Abstract

本发明涉及一种用于寄生虫卵检测的样品保存液及使用方法所述的用于寄生虫卵检测的样品保存液包括:孔雀石绿、pH缓冲剂、十二烷基磺酸钠、多聚甲醛、甲醇、乙醇、甘油、L‑丙氨酸等配制而成。本发明的有益效果主要体现在:(1)本发明保存液可以适用人和动物粪便的保存和处理,成本较低,在保持寄生虫卵完整的情况下较好地解决了样本可长期保存并便于运输,改善检测时的环境,使保存于本发明液基细胞处理液中的寄生虫卵的形态得到了良好的固定和保持,避免寄生虫卵的肿胀破裂和萎缩,便于诊断。(2)本发明用于寄生虫卵检测的样品保存液,组分中含有使视野背景柔和,减少视野疲劳的组分,便于观察。

Description

一种用于寄生虫卵检测的样品保存液
(一)技术领域
本发明涉及一种用于寄生虫卵检测的样品保存液,以及利用所述用于寄生虫卵检测的样品保存液检测粪便中寄生虫卵的方法。
(二)背景技术
寄生虫卵侵入宿主,可以产生多种危害而影响宿主的健康。寄生虫卵可以随宿主的粪便排出体外,因此可以通过粪便检测是否感染寄生虫以及感染的程度。检测寄生虫卵常用的方法,如直接检查粪便中的虫体,生理盐水涂片法,加藤法等,但这些方法均受一定条件的限制,如样本不能长期保存,检测时臭味较重,耗时等。
(三)发明内容
针对现有粪便寄生虫检测技术的不足,本发明的目的是提供一种能够快速检测,较长时间保存粪便样本,并改善检测粪便的环境,降低检测时的臭味的用于寄生虫卵检测的样品保存液,以及利用所述用于寄生虫卵检测的样品保存液检测粪便中寄生虫卵的方法。
本发明采用的技术方案是:
一种用于寄生虫卵检测的样品保存液,主要包括以下组分:
多聚甲醛50~100g/L、甲醇10~30g/L、乙醇10~50g/L,L-丙氨酸0.3~0.8g/L、L-异亮氨酸0.3~0.8g/L、磷酸盐缓冲液0.005~0.015mol/L,十二烷基硫酸钠0.1~0.3g/L,香兰素5~10g/L,孔雀石绿0.05~0.09g/L,甘油30~80g/L,调pH 6.2~6.8,余量为水。
优选的,其组成如下:
多聚甲醛60~80g/L、甲醇20~25g/L、乙醇20~30g/L,L-丙氨酸0.5~0.6g/L、L-异亮氨酸0.4~0.6g/L、磷酸盐缓冲液0.008~0.01mol/L,十二烷基硫酸钠0.15~0.2g/L,香兰素6~7g/L,孔雀石绿0.06~0.07g/L,甘油50~60g/L,调pH 6.5,余量为水。
其配制方法如下:将配方量的所述的多聚甲醛、所述的甲醇、所述乙醇、所述L-丙氨酸、所述L-异亮氨酸、所述磷酸盐缓冲液、所述十二烷基硫酸钠、所述香兰素、所述孔雀石绿和所述甘油溶解于所述水中,调节所述pH值至6.2~6.8,然后加所述水定容至一定的体积即可。
所述用于寄生虫卵检测的样品保存液中添加孔雀石绿,其目的是利于观察时视野柔和,缓和眼睛的疲劳。所述用于寄生虫卵检测的样品保存液中添加了多聚甲醛之后,粪便样本可以保存12天以上,便于携带和运输。所述的用于寄生虫卵检测的样品保存液配方方便,操作简单,可减少粪便的结块沉淀,减少残渣的干扰,并减少检测时臭味,易于从清晰的视野中观察粪便中的寄生虫卵,提高检出率。
本发明还涉及利用所述用于寄生虫卵检测的样品保存液检测粪便中寄生虫卵的方法,所述方法包括:
(1)将2~3g粪便样品置于所述的用于寄生虫卵检测的样品保存液8~16mL中,得到混合物;
(2)将步骤(1)混合物混匀,过65~80目筛(优选70目),得滤液;
(3)将步骤(2)滤液转入小口瓶中(优选一滴一滴地挤出),倒置15~20min(优选15min);
(4)从步骤(3)中倒置的瓶口取一滴溶液(优选取第一滴滤液),滴在载玻片上,盖上盖玻片,显微镜下进行观察检测。
优选的,步骤(1)中粪便样品2g,用于寄生虫卵检测的样品保存液用量为10mL。

下面结合具体实施例对本发明进行进一步描述,但本发明的保护范围并不仅限于此:
实施例1:用于寄生虫卵检测的样品保存液的配制
按如下的配方组成配制用于寄生虫卵检测的样品保存液,每1000mL组成如下:
多聚甲醛:50g;
甲醇:15mL;
乙醇:3 0mL;
L-丙氨酸:0.5g;
L-异亮氨酸:0.5g;
磷酸盐缓冲液:0.01mol/L;
十二烷基硫酸钠:0.15g;
香兰素:7.5g;
孔雀石绿:0.07g;
甘油:5g;
余量为水,调节pH值至6.7。
利用所述用于寄生虫卵检测的样品保存液检测人粪便中寄生虫卵的方法,所述方法包括:
(1)将2g粪便样品置于所述的用于寄生虫卵检测的样品保存液10mL中,得到混合物;
(2)将步骤(1)混合物混匀,过70目筛,得滤液;
(3)将步骤(2)滤液一滴一滴地挤出到小口瓶中,倒置15min;
(4)从步骤(3)中倒置的瓶口取一滴溶液(取第一滴滤液),滴在载玻片上,盖上盖玻片,显微镜下进行观察检测。
图1是经本实施例的用于寄生虫卵检测的样品保存液处理人的粪便,所检测到的蛲虫虫卵,图中背景清晰,虫卵完好,易于观察,便于用于临床检测。
实施例2:用于寄生虫卵检测的样品保存液的配制
按如下的配方组成配制用于寄生虫卵检测的样品保存液,每1000mL组成如下:
多聚甲醛:80g;
甲醇:30mL;
乙醇:30mL;
L-丙氨酸:0.6g;
L-异亮氨酸:0.6g;
磷酸盐缓冲液:0.012mol/L;
十二烷基硫酸钠:0.2g;
香兰素:8g;
孔雀石绿:0.08g;
甘油:30g;
余量为水,调节pH值至6.4。
图2是经本实施例的用于寄生虫卵检测的样品保存液处理动物的粪便(检测方法同实施例1),所检测到的虫卵,图中背景清晰,虫卵保存完整,分散均匀,易于观察,便于用于临床检测。
实施例3:用于寄生虫卵检测的样品保存液的配制
按如下的配方组成配制用于寄生虫卵检测的样品保存液,每1000mL组成如下:
多聚甲醛:70g;
甲醇:2 0mL;
乙醇:20mL;
L-丙氨酸:0.4g;
L-异亮氨酸:0.4g;
磷酸盐缓冲液:0.009mol/L;
十二烷基硫酸钠:0.12g;
香兰素:5g;
孔雀石绿:0.07g;
甘油:40g;
余量为水,调节pH值至6.5。
图3是经本实施例的用于寄生虫卵检测的样品保存液处理动物的粪便(检测方法同实施例1),所检测到的虫卵,图中背景清晰,虫卵形态维持良好,易于观察,分散均匀,便于用于临床检测。

Claims (4)

1.一种用于寄生虫卵检测的样品保存液,主要包括以下组分:
多聚甲醛50~100g/L、甲醇10~30mL/L、乙醇10~50mL/L,L-丙氨酸0.3~0.8g/L、L-异亮氨酸0.3~0.8g/L、磷酸盐缓冲液0.005~0.015mol/L,十二烷基硫酸钠0.1~0.3g/L,香兰素5~10g/L,孔雀石绿0.05~0.09g/L,甘油30~80g/L,调pH 6.2~6.8,余量为水。
2.如权利要求1所述的用于寄生虫卵检测的样品保存液,其特征在于其组成如下:
多聚甲醛60~80g/L、甲醇20~25mL/L、乙醇20~30mL/L,L-丙氨酸0.5~0.6g/L、L-异亮氨酸0.4~0.6g/L、磷酸盐缓冲液0.008~0.01mol/L,十二烷基硫酸钠0.15~0.2g/L,香兰素6~7g/L,孔雀石绿0.06~0.07g/L,甘油50~60g/L,调pH 6.5,余量为水。
3.利用权利要求1所述用于寄生虫卵检测的样品保存液检测粪便中寄生虫卵的方法,所述方法包括:
(1)将2~3g粪便样品置于所述的用于寄生虫卵检测的样品保存液8~16mL中,得到混合物;
(2)将步骤(1)混合物混匀,过65~80目筛,得滤液;
(3)将步骤(2)滤液转入小口瓶中,倒置15~20min;
(4)从步骤(3)中倒置的瓶口取一滴溶液,滴在载玻片上,盖上盖玻片,显微镜下进行观察检测。
4.如权利要求3所述的方法,其特征在于步骤(1)中粪便样品2g,用于寄生虫卵检测的样品保存液用量为10mL。
