Description

Traditional Chinese medicine composition for treating hyperlipidemia and preparation method and application thereof
Technical Field
The invention relates to a traditional Chinese medicine composition with a function of treating hyperlipidemia, and also relates to a preparation method and application of the composition.
Background
Hyperlipidemia, also known as hyperlipidemia or hyperlipoproteinemia, is a condition in which the plasma lipid concentration exceeds the normal upper limit. Blood lipid concentrations are affected by a variety of factors: cholesterol and triglycerides may increase with age; much meat is eaten more than long-term vegetable; patients with polyphagia of animal oil or cholesterol-rich food often have elevated blood cholesterol levels; the triglyceride level is often high in the case of polyphagia. In addition, hyperlipidemia is also common in a variety of diseases such as uncontrolled diabetes, hypothyroidism, atherosclerosis, coronary heart disease, stroke, nephrotic syndrome and certain inherited lipid metabolism disorders.
For the medicine for treating hyperlipemia, the western medicines mostly adopt statins, such as atorvastatin, simvastatin, rosuvastatin and the like; the traditional Chinese medicine compound has more varieties, different composing ideas, and mainly comprises the medicines for promoting blood circulation and removing blood stasis, and is supplemented with the medicines with the efficacies of clearing liver and improving vision, tonifying liver and kidney, descending turbidity and eliminating dampness, clearing heat and resolving masses and the like. For example, CN200810157509.1 discloses a Chinese medicine for treating hyperlipidemia, which is specifically prepared from processed rhizoma cyperi, radix aucklandiae, radix linderae, orange peel, pericarpium citri reticulatae viride, fructus aurantii immaturus, cortex magnoliae officinalis, fructus amomi, rhizoma nardostachyos, citron peel, radix salviae miltiorrhizae, ligusticum wallichii, peach kernel, safflower, herba lycopi, radix curcumae, rhizoma curcumae longae, rhizoma corydalis, trogopterus dung, frankincense, myrrh, lignum dalbergiae odoriferae, radix astragali, rhizoma atractylodis macrocephalae, rhizoma polygonati, cotton root, bidens tripartita, hawthorn, medicated leaven, malt, rice. For another example, CN201310691315.0 discloses a Chinese medicinal composition for treating hyperlipidemia, which is prepared from the following raw materials in parts by weight: 10-15 parts of salvia miltiorrhiza, 15-20 parts of turmeric, 12-18 parts of bighead atractylodes rhizome, 16-22 parts of poria cocos, 10-15 parts of eucommia ulmoides, 5-10 parts of wild jujube leaves, 12-18 parts of scutellaria baicalensis, 15-20 parts of curcuma aromatica, 10-15 parts of rhizoma cyperi, 5-10 parts of radix scrophulariae, 10-15 parts of loranthus parasiticus, 8-14 parts of Chinese wampee leaves, 5-10 parts of rhizoma alismatis, 6-12 parts of semen cassiae, 10-15 parts of astragalus membranaceus, 5-10 parts of coix seeds, 10-15 parts of yellow lotus flowers, 8-14 parts of fructus psoraleae, 10-15 parts of Chinese yams, 12-18 parts of rhizoma atractylodis, 15-20 parts of potentilla discolor, 6-12 parts of polygonum multiflorum. The medicines have many ingredients, complicated administration, unclear main lines and large prescription amount.
In another idea, for example, CN201510013731.4 discloses a traditional Chinese medicine health product for assisting in reducing blood lipid, angelica sinensis and astragalus membranaceus lipid-lowering particles, which adopts five medicines with blood lipid-lowering efficacy, including, by weight, 8-12 parts of angelica sinensis, 12-18 parts of astragalus membranaceus, 12-18 parts of radix puerariae, 8-12 parts of rhizoma alismatis, and 10-15 parts of hawthorn. The formulation idea of the patent document is to throw away the principle of traditional Chinese medicine formulation and directly combine a plurality of medicines with the function of reducing blood fat together for formulation.

Experimental example 1
1. Animal model making and experiment method
The animals were male SD rats of SPF grade purchased from Biotech limited, Viton, Beijing, and weighing 150-170 g.
Animals were allowed to drink water freely at room temperature (23 + -2 deg.C) and molding was started 7 days after acclimatizing feeding. Feeding the blank rats with normal drinking water and basic feed; the molded rats normally drink water, and are fed with basal feed and simultaneously fed with high-fat emulsion for intragastric administration (the formula of the high-fat emulsion is 20 wt% of lard, 5 wt% of cholesterol, 5 wt% of yolk powder, 1 wt% of propylthiouracil, 6 wt% of tween-80 and 63 wt% of distilled water), wherein the intragastric administration dose is 10ml/Kg and is once a day. After 3 weeks, blood was collected from retrobulbar venous plexus of a blank rat and a model rat, and serum was collected by centrifugation to measure the serum total cholesterol level. The total cholesterol level of the model rat is obviously increased, and the model rat is obviously different from a blank rat (P <0.05), thus indicating that the model is successfully made.
Taking 50 successfully molded rats, randomly dividing the rats into 5 groups, namely a model group, a Lipitor positive medicine group, a high-dosage group, a medium-dosage group and a low-dosage group, wherein each group comprises 10 rats; another 10 blank rats without model were used as a blank control group. From the 4 th week, the blank control group and the model group are infused with normal saline, and the dosage is 10 ml/Kg; the traditional Chinese medicine composition D (the high dose, the medium dose and the low dose are respectively equal to the crude drug amount of 18g/Kg, 9g/Kg and 4.5g/Kg) of the invention in the embodiment 4 is respectively administrated by intragastric administration, and the lipitor suspension for the positive medicine is administrated by intragastric administration in the dose of 10mg/Kg, 1 time per day and continuously administrated for 4 weeks.
After the administration is finished, all rats are fasted for 12 hours without water prohibition, blood is taken from abdominal aorta after chloral hydrate anesthesia, and serum is taken by conventional centrifugation to detect Total Cholesterol (TC), Triglyceride (TG), Low Density Lipoprotein (LDL) and High Density Lipoprotein (HDL) respectively.
2. Results of the experiment
The blood lipid changes of the rats in each group are shown in Table 1.
TABLE 1 blood lipid changes in rats of each group: (
Figure BDA0001691298460000131
n=10)
Figure BDA0001691298460000132
Note: p <0.05 compared to model group; comparison with model group # P < 0.01.
Compared with the blank control group, the model group rats have increased TC and LDL-C levels, and the difference has statistical significance (P is less than 0.05). Compared with the model group, the TC and LDL-C levels of rats in the positive control group and rats in the low-dose group are reduced, and the difference has statistical significance (P is less than 0.01 or P is less than 0.05), which indicates that the composition has obvious lipid-lowering effect.
The present invention is not limited to the above-described embodiments, and any variations, modifications, and substitutions which may occur to those skilled in the art may be made without departing from the spirit of the invention.
