Abstract
本发明涉及一种诱导和扩增源自外周血单个核细胞的自然杀伤(NK)细胞的方法,其中该方法包括:在存在细胞因子的条件下,将作为饲养细胞的经辐射的Jurkat细胞和经辐射的由EB病毒转化的连续性淋巴细胞株(EBV‑LCL)细胞与外周血单个核细胞进行共培养。根据本发明,可以从少量的外周血单个核细胞中诱导及增殖大量的NK细胞,而无需使用昂贵的设备或各种昂贵的细胞因子,从而可显著提高将所述NK细胞用于癌症时的预防和治疗的效率和效能。

技术领域
本发明涉及一种用于诱导和扩增源自外周血单个核细胞的自然杀伤细胞的方法。
背景技术
免疫反应能从病原体中保护人体,且免疫***由众多免疫相关细胞和细胞因子组成。白细胞,尤其淋巴细胞在免疫***中起重要作用。组成淋巴细胞的代表性细胞包括先天免疫***细胞和后天免疫***细胞。自然杀伤细胞(NK细胞)是一种具代表性的先天免疫细胞,已知自然杀伤细胞能以非特异性方式杀伤癌,识别并杀伤病毒、细菌等,而且可以用酶如穿孔素和颗粒酶杀伤病原体,或通过Fas-FasL相互作用杀伤病原体。据报告称,对于癌症患者而言,NK细胞的杀伤癌细胞的能力的降低与肺癌(Carrega P, et al., Cancer,2008: 112: 863-875)、肝癌(Jinushi M, et al., J Hepatol., 2005: 43; 1013-1020)、乳癌(Bauernhofer T, et al., Eur J Immunol., 2003: 33: 119-124)、子宫癌(Mocchegiani E., et al., Br j Cancer., 1999: 79: 244-250)、血癌(Tajima F., etal, Lekemia 1996: 10: 478-482)等疾病的发病紧密相连。因此,为了癌症治疗,必须要提高癌症患者的自然杀伤细胞的有关杀伤癌细胞的能力及活性。目前,尝试着利用如NK细胞的损伤癌细胞的能力来治疗实体癌(solid cancer)或血癌。
为了获得杀伤癌细胞的效果,而需要大量的NK细胞,但难以保证从癌症患者中能够获得大量血液,且血液中的NK细胞的量所占的比例也仅仅约5~20%。由于难以使用NK细胞作为免疫治疗剂,从而重点在于能够有效地扩张并增殖NK细胞。常用于扩增NK细胞的现有方法包括,利用如磁激活细胞分选(magnetic activated cell sorter,MACS)、cliniMACS或荧光激活细胞分选(fluorescence activated cell sorter,FACS)等装置从骨髓或血液的单个核细胞中分离或诱导出NK细胞。在这种方法中,进行如下操作:1)在早期,从单个核细胞中分离NK细胞,并利用细胞因子对NK细胞进行扩增培养;2)去除与单个核细胞共存的T细胞,并利用细胞因子对NK细胞进行扩增培养;及3)在骨髓中存在的干细胞中诱导NK细胞。此外,根据已有的报告,通过利用饲养细胞从外周血单个核细胞(PBMCs)中分离NK细胞的方法包括,意大利Torelli研究小组的利用源自B细胞白血病的RPMI8866细胞株的方法和日本Ishikawa研究小组的利用源自肾母瘤细胞(Wilms tumor cell)的HFWT细胞株的方法。
然而,由于需要精良的设备来选择细胞进行预扩增,并使用基因工程饲养细胞和昂贵的高浓度的各种细胞因子,从而使本领域中有报道的NK细胞的扩增方法受到了阻碍,进而普遍使用来可能过于昂贵,更加难以向病患群体实施该方法,其可用性受到了限制。
发明内容
技术问题
本发明涉及提供一种新型的用于诱导和扩增源自外周血的NK细胞的方法,从而引导出有效的策略,来获得活化的源自外周血的NK细胞,而无需使用昂贵的设备、资源或不受监管所约束。
技术方案
为了解决这些问题,本发明的一方面提供一种有选择地诱导和扩增源自外周血单个核细胞的NK细胞的方法,该方法包括以下的简化方法:在存在细胞因子的条件下,将作为饲养细胞的经辐射的Jurkat细胞和经辐射的EBV-LCL细胞与外周血单个核细胞会以限定的比例及浓度进行共培养。
除非另有具体说明,本发明中使用的所有的技术和科学术语具有与本领域的技术人员通常所理解的意义相同的意义。通常,本发明中使用的命名法和描述的实验方法是广泛所知的,并在本领域中常用到的。
本发明的发明人已经做了能从少量血液中获得大量NK细胞的研究,且发现通过用作为饲养细胞的经辐射的Jurkat细胞培养从外周血中分离的单个核细胞时可以获得增殖的NK细胞(韩国专利申请号10-2010-007877)。
此外,本发明的发明人已对利用所述专利申请所公开的方法能够有选择地增殖NK细胞的机制做了研究,且发现从单个核细胞中去除B细胞后使所述单个核细胞与经辐射的Jurkat细胞进行共培养时会显著降低NK细胞的选择性增殖,这表明B细胞在NK细胞的增殖中起重要作用(图1A和1B)。
基于这些结果,可发现,利用Jurkat细胞的NK细胞的选择性增殖取决于B细胞。从这个事实能够预期,当使用作为饲养细胞的B细胞源永生化细胞EBV-LCL时,能使更多量的NK细胞进行增殖。然而,可见当分离自外周血的单个核细胞与EBV-LCL进行共培养时,NK细胞虽有增殖,但NK细胞的纯度有显著下降。因此,本发明的发明人发现,可通过以下方式解决由单独使用Jurkat细胞或EBV-LCL细胞而引起的问题,即将经辐射的EBV-LCL细胞与经辐射的Jurkat细胞作为饲养细胞一同使用,从而可进行更高产量的单个核细胞和NK细胞的增殖。因此,本发明的发明人是基于这些事实进行研发的。
因此,根据本发明的一方面,提供一种诱导和扩增源自外周血单个核细胞的自然杀伤细胞的方法,该方法包括:在存有细胞因子的条件下,作为饲养细胞的经辐射的Jurkat细胞和经辐射的EBV-LCL细胞与外周血单个核细胞进行共培养。
在本发明中,“外周血单个核细胞”、“PBMCs”或“单个核细胞”是指从常用于抗癌免疫治疗的外周血中分离出的单个核细胞。通过利用已知方法,如聚蔗糖-泛影葡胺密度梯度法(Ficoll-Hypaque density gradient method),可从收集的人血液中获得外周血单个核细胞。
根据本发明的一个示例性实施例,“外周血单个核细胞”可从正常人、有患癌风险的患者或癌症患者中获取。这里使用的外周血单个核细胞无需一定要源自自体,也可以使用同种异体的外周血单个核细胞来诱导和增殖本发明的用于抗癌免疫治疗的NK细胞。
根据本发明的一个示例性实施例,所述外周血单个核细胞与经辐射的Jurkat细胞和经辐射的EBV-LCL细胞进行共培养时,可以获得经增殖的单个核细胞和经增殖的自然杀伤细胞。可使用所述自然杀伤细胞处理正常人、有患癌风险的患者或癌症患者,从而预防及治疗癌症。
在本发明中,所述术语“Jurkat细胞”或“Jurkat细胞株”是指血癌(永生急性T细胞白血病)细胞株,是由三藩市加利福尼亚大学的Arthur Weiss博士研发出来的。有表达各种趋化因子受体的Jurkat细胞株可以生成IL-2的细胞,由于在其表面上会高度表达作为天然杀伤细胞活性抑制因子的MHC-I类,从而认为所述Jurkat细胞株不可能成为用于抗癌免疫治疗的饲养细胞的候选者(饲养细胞)。然而,本发明的发明人为了从外周血单个核细胞向NK细胞的分化和NK细胞的增殖而筛选诸多血癌细胞后发现,所述Jurkat细胞能够用作饲养细胞(参照韩国专利申请号10-2010-0078777)。这里使用的Jurkat细胞可取自美国菌种保藏中心(American Type Culture Collection,ATCC; ATCC TIB-152)。
在本发明中,所述术语“EBV-LCL细胞”或“EBV-LCL细胞株”是指由EB病毒(EBV)转化的连续性淋巴细胞株(EBV-LCL)(D.M. Koelle et al., 1993, supra)。经常将所述EBV-LCL细胞用于研究癌变,然而未用作能从外周血中增殖单个核细胞和NK细胞的饲养细胞。本发明一个示例性实施例的EBV-LCL细胞可在通常的实验室内直接进行制备。根据本发明一个示例性实施例,直接制备并使用所述EBV-LCL细胞。EBV-LCL是一种在体外用EBV转染人B细胞而制得的B细胞株。通过用EBV转染PBMCs来制备细胞株的过程中,加入环孢霉素A以抑制对EBV起反应的T细胞。具体地,在9ml培养基中加入30×106PBMCs,并将该培养基置于T25培养瓶内。其次,将9ml的EBV上清液加入于T25培养瓶内。加入80μl环孢霉素A之后,在37℃下培养该***内容物。待培养7天后,去除一半上清液,加入新鲜的培养基。此后,加入40μl环孢霉素A。该过程中每7天重复与第七天相同的过程,直至培养28天为止。待培养28天后,可以使用所述细胞株。从第29天起,所述细胞株在未加入环孢霉素A的培养基中进行培养。
为了抑制癌细胞的增殖,所述Jurkat细胞和EBV-LCL细胞经辐射后,才能用作饲养细胞。根据本发明的一个示例性实施例,经辐射100~500Gy后,分别获得经辐射的Jurkat细胞和经辐射的EBV-LCL细胞。
在本发明中,所述术语“细胞因子”是指免疫活化细胞因子,可用来从外周血单个核细胞中诱导NK细胞。根据本发明的一个示例性实施例,所述细胞因子可以为IL-2、IL-15、IL-21、Flt3-L、SCF、IL-7、IL-12和IL18中的一种或多种的混合。特别地,由于已知作为细胞因子的IL-2、IL-15或IL-21对从外周血单个核细胞向NK细胞的分化和NK细胞的增殖具有优异的效果,从而优先使用这些细胞因子。根据本发明的一个示例性实施例,可使用IL-2,但不限于此。
在各种文献上可发现细胞因子会参与向NK细胞的诱导中的事实。从该事实可见,在未发生细胞因子受体的γc表达的小鼠体内,发现了B细胞和T细胞,但没有发现NK细胞,已知含有γc的细胞因子受体对向NK细胞的分化起重要作用(Singer, B et al., Proc.Natl. Acad. Sci. USA 92, 377-381, 1995)。所述受体的γc亚单元包括IL-2、IL-4、IL-7、IL-9、IL-15和IL-21的受体。其中,有报告称IL-2具有能够提高成熟NK细胞的增殖和活化的功能(Shibuya, A. et al., Blood 85, 3538-3546, 1995)。有报告称,在缺乏IL-2的人体和小鼠体内,所述NK细胞的数量会显著下降(DiSanto, J. P. et al., J. Exp. Med.171, 1697-1704, 1990)。另外一方面,有研究表明IL-2和IL-2Ra的缺乏会间接地影响NK细胞的数量和活化。此外,已知IL-2R链对IL-15受体的形成起一定作用。
IL-15对NK细胞的分化起一定作用。从事实可发现,在缺乏用于生成IL-15的转录因子干扰素(IFN)调节因子1的小鼠体内会缺乏NK细胞(Kouetsu et al., Nature 391,700-703, 1998),而且在缺乏IL-15或IL-15Rα的小鼠体内没有发现NK细胞。结果有报告称,IL-15可通过在NK细胞中表达的IL-15受体能够直接地增强NK细胞的生长和分化(MrozekEet al., Blood 87, 2632-2640, 1996)。
IL-21是由活化的CD4+ T细胞所分泌的细胞因子(Nature, 5:688-697, 2005),而且IL-21受体(IL-21R)在淋巴细胞,如树突状细胞、NK细胞、T细胞和B细胞中进行表达(Rayna Takaki, et al., J. Immonol 175: 2167- 2173, 2005)。IL-21在结构上与IL-2和IL-15高度相似,而且IL-2R与IL-2R、IL-15、IL-7R和IL-4R共享链(Asao et al., J.Immunol, 167: 1-5, 2001)。有报告称,IL-21可诱导从骨髓向NK细胞前体的成熟(Parrish-Novak, et al., Nature, 408: 57-63, 2000),具体来说能够提高效应因子的功能,如NK细胞的用于生成细胞因子和杀伤细胞的能力(M. Strengell, et al., JImmunol, 170, 5464-5469, 2003; J. Brady, et al., J Immunol, 172, 2048-2058,2004),而且通过增强CD8+ T细胞的效应因子功能来增强先天性和适应性免疫***的抗癌反应(Rayna Takaki, et al., J Immunol 175, 2167-2173, 2005; A. Moroz, et al.,J Immunol, 173, 900-909, 2004)。还有报告称,IL-21能够活化从人外周血中分离出的NK细胞(Parrish-Novak, et al., Nature, 408, 57, 2000),而且能从脐带血分离的造血干细胞中诱导出成熟的NK细胞(J. Brady, et al., J Immunol, 172, 2048, 2004)。
根据本发明的一个示例性实施例,所述细胞因子的使用浓度为50~1,000U/ml,例如可以为200~800U/ml、或400~600U/ml。常用于扩增NK细胞的方法需要高浓度的各种细胞因子,但根据本发明的一个示例性实施例的NK细胞的增殖方法,由于使用两种饲养细胞,即使使用低浓度的一种细胞因子,也可增殖高产量和高纯度的NK细胞。
在本发明中,可使用常用的培养基而不受限制,只要用于培养外周血单个核细胞的培养基可以诱导外周血单个核细胞而形成NK细胞并能使NK细胞增殖即可。例如,该培养基可使用RPMI、DMEM、x-vivo10、x-vivo20或cellgro SCGM培养基。此外,温度等培养条件可与外周血单个核细胞的常用培养条件相符。
根据本发明的一个示例性实施例,所述外周血单个核细胞、经辐射的Jurkat细胞和经辐射的EBV-LCL细胞进行共培养7-30天,例如可以培养10-20天。优选地,所述共培养可有效地进行10-14天。
根据本发明的一个示例性实施例,但所述外周血单个核细胞与饲养细胞进行共培养时,所述外周血单个核细胞和饲养细胞的混合比例可以为1:5~2:1。
根据本发明的一个示例性实施例,为了检测作为饲养细胞的Jurkat细胞和EBV-LCL细胞对NK细胞的诱导及增殖产生的效果,培养以下4个不同的组,分别培养预定时间:在存有IL-2的条件下培养单个核细胞的组(对照组1)、在存有IL-2的条件下培养单个核细胞和经辐射的Jurkat细胞的组(对照组2)、在存有IL-2的条件下培养单个核细胞和经辐射的EBV-LCL细胞的组(对照组3)及在存有IL-2的条件下共培养单个核细胞、经辐射的Jurkat细胞和经辐射的EBV-LCL细胞的实验组。然后,对PBMC和NK细胞计数(实施例2、图2和3)。从结果可确认,与对照组相比,存有IL-2的条件下共培养单个核细胞、经辐射的Jurkat细胞和经辐射的EBV-LCL细胞的实验组的PBMC数量增加约147倍以上(图2)。除了NK细胞的增殖之外,还能检测到NK细胞的富集程度。结果显示,待培养14天后,存有IL-2的条件下共培养单个核细胞、经辐射的Jurkat细胞和经辐射的EBV-LCL细胞的实验组中的作为NK细胞表型的CD56+和CD3细胞的比例增加至70%或以上(图4A)。
此外,本发明的一个示例性实施例的NK细胞的扩增方法可进一步包括:在后续培养的第1天至第15天,加入经辐射的Jurkat细胞、经辐射的EBV-LCL细胞和细胞因子,以在诱导及增殖源自外周血单个核细胞的NK细胞后能够维持NK细胞。由于活化的NK细胞的存活时间较短,难以用在免疫治疗而成为一个问题。因此,可以延长所述活化的NK细胞的存活时间且获得更多量的NK细胞,可在本发明的一个示例性实施例的NK细胞经增殖后进行培养第1天至第15天内,加入经辐射的Jurkat细胞、经辐射的EBV-LCL细胞和细胞因子。
根据本发明的另一方面,提供包含通过上述方法获得的源自外周血单个核细胞的NK细胞的用于预防和治疗癌症的组合物、通过上述方法获得的源自外周血单个核细胞的NK细胞在制备用于预防和治疗癌症的药物中的用途、或将有效量的通过上述方法获得的源自外周血单个核细胞的NK细胞进行给药的癌症的预防及治疗方法。
根据本发明的一个示例性实施例,对以下组进行杀伤癌细胞的评估:在存有IL-2的条件下共培养单个核细胞和经辐射的Jurkat细胞的组(对照组2)、在存有IL-2的条件下共培养单个核细胞和经辐射的EBV-LCL细胞的组(对照组3)及在存有IL-2的条件下共培养单个核细胞、经辐射的Jurkat细胞和经辐射的EBV-LCL细胞的组(实验组)。结果显示,比对照组多增殖约800倍以上的实验组的NK细胞具有强的杀伤癌细胞的能力,且未见减弱。
因此,通过本发明一个示例性实施例的方法获得的NK细胞能够有效地用于预防及治疗癌症。
根据本发明的一个示例性实施例,受试者可以为需要预防和/或治疗癌症的人。受试者可以为具有患癌风险的患者、正常人或癌症患者。
本发明的一个示例性实施例的包含源自外周血单个核细胞的NK细胞的用于预防和治疗癌症的组合物可被制成剂型,该机型的水溶液(例如,磷酸盐缓冲液、通常的注射用水溶液等)中悬浮有适当浓度的源自外周血单个核细胞的NK细胞,所述水溶液会根据需要包含适当成分。
本发明的一个示例性实施例的用于预防和治疗癌症的药物组合物可通过通常途径进行给药,如静脉注射,动脉注射,腹膜内注射,肌肉内注射或胸骨内注射。
本发明的一个示例性实施例的药物组合物中所含有的源自外周血单个核细胞的NK细胞的有效量是指,能够获得预防或治疗癌症的效果所需的量。因此,NK细胞的有效量可根据疾病种类、疾病的严重程度、组合物中含有的其他成分的种类和含量、患者的年龄、体重、一般的健康状态、性别、饮食、给药时间、给药途径、治疗期限及与NK细胞并用的药物等各种因素进行调节。本发明的一个示例性实施例的源自外周血单个核细胞的NK细胞可以,如以1×106~1×1011细胞数/kg的剂量向成人进行给药,例如以1×106~1×108细胞数/kg进行给药,且一天一次或一天多次给药,但本发明不限于此。
有益效果
根据本发明,可从少量的外周血单个核细胞中诱导及增殖大量的NK细胞,而无需使用昂贵的设备或各种昂贵的细胞因子,从而可显著提高将所述NK细胞用于癌症时的预防和治疗的效率和效能。

实施例
实施例1:源自外周血单个核细胞的NK细胞的制备
收集人血,在Ficoll(Ficoll-paqueTM PLUS, GE HealthCare)中,以2,500rpm,离心分离30分钟,从白膜层分离出外周血单个核细胞(PBMCs)。随后,用台盼蓝染色PBMCs。 然后,去除受损细胞,利用血细胞计数板仅对未染色细胞进行计数。
在37℃、5% CO2条件下,在装有人RPMI培养基的75T瓶内培养作为饲养细胞的Jurkat细胞株和EBV-LCL细胞株,该人RPMI培养基是通过向RPMI1640培养基加入10% FBS和1%青霉素/链霉素而获得的。每隔2~3天加入一次添加有500 U/ml IL-2的hRPMI培养基,每隔5天会收集细胞,然后加入新鲜的添加有IL-2的hRPMI培养基。
利用血细胞计数板测量细胞数,向浓度均为1×106/ml的Jurkat细胞株和EBV-LCL细胞株辐射100 Gy,强度为2.22 Gy/min。
将添加有500U/ml IL-2的hRPMI培养基放入24孔板的孔中,然后在37℃、提供5%CO2的孵化器中,将经辐射的Jurkat细胞株、经辐射的EBV-LCL细胞株和先前分离出的单个核细胞以1:0.5:0.5比例进行培养(实验组)。
此外,通过与上述相同的方法,制备以下各组用作对照组:未加入饲养细胞株时在存有IL-2的条件下培养单个核细胞的组(对照组1)、在存有IL-2的条件下培养单个核细胞和经辐射的Jurkat细胞的组(对照组2)及在存有IL-2的条件下培养单个核细胞和经辐射的EBV-LCL细胞的组(对照组3)。
实施例2: NK细胞的增殖能力的测量
为了确定所述饲养细胞对NK细胞增殖的作用,在培养第0天、第5天、第11天和第14天对实施例1中的实验组、对照组1、对照组2和对照组3的NK细胞进行计数,然后利用对应于作荧光标记的CD56和CD3的抗体进行染色。而后,利用流式细胞仪分析所述NK细胞,来分析NK细胞组(CD56+、CD3-)。随后,利用总的PBMCs和NK细胞的富集程度对NK细胞进行计数。
如图2所示,在培养第14天时,与对照组1(◇,PBMC)、对照组2(■,PBMC + Jurkat)或对照组3(▲,PBMC + LCL)作对比,实验组(×,PBMC + Jurkat + LCL)中的BMCs增加约147倍,该实验组中将Jurkat细胞和EBV-LCL细胞与PBMCs进行共培养。此外,如图3所示,将Jurkat细胞和EBV-LCL细胞一起与PBMCs进行共培养至第14天时,实验组的NK细胞比所述对照组增加约800倍。
实施例3:NK细胞的富集程度的测量
为了分析饲养细胞和细胞因子对NK细胞富集的作用,在培养第0天、第5天、第11天和第14天,利用作荧光标记的CD56和CD3抗体染色NK细胞,然后利用流式细胞仪分析实施例1的实验组、对照组1、对照组2和对照组3。结果显示,待培养14天后,将所述单个核细胞与Jurkat细胞和EBV-LCL细胞进行共培养时,作为NK细胞表型的CD56+和CD3细胞增加70%或以上(图4A)。图4B为能够说明在培养第14天时利用流式细胞仪所测量的NK细胞的富集程度的图。
实施例4:根据本发明的一个示例性实施例所制备的NK细胞用于杀伤癌细胞的能
力的测量
为了评估通过使用作为饲养细胞的实施例1中经辐射的Jurkat细胞株和经辐射的EBV-LCL细胞株所制备的NK细胞用于杀伤癌细胞的能力,通过利用肿瘤细胞(K562,Jurkat)作为靶细胞进行51Cr释放实验。
首先,对癌细胞K562和Jurkat细胞进行计数,用同位素51Cr作标记,然后在细胞孵化器中反应一小时。用hRPMI培养基对作同位素标记的癌细胞清洗3次,以去除同位素。利用血细胞计数板对实施例1的实验组、对照组2和对照组3中获得的细胞进行计数,然后互与作同位素标记的癌细胞以10:1、3:1或1:1的比例进行共培养4小时。待4小时后,2,500rpm下对所述细胞进行离心5分钟,而后将上清液移至试管中,用伽玛计数器测量。
如图5所示,可观察到使用IL-2和经辐射的Jurkat细胞株的对照组2(◆)中的NK细胞的杀伤癌细胞的能力与使用IL-2和经辐射的EBV-LCL细胞株的对照组3(×)和同时共培养经辐射的Jurkat细胞株和EBV-LCL细胞株的实验组(*)中的NK细胞的杀伤癌细胞的能力相近。此外可确认,即使效应细胞与靶细胞的比例(E:T)为1:1,NK细胞仍具有显著的杀伤癌细胞的能力。
由这些结果可见,本发明的一个示例性实施例的经IL-2、辐射后的Jurkat细胞和辐射后的EBV-LCL细胞处理的外周血单个核细胞会被富集而形成NK细胞,且还具有优异的杀伤癌细胞的能力。
实施例5:根据本发明的一个示例性实施例所制备的NK细胞的活化的测量
实施例4展示出,利用作为饲养细胞的实施例1中所制备的经辐射的Jurkat细胞株和经辐射的EBV-LCL细胞株来制备的NK细胞具有高度的杀伤癌细胞的能力。因此,为了评估活化标记的存在,可利用流式细胞仪分析在培养11天的NK细胞中检测各种NK细胞相关的活化受体和抑制受体的表达。
如图6所示,可见如ICAM-1、CD11a、CD48、CD2、CD49d和CD58的细胞粘附分子、如NKp30、NKp44、2B4、DNAM-1和NKG2D的活化受体及如CD69和CD25的活化标记的表达有增加;如CD183、CD184和CCR7的趋化因子受体的表达有轻微上升;而且,如CD158a、CD158b、CD94、NKB1和KIRNKAT2的抑制受体的表达有增加。
Claims (9)
1.用于诱导和扩增源自外周血单个核细胞的自然杀伤(NK)细胞的方法,其中该方法包括:在存在细胞因子的条件下,将作为饲养细胞的经辐射的Jurkat细胞和经辐射的、由EB病毒转化的连续性淋巴细胞株(EBV-LCL)细胞与外周血单个核细胞进行共培养。
2.根据权利要求1所述的方法,其特征在于,所述外周血单个核细胞是从正常人、有患癌风险的患者或癌症患者中获得的。
3.根据权利要求1或2所述的方法,其特征在于,所述经辐射的Jurkat细胞和经辐射的EBV-LCL细胞是分别通过100~500Gy的辐射处理而获得的。
4.根据权利要求1或2所述的方法,其特征在于,所述细胞因子为选自IL-2、IL-15、IL-21、Flt3-L、SCF、IL-7、IL-12和IL18中的一种或多种。
5.根据权利要求1或2所述的方法,其特征在于,所述细胞因子的使用浓度为50~1,000U/ml。
6.根据权利要求1或2所述的方法,其特征在于,所述共培养进行7~30天。
7.根据权利要求1或2所述的方法,其特征在于,在所述共培养中,所述外周血单个核细胞与饲养细胞的混合比为1:5~2:1。
8.根据权利要求1或2所述的方法,其特征在于,所述方法进一步包括:在后续培养的第1天至第15天,加入所述经辐射的Jurkat细胞、经辐射的EBV-LCL细胞和所述细胞因子,以在诱导及增殖源自外周血单个核细胞的NK细胞后维持NK细胞。
9.一种预防和治疗癌症的药物组合物,该组合物包含根据权利要求1~8中任一项所述的方法中获得的源自外周血单个核细胞的自然杀伤细胞。
