Abstract
提供了一种高载量耐碱蛋白A磁珠,其在pH 2‑14环境下均可保持其化学稳定性,并且结合免疫球蛋白IgG的能力大于50mg/mL。还提供了一种纯化和/或检测免疫球蛋白的方法,包括让含有免疫球蛋白的样品与所述高载量耐碱蛋白A磁珠接触的步骤。前述所提供的耐碱蛋白A磁珠可实现免疫球蛋白的快速纯化,节省约80%的处理时间,总体纯化成本可下降50%。除此之外,该耐碱蛋白A磁珠具备高耐碱特性,可以使用原位清洁的碱性方法对使用后的磁珠进行再生。磁珠本身的快磁响应和较好的分散性,可快速实现磁珠富集、清洗和洗脱,方便实现自动化高通量和大样品量纯化。
