Abstract

本发明涉及从小蓟炭中提取分离纯化出的量子点(纳米颗粒/纳米粒子)的方法,该方法包括:小蓟炭,经溶剂萃取或者提取,再用分子量大于1000Da的透析袋透析,或者超滤,得到小蓟炭量子点,该量子点在紫外灯365nm下呈蓝色荧光、透射电镜测量粒径<100nm,主要由C、H、O和N等元素构成;本发明还涉及此量子点在医药领域的应用,通过注射、口服或外用等途径给药后产生的止血、镇痛、镇静、抗炎、抗过敏、抗氧化、解热、解痉、降糖、降压、对抗肿瘤化疗药物的副反应等。

小蓟炭量子点(纳米颗粒/纳米粒子)的制备及其应用
技术领域
本发明属于医药技术领域,特别涉及小蓟炭量子点的制备及在医药领域的应用。
背景技术
小蓟炭为小蓟药材制成碳化物。早在医书上就记载“红见黑则止”。小蓟性味甘、苦,凉。归经归心、肝经。多用于凉血,祛瘀,止血。治吐血,衄血,尿血,血淋,便血,血崩,急性传染性肝炎,创伤出血,疔疮,痈毒。制备方法主要有以下:1、取净小蓟段放锅内,炒至棕黑色,注意存性,喷洒少许清水,取出晾干。
2、高温煅烧成炭存性
小蓟主要成分含蒙花苷,芸香苷,原儿茶酸,绿原酸(chlorogenic acid),咖啡酸,氯化钾。
发明内容
本发明首次从小蓟炭中提取分离出量子点(纳米颗粒/纳米粒子),并首次发现了小蓟炭炭量子点具有很强的止血效应、镇静、镇痛、抗炎等生物效应。
技术方案:本发明提供了从小蓟炭中提取分离出炭量子点的方法。本发明还提供了小蓟炭炭量子点的生物活性及其在医药领域中的应用。
所述制备方法,具体包括以下步骤:煅法制备小蓟炭,用溶剂从小蓟炭中提取出量子点,进一步分离纯化包括透析、超滤、层析等技术方法,得到高纯度的碳量子点。

具体实施方式
实施例1:小蓟炭碳量子点的制备
称取小蓟原药,置坩埚内,加盖,置马弗炉中,升温至250℃,保持恒温1h,待自然降温至室温,打开马弗炉取出,即为小蓟炭。
用纯水提取小蓟炭2次,合并水溶液,浓缩,用1KD透析袋透析,透析袋内的部分即为高纯度小蓟炭炭量子点的溶液。
实施例2:小蓟炭碳量子点的制备
称取小蓟原药,置坩埚内,加盖,置马弗炉中,升温至350℃,保持恒温1h,待自然降温至室温,打开马弗炉取出,即为小蓟炭。
用纯水提取小蓟炭2次,合并水溶液,浓缩,用1KD透析袋透析,透析袋内的部分即为高纯度小蓟炭炭量子点的溶液。
实施例3:小蓟炭碳量子点的制备
称取小蓟原药,置坩埚内,加盖,置马弗炉中,升温至450℃,保持恒温1h,待自然降温至室温,打开马弗炉取出,即为小蓟炭。
用纯水提取小蓟炭2次,合并水溶液,浓缩,用1KD透析袋透析,透析袋内的部分即为高纯度小蓟炭炭量子点的溶液。
实施例4小蓟炭碳量子点的止血作用
小鼠尾尖止血:
1.实验的前一晚禁食12小时,正常给水。实验当天先将老鼠称重,分为为生理盐水组;阳性药组;高剂量组;中剂量组;低剂量组。
2.将标号的小鼠分装好,待用。实验前两小时分别给药,以充分吸收药物。实验前半小时给麻醉。尾尖止血实验采用剪尾法,首先麻醉好的小鼠的尾长测量察并记录小鼠尾尖的出血时间并做好记录。
3.结果见图3,中低剂量与正常组相比均具有明显的断尾止血作用,小鼠肝脏止血:
1.实验的前一晚禁食12小时,正常给水。实验当天先将老鼠称重标号。
2.实验前两小时给药,实验前半小时麻醉,待小鼠麻醉后,将小鼠四肢固定,腹部充分的暴露,沿腹中线将小鼠腹部破开,肝脏充分的暴露,避免伤及胸腔。用生理盐水将肝脏冲洗干净,并用纱布将水吸干。将肝脏底部铺上纱布以固定,然后用2ml的针头扎肝脏的左叶距边缘约0.5cm,进针0.3cm,观察并记录出血时间。
3.结果见图3,高中低三个剂量与正常组相比均具有明显的肝脏止血作用,尤其是高剂量组比蛇凝血酶对照组的止血效果更好。

Claims (2)

1.小蓟炭碳量子点在制备止血药物中的应用,小蓟炭碳量子点的制备方法为:将小蓟置坩埚内,加盖,置马弗炉中,升温至250℃或350℃或450℃,保持恒温1h,再自然降温至室温,取出即得小蓟炭;小蓟炭用纯水提取后,再用分子量为1KDa-10KDa的透析袋透析,收集透析袋内截留的部分;或者用10KD的超滤管超滤,收集滤液部分即得小蓟炭碳量子点;所得碳量子点的特征:在紫外灯365nm下有荧光、透射电镜测量粒径为1-20nm,主要由C、O和N元素构成。
2.根据权利要求1所述的应用,其剂型为注射剂、口服液体制剂、硬胶囊、软胶囊、片剂、栓剂、水丸、蜜丸、浓缩丸、膏剂、散剂、乳剂、糊剂、涂抹剂。
