本发明属于生物技术领域,特别涉及一种棕榈酰化六肽、及其在抗肿瘤制剂、化妆品或食品中的应用。
背景技术
根据世界卫生组织(WHO)统计,全世界有3/5的人死于癌症、糖尿病、心血管疾病、慢性呼吸***疾病这4大类疾病,而癌症则是最主要的死因之一。2008年全球死于癌症的患者达760万人,占全球死亡人数的13%,其中超过70%的癌症死亡案例发生在中低收入国家,预测至2030年,全球将有超过110万人死于癌症。目前,对于癌症的治疗,药物治疗仍然是重要手段之一。因此,研究开发具有抗肿瘤活性的物质或制剂也一直是本领域技术人员的研究热点之一。
小分子肽是一类具有高活性的肽类物质,小分子肽可以以完整的形式被机体吸收;主动吸收、低耗或不需消耗能量的特点;能够直接进入血液循环并送到人体各个部位,广泛应用于生物医药领域。由于具有亲水性,蛋白质通常难以跨生物膜转运,因此,提高其膜渗透性改善其细胞内化率是重要的科学问题。通过对小分子肽进行修饰使其性能得以改善,也是本领域技术人员的研究方向之一。而开发具备抗肿瘤活性的小分子肽更是备受关注。
棕榈酰化是蛋白质翻译后脂质修饰的重要形式之一,在细胞信号传导、代谢、凋亡、疾病的发生、发展等过程中都起着重要的作用。根据连接方式不同,棕榈酰化可分为N型(棕榈酸盐通过酰胺键连接到Gly/Cys残基上)和S型(棕榈酸盐通过硫酯键共价修饰到Cys的巯基上)。
发明内容
本发明提供一种经棕榈酰化的六肽(Palm-GMCCSR),本发明的棕榈酰化六肽相比于未经棕榈酰化的六肽具有明显增强的抗肿瘤活性,而且具有抗衰老作用,可应用于生物制药和化妆品领域。
本发明所述的合成多肽缩写为Palm-GMCCSR,分子量894.24 Da,序列为:Palm-Gly-Met-Cys-Cys-Ser-Arg。其化学结构式如下:
其中,前文序列中的Palm表示英文名称为Palmitoyl,中文名称为棕榈酰基;Gly表示英文名称为Glycine,中文名称为甘氨酸的氨基酸的相应残基;Met表示英文名称为Methionine,中文名称为甲硫氨酸的氨基酸的相应残基;Cys表示英文名称为Cysteine,中文名称为半胱氨酸的氨基酸的相应残基;Ser表示英文名称为Serine,中文名称为丝氨酸的氨基酸的相应残基;Arg表示英文名称为Arginine,中文名称为精氨酸的氨基酸的相应残基。
本发明还提供上文所述的棕榈酰化六肽的纯化方法,可通过该纯化方法纯化经多肽固相合成法合成的棕榈酰化六肽,可以获得纯度大于95%的多肽,该方法包括如下步骤:
将待纯化的棕榈酰化六肽上样至色谱柱;
以流动相A和流动相B为洗脱液进行梯度洗脱,其中流动相A为含有0.08~0.12%三氟乙酸的乙腈溶液,流动相B为含有0.08~0.12%三氟乙酸的水;梯度洗脱程序为:流动相A的初始体积比例为18~22%,在上样后的第0.01min到第25min内,流动相A的体积比例上升到42~48%,在第25min到第25.1min内,流动相A的体积比例上升为100%,保持100%运行至第30min停止,检测波长为220nm;收集目标峰的多肽溶液。
进一步的,所述色谱柱为C18色谱柱;
优选的,流动相A为含有0.1%三氟乙酸的乙腈溶液,流动相B为含有0.1%三氟乙酸的水;和/或,梯度洗脱过程中,流动相A的初始体积比例为20%,在上样后的第0.01min到第25min内,流动相A的体积比例上升到45%。
本发明提供的棕榈酰化六肽可在制备抗肿瘤药物中应用。
进一步的,所述棕榈酰化六肽可在制备广谱抗肿瘤药物中应用;进一步的,所述肿瘤包括肝癌、肺癌、胃癌、乳腺癌、结肠癌。
进一步的,所述棕榈酰化六肽对HepG2、A549、SGC-7901、MCF-7、HT-29的细胞增殖抑制率达到70%以上。
本发明提供的棕榈酰化六肽还可应用于化妆品或食品中。进一步的,所述棕榈酰化六肽作为抗衰老活性成分应用于化妆品或食品中。
本发明还提供一种组合物,所述组合物中含有如上文所述的棕榈酰化六肽。进一步的,该组合物可以为抗肿瘤药物、化妆品或食品,其中棕榈酰化六肽作为其中的活性成分。
实验发现,本发明提供的经棕榈酰化的六肽Palm-GMCCSR,与修饰前的六肽GMCCSR相比,抗肿瘤活性显著增强,且表现出广谱抗癌活性。对HepG2、A549、SGC-7901、MCF-7、HT-29等癌细胞的增殖抑制作用均超过70%,其中,对A549和SGC-7901细胞的抑制活性显著强于其他细胞。
本发明提供的经棕榈酰化的抗肿瘤六肽可应用于抗肿瘤药物的制备,更进一步的,可以应用于广谱抗肿瘤药物的制备。所述肿瘤包括但不限于肝癌、肺癌、胃癌、乳腺癌、结肠癌等。本发明提供的经棕榈酰化的抗肿瘤六肽还可作为抗衰老活性成分或抗皮肤光老化活性成分应用于化妆品中,即,可以用于制备抗皮肤衰老功能的化妆品。

具体实施方式
下面结合附图对本发明的技术方案做进一步说明,但本发明的实施和保护范围不限于此。以下实施例中所用细胞或试剂若未特别说明,均为商业渠道购买获得。实施例中的实验操作若未特别说明,均为本领域的常规技术操作。下文实施例中所称六肽均为GMCCSR,棕榈酰化六肽均为Palm-GMCCSR。
通过多肽固相合成法合成Palm-GMCCSR,采用本领域常规多肽固相合成方法,具体可以通过商业化的生物合成公司来完成该多肽的合成。氨基酸序列采用本领域常规的标准Fmoc方案,下面对其进行介绍以供参考。
多肽固相合成:选用二氯三苯甲基树脂(上海杰肽生物科技有限公司),按照氨基酸序列Gly-Met-Cys-Cys-Ser-Arg的特征,先将Arg的羧基以共价键的形式与一个树脂相连,然后Arg的氨基和Ser的羧基缩水反应,处理后,再添加Cys,Ser的氨基和Cys的羧基反应,依次从右到左添加氨基酸,加好最后一个Gly氨基酸后,再切除树脂即得到六肽GMCCSR。氨基酸链合成完毕后,将活化好的棕榈酸在苯并三氮唑-N,N,N′,N′-四甲基脲六氟磷酸盐(HBTU)的催化下与肽链N端氨基反应,使用比例根据前面氨基酸的整体酸碱性更改。整个过程把棕榈酸当做一个氨基酸来参与正常反应,后面不再链接氨基酸,同时不需要洗脱步骤的脱FMOC。
棕榈酰化六肽的纯化:
采用高效液相色谱纯化通过多肽固相合成法得到最后的产品,纯化过程使用kromasil C18-5(4.6*250mm)色谱柱,流速为1.0mL/min。液相***使用两个通道,溶剂A(流动相A):含有0.1%(体积)三氟乙酸的乙腈;溶剂B(流动相B):含有0.1%(体积)三氟乙酸的水。梯度洗脱如下:A+B的体积百分比之和为100%,A初始比例为20%,在0.01min到25min内,A的比例上升到45%,在25min到25.1min内,A的比例上升为100%,保持100%运行至30min停止,检测波长220nm。收集多肽溶液,液氮速冷,然后冻干。得到纯度95%以上的产品,纯度鉴定HPLC结果见图1,并经ESI-MS鉴定结构(如图2所示)。经鉴定,多肽的化学结构如下:
对棕榈酰化前的六肽GMCCSR和棕榈酰化后的六肽Palm-GMCCSR进行二级结构分析,结果如下表1所示:

总结,由上表1可知,经过棕榈酰化后,α-螺旋和无规卷曲结构显著增加(P<0.05),β-折叠比例显著降低(P<0.05)。
多肽对肿瘤细胞的增殖抑制作用
HepG2、A549、SGC-7901、MCF-7、HT-29等癌细胞培养在完全培养基中,完全培养基由基础培养基高糖DMEM,10%胎牛血清(v/v),以及1%双抗(由青霉素和链霉素组成,v/v)组成。置于37℃、CO2体积分数为5%的饱和湿度培养箱中。每隔1d换液1次。待细胞长满培养瓶的90%左右,
用含有0.25%EDTA的胰酶将其消化下来,离心,用完全培养基悬浮细胞,用血球计数器计数,配成浓度为5×104cells/mL细胞悬液,取96孔板一块,每孔100μL,即每孔5000个细胞。在37℃、CO2体积分数为5%的饱和湿度培养箱中培养48h。同时,分别配制浓度为100μg/mL的六肽和棕榈酰化六肽溶液,每孔加入200μL样品溶液(正常对照组,则用完全培养基替代;阳性对照组,用5-氟尿嘧啶替代),孵育48h,吸弃样品,用PBS洗1遍后,采用MTT法检测细胞存活率。
实施例1
取肝癌细胞HepG2一瓶,用含有0.25%EDTA的胰酶将其消化下来,离心,用完全培养基悬浮细胞,用血球计数器计数,配成浓度为5×104cells/mL细胞悬液,取96孔板一块,每孔100μL,即每孔5000个细胞。在37℃、CO2体积分数为5%的饱和湿度培养箱中培养48h。同时,分别配制浓度为100μg/mL的六肽和棕榈酰化六肽溶液,每孔加入200μL样品溶液(正常对照组,则用完全培养基替代;阳性对照组,用5-氟尿嘧啶替代),孵育48h,吸弃样品,用PBS洗1遍后,采用MTT法检测细胞存活率。
实施例2
取肺癌细胞A549一瓶,用含有0.25%EDTA的胰酶将其消化下来,离心,用完全培养基悬浮细胞,用血球计数器计数,配成浓度为5×104cells/mL细胞悬液,取96孔板一块,每孔100μL,即每孔5000个细胞。在37℃、CO2体积分数为5%的饱和湿度培养箱中培养48h。同时,分别配制浓度为100μg/mL的六肽和棕榈酰化六肽溶液,每孔加入200μL样品溶液(正常对照组,则用完全培养基替代;阳性对照组,用5-氟尿嘧啶替代),孵育48h,吸弃样品,用PBS洗1遍后,采用MTT法检测细胞存活率。
实施例3
取胃癌细胞SGC-7901一瓶,用含有0.25%EDTA的胰酶将其消化下来,离心,用完全培养基悬浮细胞,用血球计数器计数,配成浓度为5×104cells/mL细胞悬液,取96孔板一块,每孔100μL,即每孔5000个细胞。在37℃、CO2体积分数为5%的饱和湿度培养箱中培养48h。同时,配制浓度为100μg/mL的六肽和棕榈酰化六肽溶液,每孔加入200μL样品溶液(正常对照组,则用完全培养基替代;阳性对照组,用5-氟尿嘧啶替代),孵育48h,吸弃样品,用PBS洗1遍后,采用MTT法检测细胞存活率。
实施例4
取乳腺癌细胞MCF-7一瓶,用含有0.25%EDTA的胰酶将其消化下来,离心,用完全培养基悬浮细胞,用血球计数器计数,配成浓度为5×104cells/mL细胞悬液,取96孔板一块,每孔100μL,即每孔5000个细胞。在37℃、CO2体积分数为5%的饱和湿度培养箱中培养48h。同时,配制浓度为100μg/mL的六肽和棕榈酰化六肽溶液,每孔加入200μL样品溶液(正常对照组,则用完全培养基替代;阳性对照组,用5-氟尿嘧啶替代),孵育48h,吸弃样品,用PBS洗1遍后,采用MTT法检测细胞存活率。
实施例5
取结肠癌细胞HT-29一瓶,用含有0.25%EDTA的胰酶将其消化下来,离心,用完全培养基悬浮细胞,用血球计数器计数,配成浓度为5×104cells/mL细胞悬液,取96孔板一块,每孔100μL,即每孔5000个细胞。在37℃、CO2体积分数为5%的饱和湿度培养箱中培养48h。同时,配制浓度为100μg/mL的六肽和棕榈酰化六肽溶液,每孔加入200μL样品溶液(正常对照组,则用完全培养基替代;阳性对照组,用5-氟尿嘧啶替代),孵育48h,吸弃样品,用PBS洗1遍后,采用MTT法检测细胞存活率。
实施例1-5的实验结果参见图3,从图3的实验结果可见,经过棕榈酰化修饰之后,本发明的棕榈酰化六肽的抗肿瘤活性显著增强,且表现出广谱抗癌活性。对HepG2、A549、SGC-7901、MCF-7、HT-29等癌细胞的增殖抑制作用均超过70%,且对A549和SGC-7901细胞的抑制活性显著强于其他细胞。
实施例6
取黑色素瘤细胞A375一瓶,用含有0.25%EDTA的胰酶将其消化下来,离心,用完全培养基悬浮细胞,用血球计数器计数,配成浓度为5×104cells/mL细胞悬液,取96孔板一块,每孔100μL,即每孔5000个细胞。在37℃、CO2体积分数为5%的饱和湿度培养箱中培养48h。以在化妆品中应用广泛的五胜肽为阳性对照。分别配制浓度为50μg/mL的阳性对照五胜肽(Matrixyl,Palm-KTTKS)和棕榈酰化六肽溶液,每孔加入200μL样品溶液(正常对照组,则用完全培养基替代),孵育48h,吸弃样品,用PBS洗1遍后,采用MTT法检测细胞存活率。
实验结果参见图4,结果表明,同样都是棕榈酰化多肽,棕榈酰化六肽对人黑色素瘤A375细胞的抑制作用显著强于五胜肽。由于皮肤癌变也是皮肤老化的一种形式,从而说明,棕榈酰化六肽具备在抗衰老化妆品中应用的潜力。
实施例7
取人表皮永生化细胞Hacat一瓶,用含有0.25%EDTA的胰酶将其消化下来,离心,用完全培养基悬浮细胞,用血球计数器计数,配成浓度为5×104cells/mL细胞悬液,取96孔板一块,每孔100μL,即每孔5000个细胞。在37℃、CO2体积分数为5%的饱和湿度培养箱中培养48h。同时,分别配制浓度为100μg/mL的阳性对照五胜肽(Matrixyl,Palm-KTTKS)和棕榈酰化六肽溶液,每孔加入200μL样品溶液(正常对照组,则用完全培养基替代),孵育48h,吸弃样品,用PBS洗1遍后,采用MTT法检测细胞存活率。实验结果见表2所示。

本实验测试了棕榈酰化七肽对人表皮永生化细胞(Hacat)存活率的影响,以五胜肽作为阳性对照,结果表明,棕榈酰化六肽对Hacat的毒性显著弱于阳性对照五胜肽,该棕榈酰化六肽具备在化妆品中应用的潜力。
实施例8
选取SPF级的昆明种小白鼠(或称KM小鼠),7-8周龄,全为雌性,体重25g,共56只,分为7组。购自广州南方医科大学实验动物中心,生产许可证:SCXK(粤)2011-0015,动物质量合格证为44002100005930。
饲养环境:温度23±2℃,湿度55±10%,12h光照,12h黑暗,交替,其中没有任何紫外线照射,自由供给食物和水。进行实验前先让小鼠适应环境1个星期。然后进行随机分组。
将56只KM小鼠随机分为以下4组:A为正常组(Normal control,NC)、B为模型组(Model control,MC)、C为阳性对照组(五胜肽Palm-KTTKS,Matrixyl)、D为棕榈酰化六肽(Palm-GMCCSR)预防组一共4组,每组8只。两笼一组,保持同种条件下,正常饲养,让其适应环境一周再进行后续实验。
首次用电推剪将所有小鼠的背部2.5x3cm2区域的毛发剪掉,短毛用全自动女士剃毛器轻柔剃光,在正式实验前,适应环境两天。从第一周至第十周对小鼠背部皮肤进行加药和UVB处理十周,每次处理前均进行脱毛处理,具体处理按如下要求操作:剃毛结束后,正常组给溶剂100μL,模型组给溶剂100μL,阳性对照组和棕榈酰化六肽组分别给样品100μL(10mg/mL);给药结束后,给予小鼠足够的活动空间让其单独活动2h,让样品充分渗透吸收,随后,模型组、阳性对照组和棕榈酰化六肽组均用60mJ/cm2的UVB照射,实验期间,若出现红斑、水泡和糜烂现象,立即停止照射2-3d,待症状消失再继续实验;各实验组给药频率为一周三次。
第10周末,仔细将各组小鼠背部脱毛(面积为2.5x3cm2),颈椎脱臼处死小鼠,迅速取下其背部实验处全层皮肤,剥离***和皮下脂肪,快速切取1.0cm x1.0cm组织,放入4%的多聚甲醛中固定两天以上。随后,取出固定好的皮肤组织,用流水漂洗过夜,并进行石蜡包埋,切片,染色,拍照。得到200x视野下的HE染色的照片后,随机取10个视野,取平均值来估算本样品所代表的小鼠皮肤表皮的平均厚度。Image Pro Plus 6.0图像分析软件被用于对受试鼠皮肤表皮厚度进行分析。
同时,快速切取约0.5g皮肤,用预冷生理盐水漂洗2次,拭干,称重,迅速放入预冷EP管中。用0.9%生理盐水制备10%的皮肤组织匀浆。涂片镜检,待3个以上随机视野均未见完整细胞停止匀浆,离心,去上清液备用。首先采用BCA试剂盒测定蛋白含量,谷胱甘肽过氧化物酶(GSH-Px)试剂盒测定GSH-Px酶活,羟脯氨酸试剂盒测定胶原蛋白含量,由于羟脯氨酸Hydroxyproline(Hyp)是哺乳动物一种特色氨基酸,可以根据其含量乘以系数7.46来确定胶原蛋白的含量。以上指标均严格按照说明书进行操作。
实验结果参见图5-图8。
图5中A-D为小鼠背部皮肤的宏观表征图;A1-D1为皮肤组织病理学切片检查结果(200x)。其中,A和A1为仅涂抹溶剂的正常对照组;B和B1为UVB老化的模型组;C和C1为加了阳性对照的保护组;D和D1为加了棕榈酰化六肽的保护组。
由图5可知,正常组小鼠背部皮肤光滑,色泽红润、饱满、充满弹性、未见任何松弛现象,同时,H&E染色结果表明,正常组皮肤中各层结构完整,皮下毛囊和皮脂腺形态饱满、充盈,表皮较薄,表皮与真皮连接处紧密,真皮层厚度适中,浪状胶原纤维束排列有序且分布均匀,未见病理改变。模型组小鼠背部皮肤表面灰暗,外观松弛,皱纹深重,并且表面皮层出现局部老化坏死现象。初步可以看出,所有样品处理组均比模型组小鼠皮肤有一定程度上的改善。H&E染色结果表明,模型组表皮呈现不规则增厚、细胞核碎裂、局部表皮角化不完全、基底层细胞少量细胞空泡性变、并伴有炎性细胞(淋巴细胞,单核细胞)浸润等现象。阳性对照五胜肽保护组相对于模型组而言,肤色有所改善,不再那么灰暗,皮肤的光泽度和饱满度上都有不同程度的改善,但还是能看到较为明显的晒伤的斑痕。H&E染色结果表明,阳性对照组表皮基底层少量细胞空泡性变,真皮层中度水肿伴少量炎性细胞(淋巴细胞,单核细胞)浸润。棕榈酰化六肽保护组小鼠的背部皮肤局部皮肤损伤得到改善,相较于模型组,肤色较亮,未见松弛和深重皱纹,H&E染色结果与阳性对照组类似,结果表明,该棕榈酰化六肽具备在抗衰老化妆品中应用的潜力。
图6为皮肤表皮厚度测定图。注释:*代表与正常组差异显著;#代表与模型组差异显著。由图6可知,相对于正常组,UVB处理的模型组小鼠的表皮厚度均显著增加,模型组增加最为显著;相对于模型组,阳性对照组和棕榈酰化六肽处理组小鼠的表皮厚度均显著降低。
图7为皮肤组织GSH-Px活力的测定图。其中,*代表与正常组差异显著;#代表与模型组差异显著。由图7可知,模型组相对于正常组,皮肤组织GSH-Px活力显著降低,相对于模型组,阳性对照组和棕榈酰化六肽处理组小鼠皮肤组织GSH-Px活力均显著升高。
图8为皮肤组织胶原蛋白含量测定图。其中,*代表与正常组差异显著;#代表与模型组差异显著。胶原蛋白中羟脯氨酸占13.4%,弹性蛋白中含极少量,其他蛋白中均不存在。因此,本研究采用碱水解法测定皮肤组织中羟脯氨酸的含量,从而推算出胶原蛋白的含量。由图8可知,模型组小鼠皮肤组织中胶原蛋白含量显著低于正常组,阳性对照五胜肽保护组的胶原蛋白含量显著高于模型组,同时,棕榈酰化六肽保护组的胶原蛋白含量也显著高于模型组。
由此可知,棕榈酰化六肽对UVB老化的小鼠皮肤组织的抗光老化效果可以通过增加其皮肤组织中胶原蛋白的含量来实现。
综上可见,本发明提供的棕榈酰化六肽,与修饰前的六肽相比,抗肿瘤活性显著增强,且表现出广谱抗癌活性。对HepG2、A549、SGC-7901、MCF-7、HT-29等癌细胞的增殖抑制作用均超过70%,其中,对A549和SGC-7901细胞的抑制活性显著强于其他细胞。另外,还发现本发明的七肽在抗衰老化妆品中具有应用潜力,可用于抗皮肤光老化产品中。
以上所述,仅是本发明的较佳实施例而已,并非对本发明做任何形式上的限制,故凡未脱离本发明技术方案的内容,依据本发明的技术实质对以上实施例所做的任何简单修改、等同变化与修饰,均仍属于本发明技术方案的范围内。

2.如权利要求1所述的应用,其特征在于,所述棕榈酰化六肽的纯化方法包括如下步骤:
将待纯化的棕榈酰化六肽上样至色谱柱;
以流动相A和流动相B为洗脱液进行梯度洗脱,其中流动相A为含有0.08~0.12%三氟乙酸的乙腈溶液,流动相B为含有0.08~0.12%三氟乙酸的水;梯度洗脱程序为:流动相A的初始体积比例为18~22%,在上样后的第0.01min到第25min内,流动相A的体积比例上升到42~48%,在第25min到第25.1min内,流动相A的体积比例上升为100%,保持100%运行至第30min停止,检测波长为220nm;收集目标峰的多肽溶液。
3.根据权利要求2所述的应用,其特征在于,所述色谱柱为C18色谱柱;
和/或,流动相A为含有0.1%三氟乙酸的乙腈溶液,流动相B为含有0.1%三氟乙酸的水;
