显。
中医认为,黑眼圈患者多因肝阴不足、虚火上炎所致。中医强调“五脏六腑之精气,皆注于目”,而肝开窍于目,若长期熬夜,压力偏大,或是睡眼不足等,都会造成中医所说的肝阴不足、虚火上炎,也就是气血循环差兼有上火症状,出现黑眼圈。治疗黑眼圈,中医可以依据辨证及观察上下眼睑状态、颜色,调节失调的脏腑功能和改善局部血液循环,可改善眼周的颜色黯淡。现虽有治疗黑眼圈的多种药物,但由于种种原因,其使用和疗效并不尽人意,因此,治疗黑眼圈药物上的创新势在必行。

方中人参外用可营养、光滑肌肤、防止皮肤松弛、减少皱纹、防紫外线等,同时人参活性物质还可抑制黑色素形成, 使皮肤洁白光滑。桃仁活血祛瘀;当归补血、活血止痛,丹皮清热凉血,活血化瘀,玫瑰花行气解郁、和血止痛,红花活血通经、散瘀止痛,丹参祛瘀止痛、活血清心;柠檬可去除老死细胞,明亮肤色,紧实微血管,促进胶原蛋白产生,淡化黑色素,美白眼周皮肤;白芷散风除湿、通窍止痛、消肿排脓,色白, 白色入肺, 而肺主皮毛, 皮肤上的问题, 多从肺经入手;甘草调和药性,冰片清香宣散,具有开窍醒神,清热散毒,明目退翳的功效。全方诸药外用于眼周,可活血化瘀、明目退翳,改善循环、促进色素代谢、洁白皮肤,从而达到消除黑眼圈的作用。

本发明在具体实施中,一种治疗黑眼圈的归参花精油,由以下组分:人参精油5.5mL、当归精油5.5mL、柠檬精油6.5mL、丹皮精油5.5mL、丹参精油6.5mL、白芷精油4.5mL、玫瑰花精油6.5mL、红花精油4.5mL、甘草精油5.5mL、桃仁精油5.5mL和冰片5.5g溶解、混匀制成,制备方法同实施例1。
本发明配伍科学合理,组分互相支持,具有促进眼周局部血液循环,减轻色素沉着功能,有效用于治疗黑眼圈,并经实验效果非常好,有关资料如下:
一、动物实验:
1实验材料
1.1药物试剂
2-氯苯丙氨酸,上海源叶生物科技有限公司,批号S42561;肾上腺素盐酸注射液,远大医药(中国)有限公司,批号,180406;本发明实施例1产品。小鼠TYR ELISA检测试剂盒,苏州卡尔文生物科技有限公司
1.2 实验仪器
散斑全帧实时扫描成像仪,PERIMEDAB,型号,*/pericam psi system;Cutometer®dual MPA580皮肤弹性测试仪,德国Coutage+Khazaka公司;JM 1000电子秤,余姚记铭称重校验设备有限公司;AL204电子天平,梅特勒-托利多仪器(上海)有限公司;批号:JB1T5374;高速冷冻离心机,湖南湘仪实验室仪器开发公司;批号:H2050R。
1.3 实验动物
实验动物名称:KM小鼠,18~21g,雌性 等级: SPF;提供单位:济南朋悦实验动物繁育有限公司 合格证编号:NO. 37009200017330;提供单位许可证编号:SCXY(鲁)20140007 实验室合格证:SYXK(豫)2015-0005。
2实验方法
取KM雌性小鼠60只,禁食12小时后,腹腔注射水合氯醛麻醉小鼠,使用脱毛膏除去小鼠眼部周围毛发,皮肤弹性仪测量小鼠眼睑部黑色素值,同一部位测量3次,取平均值,排除黑色素值过大的动物,将选出的40只小鼠根据黑色素值分为4组,分别为空白组、模型组、归参花精油大剂量组、归参花精油小剂量组。
除空白组外,其余组小鼠皮下注射肾上腺素盐酸注射液(0.8ml/Kg),随后腹腔注射2-氯苯丙氨酸混悬液450mg/Kg,注射肾上腺素2h后将小鼠放入0℃冷水中游泳,至其游不动时立即将小鼠取出,擦干毛发,并将其置于暖气旁,防止冻僵。小鼠活动正常后,用移液枪吸取精油涂抹于小鼠下眼睑处,剂量分别为归参花精油大剂量组10ul,归参花精油小剂量组5ul。连续造模、给药7天。第7天涂抹精油后用散斑全帧实时扫描成像仪测小鼠下眼睑血流灌注总量,用皮肤弹性仪测量小鼠眼睑部黑色素值。随后称重,摘左眼球取血,离心取血清,检测血清中酪氨酸酶(TYR)水平。取右侧下眼睑皮肤,用10%甲醛液固定作病理组织切片,HE染色,通过光镜视野下观察每5个高倍视野下黑色素颗粒细胞总数、皮肤含黑色素毛囊百分比。
3统计学处理
数据分析用SPSS 21.0统计软件包进行数据资料的统计学处理,计量资料用平均数±标准差(±s)表示,各组间比较采用单因素方差分析,方差检验齐者用最小显著差数(LSD)法,方差不齐者用Games-Howell法检验,等级资料用Radit检验。
4 实验结果
4.1 对小鼠黑眼圈模型局部黑色素含量的影响

由表1可知,与空白组相比,模型组出现明显的黑眼圈现象,小鼠眼睑处黑色素值显著升高(P<0.01),说明黑眼圈模型建立成功。与模型组相比,归参花精油大、小剂量组小鼠眼睑部黑色素值明显降低(P<0.05)。
4.2 各组动物不同处理后眼睑微循环总灌注量的变化

眼部周围血管较多,熬夜使眼睑得不到休息,血管持续紧张收缩,血流量长时间增加,引起眼周皮下组织血管淤血和水肿,滞留***影。由表2可知,与空白组相比,模型组眼睑部微循环总灌注量明显升高(P<0.05);与模型组相比,归参花精油大、小剂量可显著降低小鼠眼睑部的血流灌注量(P<0.01)。

由表4可知,与空白组相比,模型组小鼠血清中酪氨酸酶水平显著升高(P<0.01),与模型组相比,归参花精油小剂量组可使血清中酪氨酸酶水平显著降低(P<0.01),归参花精油大剂量组可使血清中酪氨酸酶水平明显降低(P<0.05)。
二、临床应用
在动物实验确保安全的情况下,经临床试用,效果非常好,有关资料如下(以实施例1为例):

应用归参花精油治疗黑眼圈15例(眼周出现明显黑色沉着,有明确的诱因为长期熬夜,压力偏大或是睡眼不足等)。
治疗方案:将归参花精油涂于眼周黑色部位,闭眼休息20-30分钟。每次用量0.2mL/cm2。连用7天。用药期间,按时休息,清淡饮食。
疗效判定标准:显效:黑眼圈黑色消退≧70%;有效:黑眼圈黑色消退≧40%;无效:黑眼圈黑色消退<40%。
统计学处理:15例中,显效8例,有效6例,无效1例,总有效率93.3%,使用效果非常好。
在对实施例1做上述实验的同时,还对实施例2、3也做了同样的实验,均取得了相同和相近似的结果,表明产品质量好,疗效稳定可靠,这里不再一一列举。
以上实验表明,本发明具有促进眼周局部血液循环,减轻色素沉着功能,有效用于治疗黑眼圈,制备方法简单,原料丰富,易生产制备,疗效确切,是治疗黑眼圈药物上的创新,有显著的经济和社会效益。

1.一种治疗黑眼圈的归参花精油,其特征在于,由以下组分:人参精油4-6mL、当归精油5-7mL、柠檬精油5-7mL、丹皮精油5-7mL、丹参精油5-7mL、白芷精油4-6mL、玫瑰花精油5-7mL、红花精油4-6mL、甘草精油4-6mL、桃仁精油5-7mL和冰片4-6g溶解、混匀制成;其中:

3.根据权利要求1所述的治疗黑眼圈的归参花精油,其特征在于,由以下组分:人参精油4.5mL、当归精油6.5mL、柠檬精油5.5mL、丹皮精油6.5mL、丹参精油5.5mL、白芷精油5.5mL、玫瑰花精油5.5mL、红花精油5.5mL、甘草精油4.5mL、桃仁精油6.5mL和冰片4.5g溶解、混匀制成。
4.根据权利要求1所述的治疗黑眼圈的归参花精油,其特征在于,由以下组分:人参精油5.5mL、当归精油5.5mL、柠檬精油6.5mL、丹皮精油5.5mL、丹参精油6.5mL、白芷精油4.5mL、玫瑰花精油6.5mL、红花精油4.5mL、甘草精油5.5mL、桃仁精油5.5mL和冰片5.5g溶解、混匀制成。
