Description

Sputum treatment method for medical examination
Technical Field
The invention relates to a sputum treatment method, in particular to a sputum treatment method for medical examination. Belongs to the technical field of medical examination.
Background
The sputum specimen collection is a common means in medical examination, and there are two conventional methods for obtaining the sputum specimen, namely a natural expectoration method or a sputum aspiration method, and the sputum specimen obtained by the two methods has poor homogeneity, wherein the distribution of bacteria and the like is also very uneven, and objective and accurate examination results cannot be obtained naturally by direct examination, so that the sputum specimen is required to be pretreated before the medical examination.
Currently, commercial sputum digestants are generally used to treat sputum specimens, such as Sputasol SR0233, manufactured by OXOID, united kingdom, which contains dithiothreitol, sodium chloride, potassium chloride, disodium hydrogen phosphate, and the like as main components. In addition, some hospitals use trypsin or chymotrypsin to digest sputum.
However, there are two important treatment points for sputum specimen, one is homogenization and the other is to protect bacteria in the sputum specimen from being damaged, and the current sputum digestive juice product cannot satisfy the two points at the same time, and still has a great space for improvement.
Disclosure of Invention
The invention aims to overcome the defects of the prior art and provide a method for treating sputum for medical examination.
In order to achieve the purpose, the invention adopts the following technical scheme:
a method for treating sputum for medical examination comprises the following steps:
(1) adding the sputum specimen into a first treatment solution with the weight being 2-3 times that of the sputum specimen, and carrying out intermittent ultrasonic treatment at 0-5 ℃ for 2-3 minutes;
(2) adding 0.5-1 time of the second treatment liquid by weight, and carrying out intermittent ultrasonic treatment at 20-30 ℃ for 1-2 minutes;
(3) smearing, dyeing and sealing;
the first treatment liquid is prepared from the following components in parts by weight: 0.3-0.4 part of 2-mercaptoethanol, 0.6-0.8 part of lauric acid monoethanolamide sodium sulfate, 12-18 parts of potassium chloride, 1-2 parts of monopotassium phosphate, 8-10 parts of dipotassium hydrogen phosphate and 1500-1800 parts of distilled water; the second treatment liquid is prepared from the following components in parts by weight: 1-2 parts of farrerol, 0.3-0.6 part of cynanchum glaucescens and 50-60 parts of ethanol; the intermittent ultrasonic treatment method comprises the step of treating once every 1 minute, wherein the treatment time is 20-30 seconds each time.
Preferably, the first treatment liquid is prepared by dissolving the formula amount of lauric acid monoethanolamide sodium sulfate in distilled water, sequentially adding potassium dihydrogen phosphate, dipotassium hydrogen phosphate and potassium chloride, stirring for dissolving, finally adding the formula amount of 2-mercaptoethanol, and stirring for uniformly mixing.
Preferably, the second treatment solution is prepared by adding farrerol and cynanchum glaucescens in a formula amount into ethanol, heating and refluxing for 2-3 hours, and filtering to obtain a filtrate.
Preferably, in the step (3), the staining is selected from hematoxylin-eosin staining or papanicolaou staining.
The invention has the beneficial effects that:
the treatment method disclosed by the invention not only realizes homogenization of the sputum specimen, but also protects bacteria in the sputum specimen from being damaged as much as possible, and has better clinical medical examination significance.
The viscous substance of the sputum is mainly glycoprotein combined by disulfide bond crosslinking, the disulfide bond is reduced to SH & lt- & gt by using 2-mercaptoethanol, but SH & lt- & gt is easily oxidized in the air and then forms a disulfide bond again, micelle protection is further formed by lauric acid monoethanolamide sodium sulfate so as to prevent re-crosslinking, and most of glycoprotein in the sputum specimen is decomposed by a reduction method.
Farrerol and cynanchum glaucescens can break the fiber of the remaining glycoprotein in the sputum, reduce the sialic acid content, reduce the viscosity of the sputum, and protect cells from being damaged.
The sputum is not transferred again in the treatment process, so that partial component loss possibly caused by the retransfer is avoided, and further, the detection result is inaccurate.
In addition, it is worth noting that the ultrasonic wave has a cavitation effect, and can generate a large amount of high-pressure and high-temperature bubbles, and the cell walls of the bacteria can be destroyed only after a period of time. The invention utilizes the resonance effect of ultrasonic waves to realize homogenization, and avoids the occurrence of cavitation by controlling intermittent ultrasonic treatment.
