Abstract

背景技术
Ⅱ型拟除虫菊酯类化合物是一大类在如农业、林业的各种各样行业中以及在住宅设置中广泛用于病虫害防治目的的化学杀虫剂。它们都是合成酯类化合物并都包含氰基(CN)基团。
Ⅱ型拟除虫菊酯化合物的非限制性的实例包括氟酯菊酯、氯氰菊酯、氟氯氰菊酯、三氟氯氰菊酯、溴氰菊酯、苯氰菊酯、氰戊菊酯、甲氰菊酯、氟氯苯菊酯、氟氰戊菊酯、氟胺氰菊酯、四溴菊酯。
Ⅱ型拟除虫菊酯化合物是有毒的,而且在某些情况下是致癌的。这是食品行业特别关注的,因为一般人群主要是通过摄入受污染的食物和饮料,如通过食用已被这种化合物处理过的受污染的水、水果和蔬菜或其提取物,而暴露于该化合物。
对食物和水污染的担忧已经导致食品制造商及其供应商需要检测水、食材和例如添加剂如调味剂的食物成分中Ⅱ型拟除虫菊酯化合物的存在。
目前有很多种分析技术用于确定如食物或水样品的样品中Ⅱ型拟除虫菊酯化合物的存在。传统的通用分析技术包括提取,例如液相提取,随后是气相色谱(GC)、与质谱(MS)联用的液相色谱(LC)、或免疫化学分析。
当对仅仅一种或两种Ⅱ型拟除虫菊酯化合物进行分析时,如上面例举的那些已知技术是有效的,但是,当需要确定多种类型Ⅱ型拟除虫菊酯化合物时,它们会变得耗时、复杂和昂贵。另外,通常在特定的情况下的II型拟除虫菊酯化合物,在某些情况下是同分异构体的水平,污染的样品必须已经是已知的,以进行准确的分析。在免疫测定分析的情况下,尤其是这样。

发明内容
现在,申请人已经发现样品中II型拟除虫菊酯化合物的存在可通过多种分析技术快速、容易、经济地确定,尤其是免疫分析,通过使样品经受将II型拟除虫菊酯化合物转变成3-苯氧基苯甲酸或4-氟-3-苯氧基苯甲酸的条件,然后分析这样处理后的样品中3-苯氧基苯甲酸和/或4-氟-3-苯氧基苯甲酸的存在。
除了可转变成4-氟-3-苯氧基苯甲酸的被称为氟氯苯菊酯和氟氯氰菊酯的II型拟除虫菊酯化合物之外,所有现在已知的II型拟除虫菊酯化合物均可转变成3-苯氧基苯甲酸。
本发明所使用的术语“样品”指可包括II型拟除虫菊酯化合物的任何媒介,固体或液体。样品的非限制性的实施例包括,水,水果和蔬菜提取物例如柑橘油,饮料,食材,食品添加剂,调味剂,香料,作物匀浆,房屋尘土,生物固体(biosolid),和土壤样品。
因此,在本发明的第一方面,提供了一种用于确定样品中Ⅱ型拟除虫菊酯化合物的存在的体外方法,该方法包括的步骤:
I.使样品经受存在于样品中的II型拟除虫菊酯化合物会转变成3-苯氧基苯甲酸和/或4-氟-3-苯氧基苯甲酸的条件
II.接着分析该样品中3-苯氧基苯甲酸和/或4-氟-3-苯氧基苯甲酸的存在

在本发明的一个方面,提供了一种用于确定样品中Ⅱ型拟除虫菊酯化合物的存在的体外方法,包括以下步骤:
I.分析样品中3-苯氧基苯甲酸和/或4-氟-3-苯氧基苯甲酸的存在
II.使样品经受存在于样品中的II型拟除虫菊酯化合物会转变成3-苯氧基苯甲酸或4-氟-3-苯氧基苯甲酸的条件
III.接着分析该样品中3-苯氧基苯甲酸和/或4-氟-3-苯氧基苯甲酸的存在或其浓度升高。
II型拟除虫菊酯化合物可通过多种已知方法使用市售的起始原料、试剂和溶剂转变成3-苯氧基苯甲酸或4-氟-3-苯氧基苯甲酸。
在一个示例性实施例中,II型拟除虫菊酯化合物在下述方法中转变成3-苯氧基苯甲酸或4-氟-3-苯氧基苯甲酸,所述方法包括以下步骤:
I.在碱性或酸性条件下水解II型拟除虫菊酯化合物以形成相应的α-氰基醇,α-羟基-3-苯氧基苯乙腈或α-羟基-4-氟-3-苯氧基苯乙腈
II.将α-羟基-3-苯氧基苯乙腈或α-羟基-4-氟-3-苯氧基苯乙腈氧化成相应的3-苯氧基苯甲醛,以及
III.氧化成3-苯氧基苯甲酸或4-氟-3-苯氧基苯甲酸

可与决定所用的水解和氧化试剂的类型和数量相关的因素可为与具体类型样品中通常包括的II型拟除虫菊酯化合物的相对比例以及样品的性质,例如由于柑橘油包含许多天然抗氧化剂,因而难以氧化包含于其中的II型拟除虫菊酯化合物。此外,柑橘油的不溶性质使得难于水解。
通常情况下,应当使用与II型拟除虫菊酯化合物的相对比例相应的大量过量的氧化性和水解试剂,通常包括于具体类型的样品中,以确保样品中II型拟除虫菊酯化合物的转变,尤其是在柑橘油样品中。
通常情况下,样品例如食物,水,水果和蔬菜提取物例如柑橘油,饮料,食材,食品添加剂,调味剂,香料,作物匀浆,房屋尘土,和土壤样品以ppb或ppm浓度包含这些化合物,并因此本领域技术人员一般可以采用在几个数量级过量的数量例如千分之几以确保样品中II型拟除虫菊酯化合物的转变。

实施例1–使用过氧化氢作为氧化剂和铂催化剂作为中和剂转变
橙油中的II型拟除虫菊酯化合物
掺入橙油以所指示的杀虫剂的最终浓度为10μM并且所有样品均一式三份运行。100μL橙油样品吸到玻璃小瓶中,然后再在稳定的压缩空气流下挥发1小时。然后向样品添加200μL二恶烷和800μL的30%w/v过氧化氢和0.4NNaOH。然后样品于50°C温育搅拌1小时。样品然后通过加入氧化铝颗粒上的0.5%的铂中和,其允许于25°C不搅拌温育2小时完成中和。然后样品在通过ELISA分析之前稀释200x至10%MeOH90%0.1M磷酸盐缓冲液中。在图2中可以看到结果。

实施例3-使用次氯酸钠作为氧化剂和抗坏血酸作为中和剂从橙 油转变拟除虫菊酯化合物

Claims (7)
1.一种用于确定样品中II型拟除虫菊酯化合物的存在的体外方法,该方法包括步骤:
(I)使样品经受存在于样品中的II型拟除虫菊酯化合物会转变成3-苯氧基苯甲酸或4-氟-3-苯氧基苯甲酸的条件,及
(II)接着分析该样品中3-苯氧基苯甲酸和/或4-氟-3-苯氧基苯甲酸的存在,以及
所述方法在所述使样品经受II型拟除虫菊酯化合物会转变成3-苯氧基苯甲酸或4-氟-3-苯氧基苯甲酸的条件之前,还包括首先确定3-苯氧基苯甲酸和/或4-氟-3-苯氧基苯甲酸的存在的步骤。
2.根据权利要求1的方法,其中II型拟除虫菊酯化合物会转变成3-苯氧基苯甲酸或4-氟-3-苯氧基苯甲酸的条件按顺序包括以下步骤:
(I)在碱性条件下使用氢氧化钠水溶液水解II型拟除虫菊酯基团以形成α-氰基醇α-羟基-3-苯氧基苯乙腈或α-羟基-4-氟-3-苯氧基苯乙腈,
(II)将α-羟基-3-苯氧基苯乙腈或α-羟基-4-氟-3-苯氧基苯乙腈氧化成3-苯氧基苯甲酸或4-氟-3-苯氧基苯甲酸,及
(III)可选地,中和步骤II中所用的氧化剂。
3.根据权利要求1的方法,其中II型拟除虫菊酯化合物会转变成3-苯氧基苯甲酸或4-氟-3-苯氧基苯甲酸的条件按顺序包括以下步骤:
(I)在碱性条件下使用氢氧化钠水溶液水解II型拟除虫菊酯基团以形成α-氰基醇α-羟基-3-苯氧基苯乙腈或α-羟基-4-氟-3-苯氧基苯乙腈,
(II)使用过氧化氢将α-羟基-3-苯氧基苯乙腈或α-羟基-4-氟-3-苯氧基苯乙腈氧化成3-苯氧基苯甲酸或4-氟-3-苯氧基苯甲酸,及
(III)可选地,中和步骤II中所用的氧化剂。
4.根据权利要求1的方法,其中II型拟除虫菊酯化合物会转变成3-苯氧基苯甲酸或4-氟-3-苯氧基苯甲酸的条件按顺序包括以下步骤:
(I)在碱性条件下使用氢氧化钠水溶液水解II型拟除虫菊酯基团以形成α-氰基醇α-羟基-3-苯氧基苯乙腈或α-羟基-4-氟-3-苯氧基苯乙腈,
(II)将α-羟基-3-苯氧基苯乙腈或α-羟基-4-氟-3-苯氧基苯乙腈氧化成3-苯氧基苯甲酸或4-氟-3-苯氧基苯甲酸,其中氧化是以酶法进行的,及
(III)可选地,中和步骤II中所用的氧化剂。
5.根据权利要求1的方法,其中II型拟除虫菊酯化合物会转变成3-苯氧基苯甲酸或4-氟-3-苯氧基苯甲酸的条件按顺序包括以下步骤:
(I)在碱性条件下使用氢氧化钠水溶液水解II型拟除虫菊酯基团以形成α-氰基醇α-羟基-3-苯氧基苯乙腈或α-羟基-4-氟-3-苯氧基苯乙腈,
(II)将α-羟基-3-苯氧基苯乙腈或α-羟基-4-氟-3-苯氧基苯乙腈氧化成3-苯氧基苯甲酸或4-氟-3-苯氧基苯甲酸,及
(III)可选地,中和步骤II中所用的氧化剂,其中中和是使用铂催化剂进行的。
6.根据权利要求1的方法,其中确定3-苯氧基苯甲酸或4-氟-3-苯氧基苯甲酸的浓度是通过ELISA进行的。
7.根据权利要求1的方法,其中所述样品是柑橘油样品。
