Abstract

本发明公开了一种草本精华提取物,其按重量计含有总黄酮60‑65%、氨基酸1.3‑1.8%、植物甾醇0.8‑1.1%、单宁0.5‑1%、维生素0.6‑1.3%、超氧化物歧化酶0.2‑0.3%,余量为杂质。本发明还公开了其制备方法,采用新鲜的、重量比3:1:1的葎草、桑叶、刺梨制备,包括原料选取、粉碎、杀菌、研磨、浸泡、溶解、过滤、干燥等工序。本发明的有益组分搭配合理、提取其过程中有益组分损失更少、提取纯度更高。

Description

一种草本精华提取物及其制备方法
技术领域
本发明涉及生物提取领域,具体涉及一种以新鲜葎草、桑叶、刺梨为原料的草本精华提取物及其制备方法。
背景技术
葎草全草含木犀草素、葡萄糖甙、胆碱及天门各酰胺,其他尚有挥发油、鞣质及树脂。全草中主成分为黄酮类化合物,已分离出秋英甙、本犀草素-7-葡萄糖甙及牡荆素。可清热解毒、利尿消肿,外用治疗痈疖肿毒、湿疹、毒蛇咬伤等。其茎、叶的乙醇浸液在液试管内对革兰阳性菌如金黄色葡萄球菌、粪链球菌、肺炎链球菌、白喉杆菌、炭疽杆菌、枯草杆菌和蜡样芽胞杆菌有明显抑制作用。
桑叶是桑科植物桑树的叶,具有极高的经济价值。我国是桑树种类最多的国家。桑叶的传统应用主要是养蚕纺丝,有研究结果表明桑叶含有大量的蛋白质、多糖、总黄酮和生物碱等成分并表现出多种生物活性。桑叶中的许多成分具有重要的应用前景,从桑叶中分离出不同组分并开发其新应用不仅可提高桑叶的附加值,而且有利于植物资源的综合利用。
刺梨果实有很高的营养价值和医疗价值。其花、叶、果、籽可入药,具有解毒,健胃,消食、活血、化淤的功能,并能起到防癌抗癌的神奇作用。特别是刺梨富含超氧化物歧化酶(简称SOD),SOD是国际公认具有抗衰、防癌作用的活性物质,还具有抗病毒、抗辐射的作用,在心血管、消化***和各种肿瘤疾病防治方面,应用十分广泛。刺梨同时含有极丰富的胡萝卜素、维生素A、B2、C、D、E、P及钙磷铁等人体必须微量元素。据1985年刺梨专家孙学惠等研究发现,刺梨具有阿托品的解痛作用。
这三种均为最为常见但功效极佳的草本植物,其对人体有着很好的保健作用,且成本低廉。在目前的应用中,通常是入药、制作为食品,其保健功效未能得到全面的发挥;且通常单一使用,其有益成分未能起到互补的作用;少数研究者也采用提取的方法来制取单一的浓缩液、膏、粉,通常都用到超声波助溶提取、大孔吸附树脂吸附、沉淀冷冻干燥等步骤的一种或几种,其中超声波助溶提取是通过其空化效应来达到破碎细胞壁、加速有益组份溶解的目的,但因为恒定的超声波的作用较为有限,提取程度不高;大孔吸附树脂吸附的效果不错,但有益组分吸附后洗脱的效率不高,损失较大;沉淀冷冻干燥会造成部分有益物质在低温下失活。
发明内容
针对现有技术中存在的上述缺陷,本发明旨在提供一种有益组分搭配合理的草本精华提取物及在提取其过程中有益组分损失更少、提取纯度更高的制备方法。
为了实现上述目的,本发明采用以下技术方案:一种草本精华提取物,其中:按重量计含有总黄酮60-65%、氨基酸1.3-1.8%、植物甾醇0.8-1.1%、单宁0.5-1%、维生素0.6-1.3%、超氧化物歧化酶0.2-0.3%,余量为杂质。
上述的草本精华提取物,其中:所述总黄酮包含芸香苷、槲皮素、异槲皮苷、木犀草素、牡荆素。
上述的草本精华提取物的制备方法,其包括以下步骤:
1)原料选取:原料为新鲜的葎草、桑叶、刺梨,配合比为重量比3:1:1;
2)粉碎:将混合原料粉碎成粗粉,细度不大于5毫米;
3)杀菌:在传送带上将原料摊薄,输送过程中采用紫外线除菌;
4)研磨:将原料粗粉加其50%重量的水,采用高强度陶瓷球对混合进行研磨,至浆料细度D90≤300微米停磨,过40目筛出磨;
5)浸泡:向滤液中加入乙醇,调节溶液的乙醇体积浓度为60-75%,搅拌后静置4-5小时;
6)溶解:对溶液施加脉冲超声波,参数为重复频率50-100HZ、功率400-500w,时间为40-50分钟;
7)过滤:滤除沉淀,留取滤液;
8)干燥:喷雾干燥,得到所述草本精华提取物。
上述的草本精华提取物的制备方法,其中:脉冲超声波的参数为重复频率60-70HZ、功率450-500w,时间为45-50分钟。
与现有技术比较,本发明的草本精华提取物由于采用了上述方案,含有的有益成分搭配合理,能广泛应用于多种领域,比如制造保健药物、牙膏、漱口水、化妆品等;具体而言,其含有总黄酮高达60-65%,来自葎草和桑叶,种类包括包含芸香苷、槲皮素、异槲皮苷、木犀草素、牡荆素等,具有很强的抗氧化和消除自由基作用,帮助人体防御辐射、消除疲劳、保护血管、防动脉硬化、扩张毛细血管、疏通微循环、活化大脑及其他脏器细胞的功能、抗脂肪氧化、抗衰老;氨基酸主要来源于桑叶和刺梨,含量也达到1.3-1.8%,种类包括人体所必需的全部20多种氨基酸;植物甾醇含量0.8-1.1%,主要来源于桑叶,具有良好的抗氧性,并在治疗溃疡、降低人体胆固醇方面具有奇效;单宁含量0.5-1%,主要来自于刺梨,单宁具有清除自由基和抑制弹性蛋白酶活力的作用,同时促进细胞新陈代谢、培养皮肤活力使其保持健康、有光泽、有弹性;维生素0.6-1.3%,主要来源于刺梨和桑叶,包含了人体必须的维生素A、B2、C、D、E、P等;超氧化物歧化酶主要来源于刺梨和桑叶,含量达到了0.2-0.3%,是国际公认具有抗衰、防癌作用的活性物质,还具有抗病毒、抗辐射的作用,在心血管、消化***和各种肿瘤疾病防治方面,应用十分广泛。
同时,与现有技术比较,本发明的制备方法由于采用了上述方案,具有以下优点:原料选取三种常见草本植物,成本低,有益成分搭配均衡合力;粉碎使原料初步破碎,便于紫外线杀菌;研磨至300微米以下,确保能使后续工序的浸泡、溶解效率更高;浸泡是为了能使乙醇的萃取作用得到充分发挥;溶解工序中采用的超声波并非传统助溶采取的恒定超声波,而是采用脉冲超声波,这使本发明的制备方法在溶解效率、溶解度方面达到了极高的水平,其原理是因为脉冲超声波既能使细胞壁在疲劳效应作用下更易破碎、有益成分更易释放,且脉冲带来的压力波是变化的,对溶液本身能起到搅拌的作用,使溶解更加彻底,同时脉冲超声波的重复频率的优选范围是60-70HZ,较高的频率疲劳效应强但空化效应和搅拌作用弱,较低的反之;过滤是为了除杂;喷雾干燥是为了得到粉末状的草本精华提取物。
具体实施方式
实施例1:
采用以下方法制备草本精华提取物:
1)原料选取:原料为新鲜的葎草、桑叶、刺梨,配合比为重量比3:1:1;
2)粉碎:将混合原料粉碎成粗粉,细度不大于5毫米;
3)杀菌:在传送带上将原料摊薄,输送过程中采用紫外线除菌;
4)研磨:将原料粗粉加其50%重量的水,采用高强度陶瓷球对混合进行研磨,至浆料细度D90≤300微米停磨,过40目筛出磨;
5)浸泡:向滤液中加入乙醇,调节溶液的乙醇体积浓度为60%,搅拌后静置4-5小时;
6)溶解:对溶液施加脉冲超声波,参数为重复频率50HZ、功率400w,时间为40分钟;
7)过滤:滤除沉淀,留取滤液;
8)干燥:喷雾干燥,得到所述草本精华提取物。
制得的草本精华提取物的含量经测定,按重量计含有总黄酮60%、氨基酸1.3%、植物甾醇0.8%、单宁0.5%、维生素0.6%、超氧化物歧化酶0.2%,余量为杂质。其中,总黄酮包含芸香苷、槲皮素、异槲皮苷、木犀草素、牡荆素及其它含量较低的种类。
实施例2:
采用以下方法制备草本精华提取物:
1)原料选取:原料为新鲜的葎草、桑叶、刺梨,配合比为重量比3:1:1;
2)粉碎:将混合原料粉碎成粗粉,细度不大于5毫米;
3)杀菌:在传送带上将原料摊薄,输送过程中采用紫外线除菌;

5)浸泡:向滤液中加入乙醇,调节溶液的乙醇体积浓度为75%,搅拌后静置4-5小时;
6)溶解:对溶液施加脉冲超声波,参数为重复频率100HZ、功率500w,时间为50分钟;
7)过滤:滤除沉淀,留取滤液;
8)干燥:喷雾干燥,得到所述草本精华提取物。
制得的草本精华提取物的含量经测定,按重量计含有总黄酮62%、氨基酸1.5%、植物甾醇1%、单宁0.7%、维生素1.1%、超氧化物歧化酶0.26%,余量为杂质。其中,总黄酮包含芸香苷、槲皮素、异槲皮苷、木犀草素、牡荆素及其它含量较低的种类。
实施例3:
采用以下方法制备草本精华提取物:
1)原料选取:原料为新鲜的葎草、桑叶、刺梨,配合比为重量比3:1:1;
2)粉碎:将混合原料粉碎成粗粉,细度不大于5毫米;
3)杀菌:在传送带上将原料摊薄,输送过程中采用紫外线除菌;
4)研磨:将原料粗粉加其50%重量的水,采用高强度陶瓷球对混合进行研磨,至浆料细度D90≤300微米停磨,过40目筛出磨;
5)浸泡:向滤液中加入乙醇,调节溶液的乙醇体积浓度为70%,搅拌后静置4-5小时;
6)溶解:对溶液施加脉冲超声波,参数为重复频率70HZ、功率480w,时间为50分钟;
7)过滤:滤除沉淀,留取滤液;
8)干燥:喷雾干燥,得到所述草本精华提取物。
制得的草本精华提取物的含量经测定,按重量计含有总黄酮65%、氨基酸1.8%、植物甾醇1.1%、单宁1%、维生素1.3%、超氧化物歧化酶0.3%,余量为杂质。其中,总黄酮包含芸香苷、槲皮素、异槲皮苷、木犀草素、牡荆素及其它含量较低的种类。

1.一种草本精华提取物,其特征在于,按重量计含有总黄酮60-65%、氨基酸1.3-1.8%、植物甾醇0.8-1.1%、单宁0.5-1%、维生素0.6-1.3%、超氧化物歧化酶0.2-0.3%,余量为杂质。
2.如权利要求1所述的草本精华提取物,其特征在于,所述总黄酮包含芸香苷、槲皮素、异槲皮苷、木犀草素、牡荆素。
3.一种如权利要求2所述的草本精华提取物的制备方法,其特征在于,包括以下步骤:
1)原料选取:原料为新鲜的葎草、桑叶、刺梨,配合比为重量比3:1:1;
2)粉碎:将混合原料粉碎成粗粉,细度不大于5毫米;
3)杀菌:在传送带上将原料摊薄,输送过程中采用紫外线除菌;
4)研磨:将原料粗粉加其50%重量的水,采用高强度陶瓷球对混合进行研磨,至浆料细度D90≤300微米停磨,过40目筛出磨;
5)浸泡:向滤液中加入乙醇,调节溶液的乙醇体积浓度为60-75%,搅拌后静置4-5小时;
6)溶解:对溶液施加脉冲超声波,参数为重复频率50-100HZ、功率400-500w,时间为40-50分钟;
7)过滤:滤除沉淀,留取滤液;
