Abstract
本发明涉及一种小儿脑发育专用型临床营养配方,其包括如下组分及其重量份数:15‑35份蛋白质、2~15份脂肪、40~60份碳水化合物、8‑12份膳食纤维、0.2‑1.2份常量元素、0.01‑0.05份微量元素、0.001‑0.02份脂溶性维生素、0.008‑0.4份水溶性维生素、4~8份膳***华、0.1‑4份药食同源成分以及0.05‑2份新资源食品;本发明还涉及上述临床营养配方的制备方法。通过上述组分的筛选和配比,本发明依据孕妇及小儿的生理特点设计,在保证蛋白质、脂肪、碳水化合物、膳食纤维的基础上,补充许多人体所需的常量元素和微量元素,并添加具有健脑益智、健胃消食、增强免疫力等功效的药食同源成分、新资源食品等特殊营养素,使得该配方营养均衡,有效地保证小儿脑发育,保证孕妇及婴幼儿的身心健康成长。

本发明涉及特殊医学用途配方食品领域,具体涉及一种小儿脑发育专用型临床营养配方及其制备方法。
背景技术
人在胚胎时期,由一个受精卵不断***增殖,就脑细胞而言,可***增殖至百多亿个,这巨大的变化无疑需要相应的营养支持。新生儿脑重量约为 370g~400g,仅为成人的25%,六个月时为成人脑重的50%,到了幼儿一岁时脑重量已达900g,相当于成人脑重的60%~70%。到了幼儿两岁以后,脑重量的增长速度虽然减缓,但脑内某些神经细胞,如小型的中间神经原,正是在儿童的这一时期发育成熟的。而神经细胞轴突与树突的发育,则可以在脑和脊髓内建立更为密集的网状组织。到了儿童12岁左右,他的脑重量已基本与成人相同。由此而看,脑部发育是否正常,是由脑神经细胞的数量与网状组织的密度决定的。脑神经细胞数目越多,则儿童的智商就有可能越高,网状组织的致密程度也是决定儿童今后智力高低的重要因素。由此看来,儿童出生后至5岁是脑发育的关键期,幼儿在这一时期若营养不足,可使幼儿大脑发生不可逆转的永久性障碍。综上所述,营养因素、环境因素、教育因素对儿童的智力的发展有很大的影响,其中营养因素起着更为重要的作用。
胎儿的脑神经***从怀孕第5、6周就开始发展,直到第8周神经管愈合完成,也就是说,从胚胎着床后,神经细胞便开始分化,接着产生各种神经细胞移动与突触结合,在怀孕第8周时,神经管便渐次愈合,但整个神经***的发育,会持续到孕期结束,甚至到宝宝出生后3年内,才会达到97%的成熟。神经***是所有器官中,发展期最长同时也是最早就开始发展的器官,这是高度分化的人类与其它物种最大的不同之处。由于目前市场上关于脑发育的营养产品种类错综复杂,例如中国专利CN101120784A公开了一种促进胎儿及婴幼儿大脑发育的健脑食品,其包括亚麻油93~97份、柠檬酸锌1~2份、视黄醇0.01~0.05份,但该产品仅仅研究单一营养成分对小儿的脑发育作用,多种营养成分联合作用的针对脑发育的营养产品相对较少,因此研发一款提高小儿脑发育的复合营养产品显得尤为重要。
发明内容
本发明的目的在于克服现有技术中的缺陷,提供一种针对小儿脑发育的专用型临床营养配方,其可满足孕妇及幼儿的日常营养需求,帮助小儿提高脑神经***,改善小儿记忆力,促进小儿大脑发育。
为实现上述目的,本发明采用如下技术方案:

步骤1)采用微囊包埋技术制备脂肪粉末、脂溶性维生素粉末和水溶性维生素粉末;
步骤2)采用粉碎、醇提、水提、喷雾干燥及过筛步骤制备药食同源成分粉末;
步骤3)将预定重量的蛋白质、碳水化合物、膳食纤维、常量元素、微量元素、膳***华以及新资源食品分别制备成粒径为150~250μm的粉末,如果需要的话可将上述原料预先进行烘晒处理;
步骤4)将步骤1)~3)制备的粉末混合均匀后杀菌备用;
步骤5)根据常规技术手段将步骤4)混合后的粉末制备成营养粉、营养乳剂、胶囊剂、片剂、丸剂或口服液中的至少一种;
其中,步骤1)-3)的顺序可相互更换。
最后,本发明涉及一种由上述小儿脑发育专用型临床营养配方制备的产品,包括营养粉、营养乳剂、胶囊剂、片剂、丸剂、口服液等。
在上述制备方法中,其中,所述药食同源成分的原料进行粉碎、醇提浓缩、水提浓缩,并将浓缩液进行喷雾干燥制成药食同源成分粉末;其中,所述维生素和脂肪分别通过微胶囊化处理制备成维生素粉末和脂肪粉末。
优选地,所述药食同源成分粉末的制备过程如下:采用粉碎机对预定重量份数的原料进行粉碎,将粉碎的原料置于容器中,加入重量为原料重量的7~8倍的 30~40%乙醇,进行浸泡、提取3次,合并醇提液并浓缩至无醇味;原药药渣挥去乙醇后,加入重量为原料重量的3~4倍的水进行浸泡,对浸泡液进行煎煮 15~20min,过滤去除原料,留取煎煮液;合并浓缩后的醇提液和煎煮液,并对其进行喷雾干燥,最后以60~80目筛分制备药食同源成分粉末。
优选地,所述维生素的微胶囊化处理的具体步骤如下:1)将脂溶性维生素作为芯材,选用羧甲基纤维素钠、明胶、卡拉胶、黄原胶、***胶、麦芽糊精、脂蛋白、玉米糖浆、大豆分离蛋白、全脂乳粉、膳食纤维、菊粉、乳清蛋白粉、低聚异麦芽糖、大豆纤维、乳糖、冰糖粉、乳化剂、稳定剂中的一种或多种作为壁材,高压均质乳化,喷雾瞬时干燥,以60~80目筛分制备脂溶性维生素粉末; 2)将水溶性维生素作为芯材,选用羧甲基纤维素钠、明胶、卡拉胶、黄原胶、***胶、麦芽糊精、脂蛋白、玉米糖浆、大豆分离蛋白、全脂乳粉、膳食纤维、菊粉、乳清蛋白粉、低聚异麦芽糖、大豆纤维、乳糖、冰糖粉、乳化剂、稳定剂中的一种或多种作为壁材,混合均匀,喷雾瞬时干燥,以60~80目筛分制备水溶性维生素粉末。
优选地,所述脂肪的微囊包埋技术的具体实施步骤如下:将脂肪、乳化剂、壁材与水混合溶解制备乳化液;将上述乳化液搅拌混合后,进行乳化均质处理,然后进行喷雾干燥,筛分制备脂肪粉末,脂肪粉末与传统油脂比较,其分散性、水溶性、乳化性、抗氧化性都有很大提高;其中,所述乳化剂选自甘油脂肪酸酯(甘油酯)、丙二醇脂肪酸酯(丙二醇酯)、山梨聚糖脂肪酸酯(山梨聚糖酸)、蔗糖脂肪酸酯(蔗糖酯)、卵磷脂、单甘脂、吐温80中的一种或多种;所述壁材选自羧甲基纤维素钠、明胶、卡拉胶、黄原胶、***胶、麦芽糊精、脂蛋白、玉米糖浆、大豆分离蛋白、全脂乳粉、膳食纤维、菊粉、乳清蛋白粉、低聚异麦芽糖、大豆纤维、乳糖、冰糖粉、乳化剂、稳定剂中的一种或多种。
本发明还提供一种上述小儿脑发育专用型临床营养配方的制备方法,其为湿法工艺,包括如下步骤:
步骤1)准备含有预定重量份数的蛋白质、脂肪、碳水化合物、膳食纤维、常量元素、微量元素、维生素、膳***华、药食同源成分以及新资源食品的原材料,在预定量的水中,按照原材料的物理性状,按照先后顺序依次添加,在添加的同时进行搅拌,制得原料的混合乳液;
步骤2)将步骤1)获得的混合乳液温度保持在40-50℃,调整均质压力使其始终保持在10-18Mpa之间,对混合乳液进行乳化均质,然后将乳化液放置蒸发器中,以预设的技术参数进行浓缩,并在85-88℃下进行杀菌,获得浓缩液;
步骤3)将步骤2)获得的浓缩液进行喷雾干燥,过60~80目筛,制备成粉末;采用上述粉末制备成营养粉、营养乳剂、胶囊剂、片剂、丸剂或口服液中的至少一种。
本发明中采用包括甘草、枸杞、橘皮、黑芝麻、益智仁、黄精、牡蛎、茯苓、山药;其中,甘草能够顺气,促进心火的发泄,帮助情绪的稳定,其对于脾虚也有一定的缓解作用,能够帮助补脾;枸杞子能够滋补肝肾、益精明目和养血、增强人体免疫力;橘皮具有调理消化***,增强肠胃功能,利胆助消化,用于脾虚饮食减少、消化不良,对抗乙酰胆碱引起的肠收缩和拮抗毛果芸香碱和氯化钡引起的肠管痉挛性收缩;黑芝麻含有多种人体必需氨基酸,能增加人体的代谢功能,还具有补充脑部智力的功效,其可促进大脑发育;益智仁能温脾止泻、开胃消食、增强儿童消化、吸收功能,平衡内环境,提高免疫功能;黄精能够益肾固精,充盈肌肉,提高身体免疫力;牡蛎含有丰富的牛磺酸,有明显的保肝利胆作用,并含有丰富微量元素和糖元,又利于促进胎儿的生长发育;茯苓能增加机体免疫功能,利尿祛湿,润肠通便,改善消化吸收,增强人体对食物的运化功能;山药能改善机体免疫功能,含有淀粉、蛋白质、粘液质等多种营养素,含有丰富的胆碱和卵磷脂,有助于提高人的大脑的记忆力。

本发明所述的小儿脑发育专用型临床营养配方中,添加紫苏油,其中的n-3 PUFAs参与膜的构建、调节基因的表达以及通过其所代谢产物发挥抗炎及抑制细胞凋亡的作用等。n-3PUFAs影响脑功能的作用机制最初多集中在对脑膜性结构的影响上,磷脂中PUFAs的变化可改变膜的流动性,影响膜受体的功能、多巴胺及血清素能等神经递质的传递,调节膜结合蛋白,如Na+/K+依赖性ATP酶的活性。通过这些改变从而为大脑提供一个高强度的物质交换及信号的转导环境,对细胞的稳定性起着重要的作用。
本发明所述的小儿脑发育专用型临床营养配方补充添加铁、锌和维生素B1。铁分布于整个脑组织,但分布不均匀,含铁最丰富的部分是锥体外系的核团,如基底节、苍白球、黑质、红核,其次是小脑及大脑的某些区域。研究发现,脑组织中主要含铁细胞是少突胶质细胞,该细胞主要功能是参与中枢神经***髓鞘形成,脑发育中缺铁使髓鞘形成受阻。少突胶质细胞数量的变化会直接影响到脑内神经元对铁的利用,故认为少突胶质细胞是脑组织铁平衡的调节体。锌是脑的重要组成成分,脑中锌的含量在端脑和大脑皮层灰质中最高。缺锌时体内氨基酸氧化过程增加,组织蛋白合成减少,生长发育受到影响。维生素B1即硫胺素在机体碳水化合物代谢中起重要作用,如果缺少硫胺素,代谢过程就会变慢或完全中断。
本发明所述的小儿脑发育专用型临床营养配方还添加牛磺酸,其可促进脑细胞DNA、RNA的合成,增加膜的磷脂酰乙醇胺含量和脑细胞对蛋白质的利用率,从而促进脑细胞尤其是海马细胞结构和功能的发育;
添加γ-氨基丁酸,其能抑制谷氨酸的脱羧反应,与α-酮戊二酸反应生成谷氨酸,使血氨降低,更多的谷氨酸与氨结合成尿素,排出体外,减低氨毒,改善肝肾功能。可以提高葡萄糖磷酸酯酶的活性,使脑细胞活动旺盛,促进脑细胞组织代谢,改善神经机能;
添加磷脂,其总磷脂质与细胞膜的生成有关,且含胆碱,是一种帮助人体制造脑部神经讯息传导物质(乙酰胆碱)重要成份。摄取丰富的卵磷脂,帮助脑髓的发育,提高脑部活动的效率。

实施例5
一种如实施例1所述的小儿脑发育专用型临床营养配方的制备方法如下:
1)制备粉末油脂(粉末脂肪):
a)制备壁材水溶液:取麦芽糊精8克、全脂乳粉11克、膳食纤维2克(抗性糊精)、菊粉0.5克、乳清蛋白粉5克、低聚异麦芽糖0.5克、大豆纤维1克、***胶0.2克、乳糖0.5克、冰糖粉2克、水50毫升搅拌均匀(有条件边加热边搅拌至60℃,保持30分钟后冷却至室温)。
b)将单甘酯取0.1克(总固形物浓度为0.2%)均匀溶于7.79毫升油脂中(紫苏籽油5.63毫升、DHA藻油微囊粉1.88毫升、花生四烯酸油脂微囊粉1.88毫升、磷脂0.38毫升)。
c)将吐温80取0.1克(总固形物浓度为0.2%)均匀溶于壁材水溶液中。
d)将添加过乳化剂单甘酯的混合植物油脂与添加过乳化剂吐温80的壁材水溶液充分混合。
e)添加稳定剂琼脂取0.1克(总固形物浓度为0.2%)进混合溶液后,用水定容至200毫升(总固形物浓度为25%),20000r/min下高速分散2分钟。
f)均质压力为25MPa,均质3次(每次5分钟),制得均质乳状液。
g)将制得的乳状液进行喷雾干燥,进风温度为195℃,出风温度为80℃。收集粉末油脂(粉末脂肪);
2)制备药食同源成分粉末:采用粉碎机对预定重量份数的原料进行粉碎,将粉碎的原料置于容器中,采用粉碎机对预定重量份数的原料进行粉碎,将粉碎的原料置于容器中,加入重量为原料重量的3~8倍的水,进行浸泡3~4小时,然后对浸泡液进行煎煮30~60min,过滤去除原料,留取煎煮液,对煎煮液进行喷雾干燥,以60~80目筛分制备药食同源成分粉末;
3)制备维生素粉末:将预定重量份数的脂溶性维生素作为芯材,选用卡拉胶、***胶作为壁材,在高压下进行2次乳化均质处理,乳化均质时间为 10~15min,然后进行喷雾瞬时干燥,以60~80目筛分制备脂溶性维生素粉末;将预定重量份数的水溶性维生素作为芯材,选用麦芽糊精、脂蛋白作为壁材,混合均匀,喷雾瞬时干燥,以60~80目筛分制备水溶性维生素粉末;
4)将预定重量的蛋白质、碳水化合物、膳食纤维、常量元素、微量元素、膳***华以及新资源食品分别制备成粒径为150~250μm的粉末;(步骤1)-4) 的顺序可相互更换)
5)将步骤1)-4)制备的粉末依次加入锥形混合机或其他形式的混合机混合均匀制得配方混合物,经过杀菌处理后,备用;
6)根据常规技术手段将步骤5)中的配方混合物制备成营养粉、营养乳剂、胶囊剂、片剂、丸剂或口服液中的至少一种。
在步骤6)中,可选择性地增加适量的食品增稠剂、食品添加剂以及相关剂型辅料。所述食品增稠剂为瓜尔胶、黄原胶、槐豆胶、魔芋胶、果胶、卡拉胶、琼脂、明胶、***胶中的至少一种,所述食品添加剂为阿斯巴甜、食用香精香料、红糖中的至少一种;更优选卡拉胶、***胶、瓜尔胶、阿斯巴甜、食用香精香料。
上述操作步骤均需在惰性气体的保护下进行,比如氮气、氩气、氦气等,以防止产生粉末***的危险。
按照所述配方可制备成粉状、颗粒状、胶囊状、片剂状、饮料等类别。
验证实施例1
选取大鼠随机平均分配到实验组、对比组和未处理组。
实验组1:喂养含有实施例1制备的营养粉剂的饲料(100g饲料加5g营养粉剂);
实验组2:喂养含有实施例1制备的营养粉剂以及鼠李糖乳杆菌、双歧杆菌、酶解珍珠粉、乌鸡粉、牡丹油、胆碱和神经节苷脂的饲料(100g饲料加5g营养粉剂),其中相对于实施例1制备的营养粉剂,鼠李糖乳杆菌、双歧杆菌、酶解珍珠粉、乌鸡粉、牡丹油、胆碱和神经节苷脂的添加量均为0.2g。
对比组:喂养含有CN101120784A实施例1制备的健脑食品的饲料(100g 饲料加1.5g健脑食品);
对照组:喂养普通饲料。
经过30天的喂养后,在喂养期间,各大鼠均未异常症状和体征,也无死亡。总食物利用率与对照组相比均无明显差异;各组血红蛋白含量、红细胞计数、白细胞计数及分类均在正常范围内,各项生化指标均在在正常范围内,各主要脏器大体解剖检测未发现明显病变。以上验证了本发明所述的营养配方并不会影响大鼠的正常生长及发育,其为可安全食用的产品。
验证实施例2
筛选小儿脑发育不全患者100名进行临床观察,并随机分为两组,实验组1、实验组2和对照组(每组50人)。实验组1使用本实施例1所得的营养液产品,实验组2使用实施例1以及鼠李糖乳杆菌、双歧杆菌、酶解珍珠粉、乌鸡粉、牡丹油、胆碱和神经节苷脂所得的营养液产品,实验组1和2的服用方法:口服, 1岁以下每次10ml,1~6岁每次20ml,7~14岁每次30ml,每日2次。对照组使用常规治疗小儿脑发育不全的药品(如脑神经节苷脂注射液、脑蛋白水解物、鼠神经生长因子、奥拉西坦注射液等)。
疗效情况判定标准:显效:经服用30天后,临床症状及体征基本消失;好转:经服用30天后,临床症状及体征明显好转;无效:经服用30天后,临床症状及体征略有改善或无改善。总有效包括显效和好转。结果如表1所示:
由上表可知,与对照组(现有制剂)相比,本发明中按实施例配制的营养配方对于小儿脑发育不全患者具有较优异的疗效,这表明本发明采用的营养配方中各组分组合配伍使用,产生了协同增效的有益效果。
由上述实施例可知,本发明所述的小儿脑发育专用型临床营养配方依据孕妇及小儿的生理特点设计,在保证蛋白质、脂肪、碳水化合物、膳食纤维的基础上,补充许多人体所需的常量元素和微量元素,并添加具有健脑益智、健胃消食、强免疫力等功效的药食同源成分、新资源食品等特殊营养素,使得该配方营养均衡,有效地保证小儿脑发育,保证孕妇及婴幼儿的身心健康成长。

其中,所述药食同源成分包括甘草、枸杞、橘皮、黑芝麻、益智仁、黄精、牡蛎、茯苓、山药;所述脂肪包括紫苏籽油、DHA藻油微囊粉、花生四烯酸油脂微囊粉、磷脂;所述新资源食品包括γ-氨基丁酸、低聚甘露糖。

3.根据权利要求1所述的一种小儿脑发育专用型临床营养配方,其特征在于,所述蛋白质包括氨基酸,所述氨基酸包括牛磺酸,所述牛磺酸的重量份数为0.1~0.3份。
4.根据权利要求1所述的一种小儿脑发育专用型临床营养配方,其特征在于,所述药食同源成分包括0.02-1份甘草、0.02-1份枸杞、0.02-1份橘皮、0.02-1份黑芝麻、0.02-1份益智仁、0.02-1份黄精、0.02-1份牡蛎、0.02-1份茯苓、0.02-1份山药。
5.根据权利要求1所述的一种小儿脑发育专用型临床营养配方,其特征在于,所述磷脂的重量份数为0.2~1份。
6.根据权利要求1所述的一种小儿脑发育专用型临床营养配方,其特征在于,所述营养配方包括麦芽糊精、水解乳清蛋白粉、紫苏籽油、低聚果糖、大豆分离蛋白、抗性糊精、DHA藻油微囊粉、花生四烯酸油脂微囊粉、海带粉、甘草、枸杞、橘皮、黑芝麻、益智仁、黄精、牡蛎、茯苓、山药、低聚甘露糖、γ-氨基丁酸、钙、铁、锌、维生素A、β-胡萝卜素、维生素D3、维生素E、维生素B1、维生素B2、维生素B6、维生素B12、维生素C、烟酸、叶酸、泛酸、牛磺酸、磷脂。
7.根据权利要求6所述的一种小儿脑发育专用型临床营养配方,其特征在于,所述营养配方还包括酶解珍珠粉、乌鸡粉、牡丹油、胆碱和神经节苷脂。
8.根据权利要求6所述的一种小儿脑发育专用型临床营养配方,其特征在于,所述营养配方还包括有益菌种微生物,所述有益菌种微生物选自肠道共生菌脆弱拟杆菌、双歧杆菌、嗜酸乳杆菌、鼠李糖乳杆菌中的至少一种。
9.一种如权利要求1~8中任一项所述的小儿脑发育专用型临床营养配方的制备方法,其特征在于,包括如下步骤:
步骤(1)预定重量的脂肪、脂溶性维生素、水溶性维生素分别采用微囊包埋技术制备并过60~80目筛制成粉末;
步骤(2)预定重量的药食同源成分通过粉碎、醇提、水提、喷雾干燥并过60-80目筛制成粉末;
步骤(3)预定重量的其余组分分别制备成粒径为150~250μm的粉末;
步骤(4)将步骤(1)~(3)制备的粉末混合均匀后杀菌备用;
步骤(5)将步骤(4)混合后的粉末制成可直接食用的配方制剂;
其中,步骤(1)~(3)的顺序可相互更换;所述配方制剂包括营养粉、营养乳剂、胶囊剂、片剂、丸剂、口服液。
10.根据权利要求1~8中任一项所述的小儿脑发育专用型临床营养配方的制备方法,其特征在于,包括如下步骤:
步骤1)准备含有预定重量份数的蛋白质和氨基酸、脂肪、碳水化合物、膳食纤维、常量元素、微量元素、维生素、药食同源成分以及新资源食品的原材料粉末,在预定量的水中,按照原材料的物理性状,按照先后顺序依次添加,在添加的同时进行搅拌,制得原料的混合乳液;
步骤2)将步骤1)获得的混合乳液进行乳化均质、浓缩杀菌,获得浓缩液;
步骤3)将步骤2)获得的浓缩液进行喷雾干燥,过筛,制备成粉末,并将上述粉末制备成营养粉、营养乳剂、胶囊剂、片剂、丸剂或口服液中的至少一种。
