Description
技术领域
本发明涉及一种慢性全身性猴帕金森病模型及其药物筛选方法。
背景技术
帕金森病(Parkinson’s disease,PD)的实验动物模型已有多种,包括小鼠、大鼠的外科损伤或6-羟多巴立体定向注射毁损的模型等。最有价值的模型动物应是和人类神经***尤其是中脑基底节区有着相似结构、功能和复杂程度的物种。灵长类PD动物模型能够反映PD症状的基本特征,是目前最接近人类PD的动物模型,为PD的基础研究提供了良好的基础和平台。
人类帕金森病是发生于中枢神经***的慢性神经变性疾病。往往由一侧肢体发病,逐渐发展到对侧肢体,症状逐渐加重。当前采用颈内动脉注射MPTP建立的恒河猴帕金森病模型,由于实验猴帕金森病样症状出现迅速且局限于一侧,与人类帕金森病的自然进程不相符合。因此,建立一种缓慢起病,并出现全身帕金森病样症状的灵长类帕金森病动物模型具有重要价值。
目前,有学者提出采用MPTP注射建立慢性全身性帕金森病模型的方法。但是,这些方法具有建模成本高,建模方法较复杂的缺点。Russ等连续29天使用总量为2.5mg/kg的MPTP注射建立了慢性帕金森病模型,模型猴在开始注射后13天开始出现帕金森病样症状,在注射结束后5天,症状消失[1]。Albanese等发现,使用慢性MPTP注射导致纹状体多巴胺水平降低50%后停止用药,8个月后,模型猴多巴胺代谢水平恢复至建模前的正常水平[2]。
发明内容
本发明的技术方案是提供了一种慢性全身性猴帕金森病模型的制备方法。本发明的另一技术方案是提供了一种治疗慢性全身性帕金森病药物筛选方法。
本发明提供了一种慢性全身性猴帕金森病模型的制备方法,是将剂量为0.2mg/kg的MPTP施用于猴,自首次施用日起,每间隔2天施用相同剂量MPTP,连续施用7次;其后改为每隔3天施用1次,连续施用7次。
其中,所述的MPTP施用方法是静脉注射。
本发明还提供了该方法制备的动物模型在筛选治疗慢性全身性帕金森病的药物中的用途。
本发明还提供了一种筛选治疗慢性全身性帕金森病的药物的方法,它包括如下步骤:
a、将剂量为0.2mg/kg的MPTP施用于猕猴,自首次施用日起,每间隔2天为施用相同剂量MPTP,连续施用7次;其后改为每隔3天施用1次,连续施用7次,建立慢性全身性猴帕金森病模型;
b、将候选药物、治疗慢性全身性帕金森病的经典药物分别施用于动物模 型;
c、与治疗慢性全身性帕金森病的经典药物比较,观察候选药物对慢性全身性帕金森病的症状的影响情况,评价潜在治疗慢性全身性帕金森病的药物。
其中所述的慢性全身性帕金森病的症状包括:基本的运动技能状况、精细运动、震颤、动作徐缓程序、木僵、运动、姿势、吃惊反应、运动减弱、平衡等。
其中,所述的MPTP施用方法是静脉注射。
本发明建模方法具有如下优点:1、建模操作简单,容易实施;2、注射药物次数少,建模成本低;3、模型维持时间长,模型症状典型;4、模型症状严重程度适中,饲养容易。
显然,根据本发明的上述内容,按照本领域的普通技术知识和惯用手段,在不脱离本发明的上述基本技术思想前提下,还可以作出其它多种形式的修改、替换和变更。

具体实施方式
实施例1本发明动物模型的制备
一、材料和方法
1.实验动物入选:
参照实验猴纳入标准(见后述),由四川平安实验动物繁育中心购入4只健康成年猕猴。年龄5-6岁,体重4.8-6.3Kg,雄性2只,雌性2只,分别以M9,M10,M11,M12代表。经检验、检疫合格后,置于国家(成都)中药安全性评估中心(GLP)大动物饲养中心饲养。
实验猴纳入标准:
①、年龄4-7岁;
②、有血液常规、生化和传染病等检测报告,且检查结果正常,无传染 病;
③、既往未接受其他实验研究;
④、体重4-7Kg;
⑤、躯干、肢体及头面部活动自如;
⑥、对外界环境及刺激反应正常。

2.建模前实验猴饲养:
满足实验要求的猴在进行建模手术前3周移入GLP中心大动物饲养中心进行适应性训练,使其适应新的生活环境。所有实验猴采用专用猴笼,单笼饲养,参照动物福利要求和医学实验伦理委员会要求,为实验猴提供全价饲料和水果,并由专职饲养人员照顾实验猴日常生活和饮食。饲养环境提供12小时照明,供应充足的食物和饮用水,并保持猴房通风、清洁,维持室内温度15-25摄氏度。定期对饲养间进行消毒处理,并对实验猴进行隔离,禁止与实验无关人员进入饲养间。
3.建模前实验猴行为评估:
实验猴在大动物饲养中心单笼饲养3周,饲养期间,固定猴笼及猴笼位置。3周后,待实验猴熟悉饲养环境、饲养人员及实验人员后,采用猴帕金森病行为评分表(详见附录I)对实验猴进行行为学评分,记录基础评分值。使用摄像机记录实验猴活动情况并存档。

4.建模前猴脑18FDG-PET-CT检查
4.1扫描时间安排:
实验猴在大动物饲养中心饲养3周后,使用四川大学华西医院核医学科 Philips PET-CT进行猴全脑18FDG-PET-CT糖代谢检测。
4.2扫描操作:
经肌肉注射***(10mg/Kg)作全身麻醉。麻醉满意后,经外周静脉注射新鲜合成的经同位素标记的显影剂:18FDG(各实验猴显影剂放射剂量见表2)。猴放置于放射屏蔽环境安静休息30分钟后,将猴放置于检查床上,妥善固定头部,开始进行全脑PET-CT扫描。由华西医院核医学科专职医学技术人员操作PET-CT设备进行图像采集,将扫描结果存档。扫描结束后,将猴放回原猴笼继续按照原饲养条件饲养。
5.建模前准备
建模前3天将实验猴移入隔离饲养间饲养,按照原饲养环境给予照明、食物和饮水。于建模手术前1晚开始禁食水,直至手术,并称量实验猴体重。
6.建模操作:

将实验猴置于隔离饲养间单笼饲养3周,按照原饲养环境给予照明、食物和饮水,并对实验猴进行血液生化检测。于每次注射MPTP溶液前1天称量实验猴体重。建模操作直接在动物隔离饲养间进行。按照第一部分描述的要求,做好隔离饲养间通风和消毒工作,确保实验人员及其它实验动物安全,避免有毒物污染环境。禁止与实验无关人员进入饲养间。自建模开始至最后一次给药后5天,所有进入饲养间的人员均按照第一部分描述的要求,穿戴个人防护用品,严格按照接触有毒物质操作规范进行建模和饲养操作,并对饲养环境作无害化处理。
6.2MPTP配制:
MPTP溶液在隔离饲养间内现场配制。严格按照个人防护要求进行。按照0.2mg/kg计算药物使用量,将所需药物用生理盐水稀释至10ml,并置于20ml注射器中。将注射器置于微量注射泵上备用。
6.3建模操作:
建模所需材料、设备以及建模操作同第二部分。自首次注射日起,每间隔2天为实验猴注射相同剂量MPTP溶液,连续注射7次;其后改为每隔3天注射1次,连续注射7次。

7.建模后饲养:
7.1模型猴在隔离饲养间饲养。提供与建模前相同的照明和饲养条件,并增加水果供应,保持饲养间的良好通风。
7.2每日定时由同一实验人员按照猴帕金森病行为学评分表,对实验猴进行行为学评分并录像。
8.建模后PET-CT检查
自首次注射MPTP 3个月后进行头部18FDG-PET-CT检查。扫描方法同前述。

9.建模后脑组织病理学检查
自首次注射MPTP 3个月后对实验猴进行脑组织病理学分析。采用灌注取材。
10.模型行为学变化分析:
对实验猴的行为学评分进行汇总,绘制模型猴行为学变化曲线图。
二、结果
1.实验猴行为学变化情况
实验猴在完成适应性饲养后,基础行为学评分均为“0”分,实验猴对周围环境感知良好,对刺激行为警惕性强,进食正常。前3次静脉注射后实验猴无明显症状。第4次静脉注射次日,M9和M12开始出现轻度活动减少。第5次注射后,所有实验猴均表现出轻度活动减少。其后,实验猴症状缓慢加重,至第7次注射后,M9和M12出现明显的活动减少和运动迟缓,面部表情减少,帕金森病样症状达到峰值(见图1,2);M10,M11症状相对较轻。所有试验猴在延长注射间隔时间至每隔3天1次后,模型猴症状与完成前7次注射后相似,且症状相对稳定,有小范围波动,实验猴表现为运动迟缓,活动减少,对刺激反应较迟钝,但躯体姿势受影响较小(见图3)。实验猴在完成7次注射后,症状达峰值,其后延长注射间隔时间后,试验猴症状保持稳定。
停止MPTP注射后,实验猴帕金森病样症状出现快速修复,至结束MPTP注射后30-40天,症状几乎全部消失。
2.实验猴脑18FDG-PET-CT检查结果
经外周静脉注射MPTP建立全身帕金森病模型后,模型猴脑双侧基底节区及额、颞叶糖代谢较建模前降低,且双侧无明显差别(如图4所示)。
3.模型组织学检查结果
3.1HE染色
实验猴基底节区神经元胞体缩小,左右侧神经元数量无显著差异。如图5,6)。
3.2TH染色
实验猴左右侧基底节区仅可见少数神经元胞浆内存在TH阳性染色颗粒, 较动脉模型中未毁损侧TH阳性细胞明显减少(见图7,8)。
3.3D2染色
D2受体染色阳性细胞显示为棕色。实验猴双侧基底节区D2受体染色阳性细胞数无显著差异(见图9,10)。
上述行为学分析以及免疫组织化学检查可证明建模成功。
综上所述,自首次注射MPTP,每间隔2天为实验猴注射相同剂量MPTP溶液,连续注射7次;然后改为每隔3天注射1次,连续注射7次,建模操作简单,模型维持时间长,症状典型;且严重程度适中,可将制备的动物模型进一步建立药物筛选平台。
实施例2利用治疗慢性全身性帕金森病的经典药物验证慢性全身性帕金森病动物模型并初步建立相关药物筛选体系
a、取实施例1制备的慢性全身性猴帕金森病模型;
b、采用目前公认的治疗慢性全身性猴帕金森病的经典药物(美多巴、息宁)、候选药物分别施用于慢性全身性猴帕金森病模型;
c、与治疗慢性全身性帕金森病的经典药物比较,观察候选药物对慢性全身性帕金森病的症状影响情况,其中所述的慢性全身性帕金森病的症状包括:基本的运动技能状况、精细运动、震颤、动作徐缓程序、木僵、运动、姿势、吃惊反应、运动减弱、平衡等;并将其量化评分,评价潜在治疗慢性全身性帕金森病的药物;也验证了本发明慢性全身性帕金森病动物模型用于药物筛选的可行性。
经过步骤c的评价,经典药物能明显改善模型猴的症状,证明本发明慢性全身性猴帕金森病模型建模成功,同时也证明了该药物筛选体系是有效的。
经过步骤c的评价,如果候选药物对上述猴慢性全身性猴帕金森病的症状有改善的,并达到类似经典治疗药物的评分标准,即证明该候选药物具有治疗慢性全身性猴帕金森病的作用。

Claims (1)
1.一种慢性全身性猴帕金森病模型的制备方法,其特征在于:将剂量为0.2mg/kg的MPTP施用于猴,自首次施用日起,每间隔2天施用相同剂量MPTP,连续施用7次;其后改为每隔3天施用1次,连续施用7次;所述的MPTP施用方法是静脉注射;所述猴为猕猴,年龄为5~6岁。
