Abstract

Description
背景
存在改进流感疫苗的需要。目前针对流感的疫苗策略集中在生成针对血凝素的稳固的抗体(体液的)反应。由于在一年的过程中循环流感病毒株中高度的抗原性漂移,对于每个流感季必须特定地重新配制疫苗株。虽然年度(或季度)流感疫苗对于不同年龄类别的变化程度是成功的,显然对于年幼和年老者特别需要更有效的保护。此外,存在再造病毒会进化的重大、永久的风险,其在称为“抗原性漂移”的过程中获得非常不同的HA(血凝素)基因。这会产生公共健康紧急情况,因为目前的流感疫苗基本上依赖于HA抗原。
流感病毒是病毒的正粘病毒科(Orthomyxoviridae)中一种有包膜的、单链、负链RNA病毒,分为3个主要类型A、B和C。流感A病毒感染多种多样的动物,包括人、鸟类、猪、马、蝙蝠等等,尽管任何特定的流感病毒的趋向性通常高度适应于特定宿主。流感B病毒感染较小数目的物种,即人和海豹,但仍然是年度流行性感冒的重要原因。大多数人流感感染是通过流感病毒A或B引起的;而感染人和猪的流感C病毒罕见导致严重的人感染或流行(Lamb)。
目前失活的流感病毒疫苗诱导抵御紧密相关的病毒株的抗体。目前得到许可的疫苗主要诱导针对血凝素(HA)的菌株-特异性的中和抗体,病毒表面上的主要抗原性决定簇其为高免疫原性的,并可预防由匹配病毒株的感染引起的疾病。然而,HA具有重要的抗原变异,其排除其单独用于经设计以提供光谱防护(broadprotection)的疫苗。为此,针对较少可变的靶物生成保护反应的可替换的疫苗策略是非常感兴趣的。
用流感A病毒的天然感染诱导体液和细胞免疫性两者。持久的细胞免疫性主要针对保守的、内部病毒蛋白,如核蛋白(NP)。在天然感染后NP抗原在人中是免疫原性的,但诱导的细胞毒性T淋巴细胞具有短的寿命(McMichaela,McMichaelb)。
针对NP的细胞免疫性是有价值的,因为其针对NP表位的不同变体,且靶向NP的DNA疫苗在动物中具有诱导的交叉-保护的免疫性(Schotsaert)。
核蛋白(NP)抗原已长期被识别为高度保守的抗原:甚至是最歧化的(divergent)流感病毒A株在它们编码的NP蛋白中共享90%同一性(Gorman,Xu)。对NP的抗原性变化是罕见的且仅仅在较小程度发生(Staneková)。
现有技术
使用核蛋白作为疫苗中的抗原
流感核蛋白用作抗原描述于1980年代(Wraith)。小鼠中针对NP的细胞免疫响应能够诱导免疫性,且能够显著地产生针对分歧的A型病毒的交叉保护。显示用由H3N2病毒纯化的NP免疫小鼠可提供免于致死性异源(H1N1)攻击的实质保护(75%),但它不能防止感染。
使用NP基因的DNA疫苗已知20年:将它们用于第一“概念证明”实验用于DNA免疫本身(Ulmer)。
NP从病毒载体的表达首次在1980年代证明(Yewdell1985),且用此载体免疫与细胞毒性T淋巴细胞针对分歧的流感病毒A株而非B株与DNA疫苗相比改进的生成相关。
自此,已显示用表达PR8核蛋白的MVA载体免疫小鼠保护它们抵御异亚型流感病毒(Altstein)的低剂量攻击。更近些,已将一种编码融合于M1蛋白的NP蛋白的病毒载体用于免疫人(Lillie,Berthoud,Antrobus)。这些研究显著地显示:当体液反应通过免疫衰老(immunosenescence)下降时,对于NP的细胞免疫响应可在较年长的人中被实质地增强(Antrobus)。
核蛋白的分泌
一些研究已表明NP蛋白起初位于核中,减少这些DNA疫苗的免疫原性(Staneková)。
改进的针对NP的细胞免疫响应可通过强制分泌NP,例如通过将tPA信号肽融合于NP基因(Luo)、通过配制DNA(Greenland,Sullivan)以及通过使用电穿孔(Laddy)以改进DNA递送来获得。
单体流感核蛋白
单体抗原在与C4bp寡聚域的融合体中的优选使用描述于专利申请WO2005/014654中。但单体化抗原使用中的风险是它们降低的免疫原性。这由Bachmann和同事用水泡性口膜炎病毒的糖蛋白G证明(Bachmann1993),并且用于流感抗原神经氨酸酶,或NA,由Fiers和同事证明(Fiers2001)。可预期的是减少或去除来自流感核蛋白的更高级结构会降低它们的免疫原性。
已显示多种突变使流感核蛋白转变,其自然寡聚化为单体形式(Ye2006)。在此2006文章中所述的NP的单体形式在更近期的文章中确认为单体的(Tarus,Ye2012)。这些文章中所述的赋予流感病毒A株的核蛋白单体的两个点突变在流感病毒B和C株的核蛋白中是保守的(参见Nakada中图3)。因此,可将相同的点突变引入流感病毒B和C株的核蛋白,从而赋予这些其他流感核蛋白单体。但是没有进行单体核蛋白的免疫原性的研究。
用NP抗原制备流感疫苗中的主要技术问题是诱导强而且持久的细胞免疫响应。所述“细胞免疫响应”是一种免疫响应,其不涉及抗体,但涉及抗原-特异性T-淋巴细胞(且特别是细胞毒性T淋巴细胞)的活化和多种细胞因子响应于抗原的释放。CD4细胞或辅助性T细胞通过分泌活化免疫响应的细胞因子而提供针对不同病原体的保护。细胞毒性T细胞(CD8)引起由于病原体的凋亡所致的死亡而不需使用细胞因子。
尽管对于针对NP的CD4或CD8响应是否对于保护是更重要的仍然有争议(Epstein),但存在共识,即对于核蛋白的细胞反应而非体液反应对于此抗原可诱导的保护是关键的(Thomas)。当T细胞表位源自高度保守的NP蛋白时,通过引发强细胞毒性T细胞响应提供保护的疫苗可为有用的(Epstein;Roy)。由T淋巴细胞介导的细胞免疫响应主要通过识别流感病毒-感染的细胞、通过抑制病毒复制以及通过加速病毒清除而发挥功能。
赋予免疫性所涉及的特异性T细胞包括CD4+和CD8+T细胞两者,并常常分别通过分泌的细胞因子和细胞裂解活性的作用来发挥它们的功能。流感NP-特异性CD8+CTL特别能够在针对小鼠中致死性流感病毒攻击的异亚型保护性免疫性起重要作用(Gschoesser),包括流感病毒从上呼吸道粘膜表面的清除(Mbawuike),促进攻击后的存活和恢复(Epstein)。最佳的基于NP的疫苗可改进CD4和CD8细胞反应两者。

发明简述
本发明涉及用于增加来自流感病毒的NP抗原的免疫原性(特别是细胞免疫原性)的方法,即,将来自流感病毒株A、B或C的NP抗原的至少一种变体融合于作为载体蛋白的鸡(chicken)C4bp寡聚化域的变体。

发明详述
在详细描述本发明之前,可理解的是本发明并不局限于具体例示的方法,并且当然可以变化。特别是,本发明涉及包含来自流感病毒的至少一个核蛋白抗原的融合蛋白,并且不限于特定的流感核蛋白。
本文引用的所有出版物、专利和专利申请,无论是下文或上文,都通过提述以它们的整体并入本文。然而,引用文本提及的出版物是为了描述和公开在所述出版物中报道的并且可能相关于本发明使用的规程、试剂和载体。
此外,除非另外指示,本发明的实施采用常规的蛋白纯化和本领域技术范围内的分子生物学技术。这些技术是技术人员已知的并且在文献中充分地解释。在所附权利要求和本发明的连续描述中,除了由于表述语言或必需含意而上下文有另外要求外,单词“包含(comprise)”、“含有(contain)”、“包涵(involve)”或“包括(include)”或变型如“包含(comprises)”、“包含(comprising)”、”含有(containing)”、“包涵(involved)”、“包括(includes)”、“包括(including)”以涵盖的意义使用,即,以详明所述特征的存在,而不排除其他特征在本发明的多个实施方案中的存在或添加。
定义如下术语用于更好地理解本发明:
流感病毒有三种类型A、B和C。此分类起初是血清学的:对流感病毒A核蛋白的抗血清与其他A型病毒的核蛋白交叉反应,但不与B型或C型病毒的那些交叉反应。流感A病毒基于它们的血凝素(H)和神经氨酸酶(N)糖蛋白的血清学交叉反应进一步分成多个亚型。
“流感核蛋白”意指流感病毒的全部三种类型(A、B和C)的核蛋白。
“载体蛋白(carrierprotein)”通常指抗原与之缀合或融合并由此赋予更多免疫原性的蛋白。本文该术语特定地在携带抗原的蛋白的涵义中使用。蛋白的功能在于增加与之缀合或融合的所述抗原的免疫原性。
“NP的变体”指所有具有序列的蛋白,所述序列与来自流感病毒株A、B和C的流感核蛋白的野生型形式具有至少90%的同一性。
“鸡C4bp寡聚化域的变体”是2013年12月11日提交的专利申请WO2007/062819和PCT/EP2013/076289(两篇文献通过提述并入本文)中所述的SEQIDNO:1的C4bp域的变体,特别是至少48个连续氨基酸的片段和/或与专利申请WO2007/062819中所述的SEQIDNO:1具有至少70%氨基酸序列同一性(的变体)。
“IMX313载体蛋白的变体”描述于2013年12月11日提交的专利申请PCT/EP2013/076289中。
鱼精蛋白由一组具有4500道尔顿的平均分子量的从鱼获得的异源肽组成。鱼精蛋白的氨基酸组成的大约67%是精氨酸。它长期用于配制胰岛素(于中性鱼精蛋白锌胰岛素(NeutralProtamineHagedorn)),或用于中和肝素。
术语“融合蛋白”指重组蛋白,其非天然存在,包含来自不同来源的已融合的两个域。更精确地,在本发明中,融合蛋白包含融合于鸡C4bp寡聚化域的载体域变体(特别是‘IMX313T’或‘IMX313P’)的流感核蛋白抗原。融合体具有产生同源产物的优势。更正式地,“缀合”可描述为遗传的:将编码原-免疫原性载体蛋白的DNA剪接至编码抗原的DNA。可将抗原融合至载体蛋白的N-或C-端。
本发明涉及免疫原性组合物,其包含流感核蛋白抗原的至少一个变体抗原和C4bp寡聚化域的变体,并引发增加的针对流感核蛋白抗原的细胞免疫响应。
根据本发明,融合于鸡C4bp寡聚化域的载体蛋白变体(特别是IMX313T或IMX313P)的核蛋白可为来自流感病毒的任何类型(A、B或C)的核蛋白。
可将核蛋白抗原融合于载体蛋白(特别是IMX313T或IMX313P)的N-或C-末端。
根据本发明,至少一个核蛋白融合于一个载体蛋白,特别是IMX313T或IMX313P;然而,两个或多个核蛋白(相同的或不同的)可融合于相同的载体蛋白。
根据本发明的优选的方面,融合于鸡C4bp寡聚化域的变体(特别是IMX313T或IMX313P)的核蛋白抗原是单体抗原。实际上,使用单体抗原是有利的,如专利申请WO2005/014654中所述,条件是单体化不减少它们的免疫原性。而且,NP在结晶中形成三聚体(Ye2006)和体内的其他寡聚体(Arranz,Moeller)。三聚或寡聚蛋白融合于七聚体蛋白如IMX313T或IMX313P面临产生空间冲突(stericclashes)的风险。另一方面,天然寡聚蛋白的单体形式具有减少的免疫原性(Fiers)。
为了获得单体核蛋白抗原,本领域的技术人员知晓可引入NP抗原的蛋白序列以诱导其单体化的不同的点突变。具体地,NP抗原呈现两个下述点突变中的至少一个:E339A和R416A。
在本发明的一个实施方案中,NP抗原来自流感病毒株A。
在本发明的一个优选的实施方案中,NP抗原包含两个点突变E339A和R416A,并且因此为单体的。
在本发明的另一个实施方案中,NP抗原呈现如SEQIDNO:2中所示的多肽序列。
对从DNA疫苗表达的抗原的增加的细胞免疫响应先前已通过将所述抗原的基因融合于编码IMX313的基因来获得(Spencer)。有利地,此域的变体呈现具有序列ZXBBBBZ的至少一个带正电的肽的C-末端取代,其中(i)Z是任何氨基酸或者是缺失的,(ii)X是任何氨基酸且(iii)B是精氨酸(R)或赖氨酸(K),如SEQIDNO:3中所示,当融合于所述抗原时,其产生改进的对抗原的免疫响应,如2013年12月11日提交的专利申请PCT/EP2013/076289中所述。IMX313载体蛋白的优选变体不诱导与鱼精蛋白交叉反应的抗体。
特别的改进的变体,称为IMX313T和IMX313P近来已在2013年12月11日提交的专利申请PCT/EP2013/076289中描述。它们的肽序列如下:

在本发明的另一个实施方案中,融合蛋白包含NP抗原,其包含信号肽。一些研究已表明NP蛋白起初位于核中,这可潜在地降低所述DNA疫苗的免疫原性。因此,通过添加信号肽使得NP抗原能够分泌是理想的。特别地,所述信号肽是tPA(组织纤维蛋白溶酶原激活剂(tissueplasminogenactivator))分泌信号肽,如(Luo)中所述。
在本发明的一个特定的实施方案中,NP抗原是单体的且包含信号肽。
在本发明的另一个实施方案中,NP抗原包含两个突变E339A和R416A,以及信号肽tPA。如实施例中所示,包含IMX313T和含有信号肽的单体NP抗原的融合蛋白,当作为DNA疫苗注射入小鼠时,诱导比Th2响应更强的Th1响应(IgG2a)(图13)。免疫学家中的共识是:Th1响应相对于Th2响应是优选的。然而,用于优先改进Th1对于抗原的响应而不使用为此目的开发的佐剂的方法不是本领域已知的。在下面的实施例中,显示IMX313T或IMX313P与流感核蛋白抗原的融合体优先改进Th1响应。
本发明还涉及包含任何包含卷曲螺旋域的载体蛋白和来自流感病毒的至少一个核蛋白(NP)抗原的融合蛋白。特别地,所述核蛋白抗原是单体。
本发明还涉及编码如上所述的融合蛋白的核酸,而且具体地:
-编码包含NP抗原和IMX313T或IMX313P的融合蛋白;
-编码包含单体NP抗原和IMX313T或IMX313P的融合蛋白;
-编码包含含有信号肽的NP抗原和IMX313T或IMX313P的融合蛋白;
-编码包含含有信号肽的单体NP抗原和IMX313T或IMX313P的融合蛋白。
作为优选的实施方案,所述核酸编码包含来自流感病毒A的单体NP抗原的融合蛋白。作为优选的实施方案,所述核酸编码融合蛋白,其不诱导与鱼精蛋白交叉反应的抗体。特别地,所述核酸呈现如SEQIDNO:6和SEQIDNO:7中所示的序列。
本发明还涉及载体(vector),其包含上面呈现的核酸,和遗传元件如启动子和增强子以确保DNA盒在宿主细胞中的表达。
本发明还涉及免疫原性组合物,其包含:
-如上呈现的融合蛋白或核酸或载体,和
-对于胞内TLRs的核酸配体,和/或任何其他疫苗佐剂。
Toll-样受体(TLRs)
先天的免疫***的细胞通过有限组的种系编码的受体检测病原体。这些先天的免疫受体识别由病原体表达的一系列保守的分子结构,即PAMPs(病原体相关的分子模式(PathogenAssociatedMolecularPatterns))。
这些源自病原体的分子一般代表对于一组病原体非常特异性的复杂分子。TLRs代表一组免疫模式识别受体,其能够在被病原体感染后立即警告免疫***。它们作为先天的免疫和适应性免疫之间的中枢组分起重要作用,并能够觉察出(scentout)许多病原体从病毒到寄生虫。第一个表征的TLR(称为Toll)显示负责在成年果蝇中的抗真菌响应而且目前已经鉴定了10个涉及病原体识别的人等同物。TLRs可基于它们的定位和它们识别的PAMPs的类型分类入不同的组。TLRs1,2,4,5和6主要在细胞表面上表达,在此它们大多识别细菌产物,而TLRs3,7,8和9定位于细胞内区室中并大多识别病毒产物和核酸。
胞内Toll-样受体
此外,为了改进免疫的方法,同样非常重要的是限制通过TLR受体的信号传导。Toll样受体(TLRs)是一类蛋白,其在先天免疫***中起关键作用。一旦微生物破坏了生物体的物理屏障,它们被TLRs识别。所识别的来自微生物特征包括病毒的双链RNA,细菌和病毒的非甲基化CpG位点岛,以及某些RNA和DNA分子。
对这些核酸有实质的兴趣,因为它们是一类Toll-样受体(下文称为TLRs),并尤其是TLR3、TLR7、TLR8、TLR9和TLR13的配体(Blasius及其中的参考文献)。这些有时分类为“胞内Toll-样受体”,但至少TLR3也存在于一些细胞表面上。TLR3表达于多种上皮细胞,包括气道、子宫、角膜、***、宫颈、胆和肠上皮细胞,而且这些细胞看起来在它们的细胞表面上表达TLR3(Akira)。
通过这些受体(特别是TLR3受体)限制信号传导的重要性是剂量依赖的。将核酸配体紧密地结合于抗原因而是必需的以防止它们在缺乏抗原时结合于TLRs。因此紧密结合的胞内TLR配体相对于结合较不紧密的配制物是高度优选的。因此,本领域的技术人员正在寻找能够高效结合TLR配体的抗原组合物,使得它们在抗原到达其将触发免疫响应的细胞之前不与抗原分离,目的在于减少由配体结合于其他地方的TLR受体而介导的潜在的不良作用。
在本申请中,且特别是实施例中,已使用如下胞内TLR配体:
-对于TLR3:多聚I:C,其为与多聚胞嘧啶核苷酸杂交的多聚肌苷酸的多核苷酸的双链体,双链RNA的类似物。每条链的链长为20个核苷酸。
-对于TLR7:称为ssRNA40的寡核苷酸,具有序列5’GsCsCsCsGsUsCsUsGsUsUsGsUsGsUsGsAsCsUsC3’其中“s”代表硫代磷酸酯键(SEQIDNO:8);
-对于TLR9:称为ODN1826的寡核苷酸,具有序列:5’tccatgacgttcctgacgtt3’(SEQIDNO:9)。
在本发明的一个特定方面,免疫原性组合物包含:
-如上所述的融合蛋白或核酸或载体,和
-polyI:C。
本发明还涉及如上所述的融合蛋白,其用作疫苗用于预防和治疗流感疾病。所述流感疫苗可用于多个应用:
-预防季节性流感;
-预防流行病状况(pandemicsituation);
-‘普遍’预防,即,针对所有类型的流感病毒免疫的疫苗;
-所有类型的流感的免疫疗法。
可用根据本发明的特定疫苗在人体或动物体中进行预防或治疗流感的方法。本领域的技术人员知晓如何将疫苗的组合物适应于各特定的应用和特定的患者。

实施例1–将IMX313***NP编码质粒
使用寡核苷酸引物IMX1289(5’caatgcagaggagtacgacaatggatccaagaagcaaggtgatgctgatg3’–SEQIDNO:10)和IMX1290(5’GTAGAAACAAGGGTATTTTTCTTtattactccttgctcagtccttgc3’–SEQIDNO:11)从质粒pIMX494扩增IMX313编码序列并将其***质粒pcDNA3-NP,如Geiser所述。
实施例2–***tPA信号肽
使用寡核苷酸IMX1305(5’cactgagtgacatcaaaatcatgGATGCAATGAAGAGAGGGC3’–SEQIDNO:12)和IMX1306(5’cgtaagaccgtttggtgccttggctagctcttctgaatcgggcatggatttcc3’–SEQIDNO:13)从载体pSG2-85A(Spencer)扩增tPA信号肽并符合读框地***多个质粒中NP编码序列的N-末端,如Geiser所述。
实施例3–产生NP的两个点突变以使其成为单体的
使用寡核苷酸引物IMX1287(5’ccattctgccgcatttgCagatctaagag3’–SEQIDNO:14)和IMX1288(5’CAAAAGGGAGATTTGCCTGTACTGAGAAC3’–SEQIDNO:15)以扩增NP基因的内部片段,并将所得的PCR产物***NP-编码质粒,如Geiser所述。因为两个寡核苷酸与NP基因不完美匹配,PCR产物的***产生两个点突变。IMX1287引物产生突变E339A(GAA至GCA),而IMX1288引物在NP基因中产生突变R416A(AGA至GCA)。
实施例4–***IMX313T
使用寡核苷酸引物IMX1289(SEQIDNO:10)和IMX051(5’GTAGAAACAAGGGTATTTTTCTTtattaggagcgacggcgacgc3’–SEQIDNO:16)从质粒pIMX497扩增IMX313T编码序列并***多个pcDNA3-NP-来源的质粒,如Geiser所述。
实施例5–用根据本发明的核酸的DNA免疫
5.1.规程
将多组五只雌性BALB/C小鼠用多个质粒DNA肌内免疫两次,间隔14天,每次注射使用20μg每个质粒。在第28天测量免疫响应以确定多个修饰的影响:+/-IMX313或IMX313T;+/-tPA信号肽;+/-单体化突变。
抗原特异性T-细胞响应使用脾细胞在第28天通过ELISPOTs测量。将分离自免疫的小鼠的纯化的脾CD4+、CD8+和总T细胞与购自Eurogentec的NPA流感肽(氨基酸366-374)共培养。
ELISPOT测定法:将平底、96-孔硝化纤维素板(Millititer;Millipore)用IFN-γmAb(15μg/ml;MABTECH,Stockholm)涂覆并在4℃温育过夜。在用PBS洗涤后,将板用10%胎牛血清在37℃封闭一小时。将每孔2x106个细胞用相关肽以2μg/ml的终浓度(NPA流感肽)在用15μg/ml抗-人IFN-γ涂覆的膜上刺激并温育20小时。在温育后,将板用PBS彻底洗涤以去除细胞,并将IFN-γmAb(1μg/ml生物素,MABTECH)加至每孔。在37℃温育2小时后,将板洗涤并用抗生蛋白链菌素-HRP(1μg/ml;MABTECH)在室温显色(develop)一小时。洗涤后,加入底物(3-氨基-9-乙基咔唑(3-amino-9-ethycarbazol)(Sigma))并温育15分钟。在进一步洗涤后,在显微镜下计数红色斑点。
为了研究体液免疫响应,我们通过特异于总IgG的ELISAs评价了抗体水平,以及分别特异于IgG1和IgG2a的ELISAs以评价Th1和Th2的相对比例。BALB/c小鼠通常以Th2-型免疫响应响应于流感疫苗,其与IgG1抗体的刺激相关。然而,病毒感染下存活的小鼠血清中存在的主要抗体同种型是IgG2a,其在Th1-型免疫响应过程中被刺激(Huber)。IgG2a抗体的刺激已与流感免疫接种的效力增加相关。
对于ELISAs,将抗原稀释至5mg/ml的浓度于0.1M碳酸钠/碳酸氢钠(pH9.6)然后用于涂覆MaxiSorb板(Nunc-Immulon,Denmark)的孔。将两倍稀释的测试血清添加至孔,并在洗涤后用缀合于辣根过氧化物酶的抗小鼠IgG2a、抗小鼠IgG或抗小鼠IgG1(Sigma)检测结合的抗体。在添加邻苯二胺(Sigma)和H2O2后测定490nm的吸光度;用1M硫酸终止反应。
结果示于图2至5。
5.2.在初步实验中,我们测试了由编码NP或融合于IMX313的NP的DNA疫苗诱导的对NP的总T细胞响应。分离自NP-IMX313免疫的小鼠的总T细胞显示与NP免疫的小鼠的那些相比显著更高的IFNγ响应,并确认IMX313增加T细胞响应的能力。
图2显示亲本NP抗原基因融合于IMX313基因改进对于NP的T细胞响应,甚至是当融合蛋白在细胞质中表达时。
5.3.为了确定ELISPOTs中检测到的IFN-γ是由CD4还是由CD8T细胞产生,我们从免疫的小鼠纯化了脾CD4+和CD8+T细胞,且将这些与流感ANP肽共培养。在用NP-IMX313免疫的组中检测到自CD8+T细胞产生IFN-γ的显著增加。产生IFN–γ的抗原特异性CD8+细胞的百分数高于相应的CD4+T的群体(图3)。
图3显示NP抗原基因融合于IMX313基因改进了对于NP抗原的CD4+和CD8+响应两者。
5.4.然后我们检查了免疫后以及末次免疫后14天对于NP的抗体响应,在血清中测量了NP-特异性IgGAb响应。NP对照小鼠和给予NP-IMX313的小鼠显示中度NP-特异性IgGAbs(图4),其在用NP-IMX313免疫的组中更高。
图4显示NP基因融合于IMX313基因改进对于NP抗原的IgG抗体响应。
5.5.还检查了血清中NP-特异性IgG1和IgG2a抗体的存在(分别为Balb/C小鼠中Th2和Th1型响应的代表)。在NP-IMX313免疫的小鼠的血清中检测到NP-特异性IgG1和IgG2a抗体同种型;然而,来自单独给予NP的小鼠的血清样本仅显示低水平的IgG1和IgG2aAb(图5)。

图6显示NP的单体化(NPm)轻微地改进其免疫原性(尽管所述改进不是统计学上显著的:NS);NPm免疫原性进一步通过融合于IMX313基因而改进;且最终单体NP融合于IMX313T基因进一步增强NP免疫原性。令人惊讶地,NP的单体化不降低其免疫原性。
图7显示,对于CD4+和CD8+响应的分析,可见如图6中相同的排序(rankordering):NP的单体化轻微改进NP的免疫原性但不显著(NS);NP的免疫原性通过融合于IMX313基因进一步改进,但是NP免疫原性的最大的改进通过将单体NP融合于IMX313T基因而获得。
图8显示对于B细胞响应可见对于T细胞响应(CD4+和CD8+两者)在图6和7中所见的相同的排序。用IMX313T得到的针对NP的总IgG响应高于以IMX313得到的。
图9显示由单体NP抗原诱导的抗体的亚类分布。对于NP,融合于IMX313基因使IgG2A响应增强大于IgG1响应,将针对NP的Th2-偏好的响应(0.8)转变为针对NP-IMX313的Th1-偏好的响应(1.51)。Th2向Th1偏好的此反转由融合于IMX313T而非融合于IMX313(1.5)来维持。IgG2a抗体在流感疫苗中的表达与病毒的清除和针对致死性流感攻击的增加的保护相关。如通过ELISA测量的两种抗体同种型的诱导增加对于疫苗功效比对于单独中和更好地相关(Huber)。

实施例6-NP抗原的分泌改进其免疫原性
制备一系列含有组织纤维蛋白溶酶原激活剂(tPA)分泌信号序列的NPDNA疫苗构建体:tPA-NP、tPA-NPm、tPA-NPm-IMX313和tPA-NPm-IMX313T。分析了tPA融合于NP对于来自免疫的动物的体液和细胞免疫响应的作用。
用含有tPA的构建体免疫的小鼠显示与NP免疫的小鼠的那些相比显著更高的IFNγ响应,并确认IMX313T和单体化突变增加T细胞响应的能力。
图10显示强制分泌NP抗原改进其免疫原性(NP对tPA-NP),无论它是否是单体的(tPA-NP对tPA-NPm)。然而,融合于IMX313显示使用单体形式的NP比使用未修饰的抗原更具免疫原性(tPA-NP-IMX313对tPA-NPm-IMX313)。而且用IMX313T取代IMX313进一步改进NP的免疫原性(tPA-NPm-IMX313对tPA-NPm-IMX313T)。
图11显示对于不同的分泌形式的NP的CD8+和CD4+响应。可见与图10中相同的排序,而且当添加IMX313时,单体化抗原的效用再次彰显。如前述附图中,当使用IMX313T而非IMX313时可见最大的免疫响应。
图12显示对于抗原NP的总IgG响应并招致与图11对于T细胞响应相同的结论:当使用IMX313T时可见最大的响应,但分泌(NP对tPA-NP)和单体化(tPA-NP-IMX313对tPA-NPm-IMX313)也是重要的贡献。
单用NP(作为NP、tPA-NP或tPA-NPm)免疫的小鼠在它们的血清中不具有或具有非常低水平的抗-NPIgG抗体(图12)。另一方面,用tPA-NP-IMX313、tPA-NPm-IMX313或tPA-NPm-IMX313T免疫的小鼠显示高水平的***性NP-特异性IgG抗体响应;再一次,tPA-NPm-IMX313T免疫的小鼠相比于所有其他组免疫的小鼠具有显著更高的(p<0.001)IgG抗体响应。这显示所有修饰(单体化突变、tPA和IMX313T)的组合赋予抗原相比于亲本序列或其他组合显著改进的免疫原性。
图13显示对于NP的B细胞响应的亚类分析,并说明了单用NP的初始Th2偏好通过IMX313并通过IMX313T反转。虽然分泌对其自身(NP对tPA-NP)、单体化(tPA-NP-IMX313对tPA-NPm-IMX313)然后是用IMX313T替代IMX313(tPA-NPm-IMX313对tPA-NPm-IMX313T)几乎不具有作用,所有均贡献于改进的Th1(IgG2a)相对于Th2(IgG1)响应。
非常重要的是tPA-NPm-IMX313T对其自身几乎等同地改进Th1和Th2响应。NP融合于IMX313显示Th1和Th2响应两者都增加,而且在响应类型上没有显著的转移。但是将IMX313T与单体化突变组合,Th1响应(IgG2a)开始占主导。免疫学家们的共识是Th1响应优选于Th2响应(图13)。

实施例7–产生重组NPm-IMX313T蛋白
表达A/WSN/33(Tarus2012b)株的野生型H1N1NP蛋白的具有C-末端6-His-标签的pET22-衍生的质粒在细菌菌株C43R中表达。此菌株通过用罕见密码子表达质粒pRARE2(Novagen)转化C43(DE3)制得。表达在TB(Terrific培养液)培养基中用IPTG诱导。过表达的蛋白如Ye所述初始纯化,并如Tarus所述用于澄清(clarification)和离子交换步骤,但在最终步骤中通过在肝素琼脂糖凝胶(HeparinSepharose)上通过亲和性并通过凝胶过滤(HiPrep26/60SephacrylS-300)纯化融合蛋白,如2013年12月11日提交的专利申请PCT/EP2013/076289中所述。
为了表达NPm-IMX313T蛋白,在两个步骤中修饰表达NP的质粒。第一,如实施例3中使用寡核苷酸引物IMX1287(5’ccattctgccgcatttgCagatctaagag3’–SEQIDNO:14)和IMX1288(5’CAAAAGGGAGATTTGCCTGTACTGAGAAC3’–SEQIDNO:15)引入单体化突变。在第二步中,使用如实施例4中相同的寡核苷酸引物以IMX313T编码序列替代6-His-标签:IMX1289(SEQIDNO:10)和IMX051(5’GTAGAAACAAGGGTATTTTTCTTtattaggagcgacggcgacgc3’–SEQIDNO:16)。然后***PCR产物替代6-His-标签,如Geiser所述。
NPm-IMX313T融合蛋白以与NP蛋白相同的方式在相同的菌株中表达,并使用相同的层析步骤纯化。
实施例8–免疫
然后进行小鼠的免疫以比较NPm-IMX313T(具有或不具有与AddaVax佐剂(Invivogen)的配制物)的免疫原性。将NP蛋白(具有或不具有与AddaVax佐剂的配制物)用作对照。
为此,将4组(五只)雌性BALB/c小鼠皮下免疫两次,间隔14天,每次注射使用20μg的每种蛋白。使用脾细胞在第28天通过ELISPOTs测量抗原-特异性T-细胞响应的诱导。将分离自免疫的小鼠的纯化的脾CD4+、CD8+和总T细胞与NP蛋白或流感ANP(366-374)肽共培养。以NP作为抗原通过ELISAs测量免疫前和第28天的抗体响应。
实施例9-IMX313T经过分泌途径时不被蛋白酶降解
通过用含有IMX313T的质粒的DNA免疫获得的结果强烈地表明当所述分子经过分泌途径(此处富含蛋白酶)时该分子的尾部不被蛋白酶切割。为了更直接地检查此问题,使用用来体内表达NPm-IMX313T的pcDNA3质粒进行CHOK1细胞的转染。该转染如别处所述(Krammer)进行。
十八至二十四小时后,转染的细胞的上清通过离心回收,并过滤然后加载于肝素琼脂糖凝胶(HeparinSepharose)柱上,如2013年12月11日提交的专利申请PCT/EP2013/076289中所述。
可见小的“峰C”,这在SDS-PAGE和Western印迹上证明含有蛋白NPm-IMX313T。
实施例10–产生重组NPm-IMX313P蛋白
为了表达NPm-IMX313P蛋白,将表达NPm-IMX313T的质粒通过用IMX313P基因取代IMX313T基因进行修饰,这通过用来自编码IMX313P的质粒的限制片段(PmlI-HindIII)交换代替编码NPM-IMX313T蛋白的质粒中相应的片段来进行。然后如实施例7中表达和纯化融合蛋白。图19显示纯化的蛋白;主要条带是单体,但是寡聚的形式也是可见的(在过量加载的凝胶上)在主要条带上方。
实施例11–产生对于IMX313P的超免疫抗血清
将一组五只雌性BALB/C小鼠用IMX313P蛋白肌内免疫六次,间隔14天,每次注射使用50μg。
通过使用修饰的ELISA方法测试血清中IgG抗体。将X级硫酸鱼精蛋白(Sigma)、IMX313或IMX313P用于涂覆微孔板的孔以捕获抗体。检测抗体为羊-抗-小鼠IgG-HRP(Sigma),其与过氧化氢反应以产生405nm的吸光度读数。
用IMX313P免疫的小鼠的所有血清呈现对于IMX313P的高效价的IgG抗体,和一些与IMX313交叉反应的抗体;但均不与鱼精蛋白交叉反应(图20)。
