背景技术
癌症是威胁人类健康的主要疾病之一,现有癌症治疗药物最大的局限在于缺乏选择性,在清除癌细胞的同时也会杀伤正常细胞。利用肿瘤细胞特异性识别分子为载体,携带有细胞毒性的药物(放、化疗药物,蛋白/肽类毒素等),是实现肿瘤靶向治疗的重要途径之一。研究证明,能特异识别肿瘤细胞表面抗原的抗体及其功能片段,能特异识别肿瘤细胞表面受体的细胞因子等是良好的药物载体(Kreitman RJ The AAPS Journal.2006;8:E532-551;Pastan I,et al Annu Rev Med.2007;58:221-37)
免疫毒素对肿瘤细胞的结合具有高亲和力和高特异性。但其分子量大,细胞穿透能力弱尤其不易进入瘤体而治疗效果欠佳(Pastan I,et al Annu RevMed.2007;58:221-37;Foerg C,et al J Pharm Sci.2008;97:144-162)。肿瘤导向肽(Tumor-homing peptide)是一类能与肿瘤细胞、肿瘤内***细胞或新生血管内皮细胞特异、高效结合的小分子肽(Laakkonen P,et al AnnN Y Acad Sci.2008;1131:37-43;Corti A,et al Blood.2008;112:2628-2635;Zwanziger D et al Curr Pharm Des.2008;14:2385-2400;Khan IU et al Anticancer Agents Med Chem.2008;8:186-199)。它们不但能高效、快速定位到肿瘤细胞,部分肽还能经癌细胞内吞进入肿瘤细胞(Ruoslahti E et al Curr Pharm Des.2005;11:3655-3660;Enback J etal Biochemical Society Transactions.2007;35:780-783)。因此,这些肿瘤导向肽被认为是潜在的高效药物载体和肿瘤影像诊断工具(Stangelberger A et al.Eur Urol.2008;53:890-900)。放射性标记的促生长素抑制素类似物(Octreotide;Novartis)已通过美国FDA认证并用于临床肿瘤诊断[Okarvi SM.Cancer Treat Rev.2008;34:13-26],而一大批类似物已进入临床前和临床试验阶段(Ahlskog J,et al QJ Nucl Med Mol Imaging.2006;50:296-309.Pelosi G et al.Q J Nucl MedMol Imaging.2006;50:272-287;Miao Y,et al Crit Rev Oncol Hematol.2008;67:213-228)。
蛙皮素(Bombesin)是一种由14个氨基酸组成的肿瘤导向肽(Anastasi Aet al Experientia.1971;27:166-169)。其特异识别的细胞表面受体包括:促神经介素B受体(BB1),促胃泌素释放肽受体(BB2),蛙皮素受体亚型3(BB3)及亚型4(BB4)(Nagalla SR et al J Biol Chem.1996;271:7731-7737;Tokita K et al J Biol Chem.2001;276:36652-36663.)。这4种受体在多种人体常见肿瘤细胞表面高表达,包括结肠癌,肾癌,卵巢癌,子宫癌,乳腺癌,胰腺癌,胃癌,头颈癌,神经母细胞瘤,食道癌,及小细胞肺癌(Reubi JC,et al Clin Cancer Res.2002;8:1139-1146.Cornelio DBet al.Annals of Oncology.2007;18:1457-1466)。在过去数十年中,蛙皮素及多种同源物与放射性同位素连接后被广泛用于实体瘤成像诊断(MainaT et al Cancer Imaging 2006;6:153-157.De Visser M et al CancerBiother Radiopharm.2008;23(2):137-157)。研究者还将阿霉素(Engel JBet al Clin Cancer Res.2005;11:2408-2415.Stangelberger A,et al.IntJ Cancer.2006;118:222-229)和喜树碱(Moody TW et al J Biol Chem.2004;279:23580-23589.Moody TW,et al.J Pharmacol Exp Ther.2006;318:1265-1272)与蛙皮素同源物连接,用于实体瘤靶向治疗取得一定效果。
近年,一些阳离子肽的抗肿瘤作用受到关注。肿瘤细胞外膜含有3-9%酸性脂类而带负电荷,阳离子肽类可通过所带正电荷结合肿瘤细胞并通过破坏细胞膜而显示杀伤活性(Papo N et al,Cancer Research,2006;66(10):5371-8).。但是,由于肿瘤细胞所带负电荷有限,一些阳离子肽类,倚靠静电反应与细胞膜的结合作用相对较弱而不能产生强烈的杀伤作用。研究发现,细胞内的线粒体膜含有大量的酸性脂类而使整个线粒体带大量负电。在导向分子的引导下,这些本身对细胞膜作用较弱的阳离子肽类一旦进入细胞,可能通过破坏线立体膜释放大量细胞色素C而引起细胞凋亡(Rege K,et al.,CancerRes 2007;67(13):6368-75)。
BMAP27(氨基酸序列:GRFKRFRKKFKKLFKKLSPVIPLLHLG,分子量3283.1Da)和BMAP28(氨基酸序列:GGLRSLGRKILRAWK KYGPIIVPIIRIG,分子量3131Da)是两种来自于牛骨髓细胞的阳离子抗菌肽。对多种细菌和真菌有强烈杀伤作用。
BMAP27和BMAP28完整分子对哺乳动物细胞膜有一定破坏作用。但去除其分子C末端的疏水结构,截短的阳离子肽段(本发明命名为B27和B28,其氨基酸序列分别是GRFKRFRKKFKKLFKKLS和GGLRSLGRKILRAWKKYG)对细胞的杀伤作用大大减弱。但该肽段对细胞线粒体仍有强烈的破坏作用(Skerlavaj B etal,J Biol Chem.1996;271:28375-81;Risso A et al.,Mol Cell Biol.2002;22:1926-1935).。因此,如果能用导向分子将其带入细胞内,其可能诱导细胞凋亡。由于蛙皮素能特异识别肿瘤细胞,用蛙皮素作为导向分子,携带阳离子肽B27和B28,构建的融合肽可能对肿瘤细胞产生选择性的、作用更强烈的杀伤。已有的研究主要利用蛙皮素及其同源物连接放、化疗药物用于实体肿瘤的诊断和治疗。尚无相关文献设计利用蛙皮素及同源物与大分子蛋白/肽构建融合肽,用于肿瘤诊断和治疗的报道。也没有利用蛙皮素及同源物与大分子蛋白/肽构建融合肽用于血液瘤治疗方面的报道。

本发明公开了一种抗肿瘤多肽,它是由蛙皮素与来源于牛骨髓细胞的阳离子抗菌肽连接而成的融合肽,其中,所述阳离子抗菌肽具有破坏线粒体的活性区域,更具体的,所述阳离子抗菌肽是BMAP27或BMAP28,优选的阳离子抗菌肽的氨基酸序列如SEQ ID NO:1或SEQ ID NO:2所示。
进一步的,所述的融合肽氨基酸序列如SEQ ID NO:3或SEQ ID NO:4所示,本发明分别命名为BB27和BB28。
本发明的另一目的是提供一种由所述抗肿瘤多肽在制备抗肿瘤药物中的新的应用。
进一步的,所述抗肿瘤药物为抗实体肿瘤的药物。
更进一步的,所述抗肿瘤药物为治疗血液瘤的药物。
更具体的,所述治疗血液瘤的药物为治疗白血病的药物。
本发明的再一个目的是提供一种药物组合物,它是以上述的抗肿瘤多肽为活性物质,加上药学上可接受的辅料制备而成。
虽然蛙皮素作为肿瘤导向肽可以与多种抗肿瘤物质连接形成具有肿瘤靶向作用的抗肿瘤药物,但是本发明第一次尝试将蛙皮素与阳离子抗菌肽连接,作为一种新的抗肿瘤活性物质,并且发明人通过对蛙皮素与多种阳离子抗菌肽连接的融合肽进行研究后发现,并非所有的阳离子抗菌肽与蛙皮素连接后都有良好的抗肿瘤效果,具体原因尚在探索中。
下面结合具体实审方式对本发明作进一步说明,但是并非对发明的限制。

具体实施方式
实施例一本发明抗肿瘤肽的制备
除特殊说明外,本发明所有细胞株均购自美国ATCC细胞库。实验中使用的细胞株包括:人脐静脉内皮细胞(HUVECs),血管内皮细胞(ECV304),人胚肺成纤维细胞(MRC-5),人***癌细胞(Du145),人乳腺癌细胞(MCF-7),人慢性骨髓原细胞(K562),人Burkitt’s B淋巴瘤细胞(Raji),人急性T淋巴瘤细胞(CEM),人急性T淋巴母细胞白血病细胞(Molt4),人急性白血病早幼粒细胞(NB4);及新鲜分离的人蜕膜基质细胞(HDSCs)。正常人外周血淋巴细胞取自实验室志愿者,急性骨髓细胞样白血病病人的外周血淋巴细胞利用低浓度Ficoll单核细胞分离液,梯度离心分离得到。所有细胞均在添加10%新生小牛血清的DMEM或RPMI1640培养基中,37℃,5%CO2培养箱中培养。
一、测定蛙皮素对实体瘤细胞和血液瘤细胞的选择性识别作用
蛙皮素采用通用的固相化学方法合成。合成产物经反向高效液相色谱HPLC分离纯化获得纯度大于90%的产品(图1A),分子量用时间飞行质谱鉴定(图1B),证明得到的产物为蛙皮素。用荧光染料FITC标记后溶解于磷酸盐缓冲液(PBS:137mM NaCl,2.68mM KCl,8.09mM Na2HPO4,1.76mM KH2PO4,pH 7.4)。取300μl肽溶液(10μM)与2×105个细胞在37℃孵育1小时,然后用PBS洗涤,用流式细胞仪进行分析。用对照肽URP为对照。蛙皮素对细胞的结合量用相对于对照肽的结合倍数表示。如图2所示,蛙皮素对实体瘤细胞,如乳腺癌细胞MCF-7,***癌细胞Du145,肝癌细胞SMMC-7721和***细胞HeLa显示了强烈的结合作用,结合量是对照肽的20-30倍。对血液瘤细胞,如NB4,CEM,K562,Molt4,Jurkate和Raji等细胞的结合量是对照肽的10-30倍。而对正常细胞,如人脐静脉内皮细胞HUVECs,人蜕膜基质细胞HDSCs,人肺成纤维细胞MRC-5,人肝细胞L02,人血管内皮细胞ECV304和人外周血单核细胞hPBMCs,蛙皮素与对照肽的结合相当,均很低。这表明蛙皮素对实体瘤细胞和血液瘤细胞均显示了强烈的结合作用,而对正常细胞不结合或低水平结合。蛙皮素对实体瘤细胞的强烈结合与前人报道的结果一致。但本发明首次发现蛙皮素可以和血液瘤细胞结合。
二、蛙皮素导向的抗肿瘤融合肽的设计及制备
根据蛙皮素对肿瘤细胞的选择性结合及阳离子肽B27、B28对细胞杀伤作用弱的特点,本发明选择Bombesin为载体分子,以阳离子肽为杀伤分子。将阳离子肽连接于载体分子氨基末端,构建抗肿瘤肽。本发明使用的阳离子肽包括B27和B28分别与载体分子连接构建的融合肽分别命名为BB27,BB28。序列见表1。
所有肽均采取通用的固相合成化学法合成,用反向高效液相色谱法进行分纯化,纯度大于90%。用时间飞行质谱检测肽分子量。BB27和BB28的纯度和分子量鉴定结果如图3,图4所示。多肽使用前用PBS缓冲液溶解,-80℃冷冻保存。

实施例二BB28的体外抗肿瘤实验
用实施例一得到的融合肽,配制成2.5、5、7.5、10、12.5、15μM的肽溶液,进行以下实验:
1)细胞毒性测试方法
贴壁细胞按10000细胞/孔接种于96孔板,培养过夜,使其贴壁。次日,弃上清培养基,替换为100μl含2%牛血清白蛋白(BSA)的培养基,加入不同浓度的肽进行反应。非贴壁细胞悬浮于2%BSA的培养基,按10000细胞/孔接入96孔板后,直接加入不同浓度肽。肽作用细胞后,还可以用CCK-8试剂盒,通过检测细胞线粒体酶活性定量测定存活细胞数量。具体操作为,细胞经肽处理后,于细胞孔中加入10μl CCK-8溶液,避光孵育4h,测定490nm下光吸收值。对照孔中加入相同体积的PBS溶液。对照组细胞存活率计为100%。
2)与蛙皮素共价连接对阳离子肽B28细胞毒性的增强作用
对蛙皮素Bombesin、B28及两者形成的融合肽BB28细胞毒性检测发现,高浓度(15μM)的Bombesin及B28对乳腺癌细胞MCF-7无明显细胞毒性,但2.5μM BB28即杀伤40%左右的细胞,5μM BB28的杀伤率超过70%(图5)。高浓度(10-15μM)B28对血液瘤细胞CEM对仅显示10-30%杀伤率。而Bombesin与B28连接形成的融合肽BB28在5μM时杀伤率达50%,7.5μM时杀伤率超过70%(图6)。利用其它肿瘤细胞进行检测也得到相同的结果。这些结果表明与蛙皮素Bombesin连接可以显著增强B28的细胞毒性。
3)蛙皮素导向的抗肿瘤肽BB28细胞杀伤选择性
进一步比较BB28对肿瘤细胞和正常细胞的杀伤活性发现,BB28在低浓度(2.5μM)对实质瘤细胞(MCF7、Du145、SMMC7721和HeLa)和血液瘤细胞(Raji,NB4,CEM,K562,Molt4和Jurkat)就产生细胞毒性。5μM BB28即对多种肿瘤细胞的杀伤率超过70%。而即便在测试的最高浓度(15μM),BB28对正常细胞hPBMC,HDSCs,HUVECs和MRC5的杀伤率均<10%(图7)。这些结果证明BB28能选择性地识别和杀伤肿瘤细胞。
4)、抗肿瘤肽BB28对肿瘤细胞的杀伤机制
(1)Annexin V/PI法监测细胞凋亡
Annexin V可和细胞膜上的磷脂酰丝氨酸结合。正常细胞的磷脂酰丝氨酸位于细胞膜内侧,Annexin V不能进入胞内而检测不到。细胞凋亡早期,虽然细胞膜还没有破,但磷脂酰丝氨酸已外翻而暴露在膜外侧,用FITC-AnnexinV即可显示。PI可与DNA结合,但其只能进入细胞膜已经破坏的细胞。因此,细胞经肽处理后,用Annexin V-PI同时染色,荧光显微镜或流式细胞仪检查,Annexin V+/PI-的细胞为凋亡早期细胞,Annexin V+/PI+细胞为凋亡晚期细胞。由此可确定多肽是否诱导细胞凋亡,并确定凋亡细胞数量。将200000个细胞悬浮于300μl液体中,经不同浓度的肽处理后用流式细胞术检测。
结果发现,血液瘤细胞CEM经0,5,10,20μM BB28处理后,早期凋亡和晚期凋亡的细胞百分比分别为:5.8/2.1;9.2/7.8;15.1/25.3;11.2/53(图8A)。表明随着肽浓度增高,凋亡细胞数量逐渐增多。
(2)线粒体膜电位变化监测细胞凋亡
线粒体膜电位的变化是细胞凋亡的重要指标。JC-1染料在正常线粒体膜上形成多聚体,显示红色荧光;当线粒体膜遭到破坏,膜电位丧失后,JC-1单体则游离于胞质内显示绿色荧光。因此,利用JC-1染色后,可通过监测细胞红色和绿色荧光比例的变化反映线粒体膜受损状况。将200000个细胞用300μl不同浓度肽处理后,用2μM JC-1染料于37℃避光染色30min,PBS洗涤2次后用流式细胞术分析。
结果如图8B所示,血液瘤细胞CEM经0,10和20μM BB28处理,JC-1染色后红色/绿色荧光比值分别为86.6/13.4;56.5/43.5;4.1/95.9。红色荧光细胞逐渐减少,绿色荧光细胞逐渐增多,表明BB28作用于CEM细胞后,显著地破坏了其线粒体膜。
(3)细胞色素C释放法监测细胞凋亡
细胞线粒体膜被破坏后,大量细胞色素C从线粒体中释放出来,从而诱导细胞凋亡。500000个细胞经50μl肽于37℃处理2h后,4℃,10000g离心20min,去除未破坏的线粒体后,小心收集上清。上清中的蛋白经15%SDS-PAGE电泳分离后,转移到PVDF膜上,再用小鼠抗人细胞色素C抗体通过Western Blot检测。用50μl裂解缓冲液(50mM Tris-HCl,pH7.6,2%CHAPS,2M Urea)裂解对照细胞,获取细胞色素C(Cc)作为标准对照。
结果如图8C所示,0,12.5,25,50μM BB28处理细胞后,从线粒体释放的细胞色素C随着肽浓度的增加而增多,进一步反映了肽对细胞线粒体膜的破坏。
综合细胞膜磷脂酰丝氨酸外翻、线粒体膜电位变化、细胞色素C的释放的检测结果,本发明构建的BB28通过线粒体依赖途径诱导肿瘤细胞凋亡。
5)抗肿瘤肽BB28对病人来源的白血病细胞的杀伤作用
从4名急性白血病人血液中分离白血病细胞,按20000细胞/孔接种入96孔板,加入不同浓度的肽处理2h后,利用CCK-8试剂测定细胞存活率。
结果发现,BB28对新鲜分离的白血病细胞显示剂量依赖的细胞毒作用(图9A)。相同浓度的BB28对不同病人来源的白血病细胞作用强度略微有差异。其中,20μM BB28能杀伤30-60%的白血病细胞。来自于1号和2号病人的白血病细胞经BB28作用后,用Annexin V和PI双染,通过流式细胞术检测细胞凋亡率。结果发现,来自于1号病人的白血病细胞经20μM BB28处理后,凋亡早期和晚期细胞分别为2.9%和54%。来自于2号病人的白血病细胞经20μMBB28处理后,凋亡早期和晚期细胞分别为6.9%和74.5%。这一结果进一步证实了BB28对来自于病人的白血病细胞具有杀伤作用(图9B)。
实施例三BB28体内抗肿瘤实验
将K562细胞(1×107)悬于100μl生理盐水,经皮下注射入6-8周龄裸鼠体内,建立肿瘤模型。
1)腹腔注射BB28的抗肿瘤效果
当瘤体长到0.03-0.05mm3时,实验动物8只随机分为2组。实验组动物经腹腔注射BB28,浓度为20mg/kg,连续5天。对照组给相同体积的磷酸盐缓冲液。每天用游标卡尺测定瘤体直径,按照公式(长x宽x宽x0.5)计算瘤体体积。实验结束后,将实验动物处死,取出瘤组织,拍照及称重。
结果如图10A所示,腹腔注射BB28能显著抑制肿瘤生长。从第15天开始,实验组和对照组肿瘤体积大小具有显著性差异并持续到实验结束。在实验结束时,对照组肿瘤体积为2.772±0.35cm3。实验组肿瘤体积大小为0.7±0.3cm3。将肿瘤组织从动物身上剥离,称重,对照组和实验组瘤体重量分别为2.68±0.53g和0.81±0.45g。实验组和对照组瘤体重量有显著性差异(P<0.05)。表明腹腔注射20mg/kg BB28具有抗肿瘤效果。
2)瘤内注射BB28的抗肿瘤效果
当瘤体长到0.1cm3时,实验动物8只随机分为2组。实验组动物瘤体内注射10mg/kg BB28,连续5天。对照组注射相同体积的磷酸盐缓冲液。每天用游标卡尺测定瘤体直径,按照公式(长x宽x宽x0.5)计算瘤体体积。实验结束后,将实验动物处死,取出瘤组织,拍照及称重。
结果如图10B所示,实验组停药一周后瘤体无明显生长。从第15天开始,实验组和对照组瘤体大小出现显著性差异。实验结束时,对照组瘤体大小为3.004±0.384cm3,实验组瘤体大小为0.168±0.089cm3。对照组和实验组瘤体重量分别为:2.49±0.78g和0.38±0.27g。两组瘤体重量也有显著性差异(P<0.05)。表明瘤内注射的10mg/kg BB28也有明显抗肿瘤效果。
实施例四BB27的体外抗肿瘤实验
1)与蛙皮素共价连接对阳离子肽B27细胞毒性的增强作用
细胞毒性测试方法同实施例二。
结果如图11所示,蛙皮素Bombesin在测试的浓度范围内,对乳腺癌细胞MCF-7均无明显杀伤作用。B27在最高浓度(15μM)时对MCF-7的杀伤率低于20%。但蛙皮素和B27共价连接形成的融合肽BB27在5μM时就导致50%左右的细胞死亡。10μM BB27的杀伤效率达到80%。图12显示,蛙皮素Bombesin在测试的浓度范围内对血液瘤细胞K562也无明显杀伤作用。B27在测试浓度范围内显示微弱的杀伤作用,最高浓度(15μM)杀伤效率大约为30%。但BB27在2.5μM时就对K562显示杀伤。5μM BB27对K562的杀伤效率达到70-80%。利用其它肿瘤细胞检测也得到类似的结果,均发现融合肽BB27的杀伤作用明显强于未连接的阳离子肽B27。这表明与蛙皮素共价连接显著提高了阳离子肽的细胞毒性。
2)BB27对肿瘤细胞的选择性杀伤
比较BB27对肿瘤细胞和正常细胞的杀伤活性发现,BB27在低浓度(2.5μM)对实质瘤细胞(MCF7,Du145,SMMC7721和HeLa)和血液瘤细胞(Raji,NB4,CEM,K562和Molt4)就产生细胞毒性。5μM BB27对肿瘤细胞杀伤率达到50%~85%。BB27对正常细胞(hPBMCs,MRC-5,HDSCs,HUVECs和ECV304)只有微弱的细胞毒作用,高浓度(10-15μM)可导致20-30%细胞死亡。但与肿瘤细胞相比,正常细胞对BB27仍然具有较高的耐受性(图13)。表明BB27对肿瘤细胞的杀伤仍具有一定的选择性。
3)BB27对肿瘤细胞的杀伤机制
BB27作用肿瘤细胞后,通过Annexin V/PI法、线粒体膜电位变化和细胞色素C释放监测细胞凋亡的方法同实施例三。
Annexin V和PI法分析显示,血液瘤细胞Raji经0,5,10,20μM BB27处理,早期凋亡和晚期凋亡的细胞百分比分别为:6.3/3.7;13.9/36.8;19.2/67.6;15.7/75.1(图14A)。表明随着肽浓度增高,凋亡细胞数量逐渐增多。
线粒体膜电位变化监测发现,血液瘤细胞Raji经20μM BB27处理,细胞红色/绿色荧光比值从85.4/14.6下降到21.6/78.4(图14B),表明BB27作用于Raji细胞后,破坏了其线粒体膜。
Western Blot检测发现,0,12.5,25,50μM BB27处理细胞后,从线粒体释放的细胞色素C随着肽浓度的增加而增多(图14C),进一步显示了BB27对细胞线粒体膜的破坏。
综合分析Annexin V/PI、线粒体膜电位变化、细胞色素C的释放的检测结果,可以判断BB27通过线粒体依赖途径诱导肿瘤细胞凋亡。
4)BB27对病人来源的白血病细胞的杀伤作用
分析方法同前。
结果发现,BB27对4名病人来源的白血病细胞有剂量依赖的细胞毒作用(图15A)。20μM BB27能导致30-60%的细胞死亡。来自于1号和2号病人的细胞经20μM BB27作用后,用Annexin V和PI双染,再通过流式细胞术检测细胞凋亡率,发现凋亡早期和晚期细胞比率分别为9.5/30.1和11.4/56.7,与CCK-8法测定结果一致(图15B)。进一步证明BB27可以有效杀伤病人来源的白血病细胞。
实施例五BB27体内抗肿瘤实验
肿瘤模型的建立方法同实施例三。
1)腹腔注射BB27的抗肿瘤效果
实验与BB28平行开展,所有实验条件相同。当瘤体长到0.03-0.05cm3时,实验组动物经腹腔注射BB27,浓度为20mg/kg,连续5天。对照组给相同体积的磷酸盐缓冲液。每天用游标卡尺测定瘤体直径,并按照公式(长x宽x宽x0.5)计算瘤体体积。实验结束后,将实验动物处死,取出瘤组织,拍照及称重。
结果如图16A所示,腹腔注射BB27能显著抑制肿瘤生长。从第15天开始,实验组和对照组肿瘤体积大小具有显著性差异并持续到实验结束。在实验结束时,对照组肿瘤体积为2.772±0.349cm3。实验组肿瘤体积大小为0.464±0.331cm3。将肿瘤组织从动物身上剥离,称重,对照组和实验组瘤体重量分别为2.68±0.53g和0.76±0.29g。实验组和对照组瘤体重量有显著性差异(P<0.05)。表明腹腔注射20mg/kg BB27具有抗肿瘤效果。
2)瘤内注射BB27的抗肿瘤效果
当瘤体长到0.1cm3时,实验组动物瘤体内注射10mg/kg BB27,连续5天。对照组注射相同体积的磷酸盐缓冲液。每天测定瘤体体积。实验结束后,将实验动物处死,取出瘤组织,拍照及称重。
结果如图16B所示,实验组停药一周后瘤体生长缓慢。从第15天开始,实验组和对照组瘤体大小出现显著性差异。实验结束时,对照组瘤体大小为3.004±0.384cm3,实验组瘤体大小为0.315±0.2cm3。对照组和实验组瘤体重量分别为:2.49±0.78g和0.35±0.51g。两组瘤体重量也有显著性差异(P<0.05)。表明瘤内注射的10mg/kg BB27也有明显抗肿瘤效果。
实施例六蛙皮素与其它阳离子抗菌肽连接构建的融合肽的抗肿瘤活性
另用实施例一的方法构建以下融合肽:

上述融合肽BF5GB和MPGB是用阳离子肽BF5(RAGLQFPV GRVHRLLRK)和MP(KKVVFKVKFKK)分别与蛙皮素连接构建而成。BF5肽段来自于阳离子抗菌肽BuforinII(TRSSRAGLQFPVGR VHRLLRK)(Gwan-Su YP,et al,FEBS Letters,1996,398:87-90)。MP(KKVVFKVKFKK)是一种人工设计的阳离子抗菌肽(HongSY,et al.,Antimicrobial Agents and Chemotherapy,1999,43(7):1704-1707)。这两种肽都有强烈的抗菌活性。按照实施例二的方法,利用合成的多肽BF5GB和MPGB进行实验,并与抗肿瘤肽BB27和BB28的进行比较。结果如图17显示,在体外条件下,BF5GB和MPGB对血液瘤细胞K562均未显示明显的肿瘤细胞生长抑制和杀伤作用。而在相同浓度下,BB27和BB28则对K562显示强烈的杀伤作用。用其它瘤细胞进行检测也得到相同的结果。这表明,并不是所有的阳离子抗菌肽与蛙皮素连接后形成的融合肽都有强烈的抗肿瘤作用。
通过上述实验可以证明,本发明的抗肿瘤多肽对实体瘤和血液肿瘤均有明显的靶向抑制作用,提高了药物的安全性,为临床用药提供了一种新途径,具有良好的应用前景。

Claims (7)
1.一种抗肿瘤多肽,其特征是:由蛙皮素与来源于牛骨髓细胞的阳离子抗菌肽连接而成的融合肽,所述阳离子抗菌肽具有破坏线粒体的活性区域;
所述蛙皮素是Bombesin,为含有13个氨基酸的多肽,其氨基酸序列为:QRLGNQWAVGHLM;
所述阳离子抗菌肽是BMAP27或BMAP28,它们的氨基酸序列如SEQ ID NO:1或SEQ ID NO:2所示。
2.根据权利要求1所述的抗肿瘤多肽,其特征是:所述融合肽的氨基酸序列如SEQ ID NO:3或SEQ ID NO:4所示。
3.权利要求1或者2所述的抗肿瘤多肽在制备抗肿瘤药物中的应用。
4.根据权利要求3所述的应用,所述抗肿瘤药物为抗实体肿瘤的药物。
5.根据权利要求3所述的应用,所述抗肿瘤药物为治疗血液瘤的药物。
6.根据权利要求5所述的应用,所述治疗血液瘤的药物为治疗白血病的药物。
7.一种药物组合物,其特征是:以权利要求1或者2所述的抗肿瘤多肽为活性物质,加上药学上可接受的辅料制备而成。
