发明概述

本文所用的术语“多潜能干细胞(pluripotent stem cell)”意指和包括细胞分化为全部三种谱系或胚层(即内胚层、外胚层和中胚层)的细胞类型的能力,但是不包括能够发育成完整人体的人全能干细胞,且不涉及人类胚胎的工业或商业目的的应用。术语多能具有本领域所理解的含义,且包括细胞分化为多种细胞类型的能力。应理解,多能细胞(multipotent cell)可以在其分化能力上比多潜能细胞(pluripotent cell)更受限。本文所用的术语诱导干细胞(“iSC”)指诱导的多潜能干细胞(“iPSC”)或诱导的多能干细胞(“iMSC”)。有时可以用术语“iPS”或“iPS细胞”代替“iPSC”;类似地,有时可以用术语“iMS”或“iMS细胞”代替“iMSC”。本文所述的适用于iPSC的方法和组合物也适用于iSC和iMSC。
术语“测量蛋白质表达”意指测定所产生的蛋白质的量(例如,在细胞孔中通过诸如抗体染色并定量该抗体所染色的细胞的特定荧光的直接方法,或间接地通过测量编码该蛋白质的mRNA的量)。
本文所用的术语“IC50”意指测试化合物导致所测量的分化标记相对于未处理的对照被抑制50%时的浓度。
术语“发育调节标记”意指其表达在发育过程中改变的基因。
术语“中内胚层标记”意指已确定发现其表达在中胚层细胞谱系中的基因。用于本发明的中内胚层标记的实例是Sox 17、EOMES和T-brachyury。

实施例
测定致畸风险的一般方法
本发明的方法通过比较处于定向分化的经处理的多潜能干细胞和对照多潜能干细胞中的中内胚层形成水平来评估药物诱导的致畸性的风险。因此,首先必须确定指导人多潜能干细胞向中内胚层分化的最佳条件。作为观点证明,使用本文实施例中所述的方案,用H9人胚胎干细胞系进行了最初的验证。
优化基线分化
首先,鉴定了在3天的分化中可重复地达到40-60%中内胚层形成的最佳初始细胞接种密度和生长因子浓度(见图2)。对生长因子处理的理想浓度和持续时间进行较少的方案优化来产生中内胚层,本领域技术人员可以将此方法适用于大多数人多潜能干细胞。虽然选择40-60%分化作为这些研究的起点,但由于该方法取决于比较经药物处理的样品与对照样品中的中内胚层形成的比例,确切的中内胚层细胞的百分比应不是关键的,只要效率在孔与孔之间一致。
在最初的验证实验过程中,检查了多种谱系标记、抗体和定量方法。 显示共表达的中内胚层谱系标记Sox17、T-brachyury和EOMES的那些实验对于监测发育破坏是有益的。还可以预测,标记Sox17/EOMES/T-brachyury阴性细胞的其他标记(如图4中所示的OCT4)的定量类似地可以用来定量指导定向分化的信号转导途径的破坏。
确定体外/体内相关性的阈值浓度
优化定向分化的条件后,确定了允许从已知不引起胎儿危害的化合物到已知的致畸化合物之间的分层的剂量反应关系。对于最初的研究,我们使用了具有已知的体内效应的30种市售化合物和40种专利化合物的系列。在第1天和第2天处理包含正在向中内胚层分化的细胞的单孔(见图3)。在单块平板上,将递增浓度的各化合物加至单孔。此外,每块平板包含用已知具有致畸性的化合物处理的孔(作为阳性对照),以及仅用溶剂处理的孔(作为阴性对照)(见图5中的示例性平板图)。
在定向分化结束时,定量中内胚层谱系标记的水平。为了证明本文所述研究的原理,通过免疫染色,然后通过流式细胞术分析、荧光显微镜检术或使用酶标仪来定量,在蛋白质水平测量了细胞中谱系标记的表达。从这些值,针对每种测试化合物计算了每种测试化合物相对于DMSO对照导致所测量的分化标记的50%抑制的浓度(IC50)。检查已知在体内具有致畸性或无致畸性的化合物的IC50值允许确定将非致畸性化合物与致畸性化合物区分开来的IC50值。对于在验证过程中检查(下文)的H9细胞系,发现20uM的浓度为预测致畸性的风险提供了>90%准确性(见图6和7中的表格)。
将该测定用于高通量筛选
一旦用目的多潜能细胞系确定了希望得到的用于测量致畸风险的阈值,即可以用该测定进行更高通量的风险评估。可以以与可接受的预测性水平相关的单个浓度三次重复来测试化合物。

方案的方法
多潜能干细胞制备:
细胞培养。在含10uM Y-27632的mTeSR培养基中解冻H9人ES细胞(来自WiCell)。在用ES细胞级基质胶(BD Biosciences)预包被的两个100mm组织培养级平皿中接种3百万个细胞。Y-27632仅在接种期间需要,应在接种后24小时内将它去除。每天更换mTeSR培养基。细胞应在3-4天内接近铺满。
在96孔板上ACCUTASE和接种
在细胞达到约50%铺满时,去除培养基,用PBS漂洗,然后每个100mm平皿加入2mlaccutase。细胞应在3分钟内分离。立即加入8ml含10uM Y-27632的mTeSR,转移20ml(来自2x100mm平皿)至50ml管中。400xg离心6分钟。弃上清液。加入10ml含Y-27632的mTeSR。上下吹吸悬浮细胞沉淀。计数活细胞的数目,并重悬至1百万个细胞/ml。按5000-50,000个细胞/96孔之间接种细胞。取决于具体细胞系和细胞的批次,可以改变此数目,以获得最佳细胞密度。对于这些实验,接种15,000个细胞/孔。每天更换mTESR培养基,更换2-5天。
多潜能干细胞定向分化和分析
第0天。用含药物的分化培养基-I开始分化
细胞应均匀地分布在96孔板中。细胞在96孔板的孔中约35%铺满时 (2-5天后)开始分化。去除培养基,用高级RPMI洗涤它4-5次。彻底洗去mTESR很重要,因为它包含非常高浓度的维持细胞处于未分化状态的多种因子。加入含有或不含测试化合物的DE分化培养基-I。500rpm涡旋平板3分钟。静置平板24小时。
第1天。更换为含药物的分化培养基-II
迅速用高级RPMI洗涤平板3次(~10分钟)。加入含有或不含化合物的DE分化培养基-II。500rpm涡旋平板3分钟。静置平板48小时。注意,细胞用药物处理了总计72小时。
第3天。终止和固定细胞
1.从96孔板去除培养基。用DPBS GENTLY洗涤孔三次,以去除顶部的碎片或死细胞。加入在37℃预热的新鲜配制的含3%甲醛的PBS。将此溶液加在细胞上15分钟。长时间固定可以引起高背景或抗原改变。用PBS洗涤三次来去除PBS。
2.用含0.1%TritonX-100的PBS透化细胞15分钟。
3.每个96孔加入30ul ImageIT-FX溶液。700rpm涡旋1分钟。室温温育30分钟。
4.用封闭缓冲液(含10%驴血清、0.3%Triton X-100、1%BSA的PBS)洗涤一次,然后加入封闭缓冲液,放置20分钟。
5.加入含Sox17抗体(1:10稀释)的封闭缓冲液。室温温育3小时。
6.用含1%BSA的PBS洗涤四次。
7.加入含DAPI或Hoechst 33242(二者终浓度都是5ug/ml)的PBS(含1%BSA)5分钟。如果随后使用含DAPI的SlowFade,则无需此步骤。
8.向96孔中的每一个加入30ul含DAPI的SlowFade。
数据分析
通过Envision酶标仪在557nm测量荧光强度。
方案和性能标准
前述实施例为示例说明,将本发明应用于人胚胎干细胞H9(Wisconsin AlumiResearch Foundation,WARF)。在将该方法应用于其他多潜能干细胞系时,必须确定影响细胞系依赖性分化效率的通常变量,如初始细胞接种密度、培养基中生长因子的浓度和分化持续时间。在上文所述的此优化方法中,我们监测了中内胚层形成的效率以及沙利度胺介导的药物效应来鉴定用于预测致畸风险的最佳条件。

Claims (17)
1.评估化合物的致畸风险的非诊断方法,其包括:
(1)在可重复地在3天的分化中达到40-60%中内胚层形成的条件下,使所述化合物与正在向中内胚层分化的多潜能干细胞接触;
(2)在分化的第三天测量一个或多个中内胚层标记的蛋白质表达,所述一个或多个中内胚层标记选自Sox17、EOMES和T-brachyury;
(3)测定所述化合物的IC50浓度,其导致所述一个或多个标记的蛋白质表达的50%抑制;和

13.权利要求1-9中任一项的方法,其中步骤(4)中的所述一种或多种已知具有致畸风险的化合物选自放线菌素D、维甲酸、阿糖胞苷、诺考达唑、鱼藤酮、异维甲酸、溴脱氧尿苷、阿霉素、dorsomorphin、沙利度胺、5-氟尿嘧啶、达沙替尼、索拉非尼、丙戊酸、舒尼替尼、齐拉西酮、米安色林、凡德他尼、己烯雌酚、6-氨基-烟酰胺、利坦色林和吉非替尼。
14.权利要求1-9中任一项的方法,其中步骤(4)中的所述一种或多种已知具有致畸风险的化合物选自沙利度胺、米安色林和利坦色林。
15.权利要求1-9中任一项的方法,其中步骤(4)中的所述一种或多种已知无致畸风险的化合物选自埃索美拉唑、叶酸、儿茶素、稠环乙脲、烟酸、阿司匹林、布洛芬、阿昔洛韦、酮舍林、链霉素、甲基多巴、糖精、咖啡因、青霉素和替加色罗。
16.权利要求1-9中任一项的方法,其中步骤(4)中的所述一种或多种已知无致畸风险的化合物选自叶酸和甲基多巴。
17.权利要求1-9中任一项的方法,其中步骤(4)中的所述一种或多种已知具有致畸风险的化合物选自沙利度胺、米安色林和利坦色林;且所述一种或多种已知无致畸风险的化合物选自叶酸和甲基多巴。
