Abstract
本发明属于监测技术领域,公开了一种初期海洋生物污损监测评价方法,基于生物膜对不同波长光线的阻碍和散射作用,借鉴朗伯比尔定律(Beer‑Lambert Law)能够测定物质浓度的基本原理,采用紫外可见分光光度法测定不同时期被附着材料的吸光度曲线,通过透光率峰值(或者曲线积分)对生物附着试样的生物量(或者生物覆盖率、叶绿素a含量等)建立标准工作曲线,再通过被测试样的吸光度数据进行计算分析,从而完成对玻璃等透明材质的生物附着情况的监测和评价,为透明材料的防生物附着性能的评价提供依据,有助于新型耐生物污损材料的开发,最终有效避免生物附着和污损造成的损失及危害。
Description
技术领域
本发明属于监测技术领域,具体涉及一种初期海洋生物污损监测评价方法。
背景技术
一般物体浸入海水中后,由于受到一系列物理、化学和生物等因素的相互作用,其表面很快会覆盖一层聚合物材料形成的薄膜,通常称为调节膜。调节膜的主要成分是蛋白质大分子等,它会使物体表面特性发生改变,成为海洋浮游生物附着、繁殖的基础。随着调节膜的形成,细菌、单细胞真核生物和多细胞真核生物依次在浸入物体表面附着,形成一个复杂的生态***。黏附在物体表面的细菌大量分泌细胞外产物,将细菌与细菌、细菌与附着底物等相互连接形成一层由细菌和海藻为主的生物膜。研究发现生物膜在随后大型海洋生物附着时起到了关键的桥梁作用,生物膜可以释放出化学信号,让海中生物感知到环境是否适合附着生存。大型海洋生物的幼虫和孢子一旦接近带有生物膜的表面后便会附着,***生长并繁殖扩大,最终形成规模较大的海洋污损生物群落。污损生物粘附在舰船底部增大了流体动力学阻力、降低了舰船机动性、增加了燃料消耗,同时,污损生物还抑制水产养殖、堵塞管道、加速金属腐蚀、影响海洋仪器仪表性能等。针对海洋污损生物群落的形成过程及附着机理,人们采取了各种各样的生物污损防除方法。根据其原理的不同可大致分为物理防污法、化学防污法和生物防污法三大类。采用微生物杀菌剂等措施控制生物污损初期微生物膜的附着和生长,可以有效避免生物附着和污损造成的损失及危害。这就需要对生物污损初期微生物膜附着情况进行在线监测,以及时采取有效措施,控制微生物膜附着和生长,防止其发展。实际上针对海洋生物污损的评价,初期主要是通过荧光显微镜及扫描电镜等方法进行检测,再就是采用实海挂片的方法观察生物附着的情况。然而这几种方法主要应用于实验室,且操作复杂,目前尚缺乏简便快速有效的技术手段,对生物污损初期微生物膜附着情况进行监测和评价。
现有技术中有通过监测传感器电极电位的变化评价微生物膜附着生长程度,但仅适用于高钝化性能的不锈钢材料表面的微生物膜附着情况评价。不同的材料其表面海洋生物附着生长的状况不同,因此对于其他材料表面生物附着状况的监测及评价技术就尤为重要。光学玻璃是海洋监测传感器上常用的材料,是仪器对海洋进行观测监测活动的窗口,直接与海水接触,容易受到海洋生物污损的侵害,从而影响海洋仪器的性能,因此需要对光学玻璃等材料的生物附着情况进行监测和评价。
吸光度(Abs):吸光度是物理学和化学的一个名词。是指光线通过溶液或物质前的入射光强度与光线通过溶液或某一物质后的透射光强度的比值(I0/I1)的以10为底的对数(即A=lg(I0/I1)),其中I0为入射光强,I1为透射光强,影响它的因素有溶剂、浓度、温度等。
透光率(Transmission):是透过透明或半透明体的光通量与其入射光通量的百分率,即T=I1/I0╳100%,即T=10-A。
污损生物覆盖率:污损生物附着基底总面积与基体材料面积的百分比率。
叶绿素a含量:附着在基体材料上的叶绿素a的含量,通过分光光度法测定,单位μg/cm2。
发明内容
本发明为克服现有技术的不足,采用下述技术方案予以实现:
一种初期海洋生物污损监测评价方法,包括以下步骤:
(1)选取监测评价对象;
(2)将监测评价对象放入海水中,通过海水中生物自然生长附着的方式获得生物附着后的监测评价对象试样。将透明材料放入海水中一段时间,通过海水中生物自然生长附着的方式获得生物附着后的透明材料的试样。根据需要,可以在不同海域、不同深度分别布置试样。设置不同的试样浸入时间,可以获得不同附着程度的试样。
(3)采用紫外可见分光光度法测量生物附着后监测评价对象的吸光度曲线,获得时间-吸光度关系标准曲线。采用紫外可见分光光度测量生物附着后透明材料的吸光度曲线,获得吸收峰的峰值或者对吸光度曲线进行积分。可以获得时间-吸光度关系标准曲线。
(4)将步骤(3)获得的标准曲线与生物污损程度情况进行对比分析,获得实际的生物污损程度情况。
步骤(3)通过获得吸收峰峰值或者对吸光度曲线积分的方式获得时间-吸光度关系标准曲线。
步骤(4)所述的生物污损程度情况是通过生物量和荧光显微镜法获得的。将获得的吸光度标准曲线结合生物量和荧光显微镜法获得的生物污损程度情况进行对比分析,获得实际的生物污损程度情况。后期可以通过生物附着后的吸光度曲线直接获取实际的生物污损程度情况。
步骤(2)可以在不同海域、不同深度分别设置监测评价对象,设置不同的浸入时间,获得不同附着程度的监测评价对象。
优选的,监测评价对象为光学玻璃。本技术方案适用于对透明材质的生物附着情况进行监测和评价,主要包括光学玻璃、有机玻璃等,因此选取需要监测和评价的透明材料,并按照要求进行加工。
本技术方案基于生物膜对不同波长光线的阻碍和散射作用,借鉴朗伯比尔定律(Beer-Lambert Law)能够测定物质浓度的基本原理,采用紫外可见分光光度法测定不同时期被附着材料的吸光度曲线,通过透光率峰值(或者曲线积分)对生物附着试样的生物量(或者生物覆盖率、叶绿素a含量等)建立标准工作曲线,再通过被测试样的吸光度数据进行计算分析,从而完成对玻璃等透明材质的生物附着情况的监测和评价,有效避免生物附着和污损造成的损失及危害,达到生物污损防除的目的。

具体实施方式
下面结合附图和具体实施方式对本发明的技术方案作进一步详细的说明。
实施例1
一种初期海洋生物污损监测评价方法,步骤如下:
(1)选取监测评价对象。选取常用的光学玻璃为监测评价对象,将光学玻璃加工成如图1所示试样。尺寸为8mm╳40mm╳1mm。
(2)将监测评价对象放入海水中,通过海水中生物自然生长附着的方式获得生物附着后的监测评价对象试样。将透明材料放入海水中一段时间(72h,120h,240h,720h),通过海水中生物自然生长附着的方式获得生物附着后的透明材料的试样。根据需要,可以在不同海域、不同深度分别布置试样。设置不同的试样浸入时间,可以获得不同附着程度的试样。
将光学玻璃试样放入海洋中,通过海水中生物自然生长附着的方式获得生物附着光学玻璃试样。本实施案例中设置的挂片时间,可以参照《GB/T 12763.6-2007海洋调查规范第六部分:海洋生物调查》中的微型生物污损调查进行。
(3)采用紫外可见分光光度法测量生物附着后监测评价对象的吸光度曲线,获得时间-吸光度关系标准曲线。采用紫外可见分光光度测量生物附着后透明材料的吸光度曲线,获得吸收峰的峰值或者对吸光度曲线进行积分。可以获得时间-吸光度关系标准曲线。
将挂片试样取出后,***带凹槽比色皿,采用紫外可见分光光度计(300-900nm)对光学玻璃试样的吸光度曲线进行测定。获得如图2所示吸光度曲线,以透光率(T)表示。测定附着后光学玻璃的污损生物附着试样的生物量(或者污损生物覆盖率、叶绿素a含量等)对不同时期试样进行标定。
(4)将步骤(3)获得的标准曲线与生物污损程度情况进行对比分析,获得实际的生物污损程度情况。通过透光率曲线峰值对生物附着试样的生物量(此处为叶绿素a含量) 建立标准工作曲线。采用叶绿素a含量进行定标,可以看出透光率曲线峰值(675nm)和叶绿素a含量线性相关(如图3所示)。因此通过透光率的测定,由标准曲线计算获得生物附着试样的叶绿素a含量,从而监测评估生物污损的程度。
(5)或者将步骤(3)获得的标准曲线与生物污损程度情况进行对比分析,获得实际的生物污损程度情况。通过不同时期的透光率曲线积分(透光率曲线对透光率100%曲线面积积分,如图4阴影部分)对应不同的生物附着试样污损生物覆盖率,建立相应的标准工作曲线。在通过测定不同样品的透光率曲线积分,由标准曲线计算获得生物附着试样的污损生物覆盖率,从而定量评估生物污损的程度。
实施例2
本发明所述方法同样适用于生物污损的现场监测。
通过实施例1中的方式建立标准工作曲线之后,通过现场实时监测获取海水中透明材料的透光率曲线峰值(675nm),可以计算获得生物附着试样的叶绿素a含量,从而实施现场监测评估材料生物污损的程度。
实施例仅说明本发明的技术方案,而非对其进行任何限制;尽管参照前述实施例对本发明进行了详细的说明,对于本领域的普通技术人员来说,依然可以对前述实施例所记载的技术方案进行修改,或者对其中部分技术特征进行等同替换;而这些修改或替换,并不使相应技术方案的本质脱离本发明所要求保护的技术方案的精神和范围。
Claims (5)
1.一种初期海洋生物污损监测评价方法,其特征在于,包括以下步骤:
(1)选取监测评价对象;
(2)将监测评价对象放入海水中,通过海水中生物自然生长附着的方式获得生物附着后的监测评价对象试样;
(3)采用紫外可见分光光度测量生物附着后监测评价对象的吸光度曲线,获得时间-吸光度关系标准曲线;
(4)将步骤(3)获得的标准曲线与生物污损程度情况进行对比分析,获得实际的生物污损程度情况。
2.根据权利要求1所述的一种初期海洋生物污损监测评价方法,其特征在于:步骤(3)通过获得吸收峰峰值或者对吸光度曲线积分的方式获得时间-吸光度关系标准曲线。
3.根据权利要求1所述的一种初期海洋生物污损监测评价方法,其特征在于:步骤(4)所述的生物污损程度情况是通过生物量和荧光显微镜法获得的。
4.根据权利要求1所述的一种初期海洋生物污损监测评价方法,其特征在于:步骤(2)可以在不同海域、不同深度分别设置监测评价对象,设置不同的浸入时间,获得不同附着程度的监测评价对象。
5.根据权利要求1-4任一所述的一种初期海洋生物污损监测评价方法,其特征在于:监测评价对象为光学玻璃。
