本发明属于环境重金属污染的修复领域,具体公开了一种提高植物对重金属吸收转运能力的复合药剂及其制备方法和应用,所述复合药剂适用于各种被重金属污染的土壤或水体。本发明提供的复合药剂包括脱落酸类激素和水杨酸,将二者同时施加至种植有植物的被污染土壤或水体中,能有效地促进植物对重金属的吸收转运能力,提高其向地上部转运系数,利于收割富集,具备广阔的应用前景。

技术领域
本发明属于环境重金属污染的修复领域,尤其涉及一种提高植物对重金属吸收转运能力的复合药剂及其制备方法和应用。所述复合药剂适用于各种被重金属污染的土壤或水体。
背景技术
随着现代科技和经济的迅猛发展,工业“三废”的排放、污水灌溉、污泥农用、农药和化肥的长期不合理施用,造成了严重的水体和土壤重金属污染,并成为一个被广泛关注的难题。特别地,对于土壤污染,2014年4月,环保部和国土资源部联合发布了全国首次土壤污染状况调查公报,公报指出,全国土壤环境状况总体不容乐观,土壤污染总超标率为16.1%,部分地区土壤污染较重,其中污染类型以重金属污染为主。从污染物超标情况看,镉、汞、砷、铜、铅、铬、锌、镍等重金属污染物赫然在列。这已严重影响到耕地质量、食品安全乃至人类的身体健康和社会的稳定。
目前,重金属污染土壤的修复技术主要包括:(1)工程修复,如客土、换土和深耕翻土等,此方法工程量大,投资费用高,不适合大面积污染土壤的治理;(2)物理修复,如电动修复和电热修复,对污染重、面积小的土壤修复效果明显,但容易导致土壤结构和理化性质破坏,降低土地的使用功能和应用价值;(3)化学修复,如化学淋洗、化学钝化等。效果较好、操作简单。但使用范围窄,容易造成二次污染。(4)生物修复,如微生物修复和植物修复,由于微生物修复技术的局限性,应用实例不多,而植物修复利用植物将重金属从土壤中富集于植物内,最终通过植物收割即可将重金属从土壤中脱离,对土壤的扰动小,对环境也更加友好,在重金属污染土壤修复中显示出了良好的应用前景。适用于各区域重金属污染土壤或水体的修复。
传统的重金属污染土壤进行植物修复技术的研究主要集中在寻找超富集植物上,利用其具有超积累某种重金属的特性,从而达到修复重金属污染土壤的目的。但是重金属超富集植物普遍存在生物量小、生长速率低以及对土壤净化时间长等缺陷,限制了植物修复的发展及推广应用。故通过提高植物修复效率,如采用大生物量植物,提高植物重金属吸收转运能力,进而使植物累积的重金属重量得到提高,缩短净化年限,促进植物修复的发展及应用迫在眉睫。
发明内容
针对现有技术中存在的不足,本发明的目的是在于提供了一种提高植物对重金属吸收能力的复合药剂,它由脱落酸(ABA)类激素和水杨酸(SA)按一定比例制成,该复合药剂是通过强化植物对重金属的吸收能力来提高植物修复效率。可广泛应用于各种被重金属污染的土壤或水体。
脱落酸和水杨酸,均是植物体内一种重要的内源信号分子,是介导环境胁迫和植物抗逆反应的重要调节物质,参与了植物应对生物胁迫与非生物胁迫反应中许多生理过程的调节。已有大部分报道称,通过单独施加外源水杨酸和脱落酸及其类似物中的任意一种能够提高植物对重金属胁迫的抗性,降低植物重金属的积累。但是对于植物修复重金属污染的土壤或水体,此方法显然只能起到缓解植物的重金属胁迫的作用,对于运用其进行修复,不具有竞争力,基本不可行。
而本发明采用脱落酸类激素和水杨酸作为复合药剂,有效地促进植物对重金属的吸收转运能力,提高其向地上部转运系数,利于收割富集。
本发明的另一个目的是在于提供了上述一种提高植物对重金属吸收转运能力的复合药剂的制备方法,操作简单,方法易行。
为了实现上述发明目的,本发明采用了以下技术措施:
一种用于提高植物对重金属吸收转运能力的复合药剂,其特征在于:所述复合药剂包括脱落酸类激素和水杨酸,所述脱落酸类激素为脱落酸和/或脱落酸的模拟物;优选的,所述复合药剂由脱落酸类激素和水杨酸组成。
所述脱落酸的模拟物优选为现有化合物8'-乙炔基ABA,其结构式如下:
与此同时,本发明还提供了一种上述复合药剂在利用植物对重金属污染区域进行修复中的应用,所述重金属污染区域为土壤或者水体,优选为水体。
所述植物可以为水稻、东南景天、高羊茅或印度芥菜,优选为高羊茅或印度芥菜。
所述重金属优选为镉或铜。
当所述重金属污染区域为水体时,所述复合药剂的施用浓度(施用到水体中的浓度)为脱落酸类激素0.5-15mg/L,水杨酸0.5-1.5mmol/L;
优选的,所述复合药剂的施用浓度为脱落酸类激素0.5-5mg/L,水杨酸0.5-1.5mmol/L;
更优选的,所述复合药剂的施用浓度为脱落酸类激素5mg/L,水杨酸0.5mmol/L。
应用时,直接向种植有植物的重金属污染区域(如水体)投加上述制备复合药剂的原料脱落酸类激素和水杨酸,使其混合均匀后达到所述施用浓度,一段时间后,收集该区域植物地上部;进一步,可灰化植物体进行重金属提取,回收利用。
与现有技术相比,本发明的优点和有益效果在于:
A、水杨酸和脱落酸按照一定比例联合添加与对照组相比,提高了植物地上部重金属含量6-15倍。
B、本发明适用的植物种类多样,无论是生物量大的高羊茅,还是超富集植物印度芥菜等都有显著效果。
C、本发明适用的重金属种类繁多,有在全国首次土壤污染状况调查公报以7%的点位超标率首居其中的镉、点位超标率为2.1%的铜等全国超标严重的重金属污染物。
D、本发明适用范围广,只需满足植物生长的条件,即可运用,包括严重污染的矿山、大面积污染的农田等土壤;排放的含重金属废水、重金属污染的河流湖泊等水体。
E、本发明提供的复合药剂,制备过程操作简单,方法易行。施用方式简单、灵活,便于使用管理,推广应用前景广阔。
F、本发明提供的复合药剂中ABA组分可由廉价的ABA模拟物代替,大大降低复合药剂的制作成本,有利于推广应用。

具体实施方式
以下结合具体实施例对本发明做进一步的详细描述,但并非是对本发明权利要求书请求保护范围的限制,凡依照本发明公开内容所作的任何本领域的等同替换,均落入本发明的保护范围。
实施例1:
供试材料在实验室的光照培养温室内进行培养,材料培养与处理如下:
(1)将30颗高羊茅(品种为猎狗五号,以下实施例同)种子播种在长、宽、高分别为10cm、10cm、8cm的穴盒中,穴盒中的基质按体积比珍珠岩:蛭石=1:1配成。在温室可控条件下(昼夜温度分别为24℃、22℃;光照强度为300μmol/(m2·s1);光周期为光照14小时,黑暗10小时),施浇1/2Hoagland营养液培养将近1个月至高羊茅幼苗长出。
(2)将12株(1)中高羊茅幼苗转入装有300mL1/2Hoagland营养液的250mL锥形瓶(锡箔纸包裹避光)中适应约10天至有新生根长出。
(3)接下来的试验设置4个处理组:对照组(不施用任何激素)、脱落酸处理(只施用脱落酸)、水杨酸处理(只施用水杨酸)、激素复合药剂处理(脱落酸和水杨酸复合处理),每个处理组进行4个重复,每个重复均含12株高羊茅,在300mL的1/2Hoagland营养液中处理,结果取平均值,以下实施例2-4均按此用量(12株高羊茅置于300mL的1/2Hoagland营养液中)进行试验。所有处理组分别在初始含镉(Cd2+)浓度为20mg/L的1/2Hoagland营养液的水培环境下进行(镉溶液用CdCl2·2.5H2O配制),处理三天。三天后,先冲洗,后取其地上部于60-70℃烘干至恒重,剪碎,混合均匀,取0.3g(镉含量计算时,以烘干至恒重后取的质量为分母),分别加8mL硝酸-过氧化氢(由65wt%硝酸与30wt%过氧化氢按照体积比例为3:1混合而得)微波消解,过膜后,用火焰原子吸收分光光度计法测各处理的重金属镉含量。试验各处理组的药剂施用浓度如下表:
注:实施例中所有处理均是通过药剂原料直接投入到含重金属离子的营养液中,施用浓度指药剂投入到含重金属离子的营养液后的浓度。以下实施例同。
结果见表1-1:

结果表明,相对于对照组的地上部,脱落酸处理的地上部镉含量降低了63%,水杨酸处理的地上部镉含量提高了1.97倍,而激素复合处理的地上部却提高了8.91倍。
实施例2:
材料培养和处理第(1)、第(2)步同实施例1。
实施例2在初始含镉浓度为20mg/L的1/2Hoagland营养液的水培环境下进行(镉溶液用CdCl2·2.5H2O配制),试验设置两个处理:对照组(不施用任何激素)和激素复合药剂处理(0.5mg/L脱落酸和1.5mmol/L水杨酸复合处理),每个处理进行4个重复,处理五天,结果取平均值。五天后,先冲洗,后取其地上部于60-70℃烘干至恒重,剪碎,混合均匀,取0.3g(镉含量计算时,以烘干至恒重后取的质量为分母),分别加8mL硝酸-过氧化氢(由65wt%硝酸与30wt%过氧化氢按照体积比例为3:1混合而得)微波消解,过膜后,用火焰原子吸收分光光度计法测各处理的重金属镉含量。结果见表2-1:

实施例3:

实施例4在初始含铜浓度为200mg/L的1/2Hoagland营养液的水培环境下进行(铜溶液用CuSO4·5H2O配制),试验设置两个处理:对照组(不施用任何激素)和激素复合药剂处理(5mg/L脱落酸和0.5mmol/L水杨酸复合处理),每个处理进行4个重复,处理四天,结果取平均值。四天后,先冲洗,后取其地上部于60-70℃烘干至恒重,剪碎,混合均匀,取0.3g(铜含量计算时,以烘干至恒重后取的质量为分母),分别加8mL硝酸-过氧化氢(由65wt%硝酸与30wt%过氧化氢按照体积比例为3:1混合而得)微波消解,过膜后,用火焰原子吸收分光光度计法测各处理的重金属铜含量。结果见表4-1:

结果表明,相对于对照组的地上部,激素复合药剂处理的地上部铜含量提高了14.65倍。
对比实施例1和实施例4可知,本发明的一种提高植物对重金属吸收的复合药剂适用的重金属种类繁多,既可用于镉污染的土壤或水体,也可用于铜污染的土壤或水体。
实施例5:
供试材料在实验室的光照培养温室内进行培养,材料培养与处理如下:
(1)将5颗印度芥菜种子播种在在长、宽、高分别为10cm、10cm、8cm的穴盒中,穴盒中的基质按体积比珍珠岩:蛭石=1:1配成。在温室可控条件下(昼夜温度分别为24℃、22℃;光照强度为300μmol/(m2·s1);光周期为光照14小时,黑暗10小时),施浇1/2Hoagland营养液培养将近20天。
(2)将(1)中2棵印度芥菜幼苗转入装有1/2Hoagland营养液的250mL锥形瓶(锡箔纸包裹避光)中适应约一周至有新生根长出。
(3)实施例5在初始含镉浓度为20mg/L的1/2Hoagland营养液的水培环境下进行(镉溶液用CdCl2·2.5H2O配制),试验设置两个处理:对照组(不施用任何激素)和激素复合药剂处理(5mg/L脱落酸和0.5mmol/L水杨酸复合处理),每个处理进行4个重复,每个重复均含2棵印度芥菜,在300mL的1/2Hoagland营养液中处理,处理三天,结果取平均值。三天后,先冲洗,后分别取其地上部和根部于60-70℃烘干至恒重,剪碎,混合均匀,各取0.3g(镉含量计算时,以烘干至恒重后取的质量为分母),分别加8mL硝酸-过氧化氢(由65wt%硝酸与30wt%过氧化氢按照体积比例为3:1混合而得)微波消解,过膜后,用火焰原子吸收分光光度计法测各处理的重金属镉含量。结果见表5-1:

结果表明,相对于对照组的地上部,激素复合药剂处理的地上部(S)镉含量提高了7.91倍,相对于对照组的根部(R),激素复合药剂处理的根部镉含量提高了9.69倍。
对比实施例1和实施例5可知,本发明的一种提高植物对重金属吸收转运能力的复合药剂适用的植物种类多样,无论是生物量大的高羊茅,还是超富集植物印度芥菜等都有显著效果。
实施例6:
供试材料在实验室的光照培养温室内进行培养,材料培养与处理如下:
(1)将30颗水稻种子(丰两优香一号)播种在长、宽、高分别为10cm、10cm、8cm的穴盒中,穴盒中的基质按体积比珍珠岩:蛭石=1:1配成。在温室可控条件下(昼夜温度分别为28℃、20℃;光照强度为210μmol/(cm2·s1);光周期为光照13小时,黑暗11小时),施浇1/2Hoagland营养液培养将近1个月至水稻幼苗长出。
(2)将12株(1)中水稻幼苗转入装有300mL 1/2Hoagland营养液的250mL锥形瓶(锡箔纸包裹避光)中适应约10天至有新生根长出。
(3)接下来的试验设置2个处理组:对照组(不施用任何激素)、激素复合药剂处理(2.5mg/L脱落酸和1.0mmol/L水杨酸复合处理),每个处理组进行4个重复,每个重复均含12株水稻,在300mL的1/2Hoagland营养液中处理,结果取平均值。所有处理组分别在初始含镉(Cd2+)浓度为20mg/L的1/2Hoagland营养液的水培环境下进行(镉溶液用CdCl2·2.5H2O配制),处理三天。三天后,先冲洗,后取其地上部于60-70℃烘干至恒重,剪碎,混合均匀,取0.3g(镉含量计算时,以烘干至恒重后取的质量为分母),分别加8mL硝酸-过氧化氢(由65wt%硝酸与30wt%过氧化氢按照体积比例为3:1混合而得)微波消解,过膜后,用火焰原子吸收分光光度计法测各处理的重金属镉含量。结果见表6-1:

结果表明,相对于对照组的地上部,激素复合药剂处理的地上部镉含量提高了8.01倍。
实施例7:
供试材料在实验室的光照培养温室内进行培养,材料培养与处理如下:
(1)从商家购买的东南景天幼苗中挑选等大小的地上部转入装有300mL 1/2Hoagland营养液的250mL锥形瓶(锡箔纸包裹避光)中适应约14天至有新生根长出。
(2)接下来的试验设置2个处理组:对照组(不施用任何激素)、激素复合药剂处理(10mg/L脱落酸和1.0mmol/L水杨酸复合处理),每个处理组进行4个重复,每个重复均含2株东南景天,在300mL的1/2Hoagland营养液中处理,结果取平均值。所有处理组分别在初始含镉(Cd2+)浓度为20mg/L的1/2Hoagland营养液的水培环境下进行(镉溶液用CdCl2·2.5H2O配制),处理三天。三天后,先冲洗,后取其地上部于60-70℃烘干至恒重,剪碎,混合均匀,取0.3g(镉含量计算时,以烘干至恒重后取的质量为分母),分别加8mL硝酸-过氧化氢(由65wt%硝酸与30wt%过氧化氢按照体积比例为3:1混合而得)微波消解,过膜后,用火焰原子吸收分光光度计法测各处理的重金属镉含量。结果见表7-1:

实施例8:
材料培养和处理第(1)、第(2)步同实施例1。
实施例8在初始含镉浓度为20mg/L的1/2Hoagland营养液的水培环境下进行(镉溶液用CdCl2·2.5H2O配制),试验设置两个处理:对照组(不施用任何激素)和激素复合药剂处理(与0.5mg/L脱落酸等摩尔浓度的ABA模拟物(8'-乙炔基ABA)和0.5mmol/L水杨酸复合处理),每个处理进行4个重复,高羊茅用量与营养液体积同实施例1,处理三天,结果取平均值。三天后,先冲洗,后取其地上部于60-70℃烘干至恒重,剪碎,混合均匀,取0.3g(镉含量计算时,以烘干至恒重后取的质量为分母),分别加8mL硝酸-过氧化氢(由65wt%硝酸与30wt%过氧化氢按照体积比例为3:1混合而得)微波消解,过膜后,用火焰原子吸收分光光度计法测各处理的重金属镉含量。结果见表8-1:

结果表明,相对于对照组的地上部,激素复合药剂处理的地上部镉含量提高了8.30倍。
对照实施例1和8可知,本发明的一种提高植物对重金属吸收转运能力的复合药剂的脱落酸组分可以被脱落酸的模拟物替代。
Claims (5)
1.一种用于提高植物对重金属吸收转运能力的复合药剂,其特征在于:所述复合药剂由脱落酸类激素和水杨酸组成,所述脱落酸类激素为脱落酸或脱落酸的模拟物,所述脱落酸的模拟物为8'-乙炔基脱落酸,所述植物为高羊茅或印度芥菜。
2.权利要求1中所述的复合药剂在利用植物对重金属污染区域进行修复中的应用,所述重金属污染区域为土壤或者水体,所述的重金属为镉或铜。
3.根据权利要求2所述的应用,其特征在于:当所述重金属污染区域为水体时,所述复合药剂施用到水体中的浓度为脱落酸类激素0.5-15 mg/L,水杨酸0.5-1.5mmol/L。
4.根据权利要求3所述的应用,其特征在于:所述复合药剂施用到水体中的浓度为脱落酸类激素0.5-5 mg/L,水杨酸0.5-1.5mmol/L。
5.根据权利要求4所述的应用,其特征在于:所述复合药剂施用到水体中的浓度为脱落酸类激素5 mg/L,水杨酸0.5 mmol/L。
