技术领域
本发明属于生物技术和医学领域,具体涉及一种有抗乳腺癌转移协同增效作用的药物组合物,包含NAC1抑制剂和肿瘤血管生成抑制剂类化合物,在抑制肿瘤,尤其是乳腺癌转移方面具有显著的协同作用。
背景技术
乳腺癌是女性最常见的恶性肿瘤之一,近年来其发病率在全球范围内呈逐年上升的趋势。全球每年有约120万妇女患乳腺癌,死亡患者约50万,发病率占全年各种恶性肿瘤的7%~10%。乳腺癌发展到晚期,患者身体内部的癌细胞一般都会发生转移和扩散,乳腺癌远处转移是导致患者死亡的主要原因,一旦发生转移则极难治疗。贝伐单抗(bevacizumab)是2004年2月被美国FDA批准上市的第一个抗血管生成药,可用于转移性乳腺癌的一线治疗。在转移性乳腺癌治疗中,贝伐单抗可明显提高无进展生存期(PFS),然而这些研究均未显著提高总体生存期(OS)。有研究报道,抗血管生成疗法有可能无意中使肿瘤更加具有侵袭性和更可能发生扩散。该研究表明:在抗血管治疗的肿瘤中缺氧区域增加了5倍,从而激活了肿瘤细胞类一系列缺氧诱导的反应;并且发现这类肿瘤细胞具有类似干细胞特性,这样这些细胞更易存活下来。
BTB/POZ家族基因NACC1基因编码细胞转录因子NAC1(nucleus accumbens-1,伏隔核1)的表达,其定位于人染色体区Ch19p13.2,是新近发现的一个新的致癌因子。NAC1在多种妇科肿瘤中(例如卵巢癌、***、子宫内膜癌、乳腺癌等)普遍高表达,而在正常组织未见表达。BTB结构域(也称为POZ结构域)是介导蛋白相互作用的重要结构域,NAC1的BZB结构域(1-129氨基酸序列)是形成NAC1二聚体复合物所必需的,形成的NAC1二聚体复合物可以参与多种生物功能调控:例如抗凋亡、促增殖、促侵袭转移、抗衰老等。
有实验表明,NAC1还可通过阻滞HDAC4出核转运,稳定核HIF-1α蛋白,促进缺氧环境下肿瘤细胞糖酵解激活;还可促进肿瘤和胚胎干细胞的自我更新。既然NAC1只是特异性地在肿瘤细胞高表达,而且参与了缺氧诱导反应和干细胞特性的维持,因此针对该分子的小分子化合物可能能够取消其抗血管治疗导致的缺氧诱导反应和干细胞特性维持,并将有可能和肿瘤血管生成抑制剂类药物联用,开辟一条肿瘤治疗新策略。
发明内容
本发明目的是提供一种协同增效的抗乳腺癌转移药物组合物及其用途。
本发明的技术方案是:
一种协同增效的抗乳腺癌转移药物组合物,其特征在于,含有:
(1)NAC1抑制剂;
(2)肿瘤血管生成抑制剂类化合物;
(3)药学、保健品学、或食品学上可接受的载体或赋形剂。
进一步的,所述NAC1抑制剂为化合物2-(4-叔丁基苯氧基)-N-(2-{[(4-叔丁基苯氧基)乙酰基]氨基}乙基)乙酰胺、或其药学、保健品学或食品学上可接受的盐或酯或衍生物、或它们的混合物。
进一步的,所述肿瘤血管生成抑制剂类化合物为贝伐单抗、阿柏西普、雷莫芦单抗、血管内皮抑素、小分子酪氨酸激酶抑制剂中的任意一种。例如索拉非尼、舒尼替尼、阿西替尼、布立尼布、帕唑替尼、阿帕替尼等,更优选贝伐单抗。
进一步的,所述NAC1抑制剂和肿瘤血管生成抑制剂类化合物之间的重量比为1:1000至1000:1,优选重量比为1:500-500:1,更优选重量比为1:100-100:1,再优选重量比为1:50-50:1。
进一步的,所述NAC1抑制剂占组合物总重量的l-95wt%,优选5-90wt%,更优选10-80wt%。
进一步的,所述组合物的剂型是片剂、胶囊、粉末剂、颗粒剂、混悬剂或注射剂。在所述组合物为单位剂型或多剂型时,所述NAC1抑制剂的含量为0.05-50000mg/剂,优选0.1-10000mg/剂,更优选0.5-5000mg/剂。
进一步的,所述组合物的用药顺序为先用NAC1抑制剂,而后使用肿瘤血管生成抑制剂类化合物。
乳腺癌细胞是表达人NAC1的肿瘤细胞,乳腺癌细胞的人NAC1表达量高于正常细胞30%~50%。上述组合物可应用于制备用于治疗抗乳腺癌细胞转移的药物。
本发明的优点是:本发明所述的协同增效的抗乳腺癌转移药物组合物,可有效抑制NAC1,控制、缓解或治愈疾病的治疗,例如乳腺癌转移等。

细胞系
MDA-MB-231细胞系:购自ATCC,这是一种阳性表达NAC1的转移性乳腺癌细胞系。
MDA-MB-231细胞系的培养方法如下:将细胞接种于含10%小牛血清的DMEM(InVitrogen公司)培养液中,置于37摄氏度、体积分数为5%的CO2培养箱中常规培养,实验前无药培养两周。
实施例
1、转移鼠模型的建立与干预
尾静脉注射乳腺癌细胞株MDA-MB-231制备乳腺癌转移鼠模型,并观察NAC1抑制剂联合贝伐单抗的作用。荧光标记的MDA-MB-231细胞传代。在细胞对数期收集细胞,做成浓度为1×106/100微升的细胞悬液,SCID鼠尾静脉下注射0.1ml细胞悬液(细胞数目为1.0×106个/只)。随机分为4组,每组5只。阴性对照组(静脉注射生理盐水组),NAC1抑制剂给药组(尾静脉注,25mg/kg),贝伐单抗给药组(腹腔注射,5mg/kg),NAC1抑制剂+贝伐单抗联合给药组。结果显示,NAC1抑制剂可以在25mg/kg显著增强贝伐单抗对MDA-MB-231细胞肺和骨的转移。
具体可参阅图1-图5,如图1所示,采用NAC1抑制剂+贝伐单抗联合给药组的小鼠在前四周时,与阴性对照组、NAC1抑制剂给药组、贝伐单抗给药组相比,无显著区别,但是在第四周后,采用阴性对照组和NAC1抑制剂给药组的小鼠的转移细胞量明显上升,采用贝伐单抗给药组的小鼠的转移细胞量低于采用阴性对照组和NAC1抑制剂给药组的小鼠,但是仍然有明显的上升,而采用NAC1抑制剂+贝伐单抗联合给药组的小鼠仍然能保持非常低的转移率。如图2所示,采用NAC1抑制剂+贝伐单抗联合给药组的小鼠的肺部转移瘤结节数极少,接近零,而其他三组给药组的小鼠的肺部转移瘤结节数均是采用NAC1抑制剂+贝伐单抗联合给药组的小鼠的肺部转移瘤结节数的20-40倍。如图3所示,肺部病理结果显示,采用NAC1抑制剂+贝伐单抗联合给药组的小鼠的肺部肿瘤转移区域(T)明显减少,而其他三组给药组的小鼠的肺部肿瘤转移区域(T)明显增多。如图4所示,采用NAC1抑制剂+贝伐单抗联合给药组的小鼠在前四周时,与阴性对照组、NAC1抑制剂给药组、贝伐单抗给药组相比,无显著区别,但是在第四周后,采用阴性对照组、NAC1抑制剂给药组和贝伐单抗给药组的小鼠的骨转移细胞量明显上升,特别是在第七周以后,上升幅度很大,而采用NAC1抑制剂+贝伐单抗联合给药组的小鼠仍然能保持非常低的转移率。如图5所示,采用NAC1抑制剂+贝伐单抗联合给药组的小鼠在90天后的存活率为100%,而采用阴性对照组的小鼠在90天后存活率为0,采用NAC1抑制剂给药组的小鼠在90天后存活率为20%,并很快下降为存活率为0。采用贝伐单抗给药组的小鼠在90天后存活率为40%,并很快下降为存活率为20%。
由此可知,NAC1抑制剂+贝伐单抗联合给药组与其他三组方式相比,可显著延长乳腺癌转移模型小鼠的生存期。
综上所述,本发明公开了一种靶向于人伏隔核1抗肿瘤协同增效的抗乳腺癌转移药物组合物及其用途,将肿瘤血管生成抑制剂类化合物联合NAC1抑制剂用药,与单独使用肿瘤血管生成抑制剂类化合物相比较,具有显著提高抗乳腺癌转移效果的作用。该发现在治疗乳腺癌转移的研究中属于极大的进步,临床应用前景良好。
应说明的是,以上实施例仅用以说明本发明的技术方案而非限制,尽管参照较佳实施例对本发明进行了详细说明,本领域的普通技术人员应当理解,可以对本发明的技术方案进行修改或者等同替换,而不脱离本发明技术方案的精神和范围,其均应涵盖在本发明的权利要求范围当中。
Claims (10)
1.一种协同增效的抗乳腺癌转移药物组合物,其特征在于,含有:
(1)NAC1抑制剂;
(2)肿瘤血管生成抑制剂类化合物;
(3)药学、保健品学、或食品学上可接受的载体或赋形剂。
2.根据权利要求1所述的一种协同增效的抗乳腺癌转移药物组合物,其特征在于:所述NAC1抑制剂为化合物2-(4-叔丁基苯氧基)-N-(2-{[(4-叔丁基苯氧基)乙酰基]氨基}乙基)乙酰胺、或其药学、保健品学或食品学上可接受的盐或酯或衍生物、或它们的混合物。
3.根据权利要求1所述的一种协同增效的抗乳腺癌转移药物组合物,其特征在于:所述肿瘤血管生成抑制剂类化合物为贝伐单抗、阿柏西普、雷莫芦单抗、血管内皮抑素、小分子酪氨酸激酶抑制剂中的任意一种。
4.根据权利要求1所述的一种协同增效的抗乳腺癌转移药物组合物,其特征在于:所述NAC1抑制剂和肿瘤血管生成抑制剂类化合物之间的重量比为1:50-50:1。
5.根据权利要求1所述的一种协同增效的抗乳腺癌转移药物组合物,其特征在于:所述NAC1抑制剂占组合物总重量的10-80wt%。
6.根据权利要求1所述的一种协同增效的抗乳腺癌转移药物组合物,其特征在于:所述组合物的剂型是片剂、胶囊、粉末剂、颗粒剂、混悬剂或注射剂。
7.根据权利要求1所述的一种协同增效的抗乳腺癌转移药物组合物,其特征在于:在所述组合物为单位剂型或多剂型时,所述NAC1抑制剂的含量为0.5-5000mg/剂。
8.根据权利要求1所述的一种协同增效的抗乳腺癌转移药物组合物,其特征在于:所述组合物的用药顺序为先用NAC1抑制剂,而后使用肿瘤血管生成抑制剂类化合物。
9.一种协同增效的抗乳腺癌转移药物组合物在制备用于治疗抗乳腺癌细胞转移的药物中的用途。
10.根据权利要求9所述的用途,其特征在于:所述乳腺癌细胞是表达人NAC1的肿瘤细胞,所述乳腺癌细胞的人NAC1表达量高于正常细胞30%~50%。
