Abstract

本发明涉及一种药物经皮促渗剂及其应用方法，属于外用给药技术领域。本发明包括1~3份冰片，1~3份樟脑，60~65份无水乙醇，20~30份油包水型乳剂，1~2份羧甲基纤维素钠，15~30份丙三醇，本发明利用脂质膜与皮肤的融合性促进药物的透皮吸收，并利用含有的高体积分数乙醇使角质细胞的流动性增强，同时利用自身的囊泡形的结构特点，将药物从基质中的溶出机制由扩散转变为溶蚀与扩散的双重作用，变形渗透至皮肤深层，有效增加药物的透皮量，同时本发明药物经皮促渗剂可有效改善药物的脂溶性，提高药物吸收率，并提高治疗效果，具有广阔的应用前景。

技术领域
本发明涉及一种药物经皮促渗剂及其应用方法，属于外用给药技术领域。
背景技术
皮肤作为人体最大的器官，是体内与外界的一个屏障***，其利用皮肤角质层中角质细胞和细胞间脂质组成的多层致密非晶体结构，其富含交联度很高的蛋白质。相邻角质形成细胞间通过质膜紧密连接，依靠特殊的跨膜蛋白，彼此对合交联，形成紧密连接结构，阻止外界因素的影响，形成药物透皮吸收的主要屏障，由于皮肤的天然屏障作用，药物的经皮促渗效果并不明显。
同时由于部分药物脂溶性差，不易直接透过生物膜，口服给药生物利用度很低，影响了药效的发挥，因此通过选择高渗透性的药物经皮促渗剂为给药载体，通过外用给药增加药物在关节部位浓度，以期提高治疗效果。 外用给药时，皮肤渗透性是影响药效发挥的重要因素，药物只有渗透到真皮层的血管和淋巴才能被人体吸收。因此，对于经皮促渗剂研究显得尤为重要。
发明内容
本发明所要解决的技术问题：针对由于皮肤的天然屏障作用，药物的经皮促渗效果不明显的问题，本发明提供了一种药物经皮促渗剂及其应用方法。
为解决上述技术问题，本发明采用的技术方案是：
一种药物经皮促渗剂，包括冰片、樟脑、乙醇、油包水型乳剂、羧甲基纤维素钠、丙三醇，其特征在于，所述油包水型乳剂为磷脂与胆固醇的***溶液，在室温下减压蒸发至凝胶状制得。
所述冰片、樟脑、乙醇、油包水型乳剂、羧甲基纤维素钠、丙三醇的重量份为1~3份冰片，1~3份樟脑，60~65份无水乙醇，20~30份油包水型乳剂，1~2份羧甲基纤维素钠，15~30份丙三醇。
所述胆固醇***溶液的质量为磷脂的2.5~10倍。
所述胆固醇***溶液的质量分数为5~10%。
所述羧甲基纤维素钠为经去离子水在70~80℃下溶胀制得。
所述的一种药物经皮促渗剂的应用方法为将药物溶解在无水乙醇中后与药物经皮促渗剂混合均匀后即可涂覆在用药部位。
所述的一种药物经皮促渗剂的应用方法中药物用量为药物经皮促渗剂质量的10~20%。
本发明的有益技术效果是：
（1）本发明利用脂质膜与皮肤的融合性促进药物的透皮吸收，并利用含有的高体积分数乙醇使角质细胞的流动性增强，同时利用自身的囊泡形的结构特点，将药物从基质中的溶出机制由扩散转变为溶蚀与扩散的双重作用，变形渗透至皮肤深层，有效增加药物的透皮量。
（2）本发明药物经皮促渗剂可有效改善药物的脂溶性，提高药物吸收率，并提高治疗效果，具有广阔的应用前景。
具体实施方式
取10~20g磷脂，加入50~100g质量分数为10%胆固醇***溶液中，在冰水浴中以300W超声波超声分散20~30min，再在室温下减压蒸发至凝胶状，得油包水型乳剂，取1~2g羧甲基纤维素钠加入20~40mL去离子水中，在70~80℃下溶胀40~50min，再加入20~30g油包水型乳剂，1~3g冰片，1~3g樟脑，60~65g无水乙醇，以300~400r/min搅拌20~30min，得混合液，将混合液与15~30g丙三醇加入研磨机中研磨1~2h，出料后置于棕色瓶密封，在0~4℃下保存，得药物经皮促渗剂，取1~2g药物加入10~20mL无水乙醇中，以300~400r/min搅拌20~30min，再加入10~20g药物经皮促渗剂，继续搅拌10~15min后涂覆在用药部位即可。
实例1
取10g磷脂，加入50g质量分数为10%胆固醇***溶液中，在冰水浴中以300W超声波超声分散20min，再在室温下减压蒸发至凝胶状，得油包水型乳剂，取1g羧甲基纤维素钠加入20mL去离子水中，在70℃下溶胀40min，再加入20g油包水型乳剂，1g冰片，1g樟脑，60g无水乙醇，以300r/min搅拌20min，得混合液，将混合液与15g丙三醇加入研磨机中研磨1h，出料后置于棕色瓶密封，在0℃下保存，得药物经皮促渗剂，取1g药物加入10mL无水乙醇中，以300r/min搅拌20min，再加入10g药物经皮促渗剂，继续搅拌10min后涂覆在用药部位即可。
实例2
取15g磷脂，加入75g质量分数为10%胆固醇***溶液中，在冰水浴中以300W超声波超声分散25min，再在室温下减压蒸发至凝胶状，得油包水型乳剂，取1.5g羧甲基纤维素钠加入30mL去离子水中，在75℃下溶胀45min，再加入25g油包水型乳剂，2g冰片，2g樟脑，63g无水乙醇，以350r/min搅拌25min，得混合液，将混合液与20g丙三醇加入研磨机中研磨1h，出料后置于棕色瓶密封，在2℃下保存，得药物经皮促渗剂，取1.5g药物加入15mL无水乙醇中，以300r/min搅拌25min，再加入15g药物经皮促渗剂，继续搅拌12min后涂覆在用药部位即可。
实例3
取20g磷脂，加入100g质量分数为10%胆固醇***溶液中，在冰水浴中以300W超声波超声分散30min，再在室温下减压蒸发至凝胶状，得油包水型乳剂，取2g羧甲基纤维素钠加入40mL去离子水中，在80℃下溶胀50min，再加入30g油包水型乳剂，3g冰片，3g樟脑，65g无水乙醇，以400r/min搅拌30min，得混合液，将混合液与30g丙三醇加入研磨机中研磨2h，出料后置于棕色瓶密封，在4℃下保存，得药物经皮促渗剂，取2g药物加入20mL无水乙醇中，以400r/min搅拌30min，再加入20g药物经皮促渗剂，继续搅拌15min后涂覆在用药部位即可。

从上表可以看出，本发明制得的药物经皮促渗剂可有效增加药物的透皮量，促进药物吸收，具有广阔的应用前景。

Claims (7)

1.一种药物经皮促渗剂，包括冰片、樟脑、乙醇、油包水型乳剂、羧甲基纤维素钠、丙三醇，其特征在于，所述油包水型乳剂为磷脂与胆固醇的***溶液，在室温下减压蒸发至凝胶状制得。
2.如权利要求1所述的一种药物经皮促渗剂，其特征在于，冰片、樟脑、乙醇、油包水型乳剂、羧甲基纤维素钠、丙三醇的重量份为1~3份冰片，1~3份樟脑，60~65份无水乙醇，20~30份油包水型乳剂，1~2份羧甲基纤维素钠，15~30份丙三醇。
3.如权利要求1所述的一种药物经皮促渗剂，其特征在于，所述胆固醇***溶液的质量为磷脂的2.5~10倍。
4.如权利要求1所述的一种药物经皮促渗剂，其特征在于，所述胆固醇***溶液的质量分数为5~10%。
5.如权利要求1所述的一种药物经皮促渗剂，其特征在于，所述羧甲基纤维素钠为经去离子水在70~80℃下溶胀制得。
6.如权利要求1~5任一项所述的一种药物经皮促渗剂的应用方法，其特征在于，所述应用为将药物溶解在无水乙醇中后与药物经皮促渗剂混合均匀后即可涂覆在用药部位。
7.如权利要求5所述的一种药物经皮促渗剂的应用方法，其特征在于，药物用量为药物经皮促渗剂质量的10~20%。
