技术领域
本发明主要涉及活性聚合物膜。更具体地说,本发明涉及一种包括内嵌无机物纳米颗粒的聚合物基质的膜。
背景技术
微生物感染或污染仍然是医药、食品加工、药品加工及环境设置中导致疾病和/或腐败的主要原因。另外,虽然所有外科手术有伤口感染的风险,但是在移植手术中或手术后医疗植入表面的细菌粘附、定植和/或生物膜的形成大大增加了感染的风险。另外,已知微生物膜在例如食品、医药、化学工业加工设备的表面上形成,同时也可以在其他的表面如灌溉设备、暴雨或污水排水管、冷却塔或船舶的表面上形成。生物膜常常会对这些设备带来严重损坏,并且微生物膜的形成会引起管道及加工设备的堵塞。
可以采用抑菌剂或杀菌剂控制微生物。抑菌剂能够抑制微生物的进一步生长,相应地能够抑制生物膜形成,或者,当在医疗环境中使用时,能够防止感染发展成全面爆发的疾病(full blown illnesses)。另一方面,杀菌剂能够通过杀死微生物起作用。少量杀菌剂对于微生物有广谱作用,并且在某些情况下,可能需要使用组合药剂控制微生物的生长。不过,大多数广谱杀菌剂不适宜用在医疗应用中,因为它们往往会对较高级生物体的组织产生有害影响。另外,尽管抗生素,通常只对细菌有活性,提供了治疗或防止人体及其他动物体中微生物感染又不会产生这种有害影响的可能性,但是,不幸地是,只有少量抗生素制剂显示广谱的活性(即,它们的杀菌活性常常只针对某种细菌种或属)。而且,多年来抗生素的广泛、频繁的使用已经导致细菌抗生素耐药性或多种耐药性菌株的演化(如,耐甲氧西林金黄色葡萄球菌(methicillin-resistant Staphylococcus aureus); MRSA)。因此,抗生素对某些细菌感染可能无效的。
因此,大量正在进行的研究工作旨在开发出新的在医疗应用中使用的抑菌剂或杀菌剂。这种需求又被减少医院获得性感染率的全球需要、西方世界的老龄化人口、对最小化医疗费用不断增加的关注,以及不断增加的被告知需要改善的医疗治疗的病人而加剧。从这个意义上来说,作为一种手术过程中或手术后抗细菌感染的有前景的方法,人们对医学移植及假肢中使用的抗菌涂层和表面的接受度不断增加。为此,替代性的抗菌技术,如纳米基涂层技术,有了良好的发展势头。医疗设备已经被视为用于包括创伤敷料、导管以及医学植入如支架和起搏器的抗菌涂层及表面技术的可能备用方式。
银的抗菌性能很多年前已经被认识到了。由于银的成本并且相对现代抗生素其给药相对困难,因此一直到近年来银作为抗菌剂也只引起了少量关注。也就是说,近年来,对于银可能作为抗菌剂使用的关注有所复苏。例如,银化合物以及银胶体已被纳入用于烧伤、急性和慢性创伤的绷带,用于治疗腿部溃疡。事实上,已知的产品,如包括1%的磺胺嘧啶银的Silvadene®乳膏,现在已是最广泛使用的用于治疗烧伤相关的感染的抗菌产品。另外,几项近期研究显示了银离子与大颗粒或纳米颗粒表面结合以减少医疗应用中微生物感染及细菌粘附和/或定植事件的能力。这些医疗应用包括,例如,烧伤治疗(Paddock et al,2007;Caruso

发明内容
第一方面,本发明提供了一种包含可渗透聚合物基质的膜,所述聚合物基质带有一种或多种官能团以及在所述聚合物基质内嵌有一种或多种无机物纳米颗粒。
优选地,无机物为银。
第二方面,本发明提供了一种用于将一膜应用到合适基质表面上的方法,所述方法包括步骤:
(ⅰ)通过等离子聚合淀积并聚合包含一种或多种官能团的单体,以在所述表面形成可渗透的聚合物基质;
(ⅱ)将无机物与所述官能团复合,以及
(ⅲ)还原所述无机物,形成一种或多种所述无机物的纳米颗粒;
其中所述一种或多种无机物纳米颗粒嵌入所述聚合物基质中。
该方法适用于制造多层膜,其中由步骤(ⅰ)到(ⅱ)制成的膜构成包括第一聚合物基质的第一膜层,所述第一聚合物基质带有一种或多种官能团以及,一种或多种无机物纳米颗粒嵌入所述第一聚合物基质中,而该方法进一步包括步骤:
(ⅳ)将第二膜层应用到第一膜层的表面。

具体实施方式
本发明申请人已经鉴定并开发了各种膜,以及制造这些膜的方法,包括其中嵌入无机物纳米颗粒的聚合物。这些膜可以提供任何数量的应用,例如,制作抗菌涂层或抗菌表面。制造这些膜的方法提供了一种快速、简单、方便的将活性聚合物膜应用至表面的工艺。
因此,第一点,本发明提供了包含具有渗透性的聚合物基质的膜,所述聚合物基质中带有一种或多种官能团以及在所述聚合物基质内嵌有一种或多种无机物纳米颗粒。
这里使用的术语“膜”包括任何物理表面层,它们可以,例如,为其应用的下层表面(如固体表面)提供某些性能。因此,上述膜的厚度或组成可以是不规则的或均一的,同时可以部分或完全包覆或覆盖下层表面。
根据第一点,所述膜包含其中嵌有纳米颗粒的聚合物基质,其对所述无机物具有渗透性(例如,无机物可以扩散进入聚合物基质,也可以从中扩散出来)。
所述聚合物基质可以是有孔的,包括大量的孔,一般是纳米孔(如孔的直径约2nm到约4μm,更优选的为约5nm到1μm),它们可以是分开的、相连的或两者的组合。无机物纳米颗粒可以位于聚合物基质的孔中。
优选地,所述膜包含等离子体聚合物基质。因此,这种聚合物基质可以通过等离子体聚合过程形成,其中至少聚合一种单体以形成聚合物基质。更优选地,所述膜包含有孔的等离子体聚合物基质。
本领域技术人员熟知包含等离子体聚合物基质的涂层通常可能是“坚硬的”,这可能由于收缩及坍塌造成涂层失效。人们怀疑这种涂层的刚性是由等离子体聚合过程中成分单体的交联引起的。所以,第一点所述的膜优选地为一薄膜,包含薄的、有弹性的聚合物基质。除了不易于失效的潜在性,这在医疗应用中是特别重要的,这种薄膜也很好的适用于“软的”应用例如隐形眼镜的表面涂层。第一点所述的膜也包含低交联度的等离子体聚合物基质,以提供机械柔韧性。
薄膜可以通过本领域技术人员熟知的任何薄膜淀积方法形成,包括但不限于,化学淀积法如电镀、化学溶液淀积(CSD)以及化学气相淀积法(CVD),物理淀积法如物理气相淀积法(PVD)、热蒸发、电子束蒸发、溅射法、脉冲激光淀积法以及阴极电弧淀积法(电弧离子蒸镀(Arc-PVD)),以及其他方法如反应溅射法、分子束外延(MBE)和拓扑衍生(topotaxy)。薄膜厚度优选地约小于4μm,更优选地在约5nm到约4μm之间,更优选地在约5nm到500nm之间,更优选地在约5nm到150nm之间,最优选地从约5nm到约25nm。
这里使用的术语“官能团”包括原子或特定的聚合物基质(或组成聚合物基质的单体)的原子基团,无机物与所述物质通过,例如,化学共价键、化学离子键或化学配位键结合(例如形成复合物)。
这里使用的术语“纳米颗粒”可以理解为“纳米”尺寸的颗粒,其可以包括各种尺寸的颗粒,并且一般平均直径或主截面尺寸范围在1到250nm,更优选地范围为5到50nm,最优选地范围为10到25nm。
这里使用的术语“无机物”包括基本纯态的无碳元素(如,铜、银或硒)以及任何主要由不是碳的元素构成的无机化合物。不过某些含碳的化合物也被认为是无机化合物,如碳酸盐、简单的碳氧化物和氰化物,以及碳的同素异形体,而针对本说明书的目的,包含碳骨架的化合物和有机金属化合物不能看成是无机化合物。
第一点所述的膜可以作为任意合适的基质表面的涂层使用,优选地是固体基质。优选地,上述表面在使用该种膜时不需要进行预先改性处理。
合适的“硬”基质包括金属、陶瓷、合成聚合物、生物聚合物以及医疗应用中常用的材料,包括不锈钢、钛、聚丙烯钛、羟磷灰石、聚乙烯、聚氨酯、有机硅聚合物和全氟聚合物。合适的“软”基质包括丙烯酸水凝胶聚合物、硅氧烷水凝胶聚合物、纤维绷带、包扎材料,以及合成敷料,如水凝胶或泡沫敷料。
另外,可以认为第一点所述的膜适合用于作为制造和包装材料使用的涂层材料,如塑料(例如,聚丙烯、聚苯乙烯、聚对苯二甲酸乙二醇酯、聚酯、聚酰胺、聚氯乙烯、聚氨酯、聚碳酸酯、聚偏二氯乙烯和聚乙烯等)、金属和金属合金(例如,不锈钢、钢、铁和锡等)。另外,光学或电子装置中使用的材料,如硅、二氧化硅、铝、铜及类似材料也可以使用第一点所述的膜涂覆。
第一点所述的膜优选地包含由单体形成的聚合物基质,所述单体包括一种或多种官能团,可与所述无机物的氧化态形成复合物。因此,优选的单体包含至少一种官能团,选自由羟基、羰基、醛基、酮基、羧基、过氧基、酰胺基、胺基、亚胺基、酰亚胺、叠氮化物、氰酸盐和硝酸盐基团组成的组。不过,更优选的是包含至少一个胺基的单体。例如,选自由挥发性胺组成的组的单体包括,例如,烯丙胺、二(二甲氨基)甲基乙烯基硅烷、二甲基氨基硅烷、吡啶和庚胺。本发明的某些实施例中,上述单体可以选自那些可聚合形成生物相容性聚合物的物质。进一步,某些实施例中,上述单体可以选自那些可以在聚合后功能化或可以共聚使得最终聚合物基质功能化的物质。
更优选地,第一点所述的膜包括选自由烯丙胺、庚胺及其他挥发性烷基胺组成的组。
在一个特别优选的实施例中,所述膜包含由正庚胺(HA)、烯丙胺(AA)或它们的组合物形成的聚合物基质。如下文实例中所述,发现正庚胺和烯丙胺在制造富含胺官能团的薄膜时非常有用。另外,已知正庚胺比其他本领域技术人员熟知的富胺单体的挥发性和毒性都更小,因此,包含由正庚胺形成的聚合物基质的膜尤其适用于医疗应用、食品和/或药物加工应用中。另外,如下文实例中所示的,如果需要,正庚胺和烯丙胺的物理化学性质适合用于改变嵌入其中的无机物纳米颗粒的浓度(concentration)及释放(release)。
第一点所述的膜可以用于对合适的带有所述无机物的基质进行功能化,优选的为固体基质。这种功能化的基质可以有很多应用,例如在医疗应用中如作为医学移植的涂层,通常作为防止细菌粘附、定植和/或生物膜形成的涂层材料,在光学中用于生成反射或抗反射涂层,以及作为电子设备的涂层表面以形成,例如,集成电路、微处理器和半导体的层绝缘体、导体。因此,所述膜可以用于功能化合适的带有无机化合物的基质,无机化合物如三氧化二砷(例如,用于涂覆砷半导体或治疗急性粒细胞性白血病)、硼酸(例如,用于杀菌或杀虫表面)、硫酸镁或氧化镁(例如,用于医疗应用的某些范围)、二氧化硅(例如,在微电子中作为电绝缘体使用)、氧化锌(例如,用在化学或生物传感器中、半导体中或作为药物营养补充剂使用)以及类似物质。另外,上述膜可以用于功能化合适的带有非碳元素如金属(例如,铜、银、金、镁和锌,或金属合金)的基质,用于形成抗菌表面或涂层(例如用于医疗装置包括可植入及不可植入的装置如替换关节、导尿管、经皮进入导管(percutaneous access catheter)、支架、和其他假体以及不可植入装置如绷带、创伤敷料、隐形眼镜和面膜以及用于呼吸医用空气及氧气的设备)或用于涂覆半导体和微处理器,例如,硒,可用于涂覆太阳能电池或用作营养补充剂,以及非金属(例如,氟,可用于涂覆假牙)。
这里上述膜包含元素金属的纳米颗粒,优选地所述金属选自铜或银(已被熟知用作抗菌剂并且给药后能够被高等生物很好地耐受的金属),或它们的混合物。这种膜优选地通过将特定金属的氧化(即,离子的)形态与官能团复合在聚合物基质中形成带金属离子的复合物来形成。之后,所述复合物被还原以形成嵌入聚合物基质中的纳米颗粒。最优选地,这种膜通过在胺-金属离子复合物形成后进行正庚胺和/或烯丙胺的等离子体聚合形成。
本发明的抗菌膜(例如,包含铜、银、硒或它们的混合物的纳米颗粒),适用于一系列上述提到的医疗应用。这种抗菌膜包含聚合物基质,其对由纳米颗粒组成的抗菌剂具有可渗透性。也就是说,抗菌剂随着时间从纳米颗粒中溶解(例如,进入已渗入所述膜的液体中)或分离,并且之后从膜中渗透出到达药剂可发挥其抗菌活性的表面和/或四周。因此,这里的抗菌膜包含例如铜或银纳米颗粒,聚合物基质优选地可被金属离子(如,Cu+和Ag+)渗透。
本发明的抗菌膜(例如,包含铜、银、硒或它们的混合物的纳米颗粒),也适用于需要抑制细菌粘附、定植和/或生物膜形成的非医疗应用中。例如,工业表面常与水流接触,特别易于生物膜形成,因此这里所述的抗菌膜适于在这种表面中应用。合适的非医疗应用的具体实例包括水处理设备的涂层、冷却塔组件的涂层、加工设备特别是食品和药品加工中的设备的涂层,以及食品及药物包装涂层。另外,本发明的抗菌膜可以用于给合适的基质,优选地为固体基质提供防腐蚀屏障(即,该膜可以提供抗腐蚀涂层)。
而且,这里所述的膜包含元素金属,特别是铜、银和金(它们的导电性众所周知)的纳米颗粒,该种膜适用于涂覆半导体或微处理器。
在一些实施例中,第一点所述的膜可以是多层膜(即,包括两层或多层膜层)。因此,该膜可包括第一和第二聚合物膜层,所述第一膜层包含聚合物基质(优选地有孔),聚合物基质带有一种或多种官能团以及在所述聚合物基质中嵌有一种或多种无机物纳米颗粒;并且其中所述第二膜层可,例如,为该膜提供附加的功能性特性和/或改进第一膜层的特性(例如,改变抗菌剂从第一膜层渗透出的速率,以使多层膜能以可持续的方式释放抗菌剂或其它的无机物),或者,使第二膜层对嵌入第一膜层的无机物基本不能渗透,从而有效地“捕捉”所述无机物。第二膜层也可以包含有用的配体如蛋白质或肽(包括对宿主细胞粘附和/或生长有利的生长因子)或其它化合物如具有抗生物粘附作用的聚酯(例如,聚乙二醇)。
因此,在一些实施例中,第一点所述的膜包括第一和第二聚合物膜层,其中所述第一膜层包含第一聚合物基质(优选地有孔),所述第一聚合物基质带有一种或多种官能团以及,在所述第一聚合物基质中嵌有一种或多种第一无机物纳米颗粒,并且其中所述第二膜层包含第二聚合物基质(优选地有孔),所述第二聚合物基质带有有一种或多种官能团(例如,金属离子复合的官能团)以及在所述第二聚合物基质内嵌有一种或多种第二无机物纳米颗粒(其中,第一和第二无机物可以是相同的或不同的);在其它的实施例中,第一点所述的膜包括第一和第二聚合物膜层,其中所述第一膜层包含第一聚合物基质(优选地有孔),所述第一聚合物基质带有有一种或多种官能团以及在所述第一聚合物基质内嵌有一种或多种无机物纳米颗粒,并且其中所述第二膜层包含第二聚合物基质,其能控制所述无机物的渗透(例如,第二聚合物基质可以对所述无机物渗透性很差或所述无机物基本不能渗透)。
另外,至少可以加入一第三膜层,其可以包含有用的配体如蛋白质或肽,或其它的化合物如聚酯,例如,发挥其特定的生物界面的作用如细胞粘附或防止细胞粘附。
任意第二或第三(等等)膜层厚度优选地小于约100nm,最优选地从约1nm到约50nm,更有选地从约5nm到约25nm。
并且,在一些实施例中,第一点所述的膜可以通过将所述膜接触氧化剂(例如,通过将所述膜置于生物液体中,或者其它水性物质如冷却塔水、废水或环境水中)来“引发”无机物从嵌在聚合物基质内的纳米颗粒中释放出来(即,通过渗透)。
第二点,本发明提供了一种将膜应用至合适基质表面上的方法,所述方法包括步骤:
(ⅰ)通过等离子体聚合法淀积并聚合包含一种或多种官能团(例如,金属离子复合的官能团)的单体,在所述表面形成具有渗透性的聚合物基质;
(ⅱ)将无机物与所述官能团复合;以及
(ⅲ)还原所述无机物,形成一种或多种所述无机物的纳米颗粒;
其中所述一种或多种无机物纳米颗粒嵌入所述聚合物基质中。
优选地,所述聚合物基质是多孔聚合物基质。
等离子体聚合适用于在任意合适的基质,如上述的硬和软的基质,的表面淀积和聚合物单体(即,用来制造聚合物基质)。另外,等离子体聚合法可以选择合适的可以适当地对表面改性(例如,提供亲水性或疏水性)的单体。而且,等离子体聚合法可以操作以使聚合物基质具有需要的交联度,反过来,(如果需要)其能够调节无机物从嵌入的纳米颗粒中任意外扩散的速率。
聚合单体的步骤优选地通过等离子体聚合法进行,例如射频辉光放电等离子体聚合法,从而形成有孔的等离子体聚合物基质。优选的单体为本发明第一点中所述的。最优选地,所述单体为正庚胺、烯丙胺或它们的组合,聚合形成的聚合物基质富含胺官能团。
将无机物与官能团复合的步骤可以采用本领域技术人员熟知的任何方法。不过,优选地,将无机物与官能团复合的步骤为将步骤(ⅰ)中形成的聚合物基质浸入包含所述无机物的溶液中。例如,为了将银离子与富胺官能团的聚合物基质复合,所述聚合物基质可以浸入AgNO3溶液中(例如,浸入0.02M溶液中1hr)。
还原无机物的步骤可以采用本领域技术人员熟知的任何方法。不过,优选地,还原无机物的步骤包括将包含复合了官能团的无机物的聚合物基质浸入还原溶液中。例如,为了还原银离子以及胺官能团复合物以形成银纳米颗粒,所述聚合物基质可以浸入NaBH4溶液中(例如浸入0.01M溶液中30min)。
可执行第二点所述的步骤(ⅰ)、(ⅱ)和(ⅲ)中的任意一个或多个(即,淀积和聚合、复合和/或还原步骤)以控制嵌入聚合物基质中的无机物纳米颗粒的量。例如,可执行步骤(ⅰ)通过改变聚合物的厚度控制纳米颗粒的量(例如,通过控制射频辉光放电(rfgd)等离子体淀积和聚合的持续时间),可执行步骤(ⅱ)通过改变聚合物基质在无机物中的浸入持续时间控制纳米颗粒的量,可执行步骤(ⅲ)通过改变聚合物基质在还原剂中的浸入持续时间控制纳米颗粒的量。
优选的,第二点所述的方法不包括任何基质表面的预先改性处理。
第二点所述的方法使得可将一膜涂覆到合适的基质表面,其中,该膜的所述聚合物基质中嵌有一种或多种无机物纳米颗粒。该纳米颗粒使膜具有需要的性质,如反射性,或可以用于无机物如抗菌银离子的持续释放。该方法也可以用于所述膜的表面改性或用于附着一另外的基团(例如,在所述表面附着其他的化学反应官能团或另外的膜层或涂层)。
第二点所述的方法适用于制造多层膜,其中由步骤(ⅰ)到(ⅲ)制成的膜构成包含第一聚合物基质的第一膜层(优选地有孔),所述第一聚合物基质带有一种或多种官能团以及在所述第一聚合物基质内嵌有一种或多种无机物纳米颗粒,该方法进一步包括步骤:
(ⅳ)将第二膜层涂覆到第一膜层的表面。

(ⅴ)在第二膜层表面涂覆第三膜层。
可以选择用于制造多层膜的合适的等离子体聚合条件(例如用于控制,例如,第二膜层的厚度,或所述无机物从第一聚合物基质的纳米颗粒中的任意外扩散速率)以适合特定多层膜以及预期应用的要求。
下面将通过非限制性实例及相关附图对本发明进行进一步描述。

材料和方法
材料
如制造商提供的使用HA、硝酸银、氢硼酸钠和玻璃基质((Sigma-Aldrich,圣路易斯,MO,美国)。使用Staphylococcus epidermidis菌株ATCC 35984进行抑菌研究,该菌株在根据制造商的说明配制的胰蛋白胨大豆肉汤(TSB)与0.25%的葡萄糖中37℃培养过夜而制备。抑菌研究后使用BacLight™细菌活力试剂盒(Invitrogen公司,加拿大)观察活和/或死细菌和/或菌落并采用荧光显微镜(Olympus BX 40,激发滤色器BP460-490,二色分光镜DM505,滤片BA515-IF)对目标放大40倍后成像。所有清洗过程采用超纯水进行(电阻率18.2欧姆)。
等离子体聚合法
等离子体聚合在Griesser,HJ(1989)所述的定做的反应器中进行,这里文献作为参考引入。简而言之,反应室包括Pyrex圆筒(Pyrex
cylinder)和两个PVC端板。每个端板中的环形槽固定一O环形密封圈,其正对上述玻璃圆筒(glass cylinder)的抛光末端。顶盖承载驱动马达和齿轮(gear),底板承载所有反应器内的部件。所述部件包括由聚四氟乙烯(Teflon)或有机玻璃(Perspex)制成的实心支架(block)。两个中心面内的支架固定铜电极,其尺寸为18mm×90mm并且间隔16mm。气体进口和出口以及电气穿通连接(feedthrough)由O形环密封。为了拆卸,顶部端部板和玻璃圆筒可以是悬空的。所述射频(rf)功率(power)由嵌入支架中并通过快插接头(push-in fitting)穿通连接的黄铜导体提供。除了反应器,rf发生器和玻璃真空管是标准设计的,包括冷阱,都是标准商业产品。
所有例子中采用13.56MHz的等离子体发生器在0.2Torr的压力下进行淀积。通过衡量某种功率下的淀积速率调节淀积时间从而获得所需的膜厚度,从前面的研究已知所述淀积速率为等离子体每持续1秒厚度为1nm。玻璃基质首先采用Piranha溶液清洗,充分地水洗并干燥。等离子体淀积前,使用40W功率将基质用氧等离子清洗40秒。
银负载方法
通过将等离子体聚合物膜浸入0.02M的AgNO3溶液中进行银负载,标准负载时间为1hr,或者在负载时间研究中为0到120min之间。
银还原方法
通过将负载银的等离子体聚合物膜浸入0.01M的NaBH4溶液中进行银还原,标准还原时间为30min,或者在还原时间研究中在0到60min之间。
UV-
可见光谱
采用Carry 5 UV-可见光谱计(Varian Australia Pty Ltd,墨尔本,澳大利亚)观察并记录UV-可见光谱。
细菌抑制研究
进行细菌抑制研究前,采用BacLight™细菌活力测定法对细菌存储培养物(bacterial stock culture)进行定量,以对活细菌进行定量。每个抗菌测试载玻片放在12微孔板的孔中(一次性细胞培养,Nunclon™表面,丹麦)并固定其中。载玻片接种107CFU/ml(近似200μl)来自肉汤培养基的Staphylococcus
epidermidis 菌株ATTCC
35984。所述板在37℃下温育4hr以使细菌菌落形成。载玻片从12-孔板中移出,并用2ml PBS(8 g/L NaCl,0.2 g/L KC1,1.44 g/L Na2HPO4
以及0.24
g KH2PO4,使用HCl 调整至pH为7.4)清洗2次。将200μl包含10%甲醛的盐溶液(0.9% w/v NaCl溶液)的固定剂滴加在每个载玻片上,并且将细菌菌落固定在载玻片表面10分钟。移除固定剂并将200μl BacLight™染色剂在暗房中滴加在每个载玻片上并以大约10次/分钟轻轻摇动15分钟。然后将染色的载玻片用盐溶液冲洗,并准备显影,将其放在玻璃载玻片表面上,通过放置油润湿载玻片表面并盖上盖玻片。载玻片通过荧光显微镜(Olympus BX 40)Z显影。

负载银纳米颗粒的
HA
等离子体膜的制造
如图1(a)所示,使用HA作为前体以生成薄的富含胺官能团的等离子体聚合物膜。开始使用定做的反应器(上面描述的)将HA淀积在干净的玻璃表面。薄等离子体聚合物膜的原子力成像显示适当地制备的膜包含纳米孔结构(Vasilev
et al,2008)。
如图1(b)所示,通过将所述膜浸入AgNO3溶液中1hr使溶液中的银离子(Ag+)与HA胺基团复合,从而将银离子嵌入所述膜中。但是不希望被原理限制,可以相信上述复合根据下述反应进行:
如图1(c)所示,通过将薄的等离子体膜浸入NaBH4溶液中将银离子还原为银纳米颗粒。该过程产生包含嵌入其中的银纳米颗粒的薄膜聚合物表面涂层。
图2(a)显示了淀积过程中玻璃基质的光学性能。淀积100nmHA等离子体膜后,玻璃基质仍然是透明的,并保持初始的光学性能。再次,浸入AgNO3溶液中(即膜上负载银离子)后,没有观察到可见的玻璃基质的光学性质改变。不过,通过将基质浸入NaBH4溶液中银离子还原成银纳米颗粒后,玻璃基质的颜色变成褐黄色。表面光学性质的改变表示嵌入膜中的银纳米颗粒的存在。
图2(b)显示了只负载了银离子的玻璃基质的吸收光谱图,与银离子还原成银纳米颗粒的吸收光谱图的对比,吸收光谱从UV光到可见光光谱波长。波长420nm处的显著吸收峰仅出现在还原后,表明嵌入膜中的银纳米颗粒的等离子体共振波长。
纳米颗粒负载浓度的调节
已经进行几项研究以确定最有效控制嵌入膜中的银纳米颗粒浓度的方法,结果表明抗菌等离子体膜可以制成多种抗菌剂量。开始,进行了一系列研究以了解改变等离子体淀积条件或抗菌负载条件对嵌入膜中的银纳米颗粒的浓度的影响。测量了在40W的淀积功率下形成的HA等离子体膜在波长约410-420nm处的等离子体共振强度,其中进行的调节有:

膜厚度。
(a)Ag+负载时间
浸入AgNO3溶液中的时间对HA等离子体膜中的银纳米颗粒的最终浓度的影响基于膜的最大吸收强度确定。除了浸入AgNO3溶液中的时间,其他所有条件保持不变。如图3(a)所示,在浸入AgNO3溶液中的120min的时间内吸收强度持续增加,吸光度最大的增量出现在开始的0-40min内。浸入60min后,吸光度持续增加,不过,速率较低。
这些结果表明HA等离子体膜浸入AgNO3溶液中的时间的增加可以促进HA胺官能团对银离子的获得,而这反过来,可以增加银离子还原后形成的银纳米颗粒的浓度。
(b)还原时间
与上面相同,还原时间对HA等离子体聚合物内形成的银纳米颗粒的最终浓度的影响通过观察膜的最大吸光度变化来评估。再次,等离子体聚合条件保持不变,在NaBH4溶液中的还原时间以规则的间隔监测60min。如图3(a)所示,开始的10min内,银纳米颗粒的形成快速进行,30min后银纳米颗粒的总浓度几乎加倍。从30min到60min,尽管观察到的纳米颗粒的形成持续增加,还原速率减缓了。
另外,膜浸入NaBH4溶液中较长时间(即超过30min)后观察到最大吸收波长改变了,因而观察到波长稍微移向更长的波长,约424nm。这种等离子体共振峰的转移,一般出现在较长的还原时间时,申请人怀疑这种情况可以解释为嵌入HA等离子体聚合膜中的银离子用于已有的纳米颗粒的持续生长,而最初的30min里,出现了成核现象,并且颗粒成长到了某种对于所有颗粒都正常的尺寸。
(c)膜厚度
膜厚度对嵌入HA聚合物膜中的银纳米颗粒的浓度的影响通过增加淀积时间来研究,淀积时间增加反过来会产生较厚的膜。如图3(c)所示,观察到一种相互关联性,据此,膜厚度增加到100nm,嵌入的银纳米颗粒的总浓度快速增加。然而,膜厚度在约100nm到200nm之间时,银纳米颗粒的浓度保持不变,随后,银纳米颗粒的浓度逐渐降低。
这些结果表明增加淀积时间会导致HA等离子体膜的化学及表面性质的逐步改变。申请人怀疑增加淀积时间导致官能团的交联增加,因此,减少了可用来形成银离子复合物的官能团的数量,并且另外减少了可用来负载银纳米颗粒的孔的数量。
抗菌释放速率的调节
除了控制嵌入膜中的银纳米颗粒的浓度,也研究了膜中银纳米颗粒的截留特性的改变,从而提供了各种抗菌释放速率的膜(即从即时释放速率到持续释放速率)。
监测了水溶液中HA-银等离子体膜的银释放速率。如图4(a)所示(由正方形表示),银纳米颗粒在水***中经过氧化,这样它们溶解同时银离子洗脱出来。银离子释放速率的控制能够使膜具有延长的或持续的抗菌银释放。因此,为了降低银从HA-银等离子体膜中释放的速率,HA-银等离子体膜的表面淀积了第二薄层HA,如图4(b)所示。

通过监测银离子在PBS中21天期间的洗脱,研究了涂覆有附加6nm或12nmHA等离子体膜的HA-银等离子体膜中银的释放情况。通过在420nm处等离子体共振峰的强度改变来观察银离子从双层膜中的洗脱。图4(a)显示了附加6nm(由圆形表示)或12nm(由三角形表示)HA等离子体膜的淀积,与没有附加HA等离子体膜的膜(由正方形表示)比较,降低的银离子释放速率,。
附加的等离子体膜的厚度增加到12nm(由三角形表示)表现出银离子释放速率的进一步降低,超过了包括6nm膜的膜(由圆形表示)。显然,银离子从HA-银等离子体膜中释放的速率能够通过涂覆附加的表面等离子体涂层有效地控制,据此银离子的释放速率能够通过涂覆更厚的表面涂层降低。
HA-
银等离子体膜的抗菌性能
为评估HA-银等离子体膜抗菌性能,HA等离子体膜、HA-银等离子体膜和涂覆了6nmHA膜的HA-银等离子体膜进行了上述的细菌抑制实验。
图5(a)显示了细菌容易吸附在不含银纳米颗粒的HA等离子体膜上,并且4hr内开始在表面形成菌落(colonise)。然而,相同的膜嵌入了银纳米颗粒时,较少的细菌能吸附到其表面上,并且极少的细菌菌落形成能被观察到。另外,在HA-银等离子体膜中观察到了浅红色菌落的出现,其表示细菌死亡(BacLightTM染色后染成红色/绿色)。图5(c)显示了通过在HA-银等离子体膜表面涂覆附加的HA等离子体膜形成的缓释膜中细菌的附着及生长。经过延长的6小时温育期后,仅观察到少量单细菌粘附在表面上,出现了107CFU/ml的死亡。另外,未观察到细菌菌落形成的迹象。
HA-银等离子体膜清楚地表现出了至少一种超过不含银纳米颗粒的膜的抑菌活性,并且同样的表现出杀菌性能。不过某些细菌能够在HA-银等离子体膜上观察到,涂覆了附加HA等离子体膜的缓释膜表现出了延长的杀菌性能,极大地减少了涂层表面的细菌数量。
据此描述的方法提供了一种通过能够淀积在实际中的任意类型的固体表面上的rfgd等离子体聚合法制造抗菌涂层的高效方法。另外,负载至等离子体膜中的银纳米颗粒的浓度以及抗菌银离子的释放速率能够通过改变淀积时间、银负载时间、还原时间来控制,或通过淀积涂覆了HA等离子体膜的附加表面来控制。

嵌有铜纳米颗粒的等离子体聚合物膜的制造
材料和方法
该种制造上述HA-银等离子体膜的方法可以容易的用于形成包括其他金属纳米颗粒的HA等离子体膜。Cu+和Se是已知的抗菌剂,因此,铜和/或硒纳米颗粒可以用于代替HA等离子体膜中的银纳米颗粒。
上述方法经过合适的改进后可以重复使用,即改变“银负载方法”以向HA等离子体膜中负载Cu+而不是Ag+。特别地,“银负载方法”中0.02M
AgNO3替换为0.02M CuNO3。可以想到尽管淀积步骤或还原步骤只需要进行小小的改变,CuNO3的浓度和铜负载时间还需要优化。
Cu+负载和释放速率将被评估并优化,HA-铜等离子体膜的抗菌活性可以如上述确定。
实例

带有银纳米颗粒的抗菌薄等离子体聚合物膜抗革兰氏阴性细菌的效力
实例1中,发现嵌有银纳米颗粒的抗菌薄等离子体聚合物膜在抑制代表性的***,即所谓的St aphylococcus
epidermidis菌株ATTCC
35984,很有效果。该实例中,研究评估了这种膜抗革兰氏阴性细菌的效果,这种细菌通常较难以抑制。
材料和方法
细菌抑制研究
进行细菌抑制研究前,采用BacLight™细菌活力检测法对Escherichia coli K12 (MG1655)菌株的存储培养物进行定量,定量其中的活细菌。根据上述实例1制备(有或没有HA重叠层膜(overlayer film)),玻璃基质上的负载了银纳米颗粒的HA等离子体膜的正方形样本(1cm2)放置在Petri培养皿中,膜侧朝下,培养皿中装有固体LB介质(富含介质)和一圆形玻璃盖片作为对照。随后,将100μl的存储培养物接种至固体介质上。各测试样本如下:
样本1 仅为HA等离子体膜
样本2 HA-银等离子体膜
样本3 包括HA-银等离子体膜和6nm HA叠层的双层膜
样本4 包括HA-银等离子体膜和12nm HA叠层的双层膜
样本5 包括HA-银等离子体膜和18nm HA叠层的双层膜
结果与讨论
包含HA-银等离子体膜的样本的Petri培养皿在正方形的银负载膜的样本四周显示出清楚的细菌抑制区域。使用光学显微镜,估测出抑菌面积。结果显示在图6中。明显地,所有负载银的样本都有效地抑制了细菌生长。对于双层样本,厚度超过12nm的HA等离子体叠层(用于降低银释放速率),抑菌面积减少了约50%。

嵌有银纳米颗粒的抗菌薄烯丙胺等离子体聚合物膜
材料和方法
材料
如制造商所提供地使用烯丙胺(AA)、硝酸银、氢硼酸钠和玻璃基质((Sigma-Aldrich,圣路易斯,MO,美国)。使用Staphylococcus epidermidis菌株ATCC 35984进行抑菌研究,菌株在根据制造商的说明配制的胰蛋白胨大豆肉汤(TSB)和0.25%的葡萄糖中于37℃下培养过夜制得。活和/或死细菌和/或菌落采用荧光显微镜观察。所有清洗过程采用超纯水进行(电阻率18.2欧姆)。
等离子体聚合法
等离子体聚合在定做的反应器(Griesser,HJ,1989)中进行,如实例1简要描述的。采用13.56MHz的等离子体发生器在0.2Torr的压力下进行淀积。通过衡量某种功率下的淀积速率来调节淀积时间从而获得所需的膜厚度(例如18nm),从前面的研究已知所述淀积速率为等离子体每持续1秒厚度增加1nm。玻璃基质首先采用Piranha溶液清洗,充分地水洗并干燥。在等离子体淀积前,使用40W功率将基质用氧等离子清洗40秒。在某些例子中,银负载及还原后,涂覆一AA叠层以制成一双层膜。所述叠层的涂覆厚度为6nm、12nm或18nm。
银负载方法
通过将等离子体聚合物膜浸入0.02M的AgNO3溶液中进行银负载,标准负载时间为1hr。
银还原方法
通过将负载银的等离子体聚合物膜浸入0.01M的NaBH4溶液中进行银还原,标准还原时间为30min。
UV-
可见光谱
采用Carry 5 UV-可见光谱计(Varian Australia Pty Ltd,墨尔本,澳大利亚)检测并记录UV-可见光谱。
细菌抑制研究
进行细菌抑制研究前,采用BacLight™细菌活力检测法对S. epidermidis菌株ATTCC 35984的存储培养物进行定量,定量其中的活细菌。每个抗菌测试载玻片放在12微孔板的孔中(一次性细胞培养,Nunclon™表面,丹麦)并且固定其中。载玻片接种107CFU/ml(近似200μl)来自存储培养物的S. epidermidis菌株。所述板在37℃下温育4hr以使细菌菌落形成。载玻片从12-孔板中移出,并如实例1中所述的进行准备,采用荧光显微镜进行细菌显影。
结果与讨论
负载银纳米颗粒的
AA
等离子体膜的制造
使用AA作为前体,采用定做的反应器在干净的玻璃表面生成薄的富含胺官能团的等离子体聚合物膜。通过将所述膜浸入AgNO3溶液中1hr,将银离子(Ag+)与溶液中的AA胺基团复合使银离子嵌入所述膜中。随后,通过将薄等离子体膜浸入NaBH4溶液中将银离子还原成银纳米颗粒。图7显示了负载有银纳米颗粒的膜的吸收光谱图(325nm到700nm);420nm处的吸收峰证明了嵌入膜中的银纳米颗粒的等离子体共振波长。
使用多层膜控制释放速率
监测银从在水溶液中的AA-银等离子体膜中释放的速率。为了降低银释放速率,第二薄层AA(6、12或18nm)淀积在AA-银等离子体膜表面。通过监测420nm处等离子体共振峰的强度改变来监测银离子在PBS中21天时间段内的洗脱。图8显示了附加的6nm(由圆形表示)或12nm(由指向上方的三角形表示)AA等离子体膜的淀积,对比未附加AA等离子体膜的膜(由正方形表示)银离子的释放速率降低。另外,附加的等离子体膜的厚度增加到18nm时(由指向下方的三角形表示),表现出进一步降低的银离子释放速率,膜上包括12nm的膜(由向上方的三角形表示)。
HA-
银等离子体膜的抗菌性能
为评估AA-银等离子体膜的抗菌性能,将AA等离子体膜、AA-银等离子体膜和涂覆了6nm AA膜的AA-银等离子体膜进行了上述的细菌抑制测试。
采用AA等离子体膜观察到的结果与实例1中所述的HA膜的结果类似。也就是说,发现细菌容易吸附在不含银纳米颗粒的AA等离子体膜上,并且在若干小时内开始在表面形成菌落。相反,当相同的膜嵌入了银纳米颗粒时,较少的细菌能吸附到其表面上,并且只能观察到极少的菌落形成。另外,AA-银等离子体膜表面包括附加的AA等离子体叠层膜,此时仅观察到少量单细菌粘附在表面上(经过延长的6小时温育期),并且未观察到菌落形成的迹象。

配体固定化到嵌有银纳米颗粒的抗菌薄
HA
等离子体聚合物膜上
本发明的抗菌膜具有一种或多种化学反应官能团,其能够提供机会以提供附加的功能特性和/或改进表面性能。为了证明这一点,在本实例中,将聚乙二醇(PEG)固定化到具有胺基的HA-银等离子体膜的表面,该膜根据实例1中所述的方法通过还原性胺化作用制得。该反应的发生仅仅是由于存在表面胺基,其能与PEG-乙醛的醛末端基团共价反应。
材料和方法
在60℃下和PBS中反应12小时将PEG“接枝”到HA-银等离子体膜表面。根据标准方法对产品进行XPS分析和检测光谱分析(survey
spectrum analysis)。
结果与讨论
在PEG固定化(“PEG接枝”)前、后进行HA-银等离子体膜的XPS光谱分析。PEG接枝前,从涂层的XPS光谱中可以清楚地鉴别出四种元素,即碳、氮、氧(来自等离子体聚合物膜)和银(来自银纳米颗粒)。高分辨率XPS光谱的C1s峰表明表面碳的主要形态是脂肪族的(C-C、C-H键)。相反,PEG接枝后,由于接枝的PEG贡献C-O键,表面的化学组分中的主要峰出现在XPS光谱的C1s区。
检测光谱中也出现了这种改变。由于PEG叠层的存在,银和氮峰的强度降低。由于PEG中存在C-O,氧含量也增加了。
另外,为了表明PEG接枝后银纳米颗粒仍留在膜层中,进行了UV-可见光谱分析(参见图9)。该图表明尽管PEG接枝中包含的反应毫无疑问地会导致银纳米颗粒的氧化,但是由于银纳米颗粒的存在,PEG接枝后,仍然存在等离子体共振吸收。不过,稍稍向较短的波长偏移以及强度的微小降低表明氧化过程已经发生了,但程度有限。
本实例的结果表明银纳米颗粒负载到HA等离子体聚合物膜上后,聚合物基质的表面仍能得到足够数量的胺官能团,并且能够用于固定化各种配体,如化合物以及生物分子(蛋白质、肽等)。

Claims (20)

6.根据权利要求1-5中的任一项所述的膜,其中无机物是基本纯态的非碳元素。
7.根据权利要求6所述的膜,其中非碳元素是铜、银、硒或它们的组合。
8.根据权利要求1-6中的任一项所述的膜,其中聚合物基质由选自烷基胺组成的组中的单体形成。
9.根据权利要求8所述的膜,其中单体是正庚胺、烯丙胺或它们的组合。
10.根据权利要求1-9中的任一项所述的膜,其中所述膜是多层膜,其包括第一和第二聚合物膜层,所述第一膜层包含一聚合物基质,该聚合物基质带有一种或多种官能团以及在所述聚合物基质内嵌有一种或多种无机物纳米颗粒;并且其中所述第二膜层为所述膜提供了附加的功能性特性和/或改进了第一膜层的性能。
11.根据权利要求10所述的膜,其中第二膜层改进了无机物从第一膜层中渗透出的渗透性,从而使得多层膜以持续的方式释放所述无机物。
12.根据权利要求10或11所述的膜,其中所述膜包括至少一第三膜层,其包含有用的配体。
13.根据权利要求10或11所述的膜,其中第二膜层的厚度从约5到约25nm。
14.根据权利要求1-13中的任一项所述的膜,其中所述膜作为涂层应用于合适的基质表面。
15.一种用于将一膜涂覆到合适的基质上的方法,所述方法包括步骤:
(i) 通过等离子体聚合法淀积并聚合包含一种或多种官能团的单体,以在所述表面形成具有渗透性的聚合物基质;
(ii)
将无机物与所述官能团复合;以及
(iii) 还原所述无机物,以形成一种或多种所述无机物的纳米颗粒;
其中所述一种或多种无机物纳米颗粒嵌入所述聚合物基质中。
16.根据权利要求15所述的方法,其中聚合物基质是多孔聚合物基质。
17.根据权利要求15或16所述的方法,其中淀积和聚合单体的步骤采用射频辉光放电(rfgd)等离子体聚合法进行。
18.根据权利要求15-17中的任一项所述的方法,其中单体是正庚胺、烯丙胺或它们的组合。
19.根据权利要求15-18中的任一项所述的方法,其中由步骤(ⅰ)到(ⅲ)制成的膜构成包含第一聚合物基质的第一膜层,所述第一聚合物基质带有一种或多种官能团以及在所述聚合物基质内嵌有一种或多种无机物纳米颗粒,该方法进一步包括步骤:
(iv) 将第二膜层涂覆到第一膜层的表面,从而产生多层膜。
20.根据权利要求19所述的方法,其中该方法进一步包括步骤:
(v)
在第二膜层表面涂覆第三膜层。
