背景

发明概述
本公开提供具有果香、萝卜和/或药草香气味香型与强烈头香的香料化合物。这些化合物可以单独使用或掺入香料组合物和/或消费品中。

2.制备香料化合物的方法
式I的香料化合物可根据本领域已知的方法制备。作为非限制性实例,式I的香料化合物可由酯原料制备。使酯与格氏试剂反应和随后的含水酸性后处理可以产生叔醇。格氏试剂可以按照本领域已知的方法制备,并且可以优选是卤化烷基镁,例如溴化烷基镁。对于每1摩尔酯,优选格利雅反应以至少2摩尔格氏试剂的摩尔比进行。格利雅反应的叔醇产物的脱水反应可以通过在酸存在下加热进行,以产生烯烃。烯烃与醇的加成反应可以得到式I的香料化合物。例如,但不限于,该加成反应可以按照本领域已知的方法进行,例如通过用强酸处理或通过羟汞化。为了示例的目的,该方法在实施例1中描述,其中初始酯是己酸乙酯,格氏试剂是溴化甲基镁,叔醇是2-羟基-2-甲基庚烷,烯烃是2-甲基-1-庚烯和2-甲基-2-庚烯的混合物,且最终的香料化合物是2-甲氧基-2-甲基庚烷。

本公开主题的香料化合物可以配制成不同的香料组合物。例如,本公开主题的香料组合物可包括一种或更多种、两种或更多种或三种或更多种上述香料化合物。
香料化合物至香料组合物的配制横跨从“简单谐香”(<10种香料化合物)到“复杂香料”(>30种香料化合物)的范围。例如,浓郁的香料组合物通常不包含少于约30种的香料化合物。这样的香料组合物还可含有至少一种香料基质、溶剂或其组合或由至少一种香料基质、溶剂或其组合组成。所述香料基质或溶剂可以是液体或固体,并且典型地不会显著改变香料化合物的嗅觉性质。

5.消费品
在某些实施方案中,可以将本公开的香料化合物和组合物配制成消费品的一部分。例如,但不限于,香料化合物和/或组合物可用于香水,古龙香水,洗发剂,漂乳液,护肤品,沐浴露,沐浴乳(body rinses),沐浴粉(body powders),空气清新剂,除臭剂,浴液,食品,零食,饮料等,如果需要与辅助材料结合使用。
如本文所体现的,式I的化合物可单独使用或掺入香料组合物中,其可有利地用于各种消费品中。例如,消费品可以是用于给合适的基材加香的那些。如本文所用,术语“基材”是指可以施用消费品的任何表面,而不会引起任何不适当的不利影响。基材可以是多种材料,包括人或动物皮肤或毛发,纸(例如加香纸),室内空气,织物,家具,餐具,硬表面和相关材料。因此,基材可包括体表,例如毛发和皮肤。

实施例
通过参考以下实施例将更好地理解本公开主题,所述实施例作为所公开主题的示例提供,而不是作为限制。
实施例1:由己酸乙酯制备2-羟基-2-甲基庚烷
在氮气氛下,通过套管向配备冷凝器、加料漏斗和入口连接器的烘干的2L三颈圆底烧瓶中加入800ml 3M溶液溴化甲基镁的***溶液(2.4mol)。向加料漏斗中加入150g(1.04mol)己酸乙酯在100mL无水***中的溶液。将圆底烧瓶浸入冰浴中,并在2小时内滴加加料漏斗中的溶液。加完后,除去冰浴,将溶液再搅拌4小时直至反应完成。将圆底烧瓶再次浸入冰浴中,通过逐滴加入1N HCl淬灭反应直至气体停止放出。此时,以更快的速率加入1NHCl,直到所有固体溶解并且有机层变为澄清和黄色。将混合物转移到分液漏斗中,除去底部水层。然后用3×500ml 1N HCl洗涤有机层。然后将含水洗涤液合并,并用500ml***再萃取。然后将合并的有机层用水和盐水洗涤,在旋转蒸发器上谨慎地除去溶剂,得到127.85g粗2-羟基-2-甲基庚烷,为黄色油状物。
2-羟基-2-甲基庚烷的脱水
将上述粗产物转移到配备简单蒸馏装置的圆底烧瓶中。为了促进脱水,将一滴硫酸加入到粗产物中,并将混合物加热至160℃。蒸馏到接收器中的产物是2-甲基-1-庚烯、2-甲基-2-庚烯和水的混合物,水沉淀到底层中。蒸馏完成后,将馏出物放入分液漏斗中,除去底部水层。然后将烯烃混合物用无水硫酸镁干燥,得到98.64g产物,将其不经进一步纯化用于下一步。
2-甲基-1(2)-庚烯甲氧基化成2-甲氧基-2-甲基庚烷
向配备加料漏斗和冷凝器和氮气入口的1L双颈圆底烧瓶中加入40mL浓硫酸和250mL无水甲醇,将所得溶液加热至35℃并搅拌15分钟。向加料漏斗中加入98.64g(0.88mol)前一步骤的2-甲基-1-庚烯和2-甲基-2-庚烯的混合物。在1小时内将该溶液逐滴加入到反应体系中,并将所得混合物在35℃下再搅拌6小时直至反应完成。然后将反应混合物转移到分液漏斗中并使其沉降。分离顶部有机层,底层用3×250ml***萃取。将这些合并的***萃取液加入到原始有机层中,用无水硫酸镁干燥。然后将溶液谨慎地在旋转蒸发器上浓缩,得到96.11g粗产物。然后将产物通过填充有不锈钢填料的1'x 2”分馏塔分馏,得到63.97g纯的2-甲氧基-2-甲基庚烷。该方法基于起始的己酸乙酯的总收率为43%(b.p.38-41℃@35Torr)。
1H NMR(500MHz,CDCl3)δ:0.86(t,3H),1.10(s,6H),1.20-1.30(m,6H),1.42(dd,2H),3.14(s,3H)。
本实施例的化合物具有多方面气味特征并且未落入一种类别。其为辛香、清新、青香和果香。
实施例2:含有2-甲氧基-2-甲基庚烷的香料组合物和洗发剂制剂
由实施例1的化合物(2-甲氧基-2-甲基庚烷)制备具有山葵莲花香型(a wasabilotus note)的香料组合物,以证明其在洗发剂中的用途。含有和不含2-甲氧基-2-甲基庚烷的制剂如下表1中所示:

洗发剂中使用0.8%的香料,并且由五位专家组成的小组对样品进行了评估。
发现与本实施例不含实施例1的化合物的组合物相比,使用实施例1的化合物((2-甲氧基-2-甲基庚烷)的本实施例的组合物具有添加的天然清鲜香型,特别是在头香,从而整体上开放了香味。清鲜度来自实施例1的化合物的青香辛香,其赋予组合物更自然的花香特征。
实施例3:使用2-甲氧基-2-甲基庚烷的漂白稳定性研究
对具有pH 12.62和起始氯(NaOCl)百分比7.99%的市售样品漂白剂中的2-甲氧基-2-甲基庚烷进行漂白稳定性测试。以0.05%施用样品。将样品储存在室温(RT)和45℃下。2周和4周后,定性地评估享乐度(即香味)。测定氯的量(%NaOCl),以便确定与7.99%的起始百分比相比氯的损耗量(%损耗)。结果如下表2中所示,其中“对照”是指单独的漂白剂且“实施例1”是指施用了2-甲氧基-2-甲基庚烷的漂白剂。

实施例4:7-甲氧基-2,4-二甲基辛-2-烯的合成
在惰性气氛中制备氢化钠(0.85g,0.035mol)在无水四氢呋喃(40mL)中的混悬液。然后滴加5,7-二甲基辛-6-烯-2-醇(5.0g,0.032mol),历时20分钟期限,在此期间,使该混合物温度升至75℃。再搅拌24小时后,加入碘甲烷(4.55g,0.032mol),将该混合物在75℃搅拌至通过气相色谱分析显示不再有起始醇存在为止。将该混合物冷却至室温,然后加入100mL***。用饱和碳酸氢钠溶液(2 x 75mL)和盐水(50mL)洗涤得到的溶液。分离有机相,用无水硫酸镁干燥,过滤,减压除去溶剂。通过蒸馏纯化粗产物,得到4.79g(88%收率)的无色油状物(b.p.35-38℃@1Torr)。
1H NMR(400MHz,CDCl3)δ:0.88(d,3H),1.07(d,3H),1.27(m,2H),1.34(m,2H),1.58(s,3H),1.65(s,3H),2.26(m,1H),3.23(m,1H),3.29(s,3H),4.86(d,1H)。
实施例5:7-甲氧基-2,4,7-三甲基辛-2-烯的合成
使用与实施例4中所示相同的方法,由碘甲烷(4.17g,0.029mol)和2,5,7-三甲基辛-6-烯-2-醇(5.0g,0.029mol)制备7-甲氧基-2,4,7-三甲基辛-2-烯。
通过蒸馏纯化粗产物,得到4.22g(78%收率)无色油状物(b.p.60-62℃@1.4Torr)。
1H NMR(400MHz,CDCl3)δ:0.90(d,3H),1.09(d,6H),1.28(m,2H),1.38(m,2H),1.57(s,3H),1.65(s,3H),2.23(m,1H),3.14(s,3H),4.85(d,1H)。
实施例6:7-(烯丙基氧基)-2,4-二甲基辛-2-烯的合成
使用与实施例4中所示相同的方法,由3-溴丙-1-烯(1.42g,0.012mol)和5,7-二甲基辛-6-烯-2-醇(2.0g,0.012mol)制备7-甲氧基-2,4,7-三甲基辛-2-烯。
通过蒸馏纯化粗产物,得到1.96g(85%收率)淡黄色油状物(b.p.46-49℃@0.15Torr)。
1H NMR(400MHz,CDCl3)δ:0.89(d,3H),1.13(d,3H),1.30(m,2H),1.47(m,2H),1.57(s,3H),1.66(s,3H),2.27(m,1H),3.38(m 1H),3.95(dm,2H),4.84(d,1H),5.13(d,1H),5.24(d,1H),5.89(m,1H)。
实施例7:7-甲氧基-2,4-二甲基辛烷的合成
使用与实施例4中所示相同的方法,由碘甲烷(2.30g,0.019mol)和5,7-二甲基辛-2-醇(3.0g,0.019mol)制备7-甲氧基-2,4-二甲基辛烷。
通过蒸馏纯化粗产物,得到2.54g(78%收率)无色油状物(b.p.43-47℃@1.5Torr)。
1H NMR(400MHz,CDCl3)δ:0.77(d,3H),0.79(d,3H),0.83(d,3H),0.96(m,1H),1.06(m,1H),1.08(d,3H),1.20-1.54(m,5H),1.60(m,1H),3.21(dt,1H),3.28(s,3H)。
实施例8:4-(烯丙基氧基)-2,6-二甲基庚烷的合成
使用与实施例4中所示相同的方法,由3-溴丙-1-烯(4.20g,0.035mol)和2,6-二甲基庚-4-醇(5.0g,0.035mol)制备4-(烯丙基氧基)-2,6-二甲基庚烷。
通过蒸馏纯化粗产物,得到4.48g(70%收率)无色油状物(b.p.70-73℃@1Torr)。
1H NMR(400MHz,CDCl3)δ:0.79(d,6H),0.82(d,6H),1.18(m,2H),1.39(dd,2H),1.68(dd,2H),3.37(bt,1H),3.94(d,2H),5.12(d,1H),5.22(d,1H),5.88(m,1H)。
实施例9:2,6-二甲基-4-((2-甲基烯丙基)氧基)庚烷的合成
使用与实施例4中所示相同的方法,由3-氯-2-甲基丙-1-烯(3.14g,0.035mol)和2,6-二甲基庚-4-醇(5.0g,0.035mol)制备2,6-二甲基-4-((2-甲基烯丙基)氧基)庚烷。
通过蒸馏纯化粗产物,得到5.29g(77%收率)无色油状物(b.p.74-75℃@0.8Torr)。
1H NMR(400MHz,CDCl3)δ:0.79(d,6H),0.81(d,6H),1.19(m,2H),1.44(m,2H),1.68(m,2H),1.72(s,3H),3.36(bt,1H),3.81(s,2H),4.80(s,1H),4.93(s 1H)。
实施例10:2-甲基-4-((2-甲基烯丙基)氧基)戊烷的合成
使用与实施例4中所示相同的方法,由3-氯-2-甲基丙-1-烯(2.22g,0.025mol)和4-甲基戊-2-醇(2.5g,0.025mol)制备2-甲基-4-((2-甲基烯丙基)氧基)戊烷。
通过蒸馏纯化粗产物,得到1.95g(51%收率)无色油状物(b.p.50-52℃@19Torr)。
1H NMR(400MHz,CDCl3)δ:0.80(d,3H),0.84(d,3H),1.11(d,3H),1.15(m,1H),1.48(m,1H),1.69(s,3H),1.72(m,1H),3.44(m,1H),3.75(d,1H),3.91(d,1H),4.80(s,1H),5.12(s,1H)。
实施例11:2-(烯丙基氧基)-4-甲基戊烷的合成
使用与实施例4中所示相同的方法,由3-溴丙-1-烯(2.96g,0.025mol)和4-甲基戊-2-醇(2.5g,0.025mol)制备2-(烯丙基氧基)-4-甲基戊烷。
通过蒸馏纯化粗产物,得到1.80g(52%收率)无色油状物(b.p.42-43℃@30Torr)。
1H NMR(400MHz,CDCl3)δ:0.79(d,3H),0.81(d,3H),1.08(d,3H),1.13(m,1H),1.45(m,1H),1.70(m,1H),3.46(m,1H),3.83(m,1H),4.01(m,1H),5.10(d,1H),5.22(d,1H),5.86(m,1H)。
实施例12:2-甲基-4-((3-甲基丁-2-烯-1-基)氧基)戊烷的合成
使用与实施例4中所示相同的方法,由1-氯-3-甲基丁-2-烯(3.07g,0.029mmol)和4-甲基戊-2-醇(3.0g,0.029mol)制备2-甲基-4-((3-甲基丁-2-烯-1-基)氧基)戊烷。
通过蒸馏纯化粗产物,得到3.05g(61%收率)无色油状物(b.p.53-55℃@5.5Torr)。
1H NMR(400MHz,CDCl3)δ:0.82(d,3H),0.84(d,3H),1.09(d,3H),1.16(m,1H),1.45(m,1H),1.62(s,3H),1.69(s,3H),1.70(m,1H),3.42(m,1H),3.83(m,1H),3.97(m,1H),5.31(bt,1H)。
实施例13:含有2-甲氧基-2-甲基庚烷的香料组合物
由实施例1的化合物(2-甲氧基-2-甲基庚烷)制备香料组合物,以掺入石榴玫瑰香洗碗液中。将制剂提供于下表3中:

3.权利要求1的香料组合物,还包含另外的香料化合物,所述另外的香料化合物选自花香化合物,美食味化合物,青香化合物,麝香化合物,辛香化合物,木香化合物及其组合。
4.权利要求1的香料组合物,还包含另外的香料化合物,所述另外的香料化合物为果香化合物。
5.权利要求1的香料组合物,还包含另外的香料化合物,所述另外的香料化合物为柑橘香化合物。
6.权利要求1的香料组合物,还包含溶剂。
7.改变香料组合物气味特性的方法,通过向所述香料组合物中添加嗅觉有效量的香料化合物来进行,所述香料化合物选自:
2-甲氧基-2-甲基庚烷;

7-甲氧基-2,4-二甲基辛烷;
