背景技术

细胞内的pH值在许多生理过程中发挥着重要作用,例如细胞增殖、酶活性调控和离子运输等。高尔基体是蛋白质分泌、运输和装配的主要场所,大量的蛋白糖基化修饰都发生在高尔基体中。作为一个弱酸性细胞器(pH 6.0-6.7),高尔基体主要通过质子泵或离子通道进行质子交换。异常的高尔基体pH会导致细胞功能障碍、糖基化紊乱,严重时会导致高尔基体结构去组织化,进而解体。因此发展一种高效靶标高尔基体、原位、实时定量检测高尔基体pH的检测手段是非常必要的。
荧光成像技术具有操作简单、无损原位成像等优点,近年来多被用于疾病的诊断与检测。其中,近红外荧光成像技术具有较低背景荧光及较低光毒性等优势,被广泛应用于细胞成像研究。光声成像技术在生物医药领域是一种新型的检测手段,它将吸收的部分光能转化为热能,发出超声信号。与荧光成像技术相比,光声成像技术可以在更高的穿透深度上实现活体水平高分辨率的无损成像检测,但是无法实现细胞成像。
发明内容
为了解决现有技术的不足,本公开的目的是提供花菁类化合物及制备方法与在检测高尔基体pH中的应用,采用该花菁类化合物作为高尔基体内pH的荧光探针,不仅具有高尔基靶向能力,而且能够实现灵敏检测高尔基体pH变化的光声-荧光双模态成像。
为了实现上述目的,本公开的技术方案为:
一方面,本公开提供了一种花菁类化合物,其化学结构如下所示:
另一方面,本公开提供了一种上述花菁类化合物的制备方法,包括以Cy7-Cl和对氨基苯磺酰胺为原料进行苯胺基团与氯的取代反应的步骤。
第三方面,本公开提供了一种上述花菁类化合物在检测pH或检测高尔基体pH中的应用。
在酸性条件下,定位基团对氨基苯磺酰胺和荧光团Cy7-Cl连接生成的-NH-会发生质子化,使得整个体系的共平面效应增加,从而引起荧光发射波长的移动。随着酸性的增强,探针的荧光发射会逐渐从750nm红移至810nm。
第四方面,本公开提供了一种上述花菁类化合物在制备靶标高尔基体pH荧光探针的应用。
第五方面,本公开提供了一种上述花菁类化合物在制备光声成像原位pH检测探针的应用。
本公开的有益效果为:
1.本公开提供了花菁类化合物作为高效靶标高尔基的荧光探针。相比起之前的高尔体定位基团,本公开中用苯磺酰胺作为高尔基体定位基团合成可定位荧光探针的策略易于合成,成本低廉,且具有良好的定位效果。
2.本公开提供的花菁类化合物用于高尔基体pH检测的比率荧光探针在不同pH的缓冲溶液中展现良好的吸收和荧光光谱响应,且具有良好的可逆性和光稳定性。
3.本公开中的花菁类化合物具有良好的生物相容性,对细胞的毒性非常低,可以用于活细胞中高尔基体的pH检测。
4.本公开中的花菁类化合物具有近红外荧光和光声响应性质,既能实现亚细胞器的定位,又能实现活体中深层组织的高空间分辨率的成像。并成功应用于炎症小鼠模型中原位pH的检测。
5.本公开中的花菁类化合物合成简单,且在酸性和碱性环境中有明显的颜色可逆变化,有望发展成一种商业化的pH试纸。

具体实施方式
应该指出,以下详细说明都是示例性的,旨在对本公开提供进一步的说明。除非另有指明,本文使用的所有技术和科学术语具有与本公开所属技术领域的普通技术人员通常理解的相同含义。
需要注意的是,这里所使用的术语仅是为了描述具体实施方式,而非意图限制根据本公开的示例性实施方式。如在这里所使用的,除非上下文另外明确指出,否则单数形式也意图包括复数形式,此外,还应当理解的是,当在本说明书中使用术语“包含”和/或“包括”时,其指明存在特征、步骤、操作、器件、组件和/或它们的组合。
为了实现灵敏检测高尔基体pH变化的光声-荧光双模态成像的目的,本公开提出了花菁类化合物及制备方法与在检测高尔基体pH中的应用。
本公开的一种典型实施方式,提供了一种花菁类化合物,其化学结构如下所示:
本公开的另一种实施方式,提供了一种上述花菁类化合物的制备方法,包括以Cy7-Cl和对氨基苯磺酰胺为原料进行苯胺基团与氯的取代反应的步骤。
其合成路线如下所示:
该实施方式的一种或多种实施例中,所述取代反应的条件为:碱性条件下,加热至不低于90℃进行反应。
该系列实施例中,采用氢氧化钠提供碱性条件。当Cy7-Cl与氢氧化钠的摩尔比为1:1~2时,能够更好的提供碱性环境进行取代反应。
该系列实施例中,反应温度为90~95℃,反应时间为12~16h。能够增加化合物的反应效率。
该实施方式的一种或多种实施例中,Cy7-Cl和对氨基苯磺酰胺的投料比为1:2~5。能够使得Cy7-Cl反应完全,提高花菁类化合物的产率。
该实施方式的一种或多种实施例中,反应体系的溶剂为N,N-二甲基甲酰胺。
为了提高花菁类化合物的纯度,该实施方式的一种或多种实施例中,将反应后的物料去除溶剂后进行柱层析提纯。
该系列实施例中,柱层析的展开剂为乙酸乙酯。
本公开的第三种实施方式,提供了一种上述花菁类化合物在检测pH或检测高尔基体pH中的应用。
在酸性条件下,定位基团对氨基苯磺酰胺和荧光团Cy7-Cl连接生成的-NH-会发生质子化,使得整个体系的共平面效应增加,从而引起荧光发射波长的移动。随着酸性的增强,探针的荧光发射会逐渐从750nm红移至810nm。
本公开的第四种实施方式,提供了一种上述花菁类化合物在制备靶标高尔基体pH荧光探针的应用。
本公开的第五种实施方式,提供了一种上述花菁类化合物在制备光声成像原位pH检测探针的应用。
为了使得本领域技术人员能够更加清楚地了解本公开的技术方案,以下将结合具体的实施例详细说明本公开的技术方案。
实施例1:
荧光探针的合成:
准确称取Cy7-Cl(0.51g,1mmol)和对氨基本磺酰胺(0.86g,5mmol)在氮气保护下加入到双口瓶,加入8mL除水的DMF作为溶剂。再加入氢氧化钠(0.04g,1mmol)提供反应的碱性环境。缓慢升温至90度,反应12小时。反应完成后,将产品减压蒸馏除去溶剂DMF,之后用硅胶柱层析法进行分离提纯,乙酸乙酯作为展开剂。提纯后得蓝色固体(0.1g,15%),记为CPH。1H NMR(400MHz,CDCl3)δ8.23(d,J=13.2Hz,1H),7.83(d,J=8.5Hz,1H),7.20(t,J=7.6Hz,2H),6.96(t,J=7.4Hz,1H),6.75(d,J=7.8Hz,1H),6.66(d,J=8.6Hz,1H),5.59(d,J=13.2Hz,1H),5.29(s,1H),3.79(d,J=6.9Hz,2H),2.55(t,J=6.0Hz,2H),2.04(s,1H),1.86–1.79(m,1H),1.55(s,6H),0.88(dd,J=8.6,4.8Hz,3H).13C NMR(101MHz,CDCl3):147.52,142.11,139.20,128.97,128.74,126.68,124.96,120.94,120.49,113.05,106.20,93.10,52.41,46.30,36.48,34.83,30.89,29.46–27.97,27.15,26.18,24.80,24.52,21.66,20.83,13.10,10.35。
CPH的效果实验:
通常,可以将染料分子溶解在生理盐水、缓冲液或由乙腈、二甲亚砜等水溶性有机溶剂中,然后加入适当缓冲液及其他有机试剂进行测试。分别探究了在不同pH的PBS缓冲溶液中,探针CPH随pH变化的吸收和荧光光谱响应,并将其用于活细胞及炎症小鼠成像实验。活细胞的染色方法是将培养好的细胞放于含有探针分子的培养液中孵育,孵育一定的时间后除去孵育液,进行共聚焦成像实验。小鼠的染色方法是将探针原位注射到小鼠的正常部位和炎症部位,一段时间后,对小鼠注射探针的部位进行光声成像。
探针CPH在不同pH缓冲液的紫外吸收、荧光发射及选可逆性实验:
探究探针CPH在不同pH的PBS(0.1M)中的紫外吸收和荧光响应性质。不同pH值的缓冲液是通过向pH为7.4的PBS中加入尽量少的氢氧化钠或者盐酸溶液进行配置。随着pH值的增大(5.0~8.7),探针CPH(50μM)的吸收光谱呈现出高灵敏度的改变。如图1a所示,在pH为5.0的缓冲介质中,探针的最大吸收峰在800nm左右,而随着pH值的增大,探针的最大吸收蓝移至610nm左右。而且由于探针在800nm处的光吸收强度(PA信号可检测)随pH的增大逐渐减弱,而690nm处的光吸收基本不变,因此探针具有比率光声成像检测pH的潜能。图1b为探针CPH(2μM)在不同pH(5.0~8.7)的缓冲液中的荧光光谱响应。在675nm的光激发下,随着pH的逐渐增大,探针于810nm处的荧光强度逐渐降低,750nm处荧光强度逐渐增强,说明探针对pH具有高灵敏度的荧光相应,可以通过比率荧光定量检测pH。
图2为探针CPH在酸性和碱性往复变化的环境中的可逆性。将探针(2μM)加入到pH为4.5的PBS中,进行荧光检测,之后不断调节其pH值在4.5和9.5之间变换,记录其光谱响应。纵坐标FI750nm/FI810nm代表探针在750nm和810nm处的荧光强度比值。结果表明在pH为4.5和9.5的缓冲中,CPH始终展现出良好的、瞬时的荧光响应,且具有良好的可逆性。
CPH的高尔基体靶向性实验:
人正常肝细胞HL-7702是由含有双抗和胎牛血清蛋白的DMEM培养液培养。之后用500nM的探针分别和定位不同亚细胞器的商业化染料(包括高尔基体、线粒体、溶酶体)共孵育细胞30min后,使用激光共聚焦显微镜进行共定位成像实验。共定位细胞成像实验如图3所示,图3a,b,c中探针CPH只与高尔基体商业化定位染料的荧光有较好的重叠。图3d,e,f是探针CPH和商业化染料在某一线性区域荧光强度的趋势图。荧光强度重叠越好,趋势越一致,说明共定位效果越好,因此该探针展现了高效靶标高尔基体的能力。
探针在不同pH的活细胞中的共聚焦荧光成像实验:
人肝癌细胞SMMC-7721是由含有双抗和胎牛血清蛋白的DMEM培养液培养。之后将细胞与CPH(500nM)在37℃下共孵育10分钟,洗去探针后用含有10μM尼日利亚菌素(一种离子导入剂)的不同pH(6.0,6.5,7.0,7.5,8.0)的PBS缓冲液(0.01M)继续孵育10分钟,然后进行共聚焦成像。由于尼日利亚菌素的介入,细胞内的pH值会根据PBS缓冲液pH的变化而变化。受限于仪器,本公开只收集了710~760nm处的荧光。如图4所示,细胞经pH为6.00的PBS孵育后,探针在细胞中呈现出微弱荧光,随着细胞孵育液pH从6.00上升到8.00的过程中,细胞中CPH的荧光逐渐增强。上述结果说明探针CPH可以高灵敏度的用于监测活细胞中的pH变化。
探针对在炎症小鼠模型中正常部位和炎症部位的原位光声成像实验:
在小鼠的左侧腹腔注射生理盐水作为控制组,右侧相同位置注射LPS(2mg/mL,400μL)去诱导腹腔炎症的产生。4小时之后,将小鼠用水合氯醛进行麻醉后进行实验。在注射LPS(右侧)及生理盐水(左侧)的部位腹腔注射探针CPH(30μM,50μL),在690nm及800nm波长下进行光声成像检测,如图5所示。结果发现,800nm波长下,小鼠LPS刺激一侧的光声信号强度明显强于注射生理盐水的一侧,且具有较低的PA690nm/PA800nm比值,说明在LPS刺激部位呈现弱酸性,pH值偏低,如图6所示。表明CPH可以作为一种荧光与光声双模态成像探针检测活体中的pH。

3.如权利要求2所述的花菁类化合物的制备方法,其特征是,所述取代反应的条件为:碱性条件下,加热至不低于90℃进行反应。
4.如权利要求3所述的花菁类化合物的制备方法,其特征是,采用氢氧化钠提供碱性条件。
5.如权利要求4所述的花菁类化合物的制备方法,其特征是,Cy7-Cl与氢氧化钠的摩尔比为1:1~2。
6.权利要求3所述的花菁类化合物的制备方法,其特征是,反应温度为90~95℃,反应时间为12~16h。
7.如权利要求2所述的花菁类化合物的制备方法,其特征是,Cy7-Cl和对氨基苯磺酰胺的投料比为1:2~5。
8.如权利要求2所述的花菁类化合物的制备方法,其特征是,反应体系的溶剂为N,N-二甲基甲酰胺。
9.如权利要求2所述的花菁类化合物的制备方法,其特征是,将反应后的物料去除溶剂后进行柱层析提纯。
10.权利要求9所述的花菁类化合物的制备方法,其特征是,柱层析的展开剂为乙酸乙酯。
11.一种权利要求1所述的花菁类化合物在检测高尔基体pH中的应用,所述应用不包括疾病的诊断和治疗。
