Abstract

背景技术
维生素受体在癌细胞中被过表达。例如,带有高亲和力(<1nM)结合到叶酸上的叶酸受体,38KD GPI在卵巢、乳腺、支气管和脑肿瘤上过表达。之后的结合受体,快速胞吞作用将叶酸传递到细胞中,在低pH下在内神经元体细胞代谢区不能被除去。重要的是,小分子、蛋白质、甚至脂质体结合到叶酸的共价配合物不能阻挡叶酸结合到它的受体上,因此,叶酸配合物能够容易地被传递并且能够通过受体-传递胞吞作用进入细胞。
由于多数细胞用无关的还原叶酸载体获得必要的叶酸,因此叶酸受体的表达约束了少数细胞类型。除了肾、肝从和胎盘,正常的组织表达叶酸受体的低的或不易察觉的水平。然而,许多恶性组织,包括卵巢、乳腺、支气管和脑肿瘤独特表达受体的提高的水平。事实上,估计95%卵巢恶性肿瘤过表达叶酸受体。
也已经报道了叶酸受体β,叶酸受体的非上皮异构重整,在活性(而不是休眠)滑液巨噬细胞上表达。因此,叶酸受体在巨噬细胞(即,活性巨噬细胞)的子***上表达。活性巨噬细胞可以通过在巨噬细胞中非特定吞噬并消灭外来病原体、通过由其它免疫细胞识别的巨噬细胞表面上的外来蛋白质退化的肽、或者通过隐藏调节T和B淋巴细胞功能的原浆转移或其它因素参与免疫反应,结果是进一步刺激免疫反应。在某些场合中活性巨噬细胞还可以为疾病的病理生理学作出贡献。例如,活性巨噬细胞可以为动脉粥样硬化、风湿、关节炎、自体免疫疾病状态和器官移植排异,以及其他疾病状态作出贡献。也示出了活性巨噬细胞在叶酸受体上过表达。叶酸受体在活性巨噬细胞和活性单核白血球上的过表达在美国专利申请No.60/696,740和美国专利申请公开No.US-2002-0192157-A1中有描述,在此引为参考。
活性巨噬细胞对疾病的贡献的实例包括动脉粥样硬化级数中的活性巨噬细胞。动脉粥样硬化是当脂肪斑纹形成在血管壁中时开始的疾病状态。脂肪斑纹的形成认为是在血管壁的内膜层中脂蛋白颗粒积累形成的结果,内膜层是内腔内皮层下面的血管壁层。脂蛋白颗粒能够与内膜层中的细胞外基质成分结合,并且能够变得难于接近血浆抗氧化剂,导致脂蛋白颗粒的氧化改性。这种氧化改性可以引发局部炎性反应,导致活性巨噬细胞和T淋巴细胞粘附到之后迁移到内膜层的内腔内皮细胞上。氧化的脂蛋白能够对内膜***例如,巨噬细胞和T细胞起到化学引诱剂的作用,或者促使脉管壁中的细胞产生化学引诱剂。然后动脉粥样硬化损伤可以形成带有由活性巨噬细胞填充的富脂芯的纤维帽。不稳定的动脉粥样硬化损伤以局部炎性为特征,破裂的和产生致命心肌梗塞的损伤以活性巨噬细胞和T细胞的渗透为特征。

发明内容

具体实施方式
本发明应用发射辐射(即,radiophore)并用于使用发射正电子段层分析技术(即,能够发射正电子辐射来产生一对向相反方向运动的湮灭质子的化合物,产生的湮灭质子是正电子与电子湮灭的产生的结果)诊断/监测方法中的化合物。radiophore一般包括交联到另一个化学结构(例如,苯环)上形成发射辐射(即,radiophore)化合物的放射同位素。然而,radiophore可以仅包括同位素。
根据本发明化合物(例如,radiophore)“用于发射正电子分析技术中”是指能够发射正电子辐射来产生一对向相反方向运动的湮灭质子的化合物,产生的湮灭质子是正电子与电子湮灭的结果。
而且,根据本发明,正电子分析技术(PET)的应用要求化合物(例如,radiophore)用于能够发射正电子辐射来产生一对向相反方向运动湮灭质子的PET发射正电子辐射中。
连接到用于PET中的radiophore的维生素或其类似物或其他配位体,用来用体外装置诊断和/或监测疾病状态。用在体外装置中的]PET检测也指“PET扫描”,并且用PET的体外检测的装置也是本领域熟知的。
本发明涉及一种用PET诊断/监测致病状态的组合物和方法,其中致病细胞独特表达、优选表达或过表达维生素受体或其他受体。本发明适用于所有产生各种病理例如肿瘤的致病细胞群。因此,致病细胞群可能是致瘤的包括良性瘤和恶性瘤的癌细胞群,或者它可能是非致瘤的。癌细胞可能是自然导致的或者是通过存在于病人germline中的突变或机体突变过程导致的,或者可能是化学-、virally-或辐射诱导的。本发明可以用于诊断/监测肿瘤例如癌、肉瘤、淋巴瘤、Hodgekin’s病、黑瘤、间皮瘤、Burkitt’s肉瘤、鼻咽癌、白血病以及骨髓瘤。癌细胞群包括,但不限于,口腔、甲状腺、内分泌、皮肤、胃、食管、喉、胰、结肠、膀胱、骨、卵巢、宫颈、子宫、乳腺、睾丸、***、直肠、肾、肝和肺癌。
致病细胞还可以是与疾病状态例如纤维肌痛、类风湿性关节炎、骨关节炎、溃疡性结肠炎、克罗恩氏病、牛皮癣、骨髓炎、多发性硬化、动脉粥样硬化、肺纤维化、结核病、***性硬化、器官移植排异(GVHD)、红斑狼疮、sjogren’s综合症、肾小球性肾炎、皮炎(例如,牛皮癣)、慢性炎以及由于损伤导致的发炎(例如,头或脊髓损伤或栓塞)相关的活性单核白血球或巨噬细胞。
在一个实施例中,提供一种诊断/监测对维生素具有易粘附点的活性巨噬细胞或活性单核白血球转达的致病状态的方法。该方法包括对病人执行评估通式L-X配合的有效量的疾病状态、允许足够的时间用于维生素结合并粘结到活性单核白血球或活性巨噬细胞上以及体外使用放射正电子段层分析技术诊断/监测疾病状态的步骤,其中L包括维生素或其类似物,基团X包括radiophore。

这一实施例涉及诊断/监测血管壁上活性动脉粥样斑的方法。在与休眠巨噬细胞相对的活性巨噬细胞表面优选表达、独特表达或过表达的受体粘接的配位体(例如,维生素或其类似物),与radiophore配合。控制配合物到评估动脉粥样硬化的病人。配合物粘接到与活性动脉粥样斑相关的活性巨噬细胞上。通过radiophore发射辐射用发射正电子段层技术体外检测。因此,配合物可以用于区分包括活性巨噬细胞的活性动脉粥样斑和存在于评估动脉粥样硬化的病人的动脉或静脉中的非活性斑。
许多不稳定的(例如,活性)动脉粥样斑能够破裂并导致急性动脉硬化并发症,但是不能产生血管腔的狭窄,特别是冠状循环。因此,这个实施例在诊断心肌梗死的风险和评估需要临床干预、病人患动脉硬化方面体现出重大进步。
根据配合物粘接到活性单核白血球或巨噬细胞上的实施例中,可以由广泛种类的配位体和radiophore形成配合物,其中广泛种类的配位体包括粘接到在活性单核白血球或活性巨噬细胞表面上过表达、独特表达或优选表达的受体上的任何配位体,活性单核白血球或活性巨噬细胞在休眠单核白血球或巨噬细胞表面上不大量表达/存在或不存在。对于活性巨噬细胞,这些配位体包括N-甲酰肽(例如,f-Met-Leu-Phe)、高活性基团因子蛋白(HMGB1)、透明质酸(hyaluronan)片断、HSP-70、toll-like受体配位体、scavenger受体配位体、coreceptors for抗原presentation、粘接到在活性巨噬细胞上标示的CD68、BER-MAC3、RFD7、CD4、CD14和HLA-D上的配位体、粘接到尿激酶纤溶酶原活化剂受体(例如,WX-360肽)、抗体或其片段上、优选粘接到活性巨噬细胞的配位体以及维生素或受体-粘接维生素类似物/衍生物。
对于单核白血球,单核白血球-粘接配位体可以是粘接到在活性单核白血球上表达或过表达受体上的任何配位体,包括CD40-、CD16-、CD14-、CD11b-和CD62-粘接配位体、5-羟基色胺、核因子KB配位体对抗剂的巨噬细胞发炎蛋白1-α,MIP-2,受体活化剂、单核白血球趋化性蛋白1-粘接配位体、化学增活受体5-粘接配位体、RANTES-粘接配位体、化学增活受体-粘接配位体以及维生素或受体-粘接维生素类似物/衍生物等。由于配位体受体在活性单核白血球或巨噬细胞上的优选表达,配合物优选能够粘接到与休眠单核白血球或巨噬细胞相比的活性单核白血球或活性巨噬细胞上。
在上述实施例中,配位体(例如,维生素或其类似物或其衍生物)可以是粘接到在与休眠单核白血球或巨噬细胞相对的癌细胞或活性单核白血球或活性巨噬细胞表面上优选表达、独特表达或过表达的受体上的任何配位体。这种配体的实例是选自由叶酸受体-粘接配位体、辅酶R受体-粘接配位体、维生素B12受体-粘接配位体、维生素B2受体-粘接配位体、维生素B1受体-粘接配位体以及其他维生素受体-粘接配位体或其类似物或衍生物。

在各个实施例中,放射同位素选自由34C1、45Ti、51Mn、61Cu、63Zn、68Ga、11C、13N、15O和18F组成的组中。
在根据本发明的通式L-X的配合物中,基团L是能够粘接到上述的癌细胞或与休眠单核白血球和巨噬细胞相对的活性单核白血球和活性巨噬细胞上的配位体。在一个示例性实施例中,癌细胞或活性单核白血球或活性巨噬细胞粘接配位体是叶酸、叶酸类似物/衍生物,或其他叶酸受体-粘接分子。
对配位体(例如,维生素或类似物或衍生物)的粘接点可以包括用于能够优选粘接到受体上的任何配位体的受体,受体在癌细胞或活性单核白血球或活性巨噬细胞表面独特表达、过表达或优选表达/存在。被癌细胞或活性单核白血球或活性巨噬细胞独特表达、过表达或优选表达的表面-存在蛋白是不存在或不以足够的浓度存在于正常细胞或休眠单核白血球或休眠巨噬细胞上的受体,该受体提供优选的检测癌细胞或活性单核白血球或活性巨噬细胞的方法。因此,在癌细胞或相对于休眠单核白血球或休眠巨噬细胞的活性单核白血球和活性巨噬细胞上被调节,或在正常细胞或休眠白血球或休眠巨噬细胞表面不表达/存在的任何受体,或在癌细胞或休眠单核白血球或休眠巨噬细胞不以足够量表达/存在的任何受体可以作为靶。在一个示例性实施例中,粘接用于本发明的配合物的点是维生素受体,例如,叶酸受体,其粘接叶酸或类似物或衍生物。
在配合物中配位体与radiophore之间的化学交联可以是直接交联也可以是通过中间交联剂的交联。如果存在交联剂,可以是本领域熟知的双相容交联剂。一般地,交联剂包括大约1到大约30碳原子,更典型地大约2到大约20碳原子。一般使用低分子量交联剂(即,具有大约30到大约500的近似分子量)。可以使用在美国专利申请系列号10/765336或60/590580中描述的任何交联剂或交联方法或化学,在此因为参考。也可以使用本领域熟知的任何交联剂或交联方法或化学。
一般,在配位体和radiophore之间、交联剂与配位体之间、交联剂与radiophore之间形成配合物的任何方式都可以用于本发明。有没有交联剂,在都可以通过配合物组分之间的配合形成配合物,例如通过氢、离子或共价键。配合物组分之间的共价键合可以产生,例如,通过酰胺、酯、二硫化物或酸、醛、羟基、氨基、琉基或叠氮基之间的离子键的形成。而且,根据本发明,交联剂可以包括结合配位体与radiophore的间接方式,例如通过间隔臂(spacer arm)或桥键分子的连接。用于结合的直接和间接方式不应当防止用于本发明方法操作的配位体粘接到癌细胞或活性单核白血球或活性巨噬细胞上的受体上。
作为示例性的化学交联,在实施例1描述方案中的乙二胺作为氟化苯酰胺(即,radiophore其中氟分子是18F)和叶酸配位体之间的间隔或交联剂。优选交联剂起到配合物的水溶性作用,或至少不实质上降低配合物的水溶性。对水溶性有利的交联剂包括水溶性聚合物例如,右旋糖酐、纤维素醚、肽交联剂或聚乙二醇。在一个实施例中,这种聚合物具有小于1,000的分子量。

通过适当的选择,交联剂通过允许配位体与有利点(the site ofinterest)例如,在肝脏的胆汁或尿中被分泌之间的癌细胞或活性单核白血球或活性巨噬细胞的结合可以限定病人的配合物分泌速率。交联剂可以利于或可以延迟配合物的新陈代谢消耗,例如通过减缓网状内皮***的吸收,特别是通过肝脏。交联剂还可以辅助防止配合物与非靶器官、细胞、液体或蛋白质之间的结合。如果,例如,配合物与血清蛋白结合,PET扫描会提供病人的血管扫描,一般与搜索的癌细胞或活性单核白血球或活性巨噬细胞的特定位置形成对照。而且,交联剂可以利于或加速配合物分泌的优选路线,例如通过尿,例如通过配合物的控制之后鼓励病人和足量的液体。
在配位体是叶酸、叶酸类似物/衍生物或其他任何叶酸受体-粘接分子的实施例中,叶酸或其类似物/衍生物可以与交联剂通过本领域熟知的通过蝶酰叠氮化合物中间体利用三氟醋酐制备γ-酯的过程来配合。这个过程合成叶酸,只通过叶酸的谷氨酸基团的γ-羧基配合到交联剂上。可选择地,叶酸类似物可以通过本领域熟知的通过谷氨酸基团的α-羧基部分或α和γ两者整体的过程来耦合。
配合物有效用于本发明方法的量依赖于许多参数,包括配合物的分子量、它的控制路线和组织分布。在本发明中配合物的“有效量”是足够粘接癌细胞或活性单核白血球或活性巨噬细胞并用于涉及活性单核白血球或活性巨噬细胞的肿瘤或疾病状态的诊断/监测中的量。被控制到评估涉及活性单核白血球或活性巨噬细的肿瘤或疾病状态的配合物的有效量可以在大约1pg/kg到大约10mg/kg、1ng/kg到大约10mg/kg、或从大约10μg/kg到大约1mg/kg或从大约100μg/kg到大约500μg/kg。
在用体外PET图像装置检测之前配合物可以控制在一个或多个剂量(例如,大约1到3个剂量)。剂量数依赖于配合物的分子量、它的控制路线和组织分布,其它因素。当用于诊断/监测涉及活性单核白血球或活性巨噬细胞的肿瘤或疾病状态时,体外检测过程一般进行大约1分钟到大约6小时的配合物的后-控制,但是只要配合物到癌细胞或活性单核白血球或活性巨噬细胞上的粘接可以被检测到并且足够时间允许没有粘接的配合物大部分从体内清除,体外检测过程可以进行任何时间的配合物后-控制。
根据本发明方法控制的配合物优选非肠道控制到病人,例如,静脉、真皮、皮下、肌注或腹腔内与药物可接受载体结合。可选择地,配合物可以通过其它医学有用过程控制例如在口头有效配方中。根据本发明,疑似有涉及活性单核白血球或活性巨噬细胞的肿瘤或疾病状态的病人,无论是否有症状,可以被评估,该病人将从使用本发明的方法的评估中受益。
用于本发明的通式为L-X的配合物用于本发明的配制诊断组合物的一个方面,该组合物包括有效量的配合物和可接受的载体。肠道配量形式的实例包括配合物的水溶液,例如,在等渗盐水、5%葡萄糖或其他熟知的药物可接受载体例如酒精、乙二醇、酯和酰胺中的溶液。
用于此处描述的方法中的配合物形成靶,因此,浓缩病人体内肿块点上或活性单核白血球或活性巨噬细胞点(例如,粘附到斑的腔内皮层上的活性巨噬细胞或存在于斑的富液心部的巨噬细胞)上的配合物。
本发明的几个方面在检测癌细胞或活性单核白血球或活性巨噬细胞方面具有优势。在一个实施例中,radiophore包括正电子发射源的元素同位素。正电子发射源从一个源原子以三个方向发射,但是发射过程以恰好相反的方向的两个部分进行。作为电子的反粒子,当从衰变的同位素出来的正电子与电子在近物质中接触时,它湮灭湮灭过程的发射能为γ射线。为了动量守恒,γ射线以相反方向运行。由于正电子具有两个用于检测的辐射射线,因此配合物在病人体内积累的位置更加容易地因此更加准确地,在合理对病人诊断的时间框架内被检测到。正电子湮灭的信号与噪音的比显著提高超过单方向γ射线。而且,通过背景-投影重合射线(back-projecting coincident rays),定位发射源的位置。
目前PET用于医学中心中作为检测肿瘤的诊断工具。在肿瘤的诊断中,可以用正电子发射源标示的葡萄糖(例如,用标示的18F氟化脱氧葡萄糖)控制病人。葡萄糖聚集在生长快的癌细胞中。可以通过PET显影剂检测肿瘤的存在。而且,通过利用PET扫描仪的背景-投影重合γ辐射确定肿瘤的位置。因此,可以结合标示的18F氟化脱氧葡萄糖使用本发明的方法来检测癌细胞。也可以结合本领域发展并已知的任何其他肿瘤诊断方法来使用本发明的方法,本领域发展并已知的任何其他肿瘤诊断方法包括使用其他已经发展的诊断剂以及利用X-射线计算层析照相法(CT)、磁共振成像(MRT)、功能性磁共振成像(FMRT)、超声波、单质子发射计算断层照相法(SPECT)。
在另一个实施例中,本发明的方法可以单独使用或与本领域已知的用于动脉粥样硬化斑的检测/分析/消融的任何其它方法相结合使用。例如,当活性斑导致血管狭窄的情况下,本发明可以与消融动脉粥样硬化斑的方法结合使用。在这种情况下,本发明的配合物不仅可以用于辨别与非活性斑向对的活性动脉硬化斑,而且可以区分动脉粥样硬化和正常组织以在消融过程中起辅助作用。因此,本发明可以用于分析动脉粥样硬化斑的生理和形态状态。例如,血管成形术包括由于斑沉积导致的狭窄的血管的非手术加宽,并且激光能,例如直接通过导管基装置中的视纤维,可以用于消融或部分除去斑沉积。用于消融斑的利用激光能的导管基装置在美国专利N0.4,817,601、4,850,351,和4,950,266中有描述,在此引为参考。
本发明可用的发射正电子同位素。适当的radiophore可以用同位素氟18F制成。也可以使用其它可用的发射正电子同位素。在选择适当同位素的值得考虑的因素包括足够的发射正电子同位素的半衰期以允许在对病人控制之前在药物可接受载体中制成诊断组合物,并且足够保持的半衰期产生足够的活性以允许体外测量,例如,通过PET扫描。而且,适当的同位素应当具有足够短的半衰期以限制病人暴露在不必要的辐射中。在示例性实施例中,具有110分钟半衰期的18F提供充分的时间用于制成诊断组合物,以及可接受的半衰变速率。
在一个示例性实施例中,同位素应当具有足够的化学活性以允许,无论是否使用交联剂,同位素能粘结到化学化合物上并依次到配位体上。应当避免具有毒性的元素同位素。具有适当半衰期的正电子衰变同位素包括:34Cl,32分钟的半衰期;45Ti,3.09小时的半衰期;51Mn,45分钟的半衰期;61Cu,3.41小时的半衰期;63Zn,38.4分钟的半衰期;68Ga,68.3分钟的半衰期;11C,20分钟的半衰期;15O,2分钟的半衰期;13N,10分钟的半衰期;或18F,110分钟的半衰期。
实施例1
叶酸配合物的合成
双向相容配合物L-X使用适当的同位素制成诊断组合物。例如在18F的情况下,适当的L-X配合物可以根据下面的合成规约制成。根据这一实施例合成的配合物具有乙二胺交联剂。

TSTU=N-羟基琥珀酰亚胺
在合成期间,有经验的从事者将按照适当的过程以浓缩和提纯p-氟化苯甲酸。一般的,但不需要通过专门提纯和浓缩氟化苯甲酸的过程,可以使添加足够的HCl以protinate分子,然后在相反相C18柱例如Waters Corp.Milford Massachusetts售出的SepPak Plus上分离。可用HCl酸化的水清洗柱以除去任何水溶性污染物。通过接下来的阴离子交换柱(例如,Dowex柱)上的进一步污染物除去可以用甲醇洗提p-氟化苯甲酸,并且通过甲醇蒸发浓缩。
混合物反相高性能液相色谱法开始。水稀释的酯溶液可以在接下来由乙二醚洗提的C18SepPak柱上浓缩。残余的水可以用无水Mg2SO4柱除去。醚蒸干之后,酯可以再溶于乙脲中。根据所描述的过程这些过程可以用于制造上述化合物。
实施例2
叶酸配合物的合成
由已知的过程开始制备P-氟化苯甲醚,如下面所述快速制成适当的配合物。浓缩和纯化过程可以如上所述,或者另外根据本领域熟知的技术。
合成可以足够快捷完成以足以保持发射正电源18F的半衰期来进行诊断试验。
实施例3
叶酸-SFB的合成
叶酸-肽的合成(树脂-赖氨酸-天冬氨酸-天冬氨酸-天冬氨酸-谷氨酸-蝶酸)
用MTT(4-甲基-3-苯甲基)和F-moc保护基团将Wang树脂连接到赖氨酸上。用20%的哌啶/DMF(二甲基甲酰胺)(10mg/g)除去F-moc保护基团。然后用氩使树脂冒泡10分钟并排出树脂。再重复两次F-moc保护基团的除去和冒泡步骤。然后用DMF清洗树脂三次。Kaiser试验对于自由胺基是正。
然后添加溶于DMF中的Fmoc-AA(Fmoc-氨基酸)、HBTU(2-(1H-苯并***-1-y1)-1,1,3,3-四甲基糖醛酸六氟磷酸酯)、HOBt(1-羟基苯并***)、DIPEA(二异丙基乙基胺)并冒泡30分钟。然后进行用DMF的清洗(三次),并重复这三个步骤。Kaiser试验是负,没有自由的胺。
然后添加氨基酸并重复上述步骤,并用DCM(二氯甲烷)清洗树脂15分钟并在1%的TEA(三氟乙酸)/DCM中冒泡以除去MTT。然后再次用二氯甲烷清洗树脂15分钟并在1%的TEA/DCM中冒泡。重复上述步骤直到黄色除去。然后用DCM清洗树脂三次并用DMF再次清洗。
对于DMF中的蝶酸(CF3),添加HOBt、HBTU和DIPEA。组合物变成褐色并冒泡过夜,然后用DMF清洗三次。Kaiser试验是负。用20%的哌啶/DMF(10mg/g)除去F-moc保护基团并使树脂冒泡10分钟并排出树脂。再重复两次F-moc保护基团的除去和冒泡步骤。然后用DMF清洗树脂三次、DCM清洗三次以及甲醇清洗三次。树脂在真空下干燥4小时。
用包括95∶2.5∶2.5的TFA∶TIS(三异丙基硅烷)∶H2O稀释TFA。将这种溶液添加到玻璃珠中,并冒泡2-3小时并排到干净的长颈瓶中。然后用相同的溶液清洗玻璃珠并排出直到玻璃珠排干净。漩涡蒸发溶液以较少溶液至2-3ml。然后用50ml的冰冷醚填充到锥形的管中。将肽混合物醚指瓶中使得肽沉淀出来。对混合物进行离心脱水并用醚清洗2-3次,在真空下以除去痕量的醚。然后将肽1%的NH4OH中来用搅拌棒混合30-40分钟出去TFA保护基团。然后冷冻TFA、用HPLC纯化并冷冻。
非-辐射活性SFB(琥珀酰亚胺基-氟化苯甲酸酯)的合成
通过Eur.J.Med.Mol.Imaging,vol.31:469-474(2004)中描述的方法合成非-辐射活性SFB。由氟化苯甲酸开始,将油浴设置在90摄氏度。制备45%的四甲基氢氧化铵(TMAH)的水溶液。将10mg4-氟苯甲酸(溶液1)添加到一个单独的瓶中。将45%的TMAH(溶液2)添加到一个单独的瓶中。然后,添加0.2ml的水和1.0ml的乙脲并将溶液2添加到溶液1中(溶液3)。用漩涡蒸发将溶液蒸干。在另一个瓶中,将14mgTSTU添加到1.2ml的乙脲中(溶液4)。将溶液4添加到溶液3中。在油浴中将混合物加热至90摄氏度2个小时。在回旋加速器中通过本领域熟知的过程合成辐射活性18F,并用于制成实施例1所示的(18F)氟化苯甲酸,然后转化为上述的18F-SFB。
叶酸-SFB的合成
在PBS中叶酸-肽与SFB结合(在乙脲中),调整叶酸-肽的pH值,通过HPLC检验配合物。
实施例4
粗叶酸-SFB的HPLC
用与Clinical Science,Vol.103:PP.4S-8S(2002)相似的条件,但具有之后的改性,通过高性能液相色谱(HPLC)分析粗叶酸-SFB。用C18柱进行反相HPLC,条件是水0.1%TFA在ACN中以77∶23的比例10分钟,60∶40的比例10分钟,50∶50的比例10分钟,40∶60的比例10分钟,并用1.0ml/分钟的流速。结果示于图1中。
实施例5
粗的和提纯的叶酸的质谱
用本领域熟知的过程通过ESI-质谱分析粗的和提纯的叶酸-SFB。结果示于图2和图3中。
实施例6
对比结合试样
为了确定叶酸-SFB是否结合到叶酸受体上,进行比较结合试样。用同等体积的组织培养基中细胞将KB细胞固定在6-并(well)平皿中。如图4所示,单独培养细胞(第一栏)、具有100nM3H-叶酸(第二栏),具有存在于100nM叶酸-SFB中的100nM3H-叶酸(第三栏)或具有存在于100μM叶酸-SFB中的100nM3H-叶酸(第四栏)。结果示出对于结合到叶酸受体上叶酸-SFB与3H-叶酸相比叶酸-SFB明确地结合到叶酸受体上。
