Abstract
本发明一种深沉发酵的银耳芽孢、桑黄菌丝体混合饮品,将桑黄菌种、银耳菌种接种到PDA斜面试管,25℃温箱里恒温避光培养5?7天;分别转接到马铃薯葡萄糖琼脂培养基平板上,培养7?9天;将培养好的桑黄菌种和银耳菌种菌丝体分别接种于三角瓶液体培养基中进行振荡培养,转速120r/min,25℃恒温避光振荡培养5?7天;将培养好的两种三角瓶菌种接种到液体发酵罐中通气恒温25℃发酵,桑黄7?9天得到菌丝体,银耳4?6天得到银耳芽孢;取样检测,按两种菌液1:1体积比混合,加入配料;巴士灭菌,即得。产品口感较好,可增强人体抵抗力。

技术领域
本发明属于菌类饮品技术领域,涉及一种深沉发酵的银耳芽孢、桑黄菌丝体混合饮品。
背景技术
桑黄菌是一种寄生于腐烂桑树上的真菌,为多年生珍稀药用真菌, 是目前国际公认的生物抗癌领域中效果最好的真菌,我国只有东北、西北和西南的少数地区能出产桑黄, 产量极为有限。桑黄的主要活性成分是桑黄多糖,同时是目前发现少有产黄酮类化合物的一种珍贵药用真菌。传统上桑黄多糖都是从子实体中提取,黄酮主要从药用植物中提取,从微生物中获得较少。但由于受到桑黄生理生态的特殊性和复杂性以及外部环境的制约,在自然界形成的子实体数量有限,人工栽培的难度也比较大,桑黄资源匮乏,使得桑黄的开发利用受到制约。虽然桑黄的人工种植还有较大困难,但其菌丝体经发酵还是可以大量得到,这就有效弥补桑黄子实体资源有限导致的其开发利用不足。
银耳味甘、淡、性平、无毒,既有补脾开胃的功效,又有益气清肠、滋阴润肺的作用。既能增强人体免疫力,又可增强肿瘤患者对放、化疗的耐受力。银耳富有天然植物性胶质,外加其具有滋阴的作用,是可以长期服用的良好润肤食品。
然而现有还没有结合上述两种原料的饮品出现。
发明内容
本发明的目的是提供一种深沉发酵的银耳芽孢、桑黄菌丝体混合饮品,生产操作简单可行,原料简单安全可靠,产品口感好,是有机、营养、保健的绿色食品。
本发明所采用的技术方案是,一种深沉发酵的银耳芽孢、桑黄菌丝体混合饮品,具体按照以下步骤进行:
步骤1,菌种活化:将桑黄菌种、银耳菌种接种到PDA斜面试管,25℃温箱里恒温避光培养5-7天;
步骤2,将已活化的桑黄菌种和银耳菌种分别转接到马铃薯葡萄糖琼脂培养基平板上,培养7-9天;
步骤3,将培养好的桑黄菌种和银耳菌种菌丝体分别接种于三角瓶液体培养基中进行振荡培养,转速120r/min,25℃恒温避光振荡培养5-7天;
步骤4,将培养好的两种三角瓶菌种接种到液体发酵罐中通气恒温25℃发酵,桑黄7-9天得到桑黄菌丝体,银耳4-6天得到银耳芽孢;
步骤5,取样检测,检测标准:纯度需达到不含杂菌,浓度需达到桑黄菌丝浓度和银耳芽孢浓度40g/L-60g/L;
步骤6,按菌丝体液和银耳芽孢1:1体积比混合,再加入配料;
步骤7,灌装,巴士灭菌,即得。
进一步的,所述步骤3中三角瓶液体配方为:葡萄糖25g,大豆粉4g,玉米粉2g,MgSO40.5g,KH2PO40.5g,水1000ml。
进一步的,所述步骤4中液体发酵罐培养基配方为:葡萄糖25g,大豆粉4g,玉米粉2g,MgSO40.5g,KH2PO40.5g,水1000ml。
本发明的有益效果是:通过无菌深沉发酵技术得到大量的银耳芽孢、桑黄菌丝体,再将两者以一定比例混合,加入其他配料得到产品。产品中含有银耳多糖及桑黄多糖,许多研究表明这两种多糖均有增强人体抵抗力,具有一定抗肿瘤作用(如:《桑黄多糖抗肿瘤作用》—张敏、纪晓光、贝祝春、郑成贵、王京燕,《中药药理与临床》2006年Z1期;《银耳多糖的药理学研究概况》--聂伟、张永祥 、周金黄,《中药药理与临床》2000年4期;)。本发明产品制备方法方便快捷,能适用各种规模的生产,生产成本低。
具体实施方式
一种深沉发酵的银耳芽孢、桑黄菌丝体混合饮品,具体按照以下步骤进行:
步骤1,菌种活化:将桑黄菌种、银耳菌种接种到PDA斜面试管,25℃温箱里恒温避光培养5-7天;
步骤2,将已活化的桑黄菌种和银耳菌种分别转接到马铃薯葡萄糖琼脂培养基平板上,培养7-9天;
步骤3,将培养好的桑黄菌种和银耳菌种菌丝体分别接种于三角瓶液体培养基中进行振荡培养,转速120r/min,25℃恒温避光振荡培养5-7天;
其中三角瓶液体配方:葡萄糖25g,大豆粉4g,玉米粉2g,MgSO40.5g,KH2PO40.5g,水1000ml,原料来源要求绿色安全无污染,氮源采用大豆粉与玉米粉,既能满足菌丝生长需求又能促进桑黄多糖、黄酮的合成;
步骤4,将培养好的两种三角瓶菌种接种到液体发酵罐中通气恒温25℃发酵,桑黄7-9天得到桑黄菌丝体,银耳4-6天得到银耳芽孢;
液体发酵罐培养基配方:葡萄糖25g,大豆粉4g,玉米粉2g,MgSO40.5g,KH2PO40.5g,水1000ml;
步骤5,取样检测,检测标准:纯度需达到不含杂菌,浓度需达到菌丝浓度40g/L到60g/L;
步骤6,按菌丝体液和银耳芽孢1:1体积比混合,再加入配料;
步骤7,灌装,巴士灭菌,即得。

1.一种深沉发酵的银耳芽孢、桑黄菌丝体混合饮品,其特征在于,具体按照以下步骤进行:
步骤1,菌种活化:将桑黄菌种、银耳菌种接种到PDA斜面试管,25℃温箱里恒温避光培养5-7天;
步骤2,将已活化的桑黄菌种和银耳菌种分别转接到马铃薯葡萄糖琼脂培养基平板上,培养7-9天;
步骤3,将培养好的桑黄菌种和银耳菌种菌丝体分别接种于三角瓶液体培养基中进行振荡培养,转速120r/min,25℃恒温避光振荡培养5-7天;
步骤4,将培养好的两种三角瓶菌种接种到液体发酵罐中通气恒温25℃发酵,桑黄7-9天得到菌丝体,银耳4-6天得到银耳芽孢;
