背景技术
在口腔中通过氧化反应产生自由基可以刺激口腔粘膜细胞。这种氧化刺激可转而导致牙龈组织降解和牙周病。已经提出将抗氧化剂作为口腔护理组合物如牙膏的成分。抗氧化剂可抑制口腔中可能发生的刺激氧化反应,从而增强口腔健康。
需要提供具有增强的抗氧化功能的改良型口腔护理制剂。
发明内容
本发明人已经证明,口腔护理组合物(且特别是牙膏组合物)中常用于产生泡沫、清洁和乳化的诸如月桂基硫酸钠表面活性剂的表面活性剂可抵消由所述组合物提供的抗氧化益处。本发明人意外地发现,包含甲基椰油酰基牛磺酸钠和泊洛沙姆(poloxamer)的特定组合的表面活性剂体系可用于替代常规的表面活性剂如月桂基硫酸钠表面活性剂,同时保持泡沫产生和清洁活性而不抵消组合物的抗氧化作用。
因此,在第一方面中,提供了一种包含抗氧化剂和表面活性剂体系的口腔护理组合物,其中所述表面活性剂体系包含以所述组合物的总重量计量为0.7重量%至1.2重量%的甲基椰油酰基牛磺酸钠和任选地,以所述组合物的总重量计量为0.4至1.2重量%的泊洛沙姆。优选地,所述组合物包含泊洛沙姆。

甲基椰油酰基牛磺酸钠

泊洛沙姆
在一种方案中,本文提供的组合物包含泊洛沙姆。泊洛沙姆是由聚氧丙烯(聚(环氧丙烷))的中心疏水链以及侧接的两条聚氧乙烯(聚(环氧乙烷))的亲水链组成的非离子三嵌段共聚物。任何泊洛沙姆均可用于所述组合物中。可用于所述组合物中的泊洛沙姆包括而不限于泊洛沙姆188、泊洛沙姆237、泊洛沙姆401和泊洛沙姆407。在优选实施方案中,泊洛沙姆为泊洛沙姆407(由BASF作为PluronicTM F127或由Croda作为SynperonicTM PE/F127供应)。泊洛沙姆407具有比其他泊洛沙姆提高的水溶性和提高的乳化性。

抗氧化剂
在一种方案中,所述组合物包含抗氧化剂。如本文中所用的术语“抗氧化剂”意指通过任何公认的体外或体内抗氧化测定或试验确定,具有抗氧化活性的任何试剂。常用抗氧化试验方法包括自由基清除法(例如,使用1,1-二苯基-2-苦肼基(DPPH方法)或2,2′-联氮双3-乙基苯并噻唑啉-6-磺酸(ATBS方法)、液体过氧化法和抗氧化还原法。

按有效降低或减轻对口腔,且特别是对口腔粘膜的刺激的水平将抗氧化剂掺入组合物中。不受限制,以所述组合物的总重量计,组合物中的抗氧化剂水平的范围可从0.01重量%至1重量%。
在优选实施方案中,所述抗氧化剂为芸香苷。本发明人意外地发现,芸香苷在口腔护理制剂中具有比常规抗氧化剂如维生素E更强的抗氧化活性。通常,以所述组合物的总重量计,芸香苷以0.1重量%至1重量%、或0.2重量%至0.8重量%、或0.3重量%至0.6重量%的量存在于所述组合物中。优选地,以所述组合物的总重量计,芸香苷以0.2重量%至0.5重量%或0.3重量%至0.5重量%的量存在于所述组合物中。
载体和其他任选成分
本文定义的组合物适合应用于口腔且通常包含口腔可接受的载体。如本文中所用的表达“口腔可接受的载体”指供口腔使用的任何安全且可接受的材料。此类材料包括水或可含保湿剂如甘油、山梨糖醇、木糖醇等的其他溶剂。在一些方面,术语“口腔可接受的载体”包括除了抗氧化剂和表面活性剂体系的表面活性剂之外的口腔护理组合物的所有组分。在其他方面,该术语是指用于将抗氧化剂和表面活性剂体系的表面活性剂,和/或任何其他功能成分递送到口腔的惰性或非活性成分。
用于本发明的口腔可接受的载体包括用于生产漱口水或口腔清洗剂、牙膏、牙胶、牙粉、锭剂、口香糖、珠粒、可食用条、片剂等常规的已知载体。所述组合物可以此类形式提供。应选择载体,以相互相容且与所述组合物的其他成分相容。
本文提供的口腔护理组合物还可包含附加成分。这些附加成分可包括但不限于表面活性剂、稀释剂、pH调节剂、其他表面活性剂、脱敏剂、牙石防治剂、粘合剂、增稠剂、清洁剂、粘着剂、泡沫调节剂、口感剂、保湿剂、甜味剂、调味剂、着色剂、抗氧化剂、氟离子源、增白剂及其混合物。这类成分和它们可掺入的量将是口腔护理领域的技术人员所知的。然而,下面提供了这些成分的非限制性实例。
本文提供的组合物任选地掺入一种或多种脱敏剂。这些包括而不限于钾盐如硝酸钾、碳酸氢钾、氯化钾、柠檬酸钾和草酸钾;辣椒素;丁香酚;锶盐;锌盐;氯盐及其组合。根据所选试剂,可以按有效量添加此类试剂,例如,以所述组合物的总重量计约0.5重量%至约20重量%。本文定义的组合物也可用于在施加于牙齿表面时通过阻断牙本质小管来治疗过敏。
本文提供的组合物可任选地包括牙石防治剂如焦磷酸盐,包括二碱金属或四碱金属焦磷酸盐如Na4P2O7、K4P2O7、Na2K2P2O7、Na2H2P2O7和K2H2P2O7,三聚磷酸钠、长链聚磷酸盐如六偏磷酸钠和环状磷酸盐如三偏磷酸钠。
本文提供的组合物还可包含粘合剂。可利用任何常规粘合剂。合适的粘合剂包括海洋胶体;羧乙烯基聚合物;卡拉胶(carrageenan);淀粉;纤维素聚合物如羟乙基纤维素、羧甲基纤维素(羧甲纤维素)、羟丙基甲基纤维素及其盐(例如,羧甲纤维素钠);天然胶如刺梧桐胶、黄原胶、***树胶和黄蓍胶;壳聚糖;胶体硅酸镁铝;胶体二氧化硅。优选地,以所述组合物的总重量计,粘合剂以0.1重量%至5重量%的量存在于所述组合物中。
可掺入到本文定义的组合物中的增稠剂包括天然和合成胶和胶体。合适的增稠剂包括天然存在的聚合物,如卡拉胶、黄原胶、以商品名Polyox出售的不同分子量的聚乙二醇和聚乙烯吡咯烷酮。相容性无机增稠剂包括无定形二氧化硅化合物和以CabotCorporation制造的商品名Cab-o-sil可获得的胶体二氧化硅化合物。其他无机增稠剂包括天然和合成粘土如锂蒙脱石粘土、硅酸锂镁(laponite)和硅酸镁铝(Veegum)。
本文定义的组合物可任选地包含一种或多种粘着剂。粘着剂可以是聚合物粘着材料。聚合物粘着材料可以是附着于哺乳动物牙齿表面和/或也可存在于牙齿表面的不均匀生物膜上的任何试剂。附着可以通过任何方式进行,如离子相互作用、范德华力(van derWaals force)、疏水-亲水相互作用等。粘着材料可以是,例如,粘附于牙齿表面的任何均聚物或共聚物(下文统称为“聚合物”)。此类聚合物可包括纤维素聚合物,例如一种或多种羟烷基纤维素聚合物,如羟丙基甲基纤维素(HPMC)、羟乙基丙基纤维素(HEPC)、羟丁基甲基纤维素(HBMC)和羧甲基纤维素(CMC)。优选地,聚合物粘着材料包含至少一种纤维素材料,例如羧甲基纤维素钠。
聚合物粘着材料可选地或另外可包括聚(环氧乙烷)聚合物(如来自Dow Chemical的POLYOX)、线性PVP和交联PVP、PEG/PPG共聚物(如BASF Pluracare L1220)、环氧乙烷(EO)-环氧丙烷(PO)嵌段共聚物(如以商标Pluronic可从BASF Corporation获得的聚合物)、酯胶、虫胶、压敏硅酮胶粘剂(如来自Dow-Corning的BioPSA)、甲基丙烯酸酯或其混合物。在一个实施方案中,共聚物包含(PVM/MA)。任选地,共聚物可选自:聚(甲基乙烯基醚/马来酸酐)或聚(甲基乙烯基醚/马来酸)或聚(甲基乙烯基醚/马来酸)半酯或聚(甲基乙烯基醚/马来酸)混合盐。
可使用任何分子量的聚合物,包括,例如分子量为50,000至500,000Da、500,000至2,500,000Da或2,500,000至10,000,000Da(按数均或重均计算)。
本文定义的口腔护理组合物还包括泡沫调节剂。泡沫调节剂通常增加产生的泡沫量,例如当使用所述组合物根据本文定义的方法刷洗口腔时。增加泡沫量的泡沫调节剂的说明性实例包括但不限于聚氧乙烯和某些聚合物,包括海藻酸盐聚合物。
以所述组合物的总重量计,起泡剂优选以0.05至0.5重量%或0.1至约0.2重量%的量存在于口腔护理组合物中。
聚氧乙烯可增加由本发明的口腔护理载体组分所产生的泡沫的量和泡沫的浓密程度。聚氧乙烯也通常被称为聚乙二醇(“PEG”)或聚氧化乙烯。适于本发明的聚氧乙烯的分子量将为200,000至7,000,000Da,且优选为600,000至2,000,000Da,且更优选为800,000至1,000,000Da。为由Union Carbide生产的高分子量聚氧乙烯的商标名。
优选地,本文提供的组合物还包含至少一种pH调节剂。此类试剂包括降低pH的酸化剂、提高pH的碱化剂和将pH控制在所需范围内的缓冲剂。pH调节剂优选包含碱化剂和/或缓冲剂。例如,可包括选自酸化剂、碱化剂和缓冲剂的一种或多种化合物以提供2至10的pH,或在各说明性实施方案中,2至8、3至9、4至8、5至7、6至10或7至9的pH。可以使用任何口腔可接受的pH调节剂,包括而不限于羧酸、磷酸和磺酸、酸式盐(例如,柠檬酸一钠、柠檬酸二钠、苹果酸一钠);碱金属氢氧化物如氢氧化钠;碳酸盐如碳酸钠、碳酸氢钠和倍半碳酸钠;硼酸盐;硅酸盐;磷酸盐(例如,磷酸一钠、磷酸三钠、焦磷酸盐)、咪唑等。一种或多种pH调节剂优选以有效地将组合物维持在口腔可接受的pH范围内的总量存在。
可以掺入到在本文定义的方法中使用的组合物中的口感剂包括在组合物使用期间赋予理想的质地或其他感觉的材料。此类试剂包括碳酸氢盐,其由于二氧化碳的泡腾和释放而赋予牙齿和牙龈“清洁感”。可以使用任何口腔可接受的碳酸氢盐,包括而不限于碱金属碳酸氢盐,如碳酸氢钠和碳酸氢钾、碳酸氢铵及其混合物。以所述组合物的总重量计,一种或多种碳酸氢盐任选地以0.1重量%至20重量%,例如1重量%至15重量%的总量存在。
本文提供的组合物可任选地包含甜味剂。可用于本发明组合物中的甜味剂包括人造甜味剂,如糖精、乙酰磺胺酸、纽甜(neotam)、环己烷氨基磺酸盐或三氯蔗糖(sucralose);天然高强度甜味剂,如索马甜(thaumatin)、甜菊苷或甘草甜素(glycyrrhizin);或糖醇如山梨糖醇、木糖醇、麦芽糖醇或甘露糖醇。按所述组合物的总重量计,这些可以按高达0.5重量%,任选地0.005重量%至0.1重量%的量存在。
本文提供的组合物可任选地包含调味剂。可用于本发明组合物中的调味剂包括精油以及各种调味醛、酯、醇以及类似物质。精油的实例包括绿薄荷油、薄荷油、茴香子油、冬青油、檫树、丁香油、鼠尾草油、桉树油、马郁兰油、肉桂油、柠檬油、莱姆酸橙(lime)油、葡萄柚油以及橙油。诸如薄荷醇、香芹酮以及茴香脑等化学品也适用。这些中,最常用的是薄荷油和绿薄荷油。以所述组合物的总重量计,调味剂可以按0.1重量%至5重量%或0.5重量%至1.5重量%的量掺入所述组合物中。
本文提供的组合物可包含至少一种着色剂。本文的着色剂包括赋予特定光泽或反射性的色素、染料和试剂,例如珠光剂。可以使用任何口腔头可接受的着色剂,包括而不限于滑石、云母、碳酸镁、碳酸钙、硅酸镁、硅酸镁铝、二氧化硅、二氧化钛、氧化锌、红、黄、棕和黑色氧化铁、亚铁氰化铁铵、锰紫、群青、钛酸云母、氯氧化铋等。以所述组合物的总重量计,一种或多种着色剂任选地以0.001重量%至20重量%、0.01重量%至10重量%、或0.1重量%至5重量%的总量存在。
防腐剂如洗必泰(chlorhexidine)、三氯生(triclosan)、季铵化合物(如苯扎氯铵)或对羟基苯甲酸酯(如对羟基苯甲酸甲酯或对羟基苯甲酸丙酯)可以掺入本发明方法中使用的组合物中。以所述组合物的总重量计,防腐剂的量通常高达0.5重量%,任选地0.05至0.1重量%。
任选地,本文定义的组合物包含氟离子源。氟离子源包括但不限于:氟化亚锡、氟化钠、氟化钾、单氟磷酸钾、单氟磷酸钠、单氟磷酸铵、氟硅酸钠、氟硅酸铵、胺氟化物如olaflur(N′-十八烷基三亚甲基二胺-Nionic,N,N′-三(2-乙醇)-二氢氟化物)、氟化铵及其组合。任选地,氟离子源包括氟化亚锡、氟化钠、胺氟化物、单氟磷酸钠及其混合物。优选地,本发明的口腔护理组合物也可含有提供氟离子或氟的成分的来源,所述来源的量足以供应约50至约5000ppm的氟离子,例如,约100至约1000ppm、约200至约500ppm或约250ppm的氟离子。可以按0.001重量%至10重量%,例如0.003重量%至5重量%、或0.01重量%至1重量%或至0.05重量%的量,向用于本发明的组合物中添加氟离子源。然而,应当理解,提供适当水平的氟离子的氟化物盐的重量将基于盐中的抗衡离子的重量而变化,并且本领域技术人员可以容易地确定这样的量。优选的氟化物盐可为氟化钠。
在优选实施方案中,所述组合物为牙膏组合物,其包含:
40重量%至60重量%的保湿剂,任选地甘油;

和任选地,0.5重量%至1重量%的泊洛沙姆。
研磨剂二氧化硅不同于增稠剂二氧化硅。通常,研磨剂(清洁)二氧化硅可表征为有约40至150cc/100g的吸油水平并且具有3mg损失/100,000转或更高的Einlelmer磨损。相反,增稠剂二氧化硅具有高于150cc/100g的吸油水平和小于2mg损失/100,000转的Einlelmer磨损。
研磨剂二氧化硅任选地为平均粒度高达约20微米的沉淀或水合二氧化硅,例如Zeodent 103、105、113、114、115或124(由J.M.Huber Chemicals Division,Havre deGrace,Md.21078销售)或Sylodent 783(由Davison Chemical Division of W.R.Grace&Company销售)。其他可能的研磨剂二氧化硅包括硅胶和沉淀无定形二氧化硅,其吸油值低于100cc/100g二氧化硅,任选地45cc/100g至低于约70cc/100g二氧化硅。这些二氧化硅是具有约3微米至约12微米和任选地约5微米至约10微米范围的平均粒度的胶态颗粒。
增稠剂二氧化硅包括Zeodent 163(具有190cc/100g的吸油水平和小于2mg损失/100,000转的Einlelmer磨损)、Zeodent 165(具有220cc/100g的吸油水平和小于2mg损失/100,000转的Einlelmer磨损)、Zeodent 167(具有235cc/100g的吸油水平和小于2mg损失/100,000转的Einlelmer磨损)(由J.M.Huber Chemicals Division,Havre de Grace,Md.21078销售)。
使用方法
本发明人已经发现,甲基椰油酰基牛磺酸钠表面活性剂,任选地与泊洛沙姆组合,对口腔粘膜没有任何刺激作用。因此,可有利地向口腔护理组合物中掺入甲基椰油酰基牛磺酸钠表面活性剂,任选地与泊洛沙姆组合,而不抵消所述组合物的抗氧化益处。
因此,在一种方案中,提供了一种包含抗氧化剂和表面活性剂体系的口腔护理组合物,其中所述表面活性剂体系包含以所述组合物的总重量计量为0.7重量%至1.2重量%的甲基椰油酰基牛磺酸钠和任选地,以所述组合物的总重量计量为0.4至1.2重量%的泊洛沙姆,并且其中所述组合物用于预防牙周病。
所述组合物可如本文所定义。在优选实施方案中,所述用途包括预防牙龈炎和/或牙龈萎缩。
在另一方案中,提供了在包含抗氧化剂的口腔护理组合物中的表面活性剂体系用于保持所述组合物的抗氧化作用的用途,其中所述表面活性剂体系包含甲基椰油酰基牛磺酸钠和任选地泊洛沙姆,其中口腔护理组合物包含以所述组合物的总重量计量为0.7重量%至1.2重量%的甲基椰油酰基牛磺酸钠和任选地,以所述组合物的总重量计量为0.4至1.2重量%的泊洛沙姆。所述组合物的抗氧化作用由抗氧化剂提供。
所述组合物可如本文所定义。在优选实施方案中,所述组合物的抗氧化作用包括口腔粘膜细胞保护作用。
还提供了保持包含抗氧化剂的口腔护理组合物的抗氧化作用的方法,该方法包括向所述组合物中掺入包含甲基椰油酰基牛磺酸钠和任选地泊洛沙姆的表面活性剂体系,其中口腔护理组合物包含以所述组合物的总重量计量为0.7重量%至1.2重量%的甲基椰油酰基牛磺酸钠和任选地,以所述组合物的总重量计量为0.4至1.2重量%的泊洛沙姆。保持抗氧化作用比使用替代表面活性剂如烷基硫酸酯表面活性剂时所观察到的程度更高。

以下实施例说明了本发明的方法及其用途。实施例是说明性的并不限制本发明的范围。
实施例
实施例1-表面活性剂对口腔粘膜细胞的作用(1)
制备不同浓度的月桂基醚硫酸钠(SLS)和甲基椰油酰基牛磺酸钠(SMCT)的水溶液。所述溶液用于激发口腔粘膜细胞。经处理的细胞用烟鲁绿B(Janus Green B)染色并使用光学显微镜观察形态变化以确定细胞是否已经被刺激或保持完好。如果细胞完好,细胞壁光滑,细胞核颜色清晰/深色,并且如果可以识别细胞器,则将细胞鉴定为正常。如果不能明确界定细胞,细胞壁模糊不清,如果存在细胞皱缩,如果细胞核和细胞质的颜色比对完好细胞观察到的更浅,并且如果无法识别细胞器,则将细胞鉴定为受刺激。表1中说明了结果。

使用SLS/SMCT对表2所说明的牙膏制剂重复实施例1的方法。

根据实施例1描述的标准,包含0.48%SLS和1.23%SMCT的制剂A显示出细胞刺激作用。包含0.48%SLS和1.71%SMCT的制剂B引起严重细胞刺激。
可以得出的结论是,SLS和SMCT的组合尽管起泡性好,但在口腔护理制剂中不适合。
实施例3-SMCT对口腔粘膜细胞的刺激作用
使用不同水平的SMCT对表3所说明的牙膏制剂重复实施例1的方法。

使用标准起泡试验(Ross Mile法)将包含0.87%SMCT的制剂C的起泡性能与包含1.71%SLS的同等制剂的起泡性能做比较。如表4中所见,制剂C产生的泡沫体积略微低于当前SLS制剂产生的泡沫体积。然而,起泡水平仍可接受。

实施例4-SMCT/泊洛沙姆407对口腔粘膜细胞的作用
泊洛沙姆407是具有极低细胞刺激作用的非离子型表面活性剂。因为泊洛沙姆407仅有非常低的细胞刺激作用并且旨在与其他阴离子表面活性剂组合使用,所以未测试纯泊洛沙姆407的细胞刺激作用。研究了包含SMCT和泊洛沙姆407的特定组合的表面活性剂体系,并通过对实施例1描述的方法测试了包含不同水平的组合SMCT/泊洛沙姆407的牙膏制剂的细胞刺激作用。下面表5中说明了测试的制剂和测试结果。

使用LPO-CC试剂盒(Kamiya Biomedical Company)测试芸香苷的抗氧化活性,并与维生素E的抗氧化活性进行比较(试剂按于乙醇中1%的量测试)。下面表7a和7b中说明了结果。光密度越低,抗氧化活性越好。过氧化氢异丙苯还原百分比越高,抗氧化活性越好。

实施例6-芸香苷的抗氧化活性(2)
制备芸香苷(0.3%)或维生素E(0.3%)的牙膏制剂。制剂包含:35重量%至60重量%的保湿剂、0.1重量%至0.4重量%的纤维素粘合剂、0.05重量%至0.4重量%的甜味剂、2重量%至4重量%的pH调节剂、15重量%至25重量%的研磨剂二氧化硅、1重量%至3重量%的增稠剂二氧化硅、2重量%至4重量%的增稠剂、1重量%至2重量%的锌盐、0.3重量%的芸香苷或维生素E和水(qs)。
口腔环境为水性。刷洗期间,芸香苷从牙膏组合物释放到水环境中。为了测定芸香苷的生物利用率,用水从制剂中萃取芸香苷并通过高效液相色谱法分析芸香苷水平。随后使制剂在室温或40℃下老化13周,并再次测量芸香苷水平。表8中说明了结果。

1.一种包含抗氧化剂和表面活性剂体系的口腔护理组合物,
其中所述抗氧化剂为0.2重量%至0.8重量%的量的芸香苷,并且
其中所述表面活性剂体系由以所述组合物的总重量计量为0.7重量%至1.2重量%的甲基椰油酰基牛磺酸钠和以所述组合物的总重量计量为0.5或1.0重量%的泊洛沙姆组成。

6.根据权利要求1所述的组合物,其中所述组合物选自漱口水、喷雾、牙膏、***条、口香糖和锭剂。
7.根据权利要求1所述的组合物,其中所述组合物为牙膏。
8.根据权利要求7所述的组合物,其中所述组合物为牙膏组合物,其包含:
40重量%至60重量%的保湿剂;
0.1重量%至0.4重量%的纤维素粘合剂;
0.05重量%至0.4重量%的甜味剂;
2重量%至4重量%的pH调节剂;
15重量%至25重量%的研磨剂二氧化硅;
1重量%至3重量%的增稠剂二氧化硅;
2重量%至4重量%的增稠剂;
5重量%至10重量%的水;并且
其中所述组合物还包含由以下组成的表面活性剂体系:
0.8重量%至1.0重量%的甲基椰油酰基牛磺酸钠;
和0.5重量%至1重量%的泊洛沙姆。
9.包含芸香苷的口腔护理组合物中的表面活性剂体系用于保持所述组合物的抗氧化作用的用途,其中所述表面活性剂体系由以所述组合物的总重量计量为0.7重量%至1.2重量%的甲基椰油酰基牛磺酸钠和以所述组合物的总重量计量为约0.5或约1.0重量%的泊洛沙姆组成。
