Abstract

技术领域
本发明涉及生物技术领域,具体地,本发明涉及Slc6a12基因及其蛋白的用途。
背景技术
急性肝衰竭是指由多种原因引起的大量肝细胞迅速丧失功能或死亡。该疾病是一类严重临床综合症,死亡率极高。造成急性肝衰竭的原因很多,如扑热息痛服用过量、肝炎病毒感染(最常见于甲型、乙型肝炎病毒)、过度饮酒、药物特异质反应、缺氧性肝损伤、自身免疫性肝炎、妊娠急性脂肪肝、威尔逊氏病、寄生虫感染等。在我国,肝衰竭的首要病因是病毒性肝炎(以乙型肝炎为主),其次是服药不当和毒性物质造成的肝损伤。而欧美国家则以药物(如扑热息痛)引起的急性肝衰竭最为常见。
目前的肝损伤模型主要分为:化学诱导法,包括LPS+半乳糖苷诱导的急性肝损伤模型、CCL4诱导的肝损伤模型、硫代乙酰胺诱导的肝损伤模型、二甲基亚硝胺诱导的肝损伤模型等;药物毒性诱导法:包括醋氨酚诱导的肝损伤模型、四环素醋氨酚诱导的肝损伤模型等;免疫诱导法,包括刀豆蛋白A诱导的肝损伤模型等;手术法:如肝脏缺血再灌注模型、胆管结扎诱导的肝损伤模型等;以及酒精造模法等。
虽然,目前肝损伤动物模型的应用较广,技术成熟,但多为毒性作用导致的损伤,致病机理与临床病例不尽相同,且靶点机制不明确。
因此本领域迫切需要开发一种可以作为研究肝损伤的发病机理以及新药筛选的有力工具的动物模型。
发明内容
本发明的目的是提供一种可以作为研究肝损伤的发病机理以及新药筛选的有力工具的动物模型。
本发明第一方面提供了一种非人哺乳动物的肝损伤动物模型的制备方法,所述方法包括以下步骤:
(a)提供非人哺乳动物的细胞,将所述细胞中的Slc6a12基因失活,得到Slc6a12基因失活的细胞;和
(b)利用步骤(a)中得到的Slc6a12基因失活的细胞,制备得到Slc6a12基因失活的肝损伤动物模型。
在另一优选例中,所述肝损伤动物模型选自下组:LPS+半乳糖苷诱导的急性肝损伤模型、CCL4诱导的肝损伤模型、硫代乙酰胺诱导的肝损伤模型、二甲基亚硝胺诱导的肝损伤模型、醋氨酚诱导的肝损伤模型、四环素醋氨酚诱导的肝损伤模型、刀豆蛋白A诱导的肝损伤模型、酒精诱导的肝损伤模型、肝脏缺血再灌注模型、胆管结扎诱导的肝损伤模型等,或其组合。
在另一优选例中,在步骤(a)中,包括如下步骤:
(a1)利用DNA同源重组技术,将所述Slc6a12基因中的外显子1至外显子3中一个或多个外显子删除或中断,任选地用筛选标记替换,得到Slc6a12基因失活的非人哺乳动物细胞。
在另一优选例中,在步骤(b)中,包括如下步骤:
(b1)将所述Slc6a12基因失活的细胞制备为嵌合囊胚;
(b2)将步骤(b1)获得的所述嵌合囊胚发育为嵌合非人哺乳动物;
(b3)将上一步骤获得的嵌合非人哺乳动物与正常野生型非人哺乳动物交配繁育,在后代中筛选获得Slc6a12基因失活的杂合子非人哺乳动物;和
(b4)通过将步骤(b3)中得到的杂合子非人哺乳动物相互交配获得Slc6a12基因失活的纯合子非人哺乳动物,从而得到Slc6a12基因失活的非人哺乳动物的动物模型。
在另一优选例中,所述步骤(b4)中还包括步骤(b4’):
对所述的纯合子非人哺乳动物进行肝损伤诱导,从而得到所述的非人哺乳动物的肝损伤动物模型。
在另一优选例中,所述的肝损伤诱导条件选自下组:

(a)在测试组中,在测试化合物存在的条件下,将测试化合物施用于本发明第一方面所述方法制备的非人哺乳动物模型,检测测试组中所述动物模型的肝衰竭严重程度Q1;并且在不施用所述测试化合物且其他条件相同的对照组中,检测对照组中所述动物模型的肝衰竭的严重程度Q2;和
(b)将上一步骤所检测的严重程度Q1和严重程度Q2进行比较,从而确定所述测试化合物是否是治疗或缓解肝损伤的潜在治疗剂;

具体实施方式
本发明人经过广泛而深入的研究,建立了一种急性肝损伤动物模型,它是Slc6a12基因被删除或失活的小鼠或其他非人哺乳动物。本发明的动物模型是一种有效的急性肝损伤动物模型,可用于研究急性肝损伤,并可以用于特定药物的筛选和测试试验。在此基础上完成了本发明。
Slc6a12基因及其蛋白
人类BGT-1基因在小鼠中的直系同源基因称为GAT-2,基因代号为Slc6a12(人类BGT-1基因的Gene ID:6539,小鼠的GAT-2基因的Gene ID:14411)它的编码产物是GABA和甜菜碱(betanine)的转运蛋白,后者为重要的渗透压调节物质。人类的BGT-1(SEQ ID NO.:2)和小鼠GAT-2(SEQ ID NO.:1)的蛋白序列同源性高达88%,前者蛋白长度为614aa,后者为628aa。BGT-1在体内的功能还没有报道。在小鼠中,GAT-2基因高表达于肝脏,但GAT-2在肝脏生理和病理过程中的作用尚不清楚。
为解析Slc6a12基因的功能,本发明采用基因剔除技术,建立了Slc6a12基因剔除小鼠,并利用该小鼠模型开展表型研究,发现Slc6a12基因缺失能够缓解细菌脂多糖和D-半乳糖胺(LPS+GalN)诱导的急性肝衰竭。结果表明,Slc6a12基因缺失小鼠可以作为研究急性肝损伤和急性肝衰竭的动物模型,而Slc6a12的编码蛋白可作为药物靶点,用于筛选急性肝损伤和急性肝衰竭的治疗药物。急性肝衰竭是临床重症,常由病毒和毒物引起,死亡率很高,无特效药物可治疗,因此研究急性肝衰竭的疾病进展机制,开发针对性药物具有重要的意义。
Slc6a12基因或其蛋白的失活剂
在本发明中,所述Slc6a12基因或其蛋白的失活剂包括全部失活或部分失活。
本发明的Slc6a12基因或其蛋白的失活剂包括(a)抑制剂,所述抑制剂的例子包括(但并不限于):小分子化合物、抗体、反义核酸、miRNA、siRNA、或其组合;(b)Slc6a12基因的敲除剂。
基因失活
对于功能未知基因的研究可采用许多方法,例如使有待研究的基因失活,分析所得的遗传修饰的表型变化,进而获得该基因的功能信息。这一研究方法的另一优点是可以将基因功能和疾病进行关联,从而在获得基因功能的同时也能获得该基因作为潜在药物或者药物靶点所能治疗的疾病信息和疾病动物模型。基因失活的方法可通过基因剔除、基因中断或基因***的方式来完成。其中,基因剔除技术是研究人类基因在整体中的功能的非常强有力的手段。
动物模型
在本发明中,提供了一种非常有效的研究肝衰竭的非人哺乳动物模型。
在本发明中,非人哺乳动物的例子包括(但并不限于):小鼠、大鼠、兔、猴等,更佳地是大鼠和小鼠。
如本文所用,术语“Slc6a12基因失活”包括一个或两个Slc6a12基因被失活的情况,即包括Slc6a12基因杂合地和纯合地失活。例如,Slc6a12基因失活的小鼠可以是杂合或纯合的小鼠。
在本发明中,可基因删除或转入外源基因(或片段)而使Slc6a12基因失活等方法制备Slc6a12基因失活的非人哺乳动物(如小鼠)。在本领域中,通过基因删除或转入外源基因而使靶基因失活的技术是已知的,这些常规技术都可用于本发明。
在本发明的另一优选例中,Slc6a12基因的失活是通过基因删除实现的。

在本发明的一具体实例中,可构建一含有外源***片段的构建物,该构建物含有与靶基因(Slc6a12)的***位点的两侧的侧翼序列同源的同源臂,从而可以通过同源重组高频地将外源***片段(或基因)***至Slc6a12基因组序列(尤其是外显子区域),造成小鼠Slc6a12基因的移码、提前终止、或敲除,从而导致Slc6a12缺失或失活。
用本发明方法获得的纯合或杂合的小鼠可育,发育正常。失活的Slc6a12基因可以孟德尔规律遗传给后代小鼠。
在一优选例中,本发明提供了一种缺失Slc6a12基因的纯合小鼠模型动物。
本发明采用基因删除的方法建立了GAT-2(Slc6a12)基因敲除小鼠,并观察了该模型在急性肝损伤中的表现,结果发现GAT-2(Slc6a12)基因敲除可以显著减轻由细菌脂多糖和D-半乳糖胺(LPS/GalN)联合引发的小鼠急性肝损伤。GalN是一种肝毒性物质,其代谢产物尿苷二磷酸可抑制尿嘧啶代谢,干扰细胞RNA合成,进而导致肝细胞凋亡。GalN常与LPS合用,以诱导急性肝衰竭模型。LPS具有很强的免疫原性,可以激活肝脏中Kupffer细胞(一种肝脏特异化的巨噬细胞)攻击肝细胞,将GalN的损伤作用进一步扩大。本发明的研究结果表明,抑制GAT-2(Slc6a12)的活性可以减轻急性肝损伤的发生,因此GAT-2(Slc6a12)可以作为一种药物作用靶点,用于筛选和研发用于治疗肝损伤,特别是急性肝损伤的药物。
候选药物或治疗剂
在本发明中,还提供了一种利用本发明的动物模型,筛选减轻或治疗肝损伤和肝衰竭的候选药物或治疗剂的方法。
在本发明中,候选药物或治疗剂是指已知具有某种药理学活性或正在被检测的可能具有某种药理学活性的物质,包括但不限于核酸、蛋白、化学合成的小分子或大分子化合物、细胞等。候选药物或治疗剂的给药方式可以是口服、静脉注射、腹腔注射、皮下注射、或椎管给药。
本发明的主要优点包括:
(a)用本发明方法获得的纯合或杂合的动物模型可育,发育正常。转基因杂合雄性小鼠具有生殖能力,失活的Slc6a12基因可以孟德尔规律遗传给后代小鼠。
(b)本发明的肝衰竭动物模型可减轻急性肝损伤的发生,因此可用于筛选和研发用于治疗肝损伤,特别是急性肝损伤的药物。
(c)本发明首次采用基因剔除的方法建立了Slc6a12基因敲除小鼠,并观察了该模型在急性肝损伤中的表现,结果发现Slc6a12基因敲除可以显著减轻由细菌脂多糖(LPS)和半乳糖苷联合引发的小鼠急性肝损伤。
(d)本发明首次揭Slc6a12可以作为一种药物作用靶点。
下面结合具体实施例,进一步阐述本发明。应理解,这些实施例仅用于说明本发明而不用于限制本发明的范围。下列实施例中未注明具体条件的实验方法,通常按照常规条件,例如Sambrook等人,分子克隆:实验室手册(New York:Cold Spring HarborLaboratory Press,1989)中所述的条件,或按照制造厂商所建议的条件。除非另外说明,否则百分比和份数是重量百分比和重量份数。
实施例中用到的材料如无特殊说明,均为市售产品。
实施例1 Slc6a12基因剔除小鼠的构建和鉴定
Slc6a12基因剔除小鼠的构建根据常规的基因打靶技术进行。通过构建打靶载体,导入到提取自小鼠囊胚的胚胎干细胞(ES)(SCR012)中,利用打靶载体上和靶基因区域同源的片段和基因组进行同源重组,将Slc6a12基因定点破坏。图1是打靶载体及同源重组打靶示意图,其中小鼠基因组中Slc6a12的外显子1-2区域被筛选基因neo取代,导致Slc6a12基因被破坏,基因起始密码子缺失。
基因打靶实验按常规操作进行,打靶载体DNA用电穿孔方法导入到小鼠胚胎干细胞(ES)中,培养在经丝裂霉素处理的小鼠胚胎成纤维细胞组成的饲养层细胞上,在G418药物作用下进行克隆筛选,对获得的克隆进行PCR鉴定,其中一对PCR引物(Slc6a12-5P:5’-GGATGGATCTGGAACTACCTTC-3’(SEQ ID NO.:6;
Slc6a1 2-3P:5’-GTGACTCTGAAGTCATCACTC-3’(SEQ ID NO.:7))分别设计在同源重组臂的外侧,另一对(neo Rh:5’-GGCCTACCCGCTTCCATTGCTC-3’(SEQ ID NO.:8);neo-Lh:5’-CCGTGCCTTCCTTGACCCTGG-3’(SEQ ID NO.:9))分别设计在筛选基因neo基因序列上,两侧PCR如均获得预期长度的扩增产物并经测序验证正确的既为阳性克隆。
用常规方法体外培育受精卵至囊胚期,作为受体囊胚。阳性ES细胞克隆扩增后,利用显微注射方法,将一定数量ES细胞注射入小鼠受体囊胚腔(C57BL/6J,购自上海灵畅生物科技有限公司请补充)中,获得嵌合囊胚,并移植到假孕小鼠输卵管中,使其着床发育,生产出嵌合小鼠,嵌合小鼠和野生型小鼠繁育,获得带有目标基因缺失的杂合子小鼠,杂合子小鼠交配后可以获得目标基因缺失的纯合子小鼠。
根据打靶策略,在验证了发生正确同源重组后,后续的小鼠的基因型鉴定可以采用下述3个引物的PCR方法,PCR引物设计如下:
gGAT2-p1:5'-GCTCTGCTTTCGGTTGG-3';(SEQ ID NO.:3)
gGAT2-p2:5'-CGCCTTCTTGACGAGTTC-3';(SEQ ID NO.:4)
gGAT2-p3:5'-ATCAGGTTAAATACAAATAGGT-3'。(SEQ ID NO.:5)
鉴定结果如图2所示。其中,实验用各基因型小鼠均来自Slc6a12+/-自交后代。根据Slc6a12基因打靶策略,设计引物,并对基因组进行PCR-电泳。不同条带结果对应不同的基因型小鼠:单一1.3kb条带为野生型(+/+);单一1.1kb条带为GAT-2基因敲除纯合子(-/-);双条带为杂合子(+/-)。
实施例2 Slc6a12在肝急性损伤中的作用
10周龄雄性野生型(WT)和Slc6a12基因剔除小鼠(slc6a12-/-),用于建立LPS+GalN诱导的急性肝衰竭模型。所有动物实验前需适应饲养环境一周以上。小鼠腹腔联合注射LPS(11μg/kg体重)+GalN(700mg/kg体重),诱导急性肝衰竭模型。GalN是一种肝毒性物质,其代谢产物尿苷二磷酸可抑制尿嘧啶代谢,干扰细胞RNA合成,进而导致肝细胞凋亡。GalN常与LPS合用,以诱导急性肝衰竭模型。LPS具有很强的免疫原性,可以激活肝脏中Kupffer细胞(一种肝脏特异化的巨噬细胞)攻击肝细胞,将GalN的损伤作用进一步扩大。该模型是目前急性肝衰竭研究中最常用的模型之一。
对于生存率实验,WT(n=14)和GAT-2-/-(n=15)小鼠诱导模型后,每15min观察一次存活情况,连续观察13h。对于Slc6a12功能实验,模型组注射LPS+GalN(WT:n=7;Slc6a12-/-:n=7)诱导急性肝衰竭模型,对照组注射等量的PBS(WT:n=3;Slc6a12-/-:n=3)。给药6h后处死小鼠。
结果表明,和野生型小鼠相比,Slc6a12基因剔除小鼠的肝衰竭严重程度明显减轻。最终存活率显示(图3),WT小鼠为21%,而Slc6a12-/-小鼠为53%,两者存在统计学意义的差异(p<0.01);作为评价肝损伤的血液指标,如图4所示,谷丙转氨酶(alaninetransarninase,ALT)和谷草转氨酶(glutamic-oxal acetic transaminase,AST)在Slc6a12-/-小鼠中比WT分别下降了51%(p<0.05)和58%(p<0.005)。
上述结果表明,Slc6a13基因剔除可以降低LPS+GalN对肝的损伤。该推论在病理检查中得到验证(图5),肝脏H&E切片结果显示:对照组小鼠肝细胞形态完整,排列规则(图5A,图5B);LPS+GalN给药后,WT小鼠出现了大量肝细胞坏死和气球样变性,并伴有大面积的肝窦充血(图5C);模型组Slc6a13-/-小鼠的细胞坏死和充血程度均轻于WT小鼠(图5D)。组织病理学评分也表明Slc6a13基因缺失减少了LPS+GalN诱导的组织病理学损伤(p<0.005)(图5E)。
上述结果说明Slc6a12基因缺失可保护LPS+GalN诱导的肝损伤,Slc6a12基因及其蛋白产物在化学物质引发的肝损伤过程中发挥作用,抑制该基因及其蛋白的活性,可以减缓肝损伤及肝衰竭的发生和发展,因此,Slc6a12可作为新的药物靶点,用于开发针对Slc6a12的可延缓急性肝损伤和肝衰竭的候选药物。
实施例3用治疗肝损伤(如急性肝损伤)的药物验证药物筛选平台
在本实施例中,给实施例1构建的模型动物小鼠注射当前临床治疗急性肝损伤的药物硫普罗宁和/或双环醇,随即对模型动物小鼠的肝损伤严重程度进行评估。
结果表明,药物硫普罗宁和/或双环醇等可显著降低模型动物小鼠的肝损伤严重程度,可显著提高模型动物小鼠的生存率,显著降低肝损伤的血液指标(如谷丙转氨酶和谷草转氨酶的含量),显著改善肝脏组织病理学症状,显著减少肝细胞的凋亡,显著降低肝脏及血液内的炎症因子或其蛋白的表达水平。
实施例4利用治疗肝损伤(如急性肝损伤)的药物筛选平台筛选候选药物
在本实施例中,计划通过给实施例1构建的模型动物小鼠注射急性肝损伤的治疗药物,随即对模型动物小鼠的肝损伤严重程度进行评估。
通过与给安慰剂的模型动物小鼠比较肝损伤严重程度的差异,能够显著降低模型动物小鼠的肝损伤严重程度、显著提高模型动物小鼠的生存率、显著降低肝损伤的血液指标(如谷丙转氨酶和谷草转氨酶的含量)、显著改善肝脏组织病理学症状、显著减少肝细胞的凋亡、显著降低肝脏及血液内的炎症因子或其蛋白的表达水平的候选药物,即为治疗急性肝损伤的潜在治疗药物。
在本发明提及的所有文献都在本申请中引用作为参考,就如同每一篇文献被单独引用作为参考那样。此外应理解,在阅读了本发明的上述讲授内容之后,本领域技术人员可以对本发明作各种改动或修改,这些等价形式同样落于本申请所附权利要求书所限定的范围。
Claims (10)
1.一种非人哺乳动物的肝损伤动物模型的制备方法,其特征在于,所述方法包括以下步骤:
(a)提供非人哺乳动物的细胞,将所述细胞中的Slc6a12基因失活,得到Slc6a12基因失活的细胞;
(b)利用步骤(a)中得到的Slc6a12基因失活的细胞,制备得到Slc6a12基因失活的肝损伤动物模型。
2.如权利要求1所述的方法,其特征在于,在步骤(a)中,包括如下步骤:
(a1)利用DNA同源重组技术,将所述Slc6a12基因中的外显子1至外显子3中一个或多个外显子删除或中断,任选地用筛选标记替换,得到Slc6a12基因失活的非人哺乳动物细胞。
3.如权利要求1所述的方法,其特征在于,所述步骤(b)中得到的Slc6a12基因失活的非人哺乳动物模型中,与野生型对照动物相比,具有选自下组的一个或多个特征:
(t1)肝损伤的严重程度降低;
(t2)非人哺乳动物的生存率提高;
(t3)谷丙转氨酶和/或谷草转氨酶的含量下降;
(t4)肝脏组织病理学症状显著改善;
(t5)肝细胞凋亡情况减少;
(t6)肝脏及血液内,炎症因子或其蛋白的表达水平降低。
4.如权利要求1所述方法制备的非人哺乳动物模型的用途,其特征在于,将该模型用作研究肝损伤的动物模型。
5.如权利要求1所述方法制备的非人哺乳动物模型的用途,其特征在于,将该模型用作研究肝损伤抵抗性程度高的动物模型。
6.如权利要求1所述方法制备的非人哺乳动物模型的用途,其特征在于,用于筛选或鉴定可减轻或治疗肝损伤的物质(治疗剂)。
7.一种筛选或确定治疗或缓解肝损伤的潜在治疗剂的方法,其特征在于,包括步骤:
(a)在测试组中,在测试化合物存在的条件下,将测试化合物施用于权利要求1所述方法制备的非人哺乳动物模型,检测测试组中所述动物模型的肝衰竭严重程度Q1;并且在不施用所述测试化合物且其他条件相同的对照组中,检测对照组中所述动物模型的肝衰竭的严重程度Q2;
(b)将上一步骤所检测的严重程度Q1和严重程度Q2进行比较,从而确定所述测试化合物是否是治疗或缓解肝损伤的潜在治疗剂;
其中,如果严重程度Q1显著低于严重程度Q2时,则表示所述测试化合物为治疗或缓解肝损伤的潜在治疗剂。
8.一种筛选或确定治疗或缓解肝损伤的潜在治疗剂的方法,其特征在于,包括步骤:
(a)在测试组中,在培养体系中,在测试化合物的存在下,培养表达Slc6a12的细胞一段时间T1,检测测试组所述培养体系中的slc6a12基因或其蛋白的表达水平E1;和/或slc6a12蛋白的活性水平V1;
并且在不存在所述测试化合物且其他条件相同的对照组中,检测对照组所述培养体系中的slc6a12基因或其蛋白的表达水平E2;和/或slc6a12蛋白的活性水平V2;
(b)将上一步骤所检测的E1、E2,和/或V1、V2进行比较,从而确定所述测试化合物是否是治疗或缓解肝损伤的潜在治疗剂;
其中,如果E1显著低于E2;和/或V1显著低于V2,则表示所述测试化合物是治疗或缓解肝损伤的潜在治疗剂。
9.一种细胞的用途,其特征在于,所述细胞中的Slc6a12基因失活或下调,用于制备构建非人哺乳动物的肝损伤动物模型的生物制剂。
10.一种Slc6a12基因或其蛋白失活剂的用途,其特征在于,用于制备一制剂或组合物,所述制剂或组合物用于治疗或缓解肝损伤。
