协同抗微生物组合物
本发明涉及杀生物剂组合,所述组合的活性大于在单个抗微生物化合物中观察的活性。
使用至少两种抗微生物化合物的组合可以拓宽潜在市场,降低使用浓度和成本,并且减少浪费。在一些情况下,即使在高使用浓度下,市售抗微生物化合物也无法提供对微生物的有效控制,这是因为其抵抗某些类型的微生物的活性较弱,或相对缓慢的抗微生物活动,或在如高温和高pH值的某些条件下的不稳定性。不同抗微生物化合物的组合有时用于提供对微生物的整体控制或在具体最终使用环境中以较低使用比率提供相同水平的微生物控制。此外,已发现协同作用是一种不可预测的现象。通常,当与具体活性组合时,类似的化合物显示不同的协同概况。可能发生的情况是不显示协同作用或在不同协同范围内发生协同作用。因为这一观察结果,难以(如果不是不可能)基于一类化合物的协同概况总结出关于一种化合物的协同概况的结论。因此,必须发现更多的协同组合和其协同范围。
协同作用的一种这类实例可见于美国专利案第5,385,896号中。这一参考文献公开膦盐与醛的组合,但这一参考文献未提出任一种本文中所要求的组合。仍需要具有增强的活性的抗微生物化合物的其它组合以提供对微生物的有效控制。本发明解决的问题是提供这类抗微生物化合物组合。
发明内容
本发明涉及协同抗微生物组合物,其包含:(a)二胺;和(b)二硫基-2,2'-双-苯甲基酰胺;其中二胺与二硫基-2,2'-双-苯甲基酰胺的重量比是10:1至1:10。
具体实施方式
如本文中所使用,除非上下文另外明确指示,否则以下术语具有指定定义。术语“抗微生物化合物”是指能够抑制微生物的生长或传播和/或杀死微生物的化合物;抗微生物化合物包括杀细菌剂、杀菌剂、抑菌剂、抑菌素、杀真菌剂、抑真菌剂、除藻剂以及抑藻剂,视所施用的剂量水平、***条件以及所需的微生物控制程度而定。术语“微生物”包括例如真菌(如酵母和霉菌)、细菌以及藻类。本说明书通篇使用以下缩写:ppm=按重量计的百万分率(重量/重量),mL=毫升。除非另外说明,否则温度以摄氏度(℃)为单位,并且提到的百分比是以重量计(重量%)。本发明的组合物中抗微生物化合物的百分比是以组合物中活性成分的总重量计,即,以抗微生物化合物本身计,不包括可能存在的任何量的溶剂、载剂、分散剂、稳定剂或其它材料。最后,包含所有范围值端点并且是可组合的(例如表示成10:1到1:10或者3:1和1:3的范围也可以包括例如10:3)。
如本文中所使用,二胺是指N-(3-氨基丙基)-N-十二烷基丙-1,3-二胺,目录号2372-82-9,并且二硫基-2,2'-双-苯甲基酰胺或“DTBMA”对应于目录号:2527-58-4。
在本发明的一些实施例中,二胺与二硫基-2,2'-双-苯甲基酰胺(DTBMA)的重量比是10:1到1:10,或9:1到1:9,或者3:1到1:3。
在本发明的一些实施例中,抗微生物组合适用于抑制水性介质中的微生物生长,例如作为罐内防腐剂。组合物还适用于控制其它水性介质中的微生物,包括(但不限于)工业水和含水介质/水被污染的培养基,如冷却水、空气洗涤器、热交换器、锅炉水、纸浆和造纸厂水、其它工业过程水介质,如:压载水、废水、金属加工流体、油气、乳胶、油漆、涂料、粘合剂、油墨、条带接合化合物、色素、基于水的浆料、个人护理和家用产品,如清洁剂、过滤***(包括逆渗透和超滤***)、抽水马桶、纺织物、皮革和皮革生产***,或与其一起使用的***。
通常,用于控制微生物生长的本发明的杀生物剂组合的量是10ppm到5,000ppm活性成分。在本发明的一些实施例中,组合物的活性成分以至少20ppm,或者至少50ppm,或者至少100ppm,或者至少150ppm,或者至少200ppm的量存在。在一些实施例中,组合物的活性成分以不超过2,000ppm,或者不超过1,000ppm,或者不超过500ppm,或者不超过400ppm,或者不超过300ppm,或者不超过250ppm,或者不超过200ppm,或者不超过100ppm,或者不超过50ppm的量存在。上文提到的浓度是以含有杀生物剂组合的液体组合物计。
本发明还涵盖一种方法,其通过将所要求的杀生物剂组合并入材料中来减少,或抑制,或预防上述使用区域中的微生物生长,尤其在罐内防腐应用中。
实例
使用Kull,F.C等人在《应用微生物学(Applied Microbiology)》9:538-541(1961)中描述的方法测定本发明的杀生物剂组合的协同作用。
用于计算协同指数(SI)的公式是
Qa/QA+Qb/QB=SI
其中
QA=化合物A的浓度(ppm),单独起作用产生终点(生长/不生长)
Qa=化合物A的浓度(ppm),呈混合物形式,其产生终点(生长/不生长)
QB=化合物B的浓度(ppm),单独起作用产生终点(生长/不生长)
Qb=化合物B的浓度(ppm),呈混合物形式,其产生终点(生长/不生长)
在这一研究中,杀生物剂终点定义为在指定接触时间下呈现至少4倍对数的细菌或酵母减少或保持最大2倍对数的细菌或酵母计数。如果终点无法确立,那么所测试的杀生物剂的最高浓度将用作供计算用的终点,并且SI将以“小于或<”值记录。
当SI具有小于1的值时,证实两种杀生物剂的协同作用。如果SI等于1,那么混合物显示累加作用,并且如果SI大于1,那么混合物显示拮抗作用。
设计最小抑制性测试(MIC)以评估可阻止细菌、酵母或真菌在所定义的培养液中生长的杀生物剂、杀生物剂掺合物或杀生物剂组合的最低浓度。在培育之后,基于可目视检测的浑浊度来评定样品。
对于杀生物剂,在稀释因子是1.5的情况下进行这一测试。
针对两种细菌菌株和一种酵母来测试以下杀生物剂产品/混合物:
最小抑制浓度(MIC)测试方案
1.在透明容器中以相同体积制备测试样品。杀生物剂稀释是在胰酶大豆培养液(TSB)中在pH 7.3下(对于细菌)和在麦芽提取物培养液(MEB)中在pH 5.4下(对于真菌菌株)进行。
对于每种杀生物剂产品和物种,通过在稀释因子是1.5的情况下进行连续稀释来一式两份地制备16个样品。
对于每种微生物菌株,添加两个不含任何杀生物剂的样品作为对照物。
2.每个测试样品用微生物悬浮液接种以分别产生约1×106CFU/ml(在细菌物种的情况下)和约1×105CFU/ml(对于真菌物种)的水平。
制备微生物悬浮液:
细菌培养物
绿脓杆菌(Pseudomonas aeruginosa) DSM编号939 ATCC编号 15442
金黄色葡萄球菌(Staphylococcus aureus) DSM编号799 ATCC编号 6538
培养物在冷冻管中在-80℃下以甘油储备液形式保存、解冻并且接着以100μl分散在TSA琼脂板上。在30℃下培育1天之后,将菌株与缓冲液(pH 7.3)一起收集。

酵母培养物

培养物在冷冻管中在-80℃下以甘油储备液形式保存、解冻并且接着以100μl分散在MEA(麦芽提取物琼脂)皮氏培养皿(petri dishes)上。

4.通过生长/不生长评定来目视进行读取。在培育之后,微生物的生长引起浑浊,澄清表示未生长。在培育之后,在培养液中显示未生长的最低浓度视为MIC值。

2.一种协同抗微生物组合物,其包含:(a)二胺;和(b)二硫基-2,2'-双-苯甲基酰胺;其中所述二胺与二硫基-2,2'-双-苯甲基酰胺的重量比是9:1到1:10。
3.一种抑制水性介质中微生物生长或控制水性介质中微生物生长的方法,其包含向所述水性介质中添加根据权利要求1所述的组合物。
4.一种抑制水性介质中微生物生长或控制水性介质中微生物生长的方法,其包含向所述水性介质中添加根据权利要求2所述的组合物。
5.一种水性介质,其包含根据权利要求1所述的协同抗微生物组合物。
6.一种水性介质,其包含根据权利要求2所述的协同抗微生物组合物。
