Abstract
将经适当处理的含海藻糖的发酵液、酶转化液及酵母提取液中的任何一种浓缩液,用氧化钙(或氢氧化钙)-二氧化碳处理,过滤除去蛋白质,多糖等大分子物质及胶质,经离子交换树脂脱盐脱色,再浓缩、结晶,可得海藻糖晶体。本工艺具有除杂质效率高,过滤速度快,结晶时间短,生产成本低等特点。
Description
本发明属于食品、医药添加剂及生物制品活性保护剂的制造方法。
英国科学家布鲁斯·罗塞博士发现了“回生植物”的秘密,这些植物受旱50年,一遇到湿润的环境仍能复活的秘密在于这种植物中含有海藻糖。海藻糖是一种稳定的非还原性双糖,在生物体内扮演抵抗逆境的角色,它能在干燥状态下保护生物体细胞内的蛋白质、脂类、糖类、核酸等组分不受破坏。目前,主要用作生物制品活性保护剂、抗冷冻剂、食品保鲜剂、甜味剂,应用前景十分广阔。现已被有些国家批准用于食品工业的干法保存食品,易失活的疫苗和药品的保藏,甚至可用来培育抗旱作物。当前,影响海藻糖应用的主要限制因素是价格比较昂贵,改进海藻糖的生产方法,是降低成本的有效途径。
五十年代有一种从酵母中提取海藻糖的方法,即将一公斤面包酵母用2.5L95%乙醇抽提,放置30分钟,不时搅拌,过滤,滤饼用70%乙醇洗三次,每次300mL,滤液及洗涤减压浓缩至300mL,加入200mL20%硫酸锌溶液,以饱和氢氧化钡溶液调pH=8(约500mL)。加20克活性炭,沸水浴中加热至70℃,过滤,溶液通过离子交换树脂柱,例如Amberlite IR-120(H+)及Duolite A-4(OH)(55cm×5cm直径),除去离子后减压浓缩成一薄浆。稀释至50mL,慢慢加入400mL95%乙醇,立即析出α,α-海藻糖结晶。先在室温放一天,以后在冰箱中放置数天。滤出结晶,以乙醇洗涤,得α,α-海藻糖。
近十几年来,人们逐渐认识到海藻糖奇特的生物功能,纷纷对海藻的生产方法进行研究。日本专利介绍的一种生产工艺为:用70~95%酒精(最好是85~95%)从酿酒酵母中提取海藻糖,过滤除去固形物后,浓缩到海藻糖的浓度为20~30%,然后用0.05~5μm(最好是0.05~0.15μm)的微滤膜及截留分子量大于5000的超滤膜过滤浓缩液,将其流过酸性和碱性离子交换树脂后,真空浓缩到海藻糖的浓度为40~85%,加入酒精进行结晶或在水相结晶。
前者的工艺采用Zn2+沉淀蛋白质,用Ba(OH)2除SO4 2-,杂质滤除速度不快;除杂质时引入的离子较多,加重了后续离子交换的负担;采用酒精作结晶溶剂,结晶在低浓度下进行,增加了溶剂回收的成本。
后者的日本专利用膜滤的方法存在的问题是在提取液浓缩后,未经任何予处理便进行膜滤,致使滤膜极易堵塞,过滤速度慢,操作费用很高。
本发明的目的是创建一种成本低、可操作性强、易于工业放大生产海藻糖、高效除杂质、快速过滤、结晶的新工艺。
本发明适用于如下三种原料液:
(1)以庶糖、葡萄糖或淀粉糖化液为原料,经微生物发酵(本发明对发酵产生海藻糖的微生物无特别限制,如氨基酸发酵菌、根粒菌等)而得并经固液分离除去了菌体及其它不溶性固形物,并浓缩成含海藻糖其浓度大于5%,且海藻糖为寡糖份的50%以上的发酵液。
(2)以庶糖、葡萄糖、麦芽糖及淀粉糖化液为原料,经酶(本发明对生成海藻糖的酶无特别限制,可以是海藻糖合成酶或海藻糖磷酸化酶和其它糖的磷酸化酶协同作用)作用所得的海藻糖含量占寡糖份的50%以上的酶转化液。
(3)以酵母的原料,用乙醇含量70~95%的提取液经提取,固液分离除去菌体,再经真空浓缩回收酒精,控制浓缩比,让卵磷脂、麦角固醇等副产物以结粒膏状从提取液中筛分或离心脱除后的提取液。
本发明与现有技术相比,最大的不同点是在上述三种经浓缩后浓度为5~30%的原料液(即发酵液、酶转化液、提取液)中加入体积为原料液2~10%的氧化钙或氢氧化钙,先在pH≥12的条件下加热反应,将蛋白质等杂质凝聚或分解为其它产物,然后通入二氧化碳气体,使与过量的氧化钙或氢氧化钙生成碳酸钙作为吸附剂和助滤剂,当pH值降至11~12时,料液具有良好的过滤性能和除杂质效果,趁热过滤,向滤液中继续通入CO2气体,使料液的pH值降至8~9,再过滤以脱除钙盐。采用此种除杂质的方法,除杂质彻底,且杂质滤除速度快,操作成本低,并为后续工序的操作提供了方便。料液经上述处理后,清液先进入强碱性离子交换树脂,再进入强酸性离子交换树脂,将其它离子和色素脱盐脱色,最后将无色透明液浓缩至40~70%的浓度,并加入少许晶种,在水溶液中只需数小时就可析出海藻糖晶体,过滤出母液,晶体经洗涤、干燥,得到海藻糖晶体。而现有技术中,在酒精溶液中结晶时,通常需要数日。
本发明的另一个特点是在用原料(3)提取液的处理工艺中,采用了控制提取液的浓缩比,使酵母提取液在浓缩阶段可以将卵磷脂、麦角固醇等结粒,并用筛分或离心的方法分出,从而获得有价值的副产物,而不是像现有技术中将这些产物当作杂质,在除杂工序中除理弃掉。
通过上述工艺,本发明的效果是获得了一条料液过滤速度快,除杂质的效果好,结晶时间短,可降低生产成本的海藻糖生产方法。
实施例
取200克活性干酵母加1立升70~95%乙醇,加热回流提取两次,每次30分钟,离心所得提取液,真空浓缩回收酒精,将浓缩液浓缩,有脂溶性固块出现,用100目筛将此副产物截留,并得约100毫升浓缩液。加入浓缩液体积2~10%的氧化钙(或氢氧化钙),加热反应,通入CO2气体,调节料液pH值至11~12,趁热过滤,向滤液中继续通CO2至pH=8~9,过滤后,清液先进入强碱性离子交换树脂D280柱,再进入强酸性离子交换树脂0.01×7柱脱盐脱色,得到无色透明溶液。将此溶液浓缩至海藻糖浓度40~70%,加少许晶种,搅拌,即析出海藻糖晶体,过滤,洗涤,干燥,得海藻糖结晶。

- 一种由提取液经浓缩、过滤、离子交换、结晶制海藻糖的方法,其特征在于以酵母为原料,用70~95%乙醇,加热回流提取两次,离心所得提取液,经真空浓缩回收酒精后,再浓缩得浓度为5~30%的浓缩液,加入此体积2~10%的氧化钙或氢氧化钙,在pH≥12时加热反应,凝聚或分解蛋白质杂质,然后通入二氧化碳,使pH值降至8~9,过滤,溶液先后通过碱性和酸性离子交换脱盐脱色,再浓缩至40~70%的浓度、结晶、过滤、洗涤、干燥,得到海藻糖晶体。
